Abstract:We respond to claims by Dong et al . that human lifespan is limited below 125 years. Using the log-linear increase in mortality rates with age to predict the upper limits of human survival we find, in contrast to Dong et al ., that the limit to human lifespan is historically flexible and increasing. This discrepancy can be explained by Dong et al .’s use of data with variable sample sizes, age-biased rounding errors, and log(0) instead of log(1) values in linear regressions. Addressing these issues eliminates the proposed 125-year upper limit to human lifespan.

Abstract:An ongoing debate in demography has focused on whether the human lifespan has a maximal natural limit. Taking a mechanistic perspective, and knowing that short telomeres are associated with diminished longevity, we examined whether telomere length dynamics during adult life could set a maximal natural lifespan limit. We define leukocyte telomere length of 5 kb as the 'telomeric brink', which denotes a high risk of imminent death. We show that a subset of adults may reach the telomeric brink within the current life expectancy and more so for a 100-year life expectancy. Thus, secular trends in life expectancy should confront a biological limit due to crossing the telomeric brink.

Abstract:Does the human lifespan have an impenetrable biological upper limit which ultimately will stop further increase in life lengths? This question is important for understanding aging, and for society, and has led to intense controversies. Demographic data for humans has been interpreted as showing existence of a limit, or even as an indication of a decreasing limit, but also as evidence that a limit does not exist. This paper studies what can be inferred from data about human mortality at extreme age. We show that in western countries and Japan and after age 110 the probability of dying is about 47% per year. Hence there is no finite upper limit to the human lifespan. Still, given the present stage of biotechnology, it is unlikely that during the next 25 years anyone will live longer than 128 years in these countries. Data, remarkably, shows no difference in mortality after age 110 between sexes, between ages, or between different lifestyles or genetic backgrounds. These results, and the analysis methods developed in this paper, can help testing biological theories of ageing and aid confirmation of success of efforts to find a cure for ageing.

Abstract:In this paper, we affirm our earlier findings of evidence for a limit to human lifespan. In particular, we assess the analyses in extreme value theory (EVT) performed by Rootz\'en and Zholud. We find that their criticisms of our work are unfounded and that their analyses are contradicted by several other papers using EVT. Furthermore, we find that even if we completely accept the conclusions about late-life human mortality reached by Rootz\'en and Zholud, their results do not actually contradict the findings presented in our original paper: whether unbounded or not, human lifespan is unlikely to greatly exceed 120 years, and the improbability of longer survival---whether it is exactly zero or merely astronomically small---acts as a de facto limit. In order to eliminate the confusion surrounding the issue, we propose the adoption of the term "limit" to denote the age at which the chance of survival is exactly zero and the term "effective limit" to denote the age at which the change of survival falls below a given threshold. Once this distinction is made, it can be demonstrated that the final result of Rootz\'en and Zholud is essentially a recapitulation of the main conclusion of our paper. Ultimately, much of the controversy surrounding the issue of a limit to human lifespan can be avoided by carefully reading the literature and applying statistics to practical human scales.

Abstract:The analysis of the demographic transition of the past century and a half, using both empirical data and mathematical models, has rendered a wealth of well-established facts, including the dramatic increases in life expectancy. Despite these insights, such analyses have also occasionally triggered debates which spill over many disciplines, from genetics, to biology, or demography. Perhaps the hottest discussion is happening around the question of maximum human lifespan, which --besides its fascinating historical and philosophical interest-- poses urgent pragmatic warnings on a number of issues in public and private decision-making. In this paper, we add to the controversy some results which, based on purely statistical grounds, suggest that the maximum human lifespan is not fixed, or has not reached yet a plateau. Quite the contrary, analysis on reliable data for over 150 years in more than 20 industrialized countries point at a sustained increase in the maximum age at death. Furthermore, were this trend to continue, a limitless lifespan could be achieved by 2102. Finally, we quantify the dependence of increases in the maximum lifespan on socio-economic factors. Our analysis indicates that in some countries the observed rising patterns can only be sustained by progressively larger increases in GDP, setting the problem of longevity in a context of diminishing returns.

Abstract:Recent discoveries show steady improvements in life expectancy during modern decades. Does this support that humans continue to live longer in future? We recently put forward the maximum survival tendency, as found in survival curves of industrialized countries, which is described by extended Weibull model with age-dependent stretched exponent. The maximum survival tendency suggests that human survival dynamics may possess its intrinsic limit, beyond which survival is inevitably forbidden. Based on such tendency, we develop the model and explore the patterns in the maximum lifespan limits from industrialized countries during recent three decades. This analysis strategy is simple and useful to interpret the complicated human survival dynamics.

Abstract:There is sustained and widespread interest in understanding the limit, if there is any, to the human life span. Apart from its intrinsic and biological interest, changes in survival in old age have implications for the sustainability of social security systems. A central question is whether the endpoint of the underlying lifetime distribution is finite. Recent analyses of data on the oldest human lifetimes have led to competing claims about survival and to some controversy, due in part to incorrect statistical analysis. This article discusses the particularities of such data, outlines correct ways of handling them, and presents suitable models and methods for their analysis. We provide a critical assessment of some earlier work and illustrate the ideas through reanalysis of semisupercentenarian lifetime data. Our analysis suggests that remaining life length after age 109 is exponentially distributed and that any upper limit lies well beyond the highest lifetime yet reliably recorded. Lower limits to 95% confidence intervals for the human life span are about 130 years, and point estimates typically indicate no upper limit at all.

Abstract:Abstract Since 1990 Jeanne Louise Calment has held the record for human longevity. She was born on 21 February 1875, became the longest-lived human on 12 May 1990 when she was 115.21 and died on 4 August 1997 at age 122.45 years. In this chapter, we use data available on 25 September 2017 on people who reached age 110, supercentenarians, to address the following questions: How likely is it that a person has reached age 122.45? How unlikely is it that Calment’s record has not yet been broken? How soon might it be broken? Assuming a constant annual probability of death of 50% after age 110, we found that the probability that a person who survived to age 110 would have lived to 122.45 by 25 September 2017 is 17.1%. Furthermore, we calculated that there was only a 20.3% chance that Calment’s record would have been broken after 1997 but before 2017. Finally, we estimated that there is less than a 50% chance that someone will surpass Calment’s lifespan before 2045. Jeanne Louise Calment’s record is exceptional but not impossible. It does not provide evidence that her lifespan is an upper limit to human lifespans.

Abstract:Although average human life expectancy is rising, the maximum lifespan is not increasing. Leading demographers claim that human lifespan is fixed at a natural limit around 122 years. However, there is no fixed limit in animals. In animals, anti-aging interventions (dietary restrictions, rapamycin, genetic manipulations) postpone age-related diseases and thus automatically extend maximum lifespan. In humans, anti-aging interventions have not been yet implemented. Instead, by treating individual diseases, medical interventions allow a patient to live longer (despite morbidity), expanding morbidity span. In contrast, slowly aging individuals (centenarians) enter very old age in good health, but, when diseases finally develop, they do not receive thorough medical care and die fast. Although the oldest old die from age-related diseases, death certificates often list "old age", meaning that diseases were not even diagnosed and even less treated. The concept of absolute compression of morbidity is misleading in humans (in truth, there is no other way to compress morbidity as by denying thorough medical care) and false in animals (in truth, anti-aging interventions do not condense morbidity, they postpone it). Anti-aging interventions such as rapamycin may potentially extend both healthspan and maximal lifespan in humans. Combining anti-aging medicine with cutting-edge medical care, regardless of chronological age, will extend maximal lifespan further.

Implausibility of radical life extension in humans in the twenty-first century

Abstract:Over the course of the twentieth century, human life expectancy at birth rose in high-income nations by approximately 30 years, largely driven by advances in public health and medicine. Mortality reduction was observed initially at an early age and continued into middle and older ages. However, it was unclear whether this phenomenon and the resulting accelerated rise in life expectancy would continue into the twenty-first century. Here using demographic survivorship metrics from national vital statistics in the eight countries with the longest-lived populations (Australia, France, Italy, Japan, South Korea, Spain, Sweden and Switzerland) and in Hong Kong and the United States from 1990 to 2019, we explored recent trends in death rates and life expectancy. We found that, since 1990, improvements overall in life expectancy have decelerated. Our analysis also revealed that resistance to improvements in life expectancy increased while lifespan inequality declined and mortality compression occurred. Our analysis suggests that survival to age 100 years is unlikely to exceed 15% for females and 5% for males, altogether suggesting that, unless the processes of biological aging can be markedly slowed, radical human life extension is implausible in this century.

Abstract:Multiple observations that organismal life span can be extended by nutritional, genetic, or pharmacological intervention has raised the prospect of transforming medicine with the goal of slowing, stopping, or even reversing age-associated disease and maintaining or restoring health and resilience in the increasing numbers of elderly across the world. The potential for such an enterprise is supported in theory by plant and animal models of negligible senescence, most notably the small, freshwater organism Hydra spp. The existence of some very long-lived species, including bowhead whale, Greenland shark, and giant tortoises, suggests that increased healthy life spans in humans, significantly higher than the current known maximum life span of about 120 years, may be possible. Here we discuss the biological restraints on human life extension based on the evolutionary basis of aging and our current genetic and molecular insights into the processes responsible for age-related loss of function and increased disease risk.

Abstract:A key but unresolved issue in the study of human mortality at older ages is whether mortality is being compressed (which implies that we may be approaching a maximum limit to the length of life) or postponed (which would imply that we are not). We analyze historical and current population mortality data between ages 50 and 100 by birth cohort in 19 currently-industrialized countries, using a Bayesian technique to surmount cohort censoring caused by survival, to show that while the dominant historical pattern has been one of mortality compression, there have been occasional episodes of mortality postponement. The pattern of postponement and compression across different birth cohorts explain why longevity records have been slow to increase in recent years: we find that cohorts born between around 1900 and 1950 are experiencing historically unprecedented mortality postponement, but are still too young to break longevity records. As these cohorts attain advanced ages in coming decades, longevity records may therefore increase significantly. Our results confirm prior work suggesting that if there is a maximum limit to the human lifespan, we are not yet approaching it.

Abstract:Humans age at different rates and families with exceptional survival provide the opportunity to understand why some people age slower than others. Unique features exhibited by centenarians include a family history of longevity, compression of morbidity with resultant extension of health span, and biomarkers such as low-circulating insulin-like growth factor 1 (IGF-1) and elevated high-density lipoprotein (HDL) cholesterol levels. Given the rarity of the centenarian phenotype, it has not been surprising that the use of discovery methods that relied on common population single nucleotide polymorphisms (SNPs) to unlock the genetic determinants of exceptional longevity have not yielded significant results. Conversely, gene sequencing has resulted in discoveries of functional gene variants that support several of the centenarian phenotypes. These discoveries have led to the strategic developments of drugs that may delay aging and prolong health span.

Abstract:Old-age survival has increased substantially since 1950. Death rates decelerate with age for insects, worms, and yeast, as well as humans. This evidence of extended postreproductive survival is puzzling. Three biodemographic insights--concerning the correlation of death rates across age, individual differences in survival chances, and induced alterations in age patterns of fertility and mortality--offer clues and suggest research on the failure of complicated systems, on new demographic equations for evolutionary theory, and on fertility-longevity interactions. Nongenetic changes account for increases in human life-spans to date. Explication of these causes and the genetic license for extended survival, as well as discovery of genes and other survival attributes affecting longevity, will lead to even longer lives.
