Abstract:Biothiols such as cysteine (Cys), homocysteine (Hcy) and glutathione (GSH) play important roles in regulating the redox balance of life process and are closely associated with human health and diseases. In order to better understand their roles in physiological process and the early diagnosis of related diseases, it is crucial for the selective detection of biothiols in biological systems. In this paper, we developed a 4-phenylselenium coumarin-nitroolefin conjugate as the fluorescent probe for the detection of biothiols. By introducing the 4-phenyselenium as the active site, probe CouSeNO2 is capable of detecting Cys/Hcy and GSH in dual fluorescence channels. For the biothiols, the first-step sensing reaction is proved to be nucleophilic substitution of 4-phenylselenium by the thiol group. Furthermore, through two-channel fluorescence imaging, probe CouSeNO2 has been successfully applied to sense the exogenous and endogenous biothiols in living cells.

Abstract:In this work, compound 1 has been rationally designed and synthesized as a new fluorescent probe for biological thiols. Notably, probe 1 has almost no background fluorescence (Phi(f) < 0.0001) in aqueous solutions; however, it exhibited fluorescence turn-on response to thiols with high sensitivity (a 246-fold fluorescence enhancement and a low detection limit of 0.22 mu M for Cys). Moreover, probe 1 showed excellent thiol specificity over other biologically relevant species. The kinetic studies indicated that the probe responded to thiols rapidly, and the pseudo-first-order rate constants of probe 1 reaction with Cys, Hcy, and GSH were determined to be 1.85842, 0.67656, and 0.51519 min(-1), respectively. A possible detection mechanism was proposed to involve the Michael addition of the thiol to the alpha,beta-unsaturated ketone, followed by a cleavage of the hemiketal group, thereby leading to the formation of a fluorescent 7-hydroxyl coumarin derivative. Furthermore, the optical responses of probe 1 to thiols were studied by TD-DFT calculations. Finally, probe 1 has been successfully applied to the detection of biological thiols in human blood serum. And the intracellular imaging applications established that probe 1 can be used to detect different concentrations of intracellular thiols in living cells.

Abstract:Hydrogen sulfide (H2S) is associated with numerous psychological and pathological processes, and related closely with human health and diseases. The detection of H2S is a key to better understand its roles. In this paper, we developed a 4-phenylselenium coumarin (PhSeCou) as a rapid-response fluorescent probe for the selective and sensitive detection of H2S. The probe PhSeCou is a stable and almost non-fluorescent molecule in a PBS buffer, and it can rapidly (< 60 s) sense H2S to give a strongly fluorescent product, Cou-H2S (Phi(f) = 0.11, centered at 563 nm), thereby achieving to a sensitive detection at an ultra-low limit of detection (8.8 nM). The fluorescent probe displays a high selectivity for H2S over biothiols and other relevant analytes. What is more, through fluorescence cell-imaging, PhSeCou was successfully applied to detect exo- and endogenous H2S in living cells.

Abstract:We designed and synthesized a novel probe Cou-Rho-SA-Cu (II) for detection of thiols based on a coumarin-rhodamine FRET system. The compound Cou-Rho-SA exhibited strong blue fluorescence as the rhodamine moiety of the compound Cou-Rho-SA was in a ring-closing form and no FRET process occurred. The probe Cou-Rho-SA-Cu (II) was prepared by addition of Cu(2+) to the solution of Cou-Rho-SA. In the absence of GSH, the complex Cou-Rho-SA-Cu (II) showed almost no fluorescence owing to the FRET process and the paramagnetic nature of Cu(2+) . However, upon addition of GSH, the rhodamine moiety of the probe Cou-Rho-SA-Cu (II) recovered to its ring-closing form, and the FRET process was prohibited to show the blue fluorescence again. Moreover, via tuning the Cu(2+) concentrations, the probe could detect the different levels of GSH.

Abstract:A novel probe incorporating tetraphenylethylene and benzoquinone into a molecule was synthesized and studied for the selective detection of biothiols. The probe was found to have a highly selective response to biothiols through a thiol-ene click-reaction with fluorescence turn-on feature. The observable color changes before and after biothiol addition indicated that the probe was suitable for naked-eye biothiol detection.

Abstract:In this work, a tetraphenylethene-coumarin hybrid fluorophore (TPE-Cou) that contains a Schiff base form is designed and synthesized. A combination of plentiful optical properties and chemical reactivity towards thiols allows TPE-Cou to work as an excellent turn-on probe of thiols with a wide linear range, revealing the great potential of this dye as a quantitative fluorescence indicator. By means of NMR and optical spectrum analyses, a mechanistic picture at the molecular level has been drawn to illustrate how this dye works as a bio-thiol-sensitive fluorescent probe.

Abstract:The sensing mechanism of a reported fluorescence probe for cysteine, homocysteine and glutathione (Yin et al., 2018) has been investigated by time-dependent density functional theory. Experimental absorption and emission spectra of the probe before and after thiol addition were reproduced well by theoretical calculations, which validated the rationality of the method. Optimized geometries showed that the probe molecule had distinctly different geometries in its ground and excited states. It corresponded to the photoisomerization process and explained the weak fluorescence of the probe molecule. Moreover, by the potential energy curve scan, photoisomerization was further confirmed to be a spontaneous process with a barrier that barely existed. Frontier orbital analysis indicated that this photoinduced isomerization of the probe molecule derived from the antibonding character for lowest unoccupied molecular orbital at its CC double bond. In contrast, probe-thiol complexes exhibited similar geometries in their ground and excited states, which was responsible for the strong fluorescence of the probe with thiols. Due to distinct excited-processes, the probe can be used to sense thiols by monitoring the fluorescent change.

Abstract:A novel fluorescent probe was developed by integrating chlorinated coumarin and benzothiazolylacetonitrile and exploited for simultaneous detection of cysteine (Cys), homocysteine (Hcy), and glutathione (GSH). Featuring four binding sites and different reaction mechanisms for different biothiols, this probe exhibited rapid fluorescence turn-on for distinguishing Cys, Hcy, and GSH with 108-, 128-, 30-fold fluorescence increases at 457, 559, 529 nm, respectively, across different excitation wavelengths. Furthermore, the probe was successfully applied to the fluorescence imaging of endogenous Cys and GSH and exogenous Cys, Hcy, and GSH in living cells.

Sulfite reacts with conjugate bond selectively, while Cys responses with aldehyde and GSH occurs substitution reaction. Different interactions cause different absorption and fluorescence responses. Abstract:A novel fluorescent probe (CA-N) was designed and synthesized for detection of biothiols. CA-N displayed a strong fluorescence in the presence of biothiols with high sensitivity, and the mechanism for detection biothiols was based on the Michael addition reaction of a thiol group to α,β-unsaturated ketones. CA-N showed low detection limit for cysteine (Cys), homocysteine (Hcy), and glutathione (GSH), which were calculated as 3.16, 0.19 and 5.15 μM, respectively. At the same time, CA-N exhibited high selectivity toward biothiols compared with other biological amino acids. In vitro cell experiments proved that CA-N had no cytotoxicity, high cell permeability and could be employed in living cell imaging for biothiols.

Abstract:A new probe based on the squaraine skeleton with a 2,4-dinitrobenzenesulfonyl unit is reported as a fluorescent probe for biothiol. It shows turn-on properties with red emission towards biothiols in acetonitrile-PBS buffer (1 : 9, v/v) solution, and its properties of rapid response, high selectivity and high sensitivity make it a potential probe with real applications. Moreover, the in vitro assays show that the probe can be used in fluorescence imaging for the detection of intracellular biothiol levels.

Abstract:Sulfite and sulfide share several similarities in terms of chemical properties, such as nucleophilic and reducing reactivities. Therefore, they may disturb the detection of each other. In order to discriminate between these two kinds of sulfur-containing species, a new probe TSSP-N-3 was developed, in which para-azidobenzenyl ketone was covalently incorporated to a coumarin fluorophore linked by a C=C double bond. Sulfite and sulfide can respectively react with the C=C double bond and the azido group to give different products, consequently, they can be differentially identified by UV-vis and fluorescence spectroscopy as well as by the naked eye. Selectivity and competition results reveal that TSSP-N-3 is a good candidate for the detection of sulfide and sulfite. The bioimaging experiment demonstrates the potential of the TSSP-N-3 probe for the differential imaging of sulfide and sulfite in living cells.

Abstract:In this work, a new reversible colorimetric and fluorescent probe for sequential recognition of copper ions and biothiols is synthesized easily. Based on the chelation-enhanced fluorescence quenching (CHEQ) effect, this probe shows high sensitivity and selectivity towards Cu2+, which can be detected by the naked eye. And this experimental phenomenon can also be realised upon addition of biothiols, restoring their initial fluorescence intensity. This probe also features a very high response speed (less than 5 seconds) and a large stokes shift (178 nm) toward Cu2+ and biothiols. Moreover, the detection limit for Cu2+ and biothiols is as low as 7.34 nM and 10.3 nM, respectively. Additionally, this ON-OFF-ON-type fluorescence recognition cycle can be repeated more than 5 times by addition of Cu2+ and biothiols in turn. Particularly, this 1-Cu2+ ensemble is further successfully applied for GSH detection in living cells.

Abstract:The article describes the design and synthesis of a near-infrared ratiometric fluorescent probe CySS for thiols,the probe CySS has heptamethine cyanine as the reporter,a disulfide group as the thiol reaction site. The properties and application of CySS were studied in detail.The results demonstrate that CySS displays high sensitivity,good selectivity,and pH-independent.Moreover,we have demonstrated that CySS is suitable for imaging thiols in living cells,providing apotentially powerful tool for thiols detection in biological systems.

Abstract:A dual-emission fluorescent probe was developed for detecting biothiols, a coumarin-chromene scaffold. In this work, the sensing mechanism of probe BDU towards biothiols was through two different pathways. For Hcy and Cys, it went through a cascade reaction followed by an isomerization, which formed a salicylic carbonate structure and coumarin substance. While for GSH, only the nucleophilic reaction was carried out without further isomerization. The probe exhibits selectivity for Hcy/Cys (Em = 470 nm) and GSH (Em = 546 nm) in PBS buffer containing 0.5 % DMSO. Besides, this probe could effectively sense biothiols in vitro and in vivo with low detecting limits, fast responses, good biocompatibility, and low toxicity. With all these excellent properties, this probe exhibits great potential value for applications in biological study.

Abstract:A benzoquinolizine coumarin-based fluorescent probe was developed for detecting thiophenols, demonstrating the superior fluorescence properties caused by the decay of the twisting effect of N,N-diethylamino group of coumarin. It discriminated thiophenols from various analytes including aliphatic thiols with good selectivity and displayed ∼700-fold fluorescence intensity enhancement and a remarkable limit of detection (4.5 nM). The new probe also can be applied to quantitative determine the concentrations of thiophenol in water samples and living cells.
