Abstract:In this work, a new reversible colorimetric and fluorescent probe for sequential recognition of copper ions and biothiols is synthesized easily. Based on the chelation-enhanced fluorescence quenching (CHEQ) effect, this probe shows high sensitivity and selectivity towards Cu2+, which can be detected by the naked eye. And this experimental phenomenon can also be realised upon addition of biothiols, restoring their initial fluorescence intensity. This probe also features a very high response speed (less than 5 seconds) and a large stokes shift (178 nm) toward Cu2+ and biothiols. Moreover, the detection limit for Cu2+ and biothiols is as low as 7.34 nM and 10.3 nM, respectively. Additionally, this ON-OFF-ON-type fluorescence recognition cycle can be repeated more than 5 times by addition of Cu2+ and biothiols in turn. Particularly, this 1-Cu2+ ensemble is further successfully applied for GSH detection in living cells.

Abstract:A 1,8-naphthalimide-Cu(II) ensemble was rationally designed and synthesized as a new turn-on fluorescent probe utilizing the 'chemosensing ensemble' method for detections of thiols (Cys, Hcy and GSH) with high selectivity over other alpha-amino acids at pH 7.4 in organic aqueous media (EtOH/HEPES, v/v = 9: 1). The recognition mechanism was attributed to the remove Cu(II) from the 1,8-naphthalimide-Cu(II) ensemble by thiols and the release of flurescence of ligand 1. Remarkable fluorescence enhancements were therefore observed in the sensing process of thiols by the 1,8-naphthalimide-Cu(II) ensemble. Furthermore, the 1,8-naphthalimide-Cu(II) ensemble was successfully applied to the fluorescence imaging of thiols in CHO cells with high sensitivity and selectivity. (C) 2013 Elsevier Ltd. Abstract:In this study, a new fluorescent probe 2-(2′-hydroxy-5′-N-maleimide phenyl)-benzothiazole (probe 1), was designed and synthesized by linking the excited state intramolecular proton transfer (ESIPT) fluorophore to the maleimide group for selective detection of thiols in aqueous solution. The fluorescence of probe 1 is strongly quenched by maleimide group through the photo-induced electron transfer (PET) mechanism, but after reaction with thiol, the fluorescence of ESIPT fluorophore is restored, affording a large Stokes shifts. Upon addition of cysteine (Cys), probe 1 exhibited a fast response time (complete within 30s) and a high signal-to-noise ratio (up to 23-fold). It showed a high selectivity and excellent sensitivity to thiols over other relevant biological species, with a detection limit of 3.78×10−8M (S/N=3). Moreover, the probe was successfully applied to the imaging of thiols in living cells.

Abstract:A new highly sensitive 1, 8-naphthalimide-based compound (probe 1) has been designed and synthesized. The colorimetric and fluorescent properties of probe 1 towards various amino acids were investigated in detail. Specific colorimetry and fluorescence response of probe 1 for thiols were found in PBS buffer solution, which can be deduced to a reaction of thio-promoted conjugate addition/cyclization sequence between probe 1 and thiols. The probe 1 can selectively recognize cysteine rather than other natural amino acids in aqueous solution with a rapid response time. Abstract:The article describes the design and synthesis of a near-infrared ratiometric fluorescent probe CySS for thiols,the probe CySS has heptamethine cyanine as the reporter,a disulfide group as the thiol reaction site. The properties and application of CySS were studied in detail.The results demonstrate that CySS displays high sensitivity,good selectivity,and pH-independent.Moreover,we have demonstrated that CySS is suitable for imaging thiols in living cells,providing apotentially powerful tool for thiols detection in biological systems.

Abstract:Based on an aldazine-copper chemosensing ensemble (NP-Cu2+), a new fluorescence chemosensor for the detection of biothiols (Cys, Hcy and GSH) was designed and synthesized. In aqueous solution, the ligand NP exhibited high selectivity toward Cu2+ ions by forming a 2:1 complex, accompanied with a dramatic fluorescence quenching and a notable bathochromic-shift of the absorbance band. Due to the high affinity of thiols and copper, the specific interaction of thiols (Cys, Hcy and GSH) with NP-Cu2+ ensemble led to the liberation of the NP. As the result, recovery of fluorescence and UV–vis absorbance was observed. The detection limits of NP-Cu2+ to Cys, Hcy and GSH were estimated to be 1.5μM, 1.8μM and 2.2μM, respectively. The fluorescence “OFF–ON” circle can be repeated to a minimum of 5 times by the alternative addition of thiols and Cu2+, implying that NP-Cu2+ is a recyclable chemosensor for thiols. Results of fluorescence microscopy imaging suggested that NP-Cu2+ has potential to be used as a powerful tool for the detection of intracellular thiols.

Abstract:A new N-butyl-4-amino-1,8-naphthalimide-based colorimetric and ratiometric fluorescent probe for the detection of biothiols (cysteine, homocysteine, and glutathione) was designed and synthesized. The probe exhibited good selectivity and sensitivity toward biothiols over other non-thiol-bearing amino acids and common anions with significant changes in both color (from colorless to yellow) and fluorescence (from blue to yellow-green). The mechanism is based on cleavage of disulfide by thiols followed by an intramolecular cyclization to give the product, N-butyl-4-amino-1,8-naphthalimide, which showed outstanding intramolecular charge transfer (ICT). Therefore, ratiometric fluorescence signal was observed. Furthermore, the probes were successfully applied for visualizing endogenous thiols in living cells.

Abstract:In view of the vital role of biothiols in many physiological processes, the development of simple and efficient probe for the detection of biothiols is of great medical significance. In this work, we demonstrate the use of 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), which respond rapidly to biothiols especially to glutathione, as a new fluorescent probe for the selective detection and bioimaging of biothiols. This new fluorescent probe can distinguish glutathione from cysteine and homocysteine easily under physiological concentration and detect glutathione quickly within three minutes. This probe exhibits high selectivity to biothiols and the detection limit was determined to be 3.08 × 10−9 M for glutathione, 8.55 × 10−8 M for cysteine, and 2.17 × 10−9 M for homocysteine, respectively. The sensing mechanism was further explored by density functional theory (DFT) and nuclear magnetic resonance (NMR) experiment; results showed that the interaction forces between the probe and biothiols were electrostatic interaction. In addition, the probe has been successfully applied to the detection of biothiols in Eca9706 cells by fluorescence confocal imaging technology.

Abstract:A new and simple fluorescent probe for detection of Cue(2+) has been synthesized and characterized. Abstract:In this work, compound 1 has been rationally designed and synthesized as a new fluorescent probe for biological thiols. Notably, probe 1 has almost no background fluorescence (Phi(f) < 0.0001) in aqueous solutions; however, it exhibited fluorescence turn-on response to thiols with high sensitivity (a 246-fold fluorescence enhancement and a low detection limit of 0.22 mu M for Cys). Moreover, probe 1 showed excellent thiol specificity over other biologically relevant species. The kinetic studies indicated that the probe responded to thiols rapidly, and the pseudo-first-order rate constants of probe 1 reaction with Cys, Hcy, and GSH were determined to be 1.85842, 0.67656, and 0.51519 min(-1), respectively. A possible detection mechanism was proposed to involve the Michael addition of the thiol to the alpha,beta-unsaturated ketone, followed by a cleavage of the hemiketal group, thereby leading to the formation of a fluorescent 7-hydroxyl coumarin derivative. Furthermore, the optical responses of probe 1 to thiols were studied by TD-DFT calculations. Finally, probe 1 has been successfully applied to the detection of biological thiols in human blood serum. And the intracellular imaging applications established that probe 1 can be used to detect different concentrations of intracellular thiols in living cells.

Abstract:A new turn-on fluorescent probe for the selective detection of thiol over other amino acids was synthesized. Probe possesses the widely-used thiol-selective 2,4-dinitrobenzenesulfonyl (DNBS) group which can react with thiol and release the fluorescein which has strong fluorescence. Fluorescein, a well known xanthene fluorescent dye, has two states at different environment. Fluorescein is in the state of spirocyclization when connected with 2,4-dinitrobenzenesulfonyl (DNBS) group which has no fluorescence. However, it is in the state of open form when it reacts with thiol which has a strong fluorescence. The transition of the two states can be used to selectively detect thiol and the color can change from colorless to yellow which can be differentiated by naked eyes. Upon the titration of thiol, the absorption band at 454nm rises gradually and the fluorescence emerges at 521nm and the detection limit can be as low as 0.16μM. All of such good properties prove it to be a good sensor for the selective detection of thiol and it shows a potential use in bioimaging applications.

Abstract:Thiols are important units in amino acids such as cysteine and peptides like glutathione. Development of chemical sensors capable of precise detection of thiols is important in cancer diagnosis and therapy. We have developed novel two-photon fluorescent turn-on probes for selective detection of thiols. The probes displayed excellent sensitivity and low detection limits. The dual-purpose probes have been demonstrated to be suitable for simultaneous imaging and proteome profiling in live cells and tumor tissues. The unique turn-on design endows the probes with excellent selectivity toward thiols in vitro and in situ, and can be further developed to support a thiol-quantification assay.
