Abstract:The macular pigment (MP) in the radially-oriented Henle fibers that surround the foveola enables the ability to perceive the orientation of polarized blue light through an entoptic phenomena known as the Haidinger's brush. The MP has been linked to eye diseases and central field dysfunctions, most notably age-related macular degeneration (AMD), a globally leading cause of irreversible blindness. Recent integration of structured light techniques into vision science has allowed for the development of more selective and versatile entoptic probes of eye health that provide interpretable thresholds. For example, it enabled the use of variable spatial frequencies and arbitrary obstructions in the presented stimuli. Additionally, it expanded the 2° retinal eccentricity extent of the Haidinger's brush to 5° for a similar class of fringe-based stimuli. In this work, we develop a spatiotemporal sensitivity model that maps perceptual thresholds of entoptic phenomenon to the underlying MP structure that supports its perception. We therefore selectively characterize the circularly-oriented macular pigment optical density (coMPOD) rather than total MPOD as typically measured, providing an additional quantification of macular health. A study was performed where the retinal eccentricity thresholds were measured for five structured light stimuli with unique spatiotemporal frequencies. The results from fifteen healthy young participants indicate that the coMPOD is inversely proportional to retinal eccentricity in the range of 1.5° to 5.5°. Good agreement between the model and the collected data is found with a Pearson $\chi^2$ fit statistic of 0.06. The presented techniques can be applied in novel early diagnostic tests for a variety of diseases related to macular degeneration such as AMD, macular telangiectasia, and pathological myopia.

Abstract:The ability to perceive polarization-related entoptic phenomena arises from the dichroism of macular pigments held in Henle’s fiber layer of the retina and can be inhibited by retinal diseases, such as age-related macular degeneration, which alters the structure of the macula. Structured light tools enable the direct probing of macular pigment density and retinal structure through the perception of polarization-dependent entoptic patterns. Here, we directly measure the visual angle of an entoptic pattern created through the illumination of the retina with a structured state of light and a perception task that is insensitive to corneal birefringence. The central region of the structured light stimuli was obstructed, with the size of the obstruction varying according to a psychophysical staircase. Two stimuli, one producing 11 azimuthal fringes and the other three azimuthal fringes, were presented to 24 healthy participants. The pattern with 11 azimuthal fringes produced an average visual angle threshold of 10° ± 1° and a 95% confidence interval (C.I.) of [6°, 14°]. For the pattern with three azimuthal fringes, a threshold extent of 3.6° ± 0.3° C.I. = [1.3°, 5.8°] was measured, a value similar to the published extent of Haidinger’s brush (4°). The increase in apparent size and clarity of entoptic phenomena produced by the presented structured light stimuli offers the potential to detect the early signs of macular disease over perception tasks using uniform polarization stimuli.

Abstract:It was previously revealed that Wnt signaling is activated in mesothelioma cells. Although epidermal growth factor receptor (EGFR) is expressed in mesothelioma cells, EGFR-tyrosine kinase inhibitors (TKIs) are not effective for mesothelioma treatment. However, in non-small cell lung cancer, the blocking of Wnt signaling has been identified to enhance the anticancer effect of EGFR-TKIs. To confirm the anticancer effect of blocking Wnt signaling in combination with EGFR-TKI treatment in mesothelioma, the present study evaluated the effect of simultaneous suppression of human dishevelled-3 (Dvl-3) expression with Dvl-3 small interfering RNA (siRNA) and of EGFR inhibition with gefitinib on mesothelioma cell viability. Mesothelioma cell lines with and without β-catenin gene expression were transfected with Dvl-3 siRNA and were cultured with gefitinib, and cell viability, colony formation and cell cycle analyses were performed. Dvl-3 siRNA downregulated the expression of Dvl-3 in mesothelioma cells. The combination of Dvl-3 siRNA with gefitinib acted synergistically to induce concomitant suppression of cell viability and colony formation, suggesting that inhibition of Wnt signaling by downregulating Dvl-3 with siRNA and inhibiting EGFR with gefitinib leads to significant antitumor effects.

Abstract:The objective of the present study was to synthesize monomethoxypolyethyleneglycol-5000 cholesteryl ester [PEG–CH] as a cost-effective substitute for polyethyleneglycol–phosphatidylethanolamine and to evaluate the influence of its incorporation in liposomal bilayers for surface modification. PEG–CH was synthesized and characterized by infrared (IR), proton nuclear magnetic resonance spectroscopy (1H NMR), and differential scanning calorimetry (DSC) studies. Influence of incorporation of PEG–CH in liposomes was evaluated on various parameters such as zeta potential, DSC, and encapsulation efficiency of a hydrophilic drug pentoxyfylline. Conventional and PEG–CH containing pentoxyfylline liposomes were formulated and their stability was evaluated at 4°C for 3 months. PEG–CH could be successfully synthesized with good yields and the structure was confirmed by IR, DSC, and 1H NMR. The incorporation of PEG–CH in liposomes resulted in reduction of the zeta potential and broadening of the DSC endotherm. Furthermore, incorporation of PEG–CH in liposomes decreased the encapsulation efficiency of pentoxifylline in liposomes when compared to conventional liposomes. Conventional and PEG–CH containing pentoxyfylline liposomes did not show any signs of pentoxyfylline degradation when stored at 4°C for 3 months.

Abstract:ABSTRACT The maintenance of visual acuity during active and passive body motion is ensured by gaze-stabilizing reflexes that aim at minimizing retinal image slip. For the optokinetic reflex (OKR), large-field visual motion of the surround forms the essential stimulus that activates eye movements. Properties of the moving visual world influence cognitive motion perception and the estimation of visual image velocity. Therefore, the performance of brainstem-mediated visuo-motor behaviors might also depend on image scene characteristics. Employing semi-intact preparations of mid-larval stages of Xenopus laevis tadpoles, we studied the influence of contrast polarity, intensity, contour shape and different motion stimulus patterns on the performance of the OKR and multi-unit optic nerve discharge during motion of a large-field visual scene. At high contrast intensities, the OKR amplitude was significantly larger for visual scenes with a positive contrast (bright dots on a dark background) compared with those with a negative contrast. This effect persisted for luminance-matched pairs of stimuli, and was independent of contour shape. The relative biases of OKR performance along with the independence of the responses from contour shape were closely matched by the optic nerve discharge evoked by the same visual stimuli. However, the multi-unit activity of retinal ganglion cells in response to a small single moving vertical edge was strongly influenced by the light intensity in the vertical neighborhood. This suggests that the underlying mechanism of OKR biases related to contrast polarity directly derives from visual motion-processing properties of the retinal circuitry.

Abstract:The interactions of spatial and chromatic processing of the short-wavelength-sensitive cone mechanism were studied in humans with patterned (checkerboard) stimuli of various spatial frequency (10, 22, 44, and 85 min of are respectively), under steady exposure to yellow light (575 nm, 390 cd/m2). Psychophysical studies and pattern-reversal visual evoked potentials were employed. Parameters of the transient pattern-reversal visual evoked potentials (pattern reversal rate of 2.4 s-1) especially observed were the latencies of P2 (P100) and N3 and the amplitude of P2-N3. It was only with the largest applicable check size (85 min of arc) that both the psychophysical studies and visual evoked potentials could succeed in satisfactorily isolating the short-wavelength-sensitive cone mechanism. Pattern-reversal visual evoked potential latencies are recommended in the evaluation of this cone mechanism because of their smaller variance and higher selectivity in isolating the short-wavelength-sensitive cone mechanism than the amplitude. The peak sensitivity of this cone mechanism was shown to be about 449 nm at the corneal level. The short-wavelength sensitive cone mechanism represented the characteristics of low spatial resolution and long latencies of the pattern-reversal visual evoked potentials.
Investigating focal macular electroretinogram function for fundus‐related perimetry application

Abstract:Aims/Purpose: This study aimed to investigate the utility of focal macular electroretinogram (ERG) function in fundus‐related perimetry (microperimetry) for assessing cryptogenic diseases such as macular dystrophy. The decline in visual sensitivity of the retina was measured using microperimetry, and the retinal layer causing this decline was identified using multifocal ERG.A focal macular ERG function was used for microperimetry instead of multifocal ERG. Methods: The developed apparatus comprised illumination and photographic optical systems separated by a mirror with a hole, a microperimetry optical system and an ERG optical system separated by a 45° cold mirror. It also consisted of an Edmund infrared camera EO‐50231, a lens, a halogen lamp, an objective lens, four double convex lenses, two aperture stops, a mirror, a 12.1‐inch high brightness (2000 cd/m 2 ) monitor and an artificial eye. Its electrical measuring equipment included a differential amplifier, a high‐pass filter, a 50‐Hz notch filter and two non‐inverting amplifiers. Moreover, the equipment consisted of the National Instruments' I/O device USB‐6216, a shielded connector block, plate electrodes, an electromagnetic shield sheet and the LabVIEW 2018 software for data retrieval. To perform the focal macular ERG, a DynaScan 49‐inch ultrahigh brightness (3500 cd/m2) liquid crystal display (LCD) was prepared. Results: The ERG software could display three stimulus spot circles with diameters of 5°, 10° and 15° on the ultrahigh brightness LCD. Additionally, a function for adjusting the size of the stimulus spot circle was added as the diameter was changed from 1° to 15°. The focal macular ERG was able to measure a wave, b wave and photopic negative response (PhNR). It was found that certain small waves on the b wave were confirmed with high probability as oscillatory potentials (OPs), while one positive wave similar to the d wave was confirmed sometimes; however, the c wave was not confirmed. Conclusions: It can be concluded that when not only the a wave, b wave and PhNR but also the OPs and d wave are confirmed, the focal macular ERG function can identify the retinal layer causing visual alterations at specific points throughout the retina.

Contribution of Bipolar Cells of Cone ON and OFF Pathways to Electroretinograms Elicited by Ultraviolet and Middle Wavelength Stimuli

Abstract:Purpose: To determine the contribution of the ON and OFF cone bipolar cell pathways to the electroretinograms (ERGs) elicited by ultraviolet (UV) and middle wavelength light in mice. Materials and methods: The experiments were performed on 8- to 10-week-old C57BL/6J mice. The ERGs elicited by single-flash and flickering UV light stimuli were compared to those elicited by green light stimuli under photopic conditions. Pharmacological agents were used to selectively block the ON and OFF pathways contributing to the ERGs. Saline was used as a control. The flicker ERGs elicited by UV light were compared to the ERGs elicited by green light after the injection of the pharmacological agents to determine the contribution of the cone ON and OFF pathways to the ERGs. Results: The photopic single-flash and flicker ERGs were more sensitive to the UV light stimuli than to those elicited by green light stimuli. The flicker ERG responses elicited by both UV and green light stimuli at stimulus frequencies lower than 15-Hz decreased after L-2-amino-4-phosphobutyric acid was injected. The ERGs elicited by UV light at 30-Hz and by green light at frequencies lower than 15-Hz decreased significantly after the intravitreal injection of cis-2, 3-piperidine-dicarboxylic acid. An analysis of the ON and OFF components of the flicker ERGs showed that there might be pharmacological differences between the UV light-sensitive responses and the green light-sensitive responses. Conclusions: These results suggest that the UV light-sensitive cones connect to both the ON and OFF bipolar cells differently than that of the green light-sensitive cones.
[Blue-sensitive mechanisms in pattern ERG and VEP]

Abstract:Pattern onset-offset ERGs and VEPs in response to square-wave stripe patterns of different wavelengths and spatial frequencies under chromatic yellow adaptation (two-channel Maxwellian view) were studied in three subjects. One channel provided a steady homogeneous adapting field of 570 nm, while the other channel, containing the patterns, was superimposed on the adapting field (33 degrees). The spectral sensitivity curve of the ERG had a prominent peak in the blue (460 nm) and a shoulder in the red and green (above 500 nm). For wavelengths below 500 nm the shape of the psychophysically determined sensitivity curve to the onset and offset of the pattern was in close agreement with the electroretinal sensitivity curves determined for a low-amplitude criterion. With almost all wavelengths there were two components in the same VEP response at the same time: a slow negative wave with maximum sensitivity at 460 nm and an earlier fast positive component with maximum sensitivity in the red-green region. When the spatial frequency of a 460 nm test pattern was varied under yellow adaptation the onset response in the ERG showed only very slight spatial tuning. In the onset response of the VEP, however, the slow component showed prominent spatial tuning (around 2 c/deg) under the same conditions.
A comparison of visual electrophysiology measures in optic neuropathy

Abstract:Aims/Purpose: Our aim is to explore the inter‐relationship of clinical PVEP and PERG measurements with each other and the optic nerve structure in children investigated for possible optic neuropathies. Methods: ISCEV Standard PVEPs and PERGs to 50′ check widths to large 30 and standard 15 degree fields were reviewed from children suspected to have optic neuropathy of various pathology. Amplitudes and peak times of PERG P50, N95, the N95:P50 amplitude ratio, the amplitude of the N95 from baseline and at 30 ms from the P50‐N95 slope were measured. Spectralis OCT RNFL sector measures and macular ganglion cell volumes were collated. Results: PERG, PVEP with OCT measurements were available for 42 patients, (83 eyes) aged 6‐17 yrs. Those with abnormal PVEPs (33 eyes) had the most severe macular RGC volume loss (mean 0.26 mm 3 in 3.45 mm) with mean LogMAR VA 0.52 (range − 0.01 to 1.00). These patients had inherited optic neuropathies, optic neuritis associated with MOGAD and RMMS. In contrast, patients with papilledema, such as associated with IIH, or presenting with headache and reduced vision and full discs had normal PVEPs. Mean VA LogMAR 0.23 (range − 1.0 to 0.9), and mean macular RGC volume 0.45 mm 3 in 3.45 mm circle. N95:P50 amplitude ratios ranged from 1.83 to 0.44 in 30 degree fields and 2.16 to 0.36 in 15 degree fields. The N95:P50 amplitude ratios, slope at 30 ms and N95 from baseline correlated with macular RGC volume, but the best PERG discriminator (AUC 0.83) in the large field was the N95 slope measured at 30 ms. RNFL values were confounded by disc oedema in many cases. Conclusions: Pattern reversal VEPs were abnormal in all patients with severe RGC volume loss. The PERG measures correlate with macular RGC volume. RNFL was confounded by disc oedema, but was helpful in highlighting temporal sector thinning in optic neuritis and inherited optic atrophy such as LHOA. References 1. Visual evoked potentials in three-dimensional color space: correlates of spatio-chromatic processing

Abstract:Visual evoked potentials (VEPs) were measured for sinusoidal gratings with spatio-chromatic modulation defined in a three-dimensional color space. The spatio-chromatic modulation of the gratings can be decomposed into contributions from an achromatic luminance varying component, an isoluminant component which modulates only the activities of L cones and M cones, and an isoluminant component corresponding to modulation of only S-cone activity. The emphasis of this report is the nature of VEPs resulting from isoluminant spatio-chromatic modulation. The VEP response was characterized along a number of spatial, temporal, and chromatic stimulus dimensions: contrast, spatial frequency, chromaticity in the isoluminant plane, chrominance/luminance ratio, orientation, and temporal frequency. Isoluminant VEPs resulting from stimuli modulating L and M cones are compared with those from S-cone modulation. When appropriate spatiotemporal conditions are employed, both types produce robust VEPs; however, the S-pathway VEPs show considerably longer latencies than do those from LM-pathway activation. The VEP results are compared to psychophysical and single unit electrophysiological observations. VEP latencies exhibit the lowpass character of psychophysical chromatic contrast sensitivity functions but VEP amplitudes show bandpass tuning along both the S and LM axes. An oblique effect, i.e. shorter latencies for horizontal and vertical gratings than for diagonal, is observed in the isoluminant VEP. S-pathway VEPs are used to demonstrate an electophysiological correlate of transient tritanopia. Normative amplitude and latency data for VEPs from selectivity stimulated chromatic mechanisms provide a baseline for clinical electrodiagnostic applications.
A distinct contribution of short-wavelength-sensitive cones to light-evoked activity in the mouse pretectal olivary nucleus

Abstract:Melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs) combine inputs from outer-retinal rod/cone photoreceptors with their intrinsic phototransduction machinery to drive a wide range of so-called non-image-forming (NIF) responses to light. Defining the contribution of each photoreceptor class to evoked responses is vital for determining the degree to which our sensory capabilities depend on melanopsin and for optimizing NIF responses to benefit human health. We addressed this problem by recording electrophysiological responses in the mouse pretectal olivary nucleus (PON) (a target of ipRGCs and origin of the pupil light reflex) to a range of gradual and abrupt changes in light intensity. Dim stimuli drove minimal changes in PON activity, suggesting that rods contribute little under these conditions. To separate cone from melanopsin influences, we compared responses to short (460 nm) and longer (600/655 nm) wavelengths in mice carrying a red shifted cone population (Opn1mw®) or lacking melanopsin (Opn4⁻/⁻). Our data reveal a surprising difference in the quality of information available from medium- and short-wavelength-sensitive cones. The majority cone population (responsive to 600/655 nm) supported only transient changes in firing and responses to relatively sudden changes in light intensity. In contrast, cones uniquely sensitive to the shorter wavelength (S-cones) were better able to drive responses to gradual changes in illuminance, contributed a distinct off inhibition, and at least partially recapitulated the ability of melanopsin to sustain responses under continuous illumination. These data reveal a new role for S-cones unrelated to color vision and suggest renewed consideration of cone contributions to NIF vision at shorter wavelengths.
Pattern-reversal electroretinogram in response to chromatic stimuli: I. Humans

Abstract:We have studied the steady-state PERG in human subjects in response to red-green plaid patterns modulated either in luminance or in chromaticity or both. By varying the relative luminance of the red and green components, a value could be obtained at which the PERG amplitude was either minimum or locally maximum. This always occurred at equiluminance, as measured by standard psychophysical techniques. PERG amplitude and phase were measured as a function of spatial and temporal frequency of sinusoidal contrast reversal. In both space and time, the response to chromatic patterns was low-pass, while that to luminance was band-pass, and extended to higher spatial and temporal frequencies. The phase of the PERG to chromatic stimuli was systematically lagged compared with that to luminance stimuli, by an amount corresponding to about 20 ms under our experimental conditions. The variation of phase with temporal frequency suggested an apparent latency of about 67 ms for color contrast compared with 47 ms for luminance. These estimates were confirmed with separate measurements of transient PERGs to abrupt contrast reversal. For both luminance and chromatic stimuli, the amplitude of PERGs increases with increasing stimulus contrast. By summing vectorially the responses to appropriate luminance and chromatic contrasts, we were able to predict with accuracy the response as a function of color ratio (ratio of red to total luminance). The above findings all agree with those reported in the accompanying paper for the monkey PERG (Morrone et al., 1994), and indicate that the differences in response latency and integration time of luminance and chromatic stimuli observed by psychophysical and VEP techniques may arise at least in part from the properties of retinal mechanisms.
The glare hypothesis of macular pigment function

Abstract:Purpose: Discomfort and reduced visual performance due to glaring light conditions are common complaints for most individuals. Past studies have shown that macular pigment (MP) reduces discomfort due to glare. In this study, we evaluated whether MP was related to visual performance under glare conditions. Methods: Thirty-six healthy subjects participated (age range, 18 to 41). Spatial profiles of MP optical density were measured using heterochromatic flicker photometry with a Newtonian-view macular densitometer. Photostress recovery and grating visibility under veiling conditions were assessed in a Maxwellian-view optical system. Both experiments used six monochromatic lights (from 440 to 620 nm) and a broadband xenon white. For the veiling glare experiment, subjects fixated a 1 degrees diameter disk containing a black and white 100% contrast grating stimulus. The intensity of an annulus (the glare source) with an 11 degrees inner and 12 degrees outer diameter was adjusted by the subject until the grating stimulus was no longer seen. For the photostress recovery experiment, the time required to detect a 1 degrees -diameter grating stimulus (detailed above) after a 5-s exposure to a 2.5 microW/cm2, 5 degrees -diameter disk was recorded. Both central and eccentric (10 degrees temporal retina) viewing conditions were assessed. Results: MP at 30' eccentricity ranged from 0.08 to 1.04 OD, and was found to dramatically reduce the deleterious effects of glare. Visual thresholds under glare conditions were strongly related to MP density (e.g., r = 0.76, p = 0.0001 when using white light). Photostress recovery time, after exposure to xenon-white light, was significantly shorter for subjects with higher MP levels (r = -0.79, p = 0.0001). Both photostress recovery and veiling glare functions were well-described by the photopic spectral sensitivity function (Vlambda). Conclusions: MP is strongly related to improvements in glare disability and photostress recovery in a manner consistent with its spectral absorption and spatial profile.
Simulating the Effects of Partial Neural Conduction Delays in the Visual Evoked Potential

Abstract:Purpose: The purpose of this study was to understand the double peaks or broadening of P100 observed in some cases of optic neuritis by inducing conduction delays in healthy eyes through stimulus luminance manipulation in analogy to the perceptual Pulfrich effect.Methods: Checkerboard pattern reversal visual evoked potentials (VEPs) with check sizes of 0.8 degrees, 0.4 degrees, and 0.2 degrees were recorded in healthy participants using two experiment variants. Variant (1) involved binocular stimulation with inter-ocular luminance difference achieved by a 1.8 neutral density (ND) filter, along with monocular control conditions. Variant (2) included monocular stimulation with hemifields having a luminance difference (half of monitor with ND filter), along with single-hemifield control conditions. In both variants, VEP curves under mixed stimulation were compared to synthesized VEPs computed from offline summation of curves from the relevant control conditions, followed by assessing P100 characteristics.Results: Despite considerable variability between participants, the binocular variant demonstrated marked differences between VEPs from mixed recordings and synthesized curves, whereas in the hemifield variant, agreement was strong. The anticipated double peak or broadened deflection pattern was observed to varying extents in participants, often contingent on check size, with nominal peak time frequently failing to indicate partial conduction delays.Conclusions: The present findings corroborate the hypothesis that nominal peak time does not always reflect conduction delays if only a subset of fiber bundles is affected. Peak shape might provide additional diagnostic evidence of a partial conduction delay.Translational Relevance: Enhancing the understanding of VEP waveform changes associated with partial conduction delays could offer diagnostic insights for optic neuritis.

Abstract:Optimal stimulus parameters for epiretinal prostheses have been investigated by analyzing retinal ganglion cell (RGC) spiking responses to white-noise electrical stimulation, through a spike-triggered average (STA) analysis technique. However, it is currently unknown as to activation of which retinal cells contribute to features of the STA. We conducted whole-cell patch clamping recordings in ON and OFF RGCs in response to white-noise epiretinal electrical stimulation by using different inhibitors of synaptic transmission in a healthy retina. An mGluR6 agonist, L-AP4, was firstly used to selectively block the output of photoreceptors (PRs) to ON bipolar cells (BCs). We subsequently fully blocked all synaptic inputs to RGCs using a combination of pharmacological agents. Our data shows that PRs dominate the ability of ON RGCs to integrate electrical pulses and form a unique STA shape, while BCs do not contribute in any way. In addition, our results demonstrate that the ability of OFF RGCs to integrate pulses is consistently impaired after blocking the PR to ON BC pathway. We hypothesise that the mechanisms underlying this co-effect are related to the narrow field AII amacrine cells connecting ON and OFF pathways.Clinical Relevance-Recent retinal studies recorded mirror-inverted STAs in ON and OFF retinal pathways, thus raising the possibility of designing a stimulation approach that can differentially activate ON and OFF pathways with electrical stimulation. However, the detailed contribution of three major retinal cell layers in forming characteristic STAs is still unclear. It is of great clinical relevance to investigate the isolated contribution of PRs to the electrically driven STA since PRs progressively degenerate in the course of retinal disease.
