Microfluidics has the potential to influence subject areas from chemical synthesis and biological analysis to optics and information technology. But the field is still at an early stage of development. Even as the basic science and technological demonstrations develop, other problems must be addressed: choosing and focusing on initial applications, and developing strategies to complete the cycle of development, including commercialization. The solutions to these problems will require imagination and ingenuity.

Abstract:Miniaturization has been the driving force of scientific and technological advances over recent decades. Recently, flexibility has gained significant interest, particularly in miniaturization approaches for biomedical devices, wearable sensing technologies, and drug delivery. Flexible microfluidics is an emerging area that impacts upon a range of research areas including chemistry, electronics, biology, and medicine. Various materials with flexibility and stretchability have been used in flexible microfluidics. Flexible microchannels allow for strong fluid-structure interactions. Thus, they behave in a different way from rigid microchannels with fluid passing through them. This unique behaviour introduces new characteristics that can be deployed in microfluidic applications and functions such as valving, pumping, mixing, and separation. To date, a specialised review of flexible microfluidics that considers both the fundamentals and applications is missing in the literature. This review aims to provide a comprehensive summary including: (i) Materials used for fabrication of flexible microfluidics, (ii) basics and roles of flexibility on microfluidic functions, (iii) applications of flexible microfluidics in wearable electronics and biology, and (iv) future perspectives of flexible microfluidics. The review provides researchers and engineers with an extensive and updated understanding of the principles and applications of flexible microfluidics.

Abstract:New fabrication technologies such as layer transfer and lamination, soft lithography, inkjet printing, and 3D printing are developed for flexible microfluidics. Flexible microfluidics is widely used, especially in wearable applications, including biosensing and motion sensing. This chapter provides information on flexible materials, fabrication technologies, and the applications of flexible microfluidics. Many materials have been developed and used in flexible microfluidics. These materials can be divided into three classes: polymer, silicone elastomer, and fabric. Each of these materials possesses inherent qualities compatible for different applications. The chapter introduces fabrication methods of traditional microfluidic devices made by polymer and silicone elastomer and the processing methods of open‐surface microfluidics made by fabric. With the fast development of flexible materials and fabrication methods, the combination of fluid manipulation and flexible electronics promotes the application of functional microfluidics, e.g. wearable microfluidics for biosensing and health care.

Abstract:Flexible electronics has extensively drawn attention in an extremely wide range of areas due to its unique advantages like portability, biocompatibility, wearability, and superior mechanical stability. Recently, microfluidic technology has been considered as an effective tool to fabricate flexible electronics because of its precise control and manipulation of fluids in microchannels. In this review, we provide a comprehensive and in-depth insight into flexible electronics from microfluidic technology, covering the basic elements as well as microfluidic fabrication of flexible electronics and various applications in biomedical engineering. Furthermore, an outlook for the future challenges and perspectives of the flexible electronics is proposed.

Abstract:This article presents a simple, low-cost method of fabrication and the applications of flexible polystyrene microfluidic devices with three-dimensional (3D) interconnected microporous walls based on treatment using a solvent/non-solvent mixture at room temperature. The complete fabrication process from device design concept to working device can be completed in less than an hour in a regular laboratory setting, without the need for expensive equipment. Microfluidic devices were used to demonstrate gas generation and absorption reactions by acidifying water with carbon dioxide (CO(2)) gas. By selectively treating the microporous structures with oxygen plasma, acidification of water by acetic acid (distilled white vinegar) perfusion was also demonstrated with the same device design.

Abstract:Acoustic actuation of fluids at small scales may finally enable a comprehensive lab-on-a-chip revolution in microfluidics, overcoming long-standing difficulties in fluid and particle manipulation on-chip. In this comprehensive review, we examine the fundamentals of piezoelectricity, piezoelectric materials, and transducers; revisit the basics of acoustofluidics; and give the reader a detailed look at recent technological advances and current scientific discussions in the discipline. Recent achievements are placed in the context of classic reports for the actuation of fluid and particles via acoustic waves, both within sessile drops and closed channels. Other aspects of micro/nano acoustofluidics are examined: atomization, translation, mixing, jetting, and particle manipulation in the context of sessile drops and fluid mixing and pumping, particle manipulation, and formation of droplets in the context of closed channels, plus the most recent results at the nanoscale. These achievements will enable applications across the disciplines of chemistry, biology, medicine, energy, manufacturing, and we suspect a number of others yet unimagined. Basic design concepts and illustrative applications are highlighted in each section, with an emphasis on lab-on-a-chip applications.

Abstract:Microfluidic devices have been widely used since 1990s for diverse manipulations of particles (a general term of beads, cells, vesicles, drops, etc.) in a variety of applications. Compared to the active manipulation via an externally imposed force field, the passive manipulation of particles exploits the flow-induced intrinsic lift and/or drag to control particle motion with several advantages. Along this direction, inertial microfluidics has received tremendous interest in the past decade due to its capability to handle a large volume of samples at a high throughput. This inertial lift-based approach in Newtonian fluids, however, becomes ineffective and even fails for small particles and/or at low flow rates. Recent studies have demonstrated the potential of elastic lift in non-Newtonian fluids for manipulating particles with a much smaller size and over a much wider range of flow rates. The aim of this article is to provide an overview of the various passive manipulations, including focusing, separation, washing and stretching, of particles that have thus far been demonstrated in non-Newtonian microfluidics.

Abstract:Microfluidics has been the key component for many applications, including biomedical devices, chemical processors, microactuators, and even wearable devices. This technology relies on soft lithography fabrication which requires cleanroom facilities. Although popular, this method is expensive and labor-intensive. Furthermore, current conventional microfluidic chips precludes reconfiguration, making reiterations in design very time-consuming and costly. To address these intrinsic drawbacks of microfabrication, we present an alternative solution for the rapid prototyping of microfluidic elements such as microtubes, valves, and pumps. In addition, we demonstrate how microtubes with channels of various lengths and cross-sections can be attached modularly into 2D and 3D microfluidic systems for functional applications. We introduce a facile method of fabricating elastomeric microtubes as the basic building blocks for microfluidic devices. These microtubes are transparent, biocompatible, highly deformable, and customizable to various sizes and cross-sectional geometries. By configuring the microtubes into deterministic geometry, we enable rapid, low-cost formation of microfluidic assemblies without compromising their precision and functionality. We demonstrate configurable 2D and 3D microfluidic systems for applications in different domains. These include microparticle sorting, microdroplet generation, biocatalytic micromotor, triboelectric sensor, and even wearable sensing. Our approach, termed soft tubular microfluidics, provides a simple, cheaper, and faster solution for users lacking proficiency and access to cleanroom facilities to design and rapidly construct microfluidic devices for their various applications and needs.

Abstract:Microfabricated integrated circuits revolutionized computation by vastly reducing the space, labor, and time required for calculations. Microfluidic systems hold similar promise for the large-scale automation of chemistry and biology, suggesting the possibility of numerous experiments performed rapidly and in parallel, while consuming little reagent. While it is too early to tell whether such a vision will be realized, significant progress has been achieved, and various applications of significant scientific and practical interest have been developed. Here a review of the physics of small volumes (nanoliters) of fluids is presented, as parametrized by a series of dimensionless numbers expressing the relative importance of various physical phenomena. Specifically, this review explores the Reynolds number Re, addressing inertial effects; the Péclet number Pe, which concerns convective and diffusive transport; the capillary number Ca expressing the importance of interfacial tension; the Deborah, Weissenberg, and elasticity numbers De, Wi, and El, describing elastic effects due to deformable microstructural elements like polymers; the Grashof and Rayleigh numbers Gr and Ra, describing density-driven flows; and the Knudsen number, describing the importance of noncontinuum molecular effects. Furthermore, the long-range nature of viscous flows and the small device dimensions inherent in microfluidics mean that the influence of boundaries is typically significant. A variety of strategies have been developed to manipulate fluids by exploiting boundary effects; among these are electrokinetic effects, acoustic streaming, and fluid-structure interactions. The goal is to describe the physics behind the rich variety of fluid phenomena occurring on the nanoliter scale using simple scaling arguments, with the hopes of developing an intuitive sense for this occasionally counterintuitive world.

Abstract:Microfluidics devices are powerful tools for studying dynamic processes in live cells, especially when used in conjunction with light microscopy. There are many applications of microfluidics devices including recording dynamic cellular responses to small molecules or other chemical conditions in perfused media, monitoring cell migration in constrained spaces, or collecting media perfusate for the study of secreted compounds in response to experimental inputs/manipulations. Here we describe a configurable low-cost (channel-based) microfluidics platform for live-cell microscopy, intended to be useful for experiments that require more precision/flexibility than simple rubber spacers, but less precision than molded elastomer-based platforms. The materials are widely commercially available, low-cost, and device assembly takes only minutes.

An Alternative Micro-Milling Fabrication Process for Rapid and Low-Cost Microfluidics

Abstract:Microfluidics is an important technology for the biomedical industry and is often utilised in our daily lives. Recent advances in micro-milling technology have allowed for rapid fabrication of smaller and more complex structures, at lower costs, making it a viable alternative to other fabrication methods. The microfluidic chip fabrication developed in this research is a step-by-step process with a self-contained wet milling chamber. Additionally, ethanol solvent bonding is used to allow microfluidic chips to be fully fabricated within approximately an hour. The effect of using this process is tested with quantitative contact profileometery data to determine the expected surface roughness in the microchannels. The effect of surface roughness on the controllability of microparticles is tested in functional microfluidic chips using image processing to calculate particle velocity. This process can produce high-quality channels when compared with similar studies in the literature and surface roughness affects the control of microparticles. Lastly, we discuss how the outcomes of this research can produce rapid and higher-quality microfluidic devices, leading to improvement in the research and development process within the fields of science that utilise microfluidic technology. The diversity in technologies as well as in applications makes this an intriguing story, but it is in the simplest of materials - paper - that we find some of the most successful applications so far.

Abstract:We describe new developments for controlled fabrication of monodisperse non-spherical particles using droplet microfluidics. The high degree of control afforded by microfluidic technologies enables generation of single and multiple emulsion droplets. We show that these droplets can be transformed to non-spherical particles through further simple, spontaneous processing steps, including arrested coalescence, asymmetric polymer solidification, polymerization in microfluidic flow, and evaporation-driven clustering. These versatile and scalable microfluidic approaches can be used for producing large quantities of non-spherical particles that are monodisperse in both size and shape; these have great potential for commercial applications.

Abstract:The advance of microfluidics started in the early 1980s. At the time, researchers realized that many processes and reactions in chemistry and biology, which typically take place on small length scales, can be defined, controlled and understood much better when using tools on equally small length scales. Reactions and reaction kinetics rely on (gradual) concentration differences and microfluidics provides the unique possibility to establish exactly such gradients of solutes, ion concentrations, pH value and so on. Nowadays the variety of specific microfluidic methods is large. In principle, they can be divided into two groups: (i) monophase flow, where miscible (e.g. aqueous) fluids are mixed, mostly by diffusion owing to the laminar flow on small length scales and (ii) multiphase flow, the most prominent example of which is probably droplet microfluidics, where water-in-oil or oil-in-water emulsions are used to encapsulate chemical or biological systems and separate them from each other, much like in 'micron-scale test tubes'. Now, 30 years later, microfluidic techniques are seriously considered for industrial applications, although some important steps in the upscaling process are still missing. The purpose of this special issue is to shed light on the different aspects in microfluidics research starting from fundamental research reaching all the way to industrial applications. The study by Toma and co-workers takes advantage of the controlled diffusive mixing when co-flowing aqueous, miscible solutions. They combine microfluidics with optical, spectroscopic and scattering techniques to study DNA packing. Nunes et al review the different regimes when replacing one of the fluids by an oil phase and varying flow rates and device geometries with a particular emphasis on using multiphase microfluidics for synthesis of particles or fibres. Going into the third dimension by fabricating microfluidic devices with several layers, producing emulsions can also be achieved by so-called 'step emulsification', the physical mechanisms behind which are described by Dangla et al. Tran and co-workers move a considerable step towards applicability of water-in-oil emulsions for biological research and review ultrahigh-throughput methods used for bio-assays. The article by Lagus et al focuses this topic specifically on single-cell experiments. Whereas it is very popular to use emulsions with drop sizes of a few tens of micrometers as 'tiny test tubes' they may also serve as templates for materials fabrication. Gundabala and co-workers combine both aspects by producing so-called 'celloidosomes', which consist of liquid drops decorated with yeast cells at the outer interface. Wang et al fabricate microcrawlers that can be thermally set in motion. Finally, Holtze gives a perspective on the possibility to upscale and apply such methods in industry. The choice of papers shows the wide and diverse applicability of microfluidics in various fields of research. While microfluidics started out as a 'niche' technique for very specific applications and as a tool in fundamental soft and biological matter research, the advancements made during recent years promise further progress in the chemical industry, biomedicine and pharmacology. Advantages such as low sample consumption, single cell accessibility and controlled experimental parameters in general may in the future be exploited for real industrial sized applications. In Journal of Physics D: Applied Physics we find a journal that is ideally positioned to give applied microfluidics research a wide readership across many disciplines. In the publication of this special issue we hope to inspire and encourage microfluidics researchers, and to promote interdisciplinary collaborations. We would like to thank all of the authors for their excellent contributions to this special issue.

Abstract:Droplet microfluidics offers exquisite control over the flows of multiple fluids in microscale, enabling fabrication of advanced microparticles with precisely tunable structures and compositions in a high throughput manner. The combination of these remarkable features with proper materials and fabrication methods has enabled high efficiency, direct encapsulation of actives in microparticles whose features and functionalities can be well controlled. These microparticles have great potential in a wide range of bio-related applications including drug delivery, cell-laden matrices, biosensors and even as artificial cells. In this review, we briefly summarize the materials, fabrication methods, and microparticle structures produced with droplet microfluidics. We also provide a comprehensive overview of their recent uses in biomedical applications. Finally, we discuss the existing challenges and perspectives to promote the future development of these engineered microparticles.

Abstract:The amount of applications associated with microfluidic devices is increasing since the introduction of Lab-on-a-chip devices in the 1990s, especially regarding biomedical and clinical fields. However, in order for this technology to leave the fundamental research and become a day-life technology (e.g., as point-of-care testing), it needs to be disposable and reasonably less expensive. Polymers, due to their several advantages, such as easier microfabrication and low-cost, fill these needs. Several methods are reported regarding microfabrication and, thus, the main aim of the present work is to provide an overview of the most relevant microfabrication techniques found in literature employing polymers, clarifying also the main advantages and disadvantages of each technique and especially considering their cost and time-consumption. Moreover, a future outlook of low-cost microfabrication techniques and standard methods is provided.

Abstract:Microfluidics have emerged as a vital tool in various scientific and engineering applications, ranging from biomedical diagnostics to chemical analysis. However, conventional fabrication methods often necessitate costly equipment and limiting accessibility, thereby hindering widespread adoption. In response, this note presents a cost-effective approach for fabricating microfluidic channels using photolithographic methods, which circumvents the need for expensive instruments. The procedure commences with the preparation of a suitable substrate, followed by the application of a photoresist layer. A photomask, containing the desired microfluidic pattern, is then used to define the layout on the photoresist-coated substrate. Upon exposure to ultraviolet (UV) light, the photoresist undergoes a chemical transformation, allowing for selective removal of either exposed or unexposed regions. Consequently, the patterned microfluidic channels are unveiled on the substrate. The versatility of this photolithographic method allows for the fabrication of intricate and customizable microchannels with relative ease and precision. Further processing steps, such as etching or surface treatment, optimize the substrate's properties for specific applications. This low cost approach not only empowers researchers and engineers to develop microfluidic devices at a fraction of the cost but also fosters broader access to photolithography related research and development. The enhanced accessibility opens doors to a myriad of applications, ranging from advanced diagnostics and drug delivery systems to portable point-of-care devices. Overall, this paper showcases a promising avenue for expanding the reach of microfluidics, fostering innovation, and driving impactful advancements in diverse scientific disciplines.
