Abstract:The holy grail for every neurophysiologist is to conclude a causal relationship between an elementary behaviour and the function of a specific brain area or circuit. Our effort to map elementary behaviours to specific brain loci and to further manipulate neural activity while observing the alterations in behaviour is in essence the goal for neuroscientists. Recent advancements in the area of experimental brain imaging in the form of longer wavelength near infrared (NIR) pulsed lasers with the development of highly efficient optogenetic actuators and reporters of neural activity, has endowed us with unprecedented resolution in spatiotemporal precision both in imaging neural activity as well as manipulating it with multiphoton microscopy. This readily available toolbox has introduced a so called all-optical physiology and interrogation of circuits and has opened new horizons when it comes to precisely, fast and non-invasively map and manipulate anatomically, molecularly or functionally identified mesoscopic brain circuits. The purpose of this review is to describe the advantages and possible pitfalls of all-optical approaches in system neuroscience, where by all-optical we mean use of multiphoton microscopy to image the functional response of neuron(s) in the network so to attain flexible choice of the cells to be also optogenetically photostimulated by holography, in absence of electrophysiology. Spatio-temporal constraints will be compared toward the classical reference of electrophysiology methods. When appropriate, in relation to current limitations of current optical approaches, we will make reference to latest works aimed to overcome these limitations, in order to highlight the most recent developments. We will also provide examples of types of experiments uniquely approachable all-optically. Finally, although mechanically non-invasive, all-optical electrophysiology exhibits potential off-target effects which can ambiguate and complicate the interpretation of the results. In summary, this review is an effort to exemplify how an all-optical experiment can be designed, conducted and interpreted from the point of view of the integrative neurophysiologist.

Synaesthesia, the arts and creativity: a neurological connection

Abstract:For over 100 years the link between synaesthesia and the arts has attracted controversy. This has been spurred by the production of auditory, literary and visual art by famous individuals who report experiences synonymous with the neurological condition. Impressive protagonists in this discussion include Arthur Rimbaud, Charles Baudelaire, Vasily Kandinsky, Vladimir Nabokov, Alexander Scriabin, Olivier Messiaen and David Hockney. Interdisciplinary debates have concerned whether synaesthesia can actively contribute to an artist's ability, whether it is a driving force or a mere idiosyncratic quirk and whether, fundamentally, it is a distinct idiopathic condition or an unusual metaphorical description of normal perception. Recent psychological and neuroscientific evidence offers a new level to the debate. Coherent patterns of a neural basis of synaesthesia have been confirmed with high spatial resolution brain imaging techniques and the link with the arts is transpiring to be more than superficial or coincidental. Moreover, the neural distinction of the synaesthete brain may prove to be a window into a neural basis of creative cognition, and therefore conducive to the expression of creativity in various media.

Abstract:This report is the second part of a comprehensive two-part series aimed at reviewing an extensive and diverse toolkit of novel methods to explore brain health and function. While the first report focused on neurophotonic tools mostly applicable to animal studies, here, we highlight optical spectroscopy and imaging methods relevant to noninvasive human brain studies. We outline current state-of-the-art technologies and software advances, explore the most recent impact of these technologies on neuroscience and clinical applications, identify the areas where innovation is needed, and provide an outlook for the future directions.

A methodological platform based on genetically encoded voltage indicators (GEVIs) that enables the monitoring of large-scale circuit dynamics has brought us closer to this ambitious goal. This Review provides an update on the current state of the art and the prospects of emerging optical GEVI imaging technologies.

Abstract:The 25-century long history of brain science can be divided into four distinct 'Eras': Era 1 (∼2000 years) started around 500 BC with reductionist studies of human and animal brains using invasive in vivo and ex vivo methods; Era 2 (∼200 years) started in the 17th century and introduced the first invasive methods to study the function of living tissues; Era 3 started around 1838 and is the ongoing era of cellular neurophysiology; finally, Era 4 (in statu nascendi) is the era of non-invasive, holistic yet mechanistic, studies of the brain. Animal experimentation is becoming increasingly more holistic as multimodal imaging and recording techniques are combined in a single experiment on the brain of awake behaving animal. This newly emerged approach can be called in vigilo (from Latin 'in awake' or 'in vigilant state'), by analogy to the earlier introduced terms in vivo or in vitro. We introduce the Special Issue "In Vigilo Veritas: New Frontiers of Optical Imaging and Electrical Recording in the Brain of Awake Behaving Mice", which features original research articles and reviews that represent some of the finest examples of a truly multimodal studies, where behavioural readouts and tasks are combined in the same longitudinal experiment and on the same mouse with the two-photon imaging, optogenetics and/or electrophysiological recordings. This exciting multi-methodological approach creates a fertile ground for breakthrough discoveries in neurophysiology and neuropsychology of an awake behaving mammalian brain.
Optical Methods for the Study of Brain Metabolism In Situ

Abstract:From its inception, the International Society of Oxygen Transport to Tissue (ISOTT) included several core members who were intrigued by the possibility of using light to probe the metabolic state of the cerebral cortex. This 50th anniversary review will focus on optical methods (UV, Visible, NIR) used to study brain oxygen metabolism in the intact brain. Chance, Lübbers, and Jöbsis did fundamental work in the 1970s and 1980s. These investigators spearheaded the use of fluorescence of pyridine nucleotides (e.g., NADH) and flavoprotein, and visible and near-infrared spectroscopy of haemoglobin amount and saturation, as well as the mitochondrial cytochrome chain. Two of our society awards are named for these pioneers and founders. By the new millennium, their many scientific descendants (although still too few for the task at hand) have been productive and moved the field forward. Technical advances in materials, detectors, devices, and computer power have made great advances possible. Physiological theory and understanding of cell and molecular mechanisms still lag, providing fertile territory for future goals. There are at least two major directions currently that will certainly continue to illuminate brain metabolism (obvious-and well worn-pun intended). These are the burgeoning NIR instrumentation variations, and second, the tremendous potential of two-photon phosphorescence lifetime microscopy. These advances have been introduced through the years and continue to be advanced at our annual ISOTT symposia.

Abstract:Our senses are constantly stimulated in our daily lives but we have only a limited understanding of how they affect our cognitive processes and, especially, our autobiographical memory. Capitalizing on a public science event, we conducted the first empirical study that aimed to compare the relative influence of the five senses on the access, temporal distribution, and phenomenological characteristics of autobiographical memories in a sample of about 400 participants. We found that the access and the phenomenological features of memories varied as a function of the type of sensory cues, but not their temporal distribution. With regard to their influence on autobiographical memory, an overlap between some senses was found, with on one hand, olfaction and taste and, on the other, vision, audition, and touch. We discuss these findings in the light of theories of perception, memory, and the self, and consider methodological implications of the sensory cuing technique in memory research, as well as clinical implications for research in psychopathological and neuropsychological populations.

Abstract:Artworks provide sets of sensory stimuli that allow special insights into cognitive processes complementing results obtained with other experimental paradigms. Examples are given from visual art and music using behavioral measures and neuroimaging technology (fMRI). The following topics are addressed: creation and maintenance of personal identity, difference or equivalence of aesthetic and moral judgments, appreciation of Eastern and Western visual art, differences in sensory processing of naturalistic and surrealistic art, importance and traps of mental frames and prejudices, effect of emotional priming on the central representation of sensory stimuli, value of single case studies, personality characteristics as predictors, and usefulness of “controlled introspection” in analyzing contents of episodic memory, in particular with respect to aesthetic and health-promoting appreciation of environments. Furthermore, the necessary distinction between anthropological universals and cultural or individual specifics is stressed in sensory processing of artworks.

Abstract:Abstract Neurons in the inferotemporal (IT) cortex respond selectively to complex visual features, implying their role in object perception. However, perception is subjective and cannot be read out from neural responses; thus, bridging the causal gap between neural activity and perception demands independent characterization of perception. Historically, though, the complexity of the perceptual alterations induced by artificial stimulation of IT cortex has rendered them impossible to quantify. To address this old problem, we tasked male macaque monkeys to detect and report optical impulses delivered to their IT cortex. Combining machine learning with high-throughput behavioral optogenetics, we generated complex and highly specific images that were hard for the animal to distinguish from the state of being cortically stimulated. These images, named “perceptograms” for the first time, reveal and depict the contents of the complex hallucinatory percepts induced by local neural perturbation in IT cortex. Furthermore, we found that the nature and magnitude of these hallucinations highly depend on concurrent visual input, stimulation location, and intensity. Objective characterization of stimulation-induced perceptual events opens the door to developing a mechanistic theory of visual perception. Further, it enables us to make better visual prosthetic devices and gain a greater understanding of visual hallucinations in mental disorders.

Abstract:Infrared neural stimulation (INS) is well characterized for the peripheral nervous system; however, translation to the central nervous system (CNS) presents a new set of challenges which require us to consider different anatomy, multiple cell types, and the physiology associated with structures in the CNS. This study presents our first attempt to translate INS to in vivo stimulation of the CNS and to image the related response. The results from this study show that INS generates intrinsic optical signals of similar magnitude and shape associated with well characterized mechanical stimuli. The implications of this work could lead to neural implants which allows for single cell stimulation making it possible to design closed loop neural prosthetics.

Abstract:In recent years, fluorescent sensors are enjoying a surge of popularity in the field of neuroscience. Through the development of novel genetically encoded sensors as well as improved methods of detection and analysis, fluorescent sensing has risen as a new major technique in neuroscience alongside molecular, electrophysiological, and imaging methods, opening up new avenues for research. Combined with multiphoton microscopy and fiber photometry, these sensors offer unique advantages in terms of cellular specificity, access to multiple targets - from calcium dynamics to neurotransmitter release to intracellular processes - as well as high capability for in vivo interrogation of neurobiological mechanisms underpinning behavior. Here, we provide a brief overview of the method, present examples of its integration with other tools in recent studies ranging from cellular to systems neuroscience, and discuss some of its principles and limitations, with the aim of introducing new potential users to this rapidly developing and potent technique.
