Abstract:A novel ultra-performance liquid chromatography electrospray ionization tandem triple quadrupole mass spectrometry method for the simultaneous determination of four water-soluble vitamins, including vitamin B5 (VB5), vitamin B8 (VB8), vitamin B9 (VB9), and vitamin B12 (VB12) in fortified infant foods is developed and validated. A reverse phase UPLC separation system consisting of a Waters ACQUITY UPLC BEH C-18 column (2.1 mm x 100 mm i.d., 1.7 microm) and a binary gradient acetonitrile-water mobile phase is applied for the separation of the four water-soluble vitamins. Formic acid is spiked into the mobile phase to enhance the ionization efficiency. Tandem MS-MS analysis is performed in multi-reaction monitoring mode (MRM). Product-ion traces at m/z 220.1 --> 89.9 for VB5, 245.1 --> 227.1 for VB8, 442.3 --> 295.2 for VB9, and 678.9 --> 147.0 for VB12 are used for quantitation of the corresponding vitamins, and traces at m/z 455.5 --> 308.0 are used for methotrexate (internal standard). Limits of quantitation (LOQs) are 0.016, 0.090, 0.020, and 0.019 microg/L for VB5, VB8, VB9, and VB12, respectively. Intra- and inter-day precisions for the determination of the four vitamins are better than 6.84% and 12.26% in relative standard deviations, and recoveries for the four vitamins are in the range of 86.0~101.5%. The developed approach is applied for the determination of the trace amounts of the vitamins in fortified milk powers and fortified rice powers.
Decontamination of Barley Seedling Powder and Effect on Its Nutritional Components by Irradiation

Abstract:Decontamination of barley seedling powder by 60 Co γ -ray and effect on its nutritional components were studied. Results showed that irradiation was very effective in killing microorganisms in barley seedling powder. The population of microorganisms, such as aerobic bacterial count, moulds and coliform bacteria, decreased with increase of irradiation dose. The D_ 10 values for aerobic bacterial count, moulds and coliform bacteria were 3.18 kGy, 4.55 kGy and 2.32 kGy, respectively. Barley seedling powder, irradiated with 11 kGy, did not have obvious changes in main chemical components, including protein, total polysaccharide, free amino acid, crude fiber, while vitamin E reduced by 38%. It may be concluded that the treatment of irradiation sterilization can increase barley seedling powder quality-guarantee period much longer. The suggested optimal doses are from 7.5 to 11 kGy.

Abstract:Objective To compare three different methods used in vitamin A determination in soft capsule and to provide reference for the assay of vitamin A and its preparations.Method UV,NP-HPLC and RP-HPLC were chosen in this research.Cyclohexane was used as solvent in the triple wavelength method of UV;Column of NP-HPLC was filled with Hypersil SiO2(250×4.6mm,5μm) and the mobile phase consisted of n-hexane and isopropyl alcohol(99.8∶0.2);Column of RP-HPLC was packed with C18(150×4.6mm,5μm) and the mobile phase was methanol-water(97∶3).Results The content of vitamin A determined by UV was 95.5%,RSD=1.3%(n=5);112.5%,RSD=3.2%(n=5)by NP-HPLC and 102.3%,RSD=1.0%(n=5) by RP-HPLC.Conclusion Both UV and RP-HPLC are convenient measurements for vitamin A;NP-HPLC shows advantages in the test of impurities,while its stability is relatively poor.

Abstract:Few studies on the irradiation effects on dehydration characteristics and quality of dried-product ha ve been reported in the past three decades. This study focuses on drying characteristics of bio-products by hot-air after they are pretreated by gamma irradiation. Experiments were made to study the influence of different irradiation dose, air temperature, slice thickness of apple on the dehydration rate, and appearance quality (L-values), vitamin C content, and the rehydration ratio of dried apple. A conclusion is reached that the three factors of irradiation dose, hot-air temperature, and initial slice thickness significantly affected the four indices of appearance quality (L-values), vitamin C content, dehydration rate and rehydration ratio.

Abstract:Sugars are major constituents in many food systems. In this study, we tried to establish a rapid identification method to detect irradiated food which contained sugars by analyzing ultraweak luminescence of the food. The results indicated that the ultraweak luminescence behavior of glucose and sucrose changed greatly after gamma ray irradiation. Under dry condition, there were little changes in ultra luminescence intensities with the irradiated samples of glucose and sucrose, however, they did show marked changes in their ultra luminescence intensities when water was added during the measurement. It seemed that photon numbers of luminescence peak could be correlated to the dose delivered to the samples. The difference in the luminescence behavior of glucose and su- crose may provide an effective way to differentiate irradiated food.

Abstract:Ion chromatography method(IC) in conjunction with photochemical fluorimetry(PCF) was developed to determine riboflavin in health protection products.Chromatographic separation of the treated sample was performed on Dionex IonPac AG11-HC(4 mm×50 mm) guard column and IonPac AS11-HC(4 mm×250 mm) analytical column with 40 mmol·L-1 NaOH as eluent.The column effluents were subjected on-line to homemade UV irradiation(254 nm)reactor,riboflavin was transformed to a strongly fluorescent compound and detected by spectrofluorimetor.Under the optimum conditions,the relative standard deviations(RSDs) of retention time and peak area of 50 mg·L-1 riboflavin were 0.6% and 2.1%(n=6).The concentration of riboflavin showed good linear relationship with peak area in range of 10~100 mg·L-1 with r=0.993,the limit of detection(S/N=10)was 0.5 mg·L-1 and the recovery was(101.46±2.51)%.

Abstract:Terahertz time-domain spectroscopy (THz-TDS) is a new coherent spectral technique based on femtosecond laser technology. The radiation band belongs to far infrared. In this study, the optical characteristics of riboflavin and nicotinic acid in the frequency range 0.2 similar to 2.0 THz were investigated by THz-TDS, and their absorption spectra and the refraction spectra were obtained at room temperature. The results indicate that riboflavin and nicotinic acid have distinct absorption features which have large differences in such a frequency range. The average refractive indices of riboflavin and nicotinic acid are 1.70 and 1.65, respectively. This research provides a new method for the identification of vitamin molecules and experimental research in a wider effective spectral range.

Abstract:A high performance liquid chromatographic method with post-column photochemical derivation and fluorescence detection was developed for the analysis of thiamine in foods. A C-18 column was used for separation, and elution was performed with a pH 4.0 phosphate buffer (0.1 mol/L) containing 2% acetonitrile. When the eluate was on-line converged with a mixed 0.75% Na2SO3-4% NaOH solution, thiamine in the eluate was transferred to an intensive fluorescence compound by post-column photochemical derivation that occurred in a knotted polytetrafluoroethylene reactor, and detected by a spectrofluorimetor. Under optimized conditions, a detection limit of 3.8 mu g/L thiamine was achieved with the injection volume of 20 mu L, and a linear calibration curve was obtained in the range of 0.033 similar to 10 mg/L thiamine. RSD was in the level of 1.8%, and the recoveries of spiked analyte were in the range of 97% similar to 102%. The developed method was applied to determine the thiamine contents in pork liver and milk powder, and satisfactory results were obtained.

Abstract:Ultraviolet-visible spectrophotometry was used to determinate the content of bromate in mineral water and drinking water.The effect of reaction time,dosage of hydrochloric acid,dosage of potassium iodide,working curve and recovery were studied,and the content of bromate in mineral water anddrinkingwater was testedinabsenceofthelightas 10.33 and 9.33 μg/L,respectively.The recovery of mineral water is 96.03%~105.59%,while the recovery of drinking water is 95.89%~96.12%.The precision and detection limit of this method are 0.36 and 0.108,respectively.

Abstract:The objectives of this study were to obtain the effect of gamma irradiation on the quality of dried potato. Experiments were conducted to study the influence of different doses, air temperatures, slice thickness of potatoes on the dehydration rate, appearance quality (L-values), vitamin C content, and the rehydration ratio of dried potatoes. The greater the dose, the higher the dehydration rate, the lesser the vitamin C content, and the lower the rehydration ratio. The L-values for low-dose irradiation was greater than that for non-irradiated potatoes.

Abstract:OBJECTIVE To determine the contents of Water-soluble Vitamin for Injection by HPCE.METHODS The separation was performed on a fused-silica capillary column:67 cm(efficiency length 55 cm×50 μm),The mobile phase consisted of 50 mmol·L-1 SDS——50 mmol·L-1 Na2B4O7·10H2O solution(pH=8.33) eletroinjection:6.89 KPa×10 s;with running voltage of 15 kV.The detection wavelength was 214 nm.The column temperature was 25 ℃.RESULTS The linear ranges(n=5) for the determination of nicotinamide,Vitamin B6,D-biotin,Riboflavin 5′-phosphate sodium,Vitamin B12,Vitamin C,D-pantothenic acid sodium salt,folic acid and Vitamin B1 were 4.16~520.0 mg·L-1(r=0.999 1),0.40~50.0 mg·L-1(r=0.999 7),0.42~52.0 mg·L-1(r=0.999 3),0.40~50.0 mg·L-1(r=0.998 2),0.39～49.0 mg·L-1(r=0.999 6),8.00～1 000.0 mg·L-1(r=0.992 0),0.83～104.0 mg·L-1(r=0.999 5),0.42～53.0 mg·L-1(r=0.999 0),0.41～51.0 mg·L-1(r=0.999 8),respectively.The precision was good(with RSDs of 1.75%,1.04%,1.6%,2.84%,1.85%,3.26%,1.58%,2.5% and 1.92%,respectively(n=5).The detection limits were 0.60,0.16,0.24,0.23,0.21,0.45,0.41,0.20 and 0.18 mg·L-1,respectively.CONCLUSION The method of determining contents of water-soluble vitamin for injection by HPCE is easy to operate and the results of the determination are accurate.

Abstract:ABSTRACT A sensitive chemiluminescence (CL) induced by light for the determination of riboflavin with flow injection analysis is developed. Riboflavin shows a strong enhancement effect on the CL reaction of luminol oxidized by periodate after the photochemical reaction of riboflavin in alkaline solution, and the enhancing effect of riboflavin disappears under dark conditions. Oxygen can further improve the enhancing effect of the riboflavin in this CL system, and the effects were also investigated. And based on this phenomenon, a very sensitive method of various surfactants, β-cyclodextrin and organic solvents on this CL system for the determination of riboflavin was described. The range of the linear response for riboflavin using this method is ca. 2.4×10−8 ~ 1.0 × 10−6 mol/L, and the detection limit (S/N = 3) is l.0×10−8 mol/L. The applicability of the method was demonstrated by the determination of riboflavin in pharmaceutical preparation.

Abstract:ABSTRACTWheat, pretreated by 60Co gamma irradiation, was dried by hot air with irradiation dosage 0–3 kGy, drying temperature 40–60C, and initial moisture contents 19–25% (d.b.). The drying time, average dehydration rate, wet gluten content (WGC), moisture content of wet gluten (MCWG) and titratable acidity (TA) were evaluated based on a quadratic rotation‐orthogonal composite experimental design with three variables (at five levels). The drying time, average dehydration rate, WGC, MCWG and TA were sensitive to irradiation dosage, drying temperature and initial moisture content. Irradiation dosage is the most dominating factor on the quality of wheat flour (WGC, MCWG and TA); drying temperature is the most dominating factor on drying characteristics (drying time and average dehydration rate). Irradiation dosage had a significant effect on drying time, WGC, MCWG, average dehydration rate and TA.PRACTICAL APPLICATIONSGamma irradiation had been employed to decontaminate and to extend the shelf life of food and has been widely used for agricultural products for long‐period storage. The previous studies emphasize particularly on that dried samples were irradiated by gamma irradiation after drying. However, no information has been available on effect of gamma irradiation on drying characteristics and quality of wheat.The results of this study will show irradiation dosage had a significant effect on drying time, average dehydration rate, wet gluten content, moisture content of wet gluten and titratable acidity. This implies that as a pretreatment before drying may be a method to apply for not only increasing the dehydration rate, but also changing the quality of wheat.

Abstract:Introduction: The measurement of water-soluble vitamins is essential to diagnose and monitor various vitamin deficiencies. Establishing stability limits for these vitamins is crucial to ensure accurate laboratory testing. This study aimed to assess the stability of commonly measured water-soluble vitamins under different storage conditions to improve the accuracy of water-soluble vitamin measurement. Methods: The stabilities of thiamine, riboflavin, nicotinamide, pantothenic acid, pyridoxic acid and pyridoxal, biotin, 5-methyltetrahydrofolic acid (5-MTHF), and ascorbic acid were measured using liquid chromatography-tandem mass spectrometry. We investigated three pre-analytical factors: the effect of different temperatures and time durations on serum stability, variation between serum and plasma samples, and the impact of transferring samples to an ice bath before serum separation. We evaluated stability based on differences from the baseline. Results: Thiamine, pyridoxal, and ascorbic acid in serum exhibited instability at room temperature and 2–8°C. Riboflavin and 5-MTHF in serum were only stable for up to 48 and 72 h at 2–8°C. However, when stored at -20°C, all water-soluble vitamins remained stable for up to 72 h, whereas at -80°C, stability was maintained for up to 7 days. All vitamins in whole blood, except nicotinamide, were stable for up to 2–4 h when stored in an ice bath. Conclusions: Water-soluble vitamins, such as thiamine, riboflavin, pyridoxal, and ascorbic acid, are unstable at room temperature and 2–8°C. All vitamins were stable for up to 7 days and stored at -80°C. The ice bath improved the stability of whole blood samples before centrifugation. Thus, laboratories should ensure appropriate storage conditions to maintain pre-analytical quality for vitamin measurements.

Simultaneous Determination of Twelve Water- and Fat-Soluble Vitamins by High-Performance Liquid Chromatography with Diode Array Detection

Abstract:A simple and rapid method for the simultaneous determination of nine water-soluble vitamins was developed by high performance liquid chromatography with diode array detection. The water-soluble vitamins were analyzed by HPLC on a μBondapak C 18 column (300 × 3.9 mm, 10 μm) with methanol-0.1 mol/L KH 2 PO 4 buffer (pH 7.0) as mobile phase (1.5 mUmin) in gradient mode as follows: in the range 0-7.0 min, 10:90; and in the range 8.0-15.0 min, 40:60 (v/v). All nine water-soluble vitamins were separated in a single HPLC run within less than 15 min. The linearity of the calibration graphs was compound-dependent with correlation coefficients of 0.9986 to 0.9999 (n = 6). The detection limits ranged from 0.02 μg/mL to 0.5 μg/mL. The method was successfully applied to the determination of vitamins in pharmaceutical preparations. The recoveries were from 95% to 102% and the relative standard deviations were in the range of 0.8% to 4.2%.
Effect of 60Co gamma irradiation pre-treatment on drying characteristic of wheat

Abstract:Wheat, pretreated by gamma irradiation, was dried by hot-air. Experiments were conduced to study the influence of three factors (different irradiation dosages, air temperatures, and initial moisture contents of wheat) on the drying characteristics and surface temperature of wheat. Conclusions were reached that: the drying characteristics and surface temperature were all affected by irradiation, and the drying rate and the surface temperature of wheat increased with the increase of irradiation dose. Study on the cell structure of wheat grain after irradiated by electron microscope, and it was found that the change of drying characteristics and surface temperature of wheat were due to the destruction of the cell structure made by irradiation. And the hot air temperatures and initial moisture contents of wheat can also affect the drying characteristics of wheat. The effects of dehydration rate and moisture content on surface temperature under different doses of gamma irradiation were analyzed.

Abstract:In this paper, the riboflavin (RF) was used to study the interaction and sonodynamic damage to bovine serum albumin (BSA) by fluorescence and UV–vis spectroscopy. The results showed that the RF could efficiently bind to BSA in aqueous solution. Under ultrasonic irradiation, the RF could obviously damage the BSA. In addition, synchronous fluorescence spectroscopy revealed that the RF showed more accessible to tryptophan (Trp) residues than to tyrosine (Tyr) residues. Also, it damaged Trp residues more seriously than Tyr residues under ultrasonic irradiation. At last, the generation of reactive oxygen species (ROS) in sonodynamic process was estimated by the method of Oxidation–Extraction Spectrometry (OES). And then, several radical scavengers were used to determine the kind of ROS. It was found that at least the singlet oxygen (1O2) and hydroxyl radicals (OH) were generated.

The Activity of Certain Water-Soluble Vitamins After Exposure to Gamma Radiations in Dry Mixtures and in Solutions

Abstract:The effect of ionizing radiations on choline, folic acid, thiamine, riboflavin, pyridoxine and pantothenic acid was investigated using baby chicks as experimental animals. The vitamins were irradiated under various conditions with a dose of 2.79 × 106 rad of gamma rays. There was no evidence that any of the vitamins were destroyed when the synthetic diet containing the vitamins or when a mixture of the vitamins with vitamin-free casein was irradiated. Choline was not destroyed when an aqueous solution containing 300 mg of choline chloride per ml was irradiated. Twenty-five to 50% of the folic acid in a solution containing 50.0 µg/ml was lost by the irradiation process. There was no evidence of destruction of thiamine, riboflavin, pyridoxine, pantothenic acid and folic acid when the vitamins were mixed with casein and irradiated dry, but there was some destruction of each vitamin when irradiation was carried out in an aqueous solution. The loss of thiamine in the solution was largest and the loss of pyridoxine was slightly higher than that of riboflavin and pantothenic acid.

Abstract:Volvariella volvacea is a mushroom known for its high palatability and nutritional value. However, it is susceptible to spoilage thus making it challenging to preserve and keep fresh after harvest, resulting in constraints in long-distance transportation and long-term storage. This study aimed to investigate the feasibility of using irradiation and sodium dehydrogenate (SD) as a preservative in the preservation process of V. volvacea. The effects of three treatments of 0.8 kGy 60Coγ irradiation (B), 0.04% SD (C), combined with 0.04% SD and 0.8 kGy 60Coγ irradiation (A) on the postharvest freshness of V. volvacea were investigated. The assessment indices for V. volvacea, including appearance, browning rate, weight loss, respiration rate, MDA content, antioxidant enzyme activities, vitamin C (Vc), and soluble protein content, were measured and compared. The three treatments were compared to determine the changes in storage time over 7 days post-harvest. The results demonstrated that the hardness of the fruiting body exhibited a significant increase of 81.19%, 97.96% and 168.81% in comparison to the control, B and C, respectively, following the application of the treatment A. Compared to the control group, the soluble protein content was significantly increased by 20.28%. Respiration intensity and browning rate were significantly lower in the control treatment, decreasing by 35.07% and 45.49% respectively. On the 6th day of storage, the activities of SOD and POD increased by 81.06% and 73.71%, respectively, compared to the control, which significantly delayed the senescence of the fruiting bodies. The Vc content was significantly increased by 50.27%, 133.90%, and 101.39% in treatment B, which received 0.8 kGy 60Coγ irradiation alone, compared to the control, treatment A, and treatment C, respectively. The treatment C alone significantly reduced respiratory intensity and MDA variables by 39.55% and 31.01%, respectively, compared to the control. The findings can provide theoretical references and technical support for extending the preservation period of V. volvacea after harvesting by using irradiation and sodium dehydrogenate as a preservative.
