Abstract:Acoustic actuation of fluids at small scales may finally enable a comprehensive lab-on-a-chip revolution in microfluidics, overcoming long-standing difficulties in fluid and particle manipulation on-chip. In this comprehensive review, we examine the fundamentals of piezoelectricity, piezoelectric materials, and transducers; revisit the basics of acoustofluidics; and give the reader a detailed look at recent technological advances and current scientific discussions in the discipline. Recent achievements are placed in the context of classic reports for the actuation of fluid and particles via acoustic waves, both within sessile drops and closed channels. Other aspects of micro/nano acoustofluidics are examined: atomization, translation, mixing, jetting, and particle manipulation in the context of sessile drops and fluid mixing and pumping, particle manipulation, and formation of droplets in the context of closed channels, plus the most recent results at the nanoscale. These achievements will enable applications across the disciplines of chemistry, biology, medicine, energy, manufacturing, and we suspect a number of others yet unimagined. Basic design concepts and illustrative applications are highlighted in each section, with an emphasis on lab-on-a-chip applications.

Abstract:Acoustofluidics offers contact-free manipulation of particles and fluids, enabling their uses in various life sciences, such as for biological and medical applications. Recently, there have been extensive studies on acoustic streaming-based acoustofluidics, which are formed inside a liquid agitated by leaky surface acoustic waves (SAWs) through applying radio frequency signals to interdigital transducers (IDTs) on a piezoelectric substrate. This paper aims to describe acoustic streaming-based acoustofluidics and provide readers with an unbiased perspective to determine which IDT structural designs and techniques are most suitable for their research. This review, first, qualitatively and quantitatively introduces underlying physics of acoustic streaming. Then, it comprehensively discusses the fundamental designs of IDT technology for generating various types of acoustic streaming phenomena. Acoustic streaming-related methodologies and the corresponding biomedical applications are highlighted and discussed, according to either standing surface acoustic waves or traveling surface acoustic waves generated, and also sessile droplets or continuous fluids used. Traveling SAW-based acoustofluidics generate various physical phenomena including mixing, concentration, rotation, pumping, jetting, nebulization/atomization, and droplet generation, as well as mixing and concentration of liquid in a channel/chamber. Standing SAWs induce streaming for digital and continuous acoustofluidics, which can be used for mixing, sorting, and trapping in a channel/chamber. Key challenges, future developments, and directions for acoustic streaming-based acoustofluidics are finally discussed.
Quantitative assessment of parallel acoustofluidic device

Abstract:The advantage of ultrasonic fields in harmless and label-free applications intrigued researchers to develop this technology. The capability of acoustofluidic technology for medical applications has not been thoroughly analyzed and visualized. Toward efficient design, in this research, flowing fluid in a microchannel excited by acoustic waves is fully investigated. To study the behavior of acoustic streaming, the main interfering parameters such as inlet velocity, working frequency, displacement amplitude, fluid buffer material, and hybrid effect in a rectangular water-filled microchannel actuated by standing surface acoustic waves are studied. Governing equations for acoustic field and laminar flow are derived employing perturbation theory. For each set of equations, appropriate boundary conditions are applied. Results demonstrate a parallel device is capable of increasing the inlet flow for rapid operations. Frequency increment raises the acoustic streaming velocity magnitude. Displacement amplitude amplification increases the acoustic streaming velocity and helps the streaming flow dominate over the incoming flow. The qualitative analysis of the hybrid effect shows using hard walls can significantly increase the streaming power without depleting excessive energy. A combination of several effective parameters provides an energy-efficient and fully controllable device for biomedical applications such as fluid mixing and cell lysis.

Abstract:Microfluidics that permits to manipulate fluids at the micrometer or submicrometer scale offers fundamentally new opportunities in chemical and biomedical engineering. The past decade has witnessed considerable progress in the development of microfluidic systems for use in micro-/nanomaterials synthesis, biosensing, separation, catalysis, tissue engineering, and so on. Among many microfluidic mixers to enable those functions, the fusion of acoustics and microfluidics (termed acoustofluidics) represents an emerging promising platform. Acoustofluidics can manipulate fluids spatiotemporally by using the interaction of sound waves with solids, liquids, and/or gasses in a confined space for active and intensive mixing. This review accesses the utility of acoustofluidic technology in chemical and biomedical engineering for both academic research and industrial applications. We summarize the established categories of acoustic micromixers (sharp-edge, bubble, surface acoustic wave, resonant acoustic mixing, etc.), discuss the underlying principles, highlight the current achievements for diverse lab-on-a-chip application fields (chemical synthesis, biosensing, protein engineering, separation, cell lysis, etc.), and finally set out the practical challenges facing acoustofluidics to inspire future research in this field. Abstract:Acoustofluidics is a term describing the class of phenomena in which mechanical or acoustic vibrations induce a deformation or a flow in a fluid. Many deficiencies in our understanding of these phenomena remain to be addressed, with respect to the fundamental theoretical framework as well as in numerous applications. In this regard, the frequency of external forcing is a key parameter. Owing to the low cost, substantial magnitude, and versatility associated with acoustofluidic phenomena at audible frequencies, studies of these phenomena in the audible range have emerged with increasing amount in recent years and have attracted considerable attention. However, compared with studies focusing on the ultrasonic frequency domain, critical features and information specific to audible acoustofluidics remain dispersed across many independent publications, and a systematic integration of the literature on this topic is necessary. Accordingly, this review summarizes the basic theory and methods for generating vibrations in the audible range, presents various applications thereof in biology, chemistry, and other fields, and provides a high-level overview of the current status of the topic to motivate developing interesting proposals for further research in this field of study.

Acoustofluidic precise manipulation: Recent advances in applications for micro/nano bioparticles

Abstract:Acoustofluidic technologies that integrate acoustic waves and microfluidic chips have been widely used in bioparticle manipulation. As a representative technology, acoustic tweezers have attracted significant attention due to their simple manufacturing, contact-free operation, and low energy consumption. Recently, acoustic tweezers have enabled the efficient and smart manipulation of biotargets with sizes covering millimeters (such as zebrafish) and nanometers (such as DNA). In addition to acoustic tweezers, other related acoustofluidic chips including acoustic separating, mixing, enriching, and transporting chips, have also emerged to be powerful platforms to manipulate micro/nano bioparticles (cells in blood, extracellular vesicles, liposomes, and so on). Accordingly, some interesting applications were also developed, such as smart sensing. In this review, we firstly introduce the principles of acoustic tweezers and various related technologies. Second, we compare and summarize recent applications of acoustofluidics in bioparticle manipulation and sensing. Finally, we outlook the future development direction from the perspectives such as device design and interdisciplinary.

Abstract:Acoutofluidics is an increasingly developing and maturing technical discipline. With the advantages of being label-free, non-contact, bio-friendly, high-resolution, and remote-controllable, it is very suitable for the operation of living cells. After decades of fundamental laboratory research, its technical principles have become increasingly clear, and its manufacturing technology has gradually become popularized. Presently, various imaginative applications continue to emerge and are constantly being improved. Here, we introduce the development of acoustofluidic actuation technology from the perspective of related manipulation applications on living cells. Among them, we focus on the main development directions such as acoustofluidic sorting, acoustofluidic tissue engineering, acoustofluidic microscopy, and acoustofluidic biophysical therapy. This review aims to provide a concise summary of the current state of research and bridge past developments with future directions, offering researchers a comprehensive overview and sparking innovation in the field.

Abstract:Microfluidic separation technology has garnered significant attention over the past decade where particles are being separated at a micro/nanoscale in a rapid, low-cost, and simple manner. Amongst a myriad of separation technologies that have emerged thus far, acoustic microfluidic separation techniques are extremely apt to applications involving biological samples attributed to various advantages, including high controllability, biocompatibility, and non-invasive, label-free features. With that being said, downsides such as low throughput and dependence on external equipment still impede successful commercialization from laboratory-based prototypes. Here, we present a comprehensive review of recent advances in acoustic microfluidic separation techniques, along with exemplary applications. Specifically, an inclusive overview of fundamental theory and background is presented, then two sets of mechanisms underlying acoustic separation, bulk acoustic wave and surface acoustic wave, are introduced and discussed. Upon these summaries, we present a variety of applications based on acoustic separation. The primary focus is given to those associated with biological samples such as blood cells, cancer cells, proteins, bacteria, viruses, and DNA/RNA. Finally, we highlight the benefits and challenges behind burgeoning developments in the field and discuss the future perspectives and an outlook towards robust, integrated, and commercialized devices based on acoustic microfluidic separation.

Abstract:In this review, apart from the basic constitutions, mechanisms and materials of acoustofluidics, the research updates recent advancements and innovations in the designs of acoustofluidic devices, such as BAW‐based devices, SAW‐based devices, microfluidic systems of acoustofluidics and the integration of acoustofluidics with other technologies. The related separation performances, benefits, and drawbacks of different designs of acoustofluidics have also been covered. Separation of micro/nanoparticles, such as cellular, subcellular and biomolecular, has attracted increasing attention because of their remarkable potential applications in various fields, including chemistry, physics, medicine, etc. Among different micro/nanoparticle separation methods, acoustofluidics, which combines acoustics and microfluidics, has drawn the interest of researchers due to its biocompatibility, high efficiency and free labeling. In this review, the basic constitutions, mechanisms, and materials of acoustofluidics are described. Subsequently, sorts of delicately designed acoustofluidic devices, including diverse bulk acoustic wave (BAW) microfluidics and surface acoustic wave (SAW) microfluidics, are discussed, covering principles, advantages, limitations and applications in separation. Besides the introduction of advances of micro/nanoparticle separation in the BAW microfluidics, the SAW microfluidics are elaborated in detail with a focus on various configurations of interdigital transducers (IDTs), comprising straight IDT, slanted‐finger IDT, chirped IDT and focused IDT. Microfluidic systems of the acoustofluidics involve the forms of straight channels, serpentine channels, and droplets. Additionally, besides simply structured acoustofluidics, acoustofluidics integrated with other structures are also mentioned. Finally, the prospects and limitations of acoustofluidics in micro/nanoparticle separation are also discussed. The acoustofluidics reviewed here is envisioned as a versatile tool for micro/nanoparticle separation at the cellular, sub‐cellular, and biomolecular levels.

Abstract:Effective enrichment and separation of particles and mixing of multiphase microflows can be technically challenging in microfluidic sample processing and analysis. Acoustofluidic techniques have advantages such as high biocompatibility, versatility, and simple device design to meet these demands. Here, we present a novel tri-symmetrical acoustofluidic centrifuge based on traveling surface acoustic waves (TSAWs), achieving particle enrichment and bidirectional size-based separation, pushing away the larger particles while enriching the smaller ones, in a rapid time (<6 s). A microfluidic chamber was designed for further extraction of treated particles and cells. Experimental results show that our device can achieve enrichment of particles in a range of sizes and high purity separation (e.g. >90 % in 5 µm and 40 µm sample), demonstrating its sample processing capabilities. The acoustic treatment did not affect cells (consistent with the control group above 80 %). TSAW-induced acoustic streaming enabled efficient mixing (mixing efficiency χ >85 %) of fluorescent multi-phase microflows within a short time (<0.5 s). The underlying physical mechanisms have been verified by numerical simulations.

Abstract:Acoustofluidic techniques are increasingly used to manipulate nano- and micro-particles in microfluidics. A wide range of acoustofluidic devices consisting of microchannels and acoustic sources have been developed for applications in biochemistry and biomedicine. In this work, two hybrid acoustofluidic devices are developed and modelled, including a double surface acoustic wave (SAW) transducer and a PZTSAW transducer. The numerical study to these devices demonstrates a higher acoustic pressure present in the microchannel resulting in larger particle velocities in migration to the pressure nodes. The amplitude of the acoustic pressure and the pattern of the pressure distribution can be controlled in the hybrid transducers. By sweeping the height and width of the microchannel, one can identify an optimum dimension to produce intensive acoustic pressure in the PZT-SAW transducer. The particle trajectories reveal that both the SAW-SAW and PZT-SAW configurations produce significantly higher acoustic pressure and particle velocity in the microchannel. This work provides new insights to design acoustofluidic devices with more than one SAW transducer to effectively manipulate micro- and nano-particles.(c) 2023 Elsevier Ltd. All rights reserved.

Abstract:The precise manipulation of acoustic fields in microfluidics is of critical importance for the realization of many biomedical applications. Despite the tremendous efforts devoted to the field of acoustofluidics during recent years, dexterous control, with an arbitrary and complex acoustic wavefront, in a prescribed, microscale region is still out of reach. Here, we introduce the concept of acoustofluidic waveguide, a three-dimensional compact configuration that is capable of locally guiding acoustic waves into a fluidic environment. Through comprehensive numerical simulations, we revealed the possibility of forming complex field patterns with defined pressure nodes within a highly localized, pre-determined region inside the microfluidic chamber. We also demonstrated the tunability of the acoustic field profile through controlling the size and shape of the waveguide geometry, as well as the operational frequency of the acoustic wave. The feasibility of the waveguide concept was experimentally verified via microparticle trapping and patterning. Our acoustofluidic waveguiding structures can be readily integrated with other microfluidic configurations and can be further designed into more complex types of passive acoustofluidic devices. The waveguide platform provides a promising alternative to current acoustic manipulation techniques and is useful in many applications such as single-cell analysis, point-of-care diagnostics, and studies of cell-cell interactions.

Abstract:In recent years, the microfluidics technology based on the surface acoustic waves (SAW) has attracted a wide attention due to its many advantages such as simple and efficient generation and control of the SAW, multiple interacting forms between the SAW and the fluids, simple manufacturing process, and easy to integrate and detect.At present, as far as the biochemical analysis and the medical detection are concerned, the research of the SAW based microfluidic technology is mainly focused on the cells/particles sorting, the separation, mixing and concentration, the acoustic heating, the acoustic atomization, and the biological sensing.Some prototypes have almost reached the maturity stage, to be developed into portable devices with a huge prospective market.In this paper, the research of the microfluidic technology based on the SAW for the cells/particles sorting, separation, mixing and concentration, the acoustic heating, the acoustic atomization, the biological sensing reported in the last twenty years is reviewed, as well as the related research trend from a single effect study of acoustoforce, acoustothermal and acoustoelectric to the study of multiple physical effects, from two dimensional and micro manipulation to three dimensional and nano manipulation, and from planar device to flexible device.

Abstract:Interdigitated transducers (IDTs) were originally designed as delay lines for radars. Half a century later, they have found new life as actuators for microfluidic systems. By generating strong acoustic fields, they trigger nonlinear effects that enable pumping and mixing of fluids, and moving particles without contact. However, the transition from signal processing to actuators comes with a range of challenges concerning power density and spatial resolution that have spurred exciting developments in solid-state acoustics and especially in IDT design. Assuming some familiarity with acoustofluidics, this paper aims to provide a tutorial for IDT design and characterization for the purpose of acoustofluidic actuation. It is targeted at a diverse audience of researchers in various fields, including fluid mechanics, acoustics, and microelectronics.

Abstract:The development of microfluidic devices for lab-on-a-chip applications has largely been limited by a lack of flexible control and intensive mixing. The fusion of acoustics and microfluidics, termed acoustofluidics, has been demonstrated to be a promising tool for precisely manipulating micro-/nanoscale fluids and objects. When introducing sharp-edges inside the microchannel of acoustofluidic devices, the sound waves can drive highly controllable streaming flows via oscillating these microsolid structures. Considering its precise fluid control, simple device design, greatly reduced reaction duration, contactless operation, and good biocompatibility, sharp-edge acoustofluidics has demonstrated superior on-chip mixing and pumping performance for many application fields. Herein, we provide a comprehensive overview of the research and development of sharp-edge acoustofluidics. In particular, we discuss the design principles and underlying physics of sharp-edges, summarize the existing different types of sharp-edge microreactors (including sidewall wedge, sidewall rod, off-sidewall sharp-edges, and inlet conjunction needle), and highlight established applications of sharp-edge acoustofluidic technology in materials synthesis, bioliquefaction, enzyme bioassay, biomanipulation, and cell lysis. Finally, we point out the current challenges and future directions for inspiring further research in this field. The scope of this work is not only to provide an in-depth understanding of the state-of-the-art sharp-edge acoustofluidics, but also to inspire research efforts on the development of advanced on-chip platforms to satisfy the demanding needs of both academic and industrial communities. Abstract:Particles, ranging from submicron to nanometer scale, can be broadly categorized into biological and non‐biological types. Submicron‐to‐nanoscale bioparticles include various bacteria, viruses, liposomes, and exosomes. Non‐biological particles cover various inorganic, metallic, and carbon‐based particles. The effective manipulation of these submicron to nanoparticles, including their separation, sorting, enrichment, assembly, trapping, and transport, is a fundamental requirement for different applications. Acoustofluidics, owing to their distinct advantages, have emerged as a potent tool for nanoparticle manipulation over the past decade. Although recent literature reviews have encapsulated the evolution of acoustofluidic technology, there is a paucity of reports specifically addressing the acoustical manipulation of submicron to nanoparticles. This article endeavors to provide a comprehensive study of this topic, delving into the principles, apparatus, and merits of acoustofluidic manipulation of submicron to nanoparticles, and discussing the state‐of‐the‐art developments in this technology. The discourse commences with an introduction to the fundamental theory of acoustofluidic control and the forces involved in nanoparticle manipulation. Subsequently, the working mechanism of acoustofluidic manipulation of submicron to nanoparticles is dissected into two parts, dominated by the acoustic wave field and the acoustic streaming field. A critical analysis of the advantages and limitations of different acoustofluidic platforms in nanoparticles control is presented. The article concludes with a summary of the challenges acoustofluidics face in the realm of nanoparticle manipulation and analysis, and a forecast of future development prospects.

Submicron Particle Concentration and Patterning with Ultralow Frequency Acoustic Vibration

Abstract:Acoustofluidics have been widely used for particle and cell manipulations. Given the scaling of acoustic radiation forces and acoustic streaming flow velocities with increasing frequency, existing acoustofluidic manipulation of submicron particles require actuation at MHz and even GHz frequencies. In this work, we explore a novel acoustofluidic phenomenon, where an ultralow frequency (800 Hz) acoustic vibration is capable of concentrating and patterning submicron particles at two poles of each pillar in an array embedded in a microfluidic device. This unprecedented phenomenon is attributed to a collective effect of acoustic streaming induced drag force and non-Newtonian fluid induced elastic lift force, arising from symmetric acoustic microstreaming flows around each pillar uniformly across the entire pillar array. To our knowledge, this is the first demonstration that particles can be manipulated by an acoustic wave with a wavelength that is 6 orders of magnitude larger than the particle size. This ultralow frequency acoustofluidics will enable a simple and cost-effective solution to effective and uniform manipulation of submicron biological particles in large scales, which has the potential to be widely exploited in clinical and biomedical fields.

Abstract:Surface acoustic wave (SAW) devices form an important class of acoustofluidic devices, in which the acoustic waves are generated and propagate along the surface of a piezoelectric substrate. Despite their wide-spread use, only a few fully three-dimensional (3D) numerical simulations have been presented in the literature. In this paper, we present a 3D numerical simulation taking into account the electromechanical fields of the piezoelectric SAW device, the acoustic displacement field in the attached elastic material, in which the liquid-filled microchannel is embedded, the acoustic fields inside the microchannel, as well as the resulting acoustic radiation force and streaming-induced drag force acting on micro- and nanoparticles suspended in the microchannel. A specific device design is presented, for which the numerical predictions of the acoustic resonances and the acoustophoretic repsonse of suspended microparticles in 3D are successfully compared with experimental observations. The simulation provides a physical explanation of the the observed qualitative difference between devices with an acoustically soft and hard lid in terms of traveling and standing waves, respectively. The simulations also correctly predict the existence and position of the observed in-plane streaming flow rolls. The presented simulation model may be useful in the development of SAW devices optimized for various acoustofluidic tasks.
