Abstract:Ultrasonic manipulation is widely used as a noncontact technology and recently small particles rotating on a vibrating substrate has been observed. In this report,a novel methodology which compounds the procedures of ultrasonic trapping, transporting and rotating micro-particles in fluid using a single device is investigated. Irregular micro-particles in a standing wave field experience both acoustic radiation force and torque, which drive the particles to pressure nodes and keep them in a balance posture. A prototype device has also been built according to this theory, which six phase-controlled piezoelectric transducers whose sound beam axes are arranged with an angle of 60 deg in the x-y plane are used to generate ultrasonic standing waves with arbitrary node. The transducers are divided to two groups, so the wave field can be rotated by switching between the two groups. The synthesized sound field is scanned using a needle hydrophone and 200 μm irregular SiO2 particles are used to perform the compound manipulation ability of our device. The experimental results show good agreement with the theoretical calculation and the possible reason accounting for the small deviations is also discussed. This method may provide more complex and elaborate applications in micro-assembling and cell biomechanics.

Abstract:State of the art acoustofluidics typically treat micro-particles in a multi-wavelength range due to the scale limitations of the established ultrasound field. Here, we report a spatial selective acoustofluidic device that allows trapping micro-particles and cells in a wavelength scale. A pair of interdigital transducers with a concentric-arc shape is used to compress the beam width, while pulsed actuation is adopted to localize the acoustic radiation force in the wave propagating direction. Unlike the traditional usage of geometrical focus, the proposed device is designed by properly superposing the convergent section of two focused surface acoustic waves. We successfully demonstrate a single-column alignment of 15-μm polystyrene particles and double-column alignment of 8-μm T cells in a wavelength scale. Through proof-of-concept experiments, the proposed acoustofluidic device shows potential applications in on-chip biological and chemical analyses, where localized handing is required.
Inter-Particle Effects with a Large Population in Acoustofluidics

Abstract:The ultrasonic manipulation of cells and bioparticles in a large population is a maturing technology. There is an unmet demand for improved theoretical understanding of the particle-particle interactions at a high concentration. In this study, a semi-analytical method combining the Jacobi-Anger expansion and two-dimensional finite element solution of the scattering problem is proposed to calculate the acoustic radiation forces acting on massive compressible particles. Acoustic interactions on arrangements of up to several tens of particles are investigated. The particle radius ranges from the Rayleigh scattering limit (ka << 1) to the Mie scattering region (ka approximate to 1). The results show that the oscillatory spatial distribution of the secondary radiation force is related to the relative size of co-existing particles, not the absolute value (for particles with the same radius). In addition, the acoustic interaction is non-transmissible for a group of identical particles. For a large number of equidistant particles arranged along a line, the critical separation distance for the attraction force decreases as the number of particles increases, but eventually plateaus (for 16 particles). The range of attraction for the formed cluster is stabilized when the number of aggregated particles reaches a certain value.
Relative Position Control and Coalescence of Independent Microparticles Using Ultrasonic Waves

Abstract:Controlling the relative positions and coalescence of independent cells or microparticles is of particular importance for studying many physical phenomena, biological research, pharmaceutical tests, and chemical material processing. In this work, contactless maneuvering of two independent microparticles initially lying on a rigid surface was performed at a stable levitation height within a water-filled ultrasonic chamber. Three lead zirconate titanate transducers with 2 MHz thickness resonance frequency were obliquely mounted in a homemade device to form a sound field in a half space. By modulating the excitation voltage of a single transducer and the subsequent combination of amplitude and phase modulation, two separate 80 μm diameter silica beads were picked up from the chamber bottom, approached, and then coalesced to form a cluster in different ways. Both particles simultaneously migrated towards each other in the former process, while more dexterous movement with single-particle migration was realized for the other process. There is good agreement between the measured trajectories and theoretical predictions based on the theory of the first-order acoustic radiation force. The method introduced here also has the ability to form a cluster at any desired location in the chamber, which is promising for macromolecule processing ranging from the life sciences to biochemistry and clinical practice.

Abstract:The accurate and efficient manipulation of particles and droplets is crucial for the fields of disease diagnosis, target drug delivery, and sample purification, specific microstructure-based particle and droplet manipulation have been widely utilized in bioassays. However, current techniques are mainly based on the fixed microstructures, which is extremely difficult to achieve dynamic modulation by adjusting microstructure morphology and excitation conditions. Herein, we present a novel particle capture and droplet manipulation methodology by using morphologically reconfigurable magnetic micropillar arrays incorporate with acoustic streaming effects, which enables reconfigurable particle capturing, cargo transportation, and droplet manipulation. The mechanism is to assemble NdFeB magnetic powders by magnetic gradient fields to form morphologically variable magnetic micropillar arrays with porous surfaces, and the localized acoustic streaming effects generated by surface acoustic waves (SAWs) enable micropillar arrays to capture nearby particles onto the surfaces or to drive perturbations in droplets in the vicinity. To demonstrate the capabilities of this technology, we have achieved the high-volume acoustofluidic particle capture and loading (0.214 MCF), targeted complex maze navigation, efficient droplets manipulation (≤ 3.96mm/s), and acoustofluidic mixing acceleration (≤ 2.5s). Therefore, this technique enables reconfigurable and versatile manipulation of particles and droplets in biomedical applications.

Abstract:Acoustic actuation of fluids at small scales may finally enable a comprehensive lab-on-a-chip revolution in microfluidics, overcoming long-standing difficulties in fluid and particle manipulation on-chip. In this comprehensive review, we examine the fundamentals of piezoelectricity, piezoelectric materials, and transducers; revisit the basics of acoustofluidics; and give the reader a detailed look at recent technological advances and current scientific discussions in the discipline. Recent achievements are placed in the context of classic reports for the actuation of fluid and particles via acoustic waves, both within sessile drops and closed channels. Other aspects of micro/nano acoustofluidics are examined: atomization, translation, mixing, jetting, and particle manipulation in the context of sessile drops and fluid mixing and pumping, particle manipulation, and formation of droplets in the context of closed channels, plus the most recent results at the nanoscale. These achievements will enable applications across the disciplines of chemistry, biology, medicine, energy, manufacturing, and we suspect a number of others yet unimagined. Basic design concepts and illustrative applications are highlighted in each section, with an emphasis on lab-on-a-chip applications.

Abstract:Background and Objectives: Acoustofluidic manipulation of particles and biological cells has been widely applied in various biomedical and engineering applications, including effective separation of cancer cell, point-of-care diagnosis, and cell patterning for tissue engineering. It is often implemented within a polydimethylsiloxane (PDMS) microchannel, where standing surface acoustic waves (SSAW) are generated by sending two counter-propagating ultrasonic waves on a piezoelectric substrate. Methods: In this paper, we develop a full cross-sectional model of the acoustofluidic device using finite element method, simulating the wave excitation on the substrate and wave propagation in both the fluid and the microchannel wall. This model allows us to carry out extensive parametric analyses concerning the acoustic properties of the fluid and the microchannel wall, as well as the dimensions of the channel, to explore their influences on the acoustic field, fluid flow and microparticle aggregation. Results: Our findings demonstrate an order-of-magnitude enhancement in acoustic pressure amplitude and aggregation speed and a reduction in the particle threshold radius to submicron levels, which can be achieved through adjustments to the channel height and the difference in acoustic impedance between the channel wall and the fluid. The optimum channel heights are determined, which depend on the acoustic properties of the channel wall. The particle trajectories, movements along pressure nodal planes, and terminal positions are identified, with relative strength between the radiation force and the streaming force compared in different combinations of parameters. Conclusions: This work demonstrates that finetuning the dimensions and acoustic properties of the fluid and microchannel wall in acoustofluidic device can greatly enhance particle aggregation throughput and reduce constraints on particle size. Our findings offer valuable insights into device design and optimization.

Abstract:Particles, ranging from submicron to nanometer scale, can be broadly categorized into biological and non‐biological types. Submicron‐to‐nanoscale bioparticles include various bacteria, viruses, liposomes, and exosomes. Non‐biological particles cover various inorganic, metallic, and carbon‐based particles. The effective manipulation of these submicron to nanoparticles, including their separation, sorting, enrichment, assembly, trapping, and transport, is a fundamental requirement for different applications. Acoustofluidics, owing to their distinct advantages, have emerged as a potent tool for nanoparticle manipulation over the past decade. Although recent literature reviews have encapsulated the evolution of acoustofluidic technology, there is a paucity of reports specifically addressing the acoustical manipulation of submicron to nanoparticles. This article endeavors to provide a comprehensive study of this topic, delving into the principles, apparatus, and merits of acoustofluidic manipulation of submicron to nanoparticles, and discussing the state‐of‐the‐art developments in this technology. The discourse commences with an introduction to the fundamental theory of acoustofluidic control and the forces involved in nanoparticle manipulation. Subsequently, the working mechanism of acoustofluidic manipulation of submicron to nanoparticles is dissected into two parts, dominated by the acoustic wave field and the acoustic streaming field. A critical analysis of the advantages and limitations of different acoustofluidic platforms in nanoparticles control is presented. The article concludes with a summary of the challenges acoustofluidics face in the realm of nanoparticle manipulation and analysis, and a forecast of future development prospects.

Abstract:Hypothesis: The accurate and dynamic manipulation of multiple micro-sized objects has always been a technical challenge in areas of colloid assembly, tissue engineering, and organ regeneration. The hypothesis of this paper is the precise modulation and parallel manipulation of morphology of individual and multiple colloidal multimers can be achieved by customizing acoustic field. Experiments: Herein, we present a colloidal multimer manipulation method by using acoustic tweezers with bisymmetric coherent surface acoustic waves (SAWs), which enables contactless morphology modulation of individual colloidal multimers and patterning arrays by regulating the shape of acoustic field to specific desired distributions with high accuracy. Rapid switching of multimer patterning arrays, morphology modulation of individual multimers, and controllable rotation can be achieved by regulating coherent wave vector configurations and phase relations in real time. Findings: To demonstrate the capabilities of this technology, we have firstly achieved eleven patterns of deterministic morphology switching for single hexamer and precise switching between three array modes. In addition, the assembly of multimers with three kinds of specific widths and controllable rotation of single multimers and arrays were demonstrated from 0 to 22.4 rpm (tetramers). Therefore, this technique enables reversible assembly and dynamic manipulation of particles and/or cells in colloid synthesis applications.

Abstract:Effective enrichment and separation of particles and mixing of multiphase microflows can be technically challenging in microfluidic sample processing and analysis. Acoustofluidic techniques have advantages such as high biocompatibility, versatility, and simple device design to meet these demands. Here, we present a novel tri-symmetrical acoustofluidic centrifuge based on traveling surface acoustic waves (TSAWs), achieving particle enrichment and bidirectional size-based separation, pushing away the larger particles while enriching the smaller ones, in a rapid time (<6 s). A microfluidic chamber was designed for further extraction of treated particles and cells. Experimental results show that our device can achieve enrichment of particles in a range of sizes and high purity separation (e.g. >90 % in 5 µm and 40 µm sample), demonstrating its sample processing capabilities. The acoustic treatment did not affect cells (consistent with the control group above 80 %). TSAW-induced acoustic streaming enabled efficient mixing (mixing efficiency χ >85 %) of fluorescent multi-phase microflows within a short time (<0.5 s). The underlying physical mechanisms have been verified by numerical simulations.

Abstract:Particles and cells can be patterned and moved (i.e., manipulated) precisely using acoustically driven techniques. To date, application of acoustic particle manipulation has been limited to plain surfaces. There is much potential for applying acoustic manipulation techniques to surfaces with microfabricated structures for high-throughput sensing. But adding thin film structures could alter manipulation characteristics compared to a plain surface. Using a two-chip setup that allows the wave generating device to be reused, we study the feasibility of acoustofluidic micro-manipulation on a surface-micromachined silicon (SMS) chip. The SMS chip is a complex superstrate with generic thin-film structures fabricated by patterning and etching multiple layers of thin films, with properties meant to represent a broad range of microfabricated devices. We report notable alterations in the particle separation distances on the SMS chip compared to a bare silicon superstrate, which we attribute to a change in wave type through a comparison of different superstrates prepared. We demonstrate a high cell viability after acoustic manipulation of live cells on the SMS chip. The results herein demonstrate the possibility of integrating a suite of microfabricated sensors on a chip with acoustically driven manipulation capabilities for multiplexed sensing and analysis for bio-applications.

Abstract:Contactless manipulation of particles and cells using acoustic forces that can be adjusted in real time is now important in $e.g.$ biomedical sensors, imaging devices, and diagnostic tools. Dynamic manipulation typically requires huge phased arrays with complex electrical control, or a moving source with an inflexible displacement platform, but here the authors develop a method using an acoustic field modulated by a phononic crystal plate (PCP) in an acoustofluidic channel. PCP resonance-based dynamic manipulation via a single source, by switching the frequency, has the advantages of being simple, disposable, scalable, and combinable with a microfluidic chip.

Abstract:Precisely controllable transport and rotation of microparticles and cells has great potential to enable new capabilities for single-cell level analysis. In this work, we present versatile ultrasonic microstreaming based manipulation that enables active and precise control of transport and rotation of individual microscale particles and biological cells in a microfluidic device. Two different types of ultrasonic microstreaming flow patterns can be produced by oscillating embedded microstructures in circular and rectilinear vibration modes, which have been validated by both numerical simulation and experimental observation. We have further showcased the ability to transport individual microparticles along the outlines of complex alphabet letters, demonstrating the versatility and simplicity of single-particle level manipulation with bulk vibration.
A Contactless Methodology of Picking Up Micro-Particles from Rigid Surfaces by Acoustic Radiation Force

Abstract:Controlled movement and pick up of small object from a rigid surface is a primary challenge in many applications. In this paper, a contactless methodology of picking up micro-particles within deionized water from rigid surfaces by acoustic radiation force is presented. In order to achieve this, an acoustic radiation force was generated by 1.75 MHz transducers. A custom built setup facilitates the optimization of the sound field by varying the parameters such as sound source size and source position. The three-dimensional pressure distributions are measured and its relative sound field is also characterized accordingly. The standing wave field has been formed and it is mainly composed of two obliquely incident plane waves and their reflectors. We demonstrated the gripping and positioning of silica beads, SiO2, and aluminum micro-particles of 100 μm to 500 μm in size with this method using acoustic radiation force. The acoustic radiation force generated is well controlled, contactless, and in the tens of nano-Newton range which allowed us to manipulate relative big micro objects such as MEMS components as well as moving objects such as living cells. The proposed method provided an alternative form of contactless operating environment with scalable dimensions suitable for the manipulating of small objects. This permits high-throughput processing and reduction in time required for MEMS assembling, cell biomechanics, and biotechnology applications.

Abstract:Acoustic tweezers based on travelling surface acoustic waves (TSAWs) have the potential for contactless trajectory manipulation and motion-parameter regulation of microparticles in biological and microfluidic applications. Here, we present a novel design of a tri-directional symmetrical acoustic tweezers device that enables the precise manipulation of linear, clockwise, and anticlockwise trajectories of microparticles. By switching the excitation combinations of interdigital electrodes (IDTs), various shape patterns of acoustic pressure fields can be formed to capture and steer microparticles accurately according to pre-defined trajectories. Numerical simulations and experimental tests were conducted in this study. By adjusting the input electric signals and the fluid's viscosity, the device is able to manipulate microparticles of various forms as well as brine shrimp egg cells with the accurate modulation of motion parameters. The results show that the proposed programmable design possesses low-cost, compact, non-contact, and high biocompatibility benefits, with the capacity to accurately manage microparticles in a range of motion trajectories, independent of their physical and/or chemical characteristics. Thus, our design has strong potential applications in chemical composition analysis, drug delivery, and cell assembly.
Capillary-based, multifunctional manipulation of particles and fluids via focused surface acoustic waves

Abstract:Surface acoustic wave (SAW)-enabled acoustofluidic technologies have recently atttracted increasing attention for applications in biology, chemistry, biophysics, and medicine. Most SAW acoustofluidic devices generate acoustic energy which is then transmitted into custom microfabricated polymer-based channels. There are limited studies on delivering this acoustic energy into convenient commercially-available glass tubes for manipulating particles and fluids. Herein, we have constructed a capillary-based SAW acoustofluidic device for multifunctional fluidic and particle manipulation. This device integrates a converging interdigitated transducer to generate focused SAWs on a piezoelectric chip, as well as a glass capillary that transports particles and fluids. To understand the actuation mechanisms underlying this device, we performed finite element simulations by considering piezoelectric, solid mechanic, and pressure acoustic physics. This experimental study shows that the capillary-based SAW acoustofluidic device can perform multiple functions including enriching particles, patterning particles, transporting particles and fluids, as well as generating droplets with controlled sizes. Given the usefulness of these functions, we expect that this acoustofluidic device can be useful in applications such as pharmaceutical manufacturing, biofabrication, and bioanalysis.

Abstract:Recently, acoustic levitation for non-contact micro-particle manipulation has been attracting great interest in physical, biological, and medical applications. Among the state-of-the-art manipulation technologies, single beam acoustic tweezing exhibits advantages of providing stronger trapping force and deeper penetration depth in tissues, inducing less tissue damage, and a simple configuration involving only one device. However, particle trapping by the single beam acoustic tweezer could only be operated on a smooth two-dimensional substrate, which limits the potential for real applications. Here, we report an initial attempt to acoustically levitate an individual micro-particle stably in water and manipulate the levitated micro-particle arbitrarily two-dimensionally by simply employing a 60-MHz focused ring ultrasonic transducer. The proposed working mechanism agrees well with the phenomenon. This approach could not only acoustically levitate and manipulate a micro-particle on a culture dish and on a mylar film, but could also work properly in levitating and manipulating a micro-particle placed inside the polyimide tube. This simple and low-cost approach is extremely useful for effective non-contact micro-particle manipulation without having critical concerns on the substrate properties.

Abstract:In this study, a novel strategy to generate sophisticated acoustic streaming vortices, which would be available for rotational manipulation of micro-/nano-scale objects, is proposed and simulated. All structural units in the microfluidic chamber are symmetric in design, and all radiation surfaces have the same settings of input frequency, oscillation amplitude, and initial phase. Different kinds of asymmetric acoustofluidic patterns can be generated in the originally static microfluidic chamber only because of the asymmetric arrangement of multiple radiation surfaces in space. The calculation results of kaleidoscopic acoustofluidic fields together with particle movement trajectories induced by cross structures with different radiation surface distributions further demonstrate the versatile particle manipulation capabilities of these functional microfluidic devices. In comparison to the existing oscillation modulation method, which requires multiple radiation surfaces with different initial phases, acoustofluidic devices with a same initial phase of all radiation surfaces can significantly reduce the required number of auxiliary signal generators and power amplifiers. The proposed generation method of acoustofluidic patterns is promising for microfluidic mixing without rotating machinery, driving operation of microrobots, and rotational manipulation of biological samples.

Abstract:Surface acoustic wave-based particle/bioparticle manipulation has emerged as a promising tool for disease diagnosis. The effects of the titled angle of the acoustic field θ and the microchannel aspect ratios β on the particle migration mode, the force of particle, and the three-dimensional focusing behavior are studied by using simulation and high-speed microscopic visualizations experiments. The acoustic field tilt range is from 0° to 15°, and the wavelength is 160 μm. Particle migration trajectory is observed from high-speed photographic images. Compared with most parallel acoustic fields, the particle migration efficiency of the tilted acoustic field is higher because the acoustic radiation force (Fr) continues to act on the particles in the lateral direction. The tilted angle of the acoustic field is not a fixed value (usually 15°), and there is an optimal angle to match the maximum lateral migration of the target particles. A model is put forward to predict the optimal acoustic field tilt-angle for acoustofluidic devices, which can achieve 96% separation of 15 μm target particles. The change in the direction of the Fr drives the particles to create two typical migration states during the lateral migration process, named continuous migration and intermittent migration. The phenomenon of multi-layer particle focus in the vertical Z-direction of the microchannel is experimentally observed for the first time, which mainly depends on whether the microchannel has enough height to make multiple acoustic pressure nodes in the vertical direction. Two or even three layers of particle focus lines can be observed in the vertical direction at the microchannel aspect ratios β > 0.5. The research results provide new insight into the high-throughput development of microfluidic devices.

Abstract:Acoustic manipulation of microparticles and cells has attracted growing interest in biomedical applications. In particular, the use of acoustic waves to concentrate particles plays an important role in enhancing the detection process by biosensors. Here, we demonstrated microparticle concentration within sessile droplets placed on the hydrophobic surface using the flexural wave. The design benefits from streaming flow induced by the Lamb wave propagated in the glass waveguide to manipulate particles in the droplets. Microparticles will be concentrated at the central area of the droplet adhesion plane based on the balance among the streaming drag force, gravity, and buoyancy at the operating frequency. We experimentally demonstrated the concentration of particles of various sizes and tumor cells. Using numerical simulation, we predicted the acoustic pressure and streaming flow pattern within the droplet and characterized the underlying physical mechanisms for particle motion. The design is more suitable for micron-sized particle preparation, and it can be valuable for various biological, chemical, and medical applications.
