Abstract:In recent years, nitrosamines have been discovered in some types of drug products that becomes a current regulatory hotspot, and have attracted a lot attention from both regulatory authorities and industry. This manuscript provided an industry perspective on the nitrosamines research. A liquid chromatography coupled with tandem mass spectrometry(LC-MS/MS)method was developed and applied for the quantification of N-nitrosodimethylamine (NDMA) in metformin hydrochloride sustained-release tablets (MET). The key factors resulting in the NDMA formation in MET were identified through forced degradation and drug-excipient studies, which included high temperature, dimethylamine, strong alkali and oxidation conditions, peroxide and alkaline components contained in the formulation as well as the nitrite and nitrate impurities that might be presented in certain excipients. Further, API particle size and water content of the drug product would also affect the growth rate of NDMA. Therefore, the following mitigation strategies to reduce the risk of nitrosamines in the finished drug product are proposed in this manuscript: 1) avoid the use of excipients containing nitrite, nitrate and peroxide impurities; 2) avoid high temperature and strong alkaline environment in the production and storage condition; 3) maintain an appropriate water content level in the formulation. Based on the above principles, it was recommended to add antioxidant or incorporate excipient such as Na2CO3 to modify the formulation pH to weak basic environment in the formulation of MET, which can could effectively prevent formation of NDMA in the stability process.

Abstract:The mitigation of nitrosamine formation in drug products has been studied and approaches such as using formulations with pH modifiers and antioxidants have been shown to decrease the formation of nitrosamines. However, more studies are needed to explore the effectivness of mitigation strategies with different drug models and formulations. The primary objective of this work was to assess the role of different antioxidants and pH modifiers in tablet formulations to mitigate the formation of NDMA, prepared in-house, using metformin hydrochloride as a model drug. A study design for manufacturing metformin hydrochloride formulations was created to evaluate potential mitigation stratigies. The formulations were prepared by wet granulation that included a sodium nitrite spike and various antioxidants such as ascorbic acid, caffeic acid and ferulic acid at various concentrations that may inhibit nitrosamine formation. The study design also included pH modifiers such as hydrochloric acid and sodium carbonate. The metformin hydrochloride formulations were placed under stability conditions that included humidity, temperature and time over a six month period. NDMA inhibition was found to be most effective in formulations with basic pH, followed by the addition of tested antioxidants with 0.1% concentrations in the formulations. All tested antioxidants showed complete mitigation in formulations with 0.5% and 1% concentrations. In summary, basic pH and the inclusion of antioxidants exhibited the potential to mitigate the formation of NDMA in metformin hydrochloride tablets.

Abstract:The authors wish to correct three product structures in their Communication. The three structures (2ag–2ai) in Table 2, which were originally identified as γ-aminoketone hydrochlorides, are actually their intramolecular dehydrated condensation derivative hydrochlorides. The original structures and the corrected structures are shown below. The reaction of ketoxime ethers 1ag–1ai first delivered linear γ-aminoketone hydrochlorides (the original structures), which can spontaneously undergo condensation to form cyclic imine hydrochlorides. The obtained products 2ag–2ai were initially characterized as γ-aminoketone hydrochlorides on the basis of NMR spectra and HRMS data. However, single-crystal X-ray diffraction analysis of 2ah shows that it has the structure of a cyclic imine hydrochloride. There is an equilibrium between the linear γ-aminoketone hydrochloride and its dehydrated cyclic imine hydrochloride in solvent (D2O, DMSO-D6, and CD3OD), which tpredominantly favors dehydration. Although γ-aminoketone hydrochlorides can be detected by HRMS, only cyclic imine hydrochlorides were isolated in pure form. The related description “Significantly, this tactic was also feasible to 1,5-amino migration by employing ketoxime ethers 1ag–1ai and the desired trifluoromethyl substituted γ-aminoketones 2ag–2ai were successfully generated in moderate yields.” should be corrected to “Significantly, this tactic was also feasible to 1,5-amino migration by employing ketoxime ethers 1ag–1ai, and the desired trifluoromethyl substituted γ-aminoketones were successfully generated in moderate yields; the latter readily underwent intramolecular condensation to afford cyclic imine hydrochlorides 2ag–2ai.” HRMS data of 2ag–2ai in the form of dehydrated cyclic structures, X-ray diffraction structure of 2ah, and characterization data of the unhydrolyzed 1,5-amino migration products 2ag’–2ai’ have been updated in the Supporting Information provided with this Corrigendum.
