Abstract:Near-infrared (NIR)-absorbing organic nanoparticles (ONPs) are emerging candidates for "one-for-all" theranostic nanomaterials with considerations of safety and formulation in mind. However, facile fabrication methods and improvements in the photothermal conversion efficiency (PCE) and photostability are likely needed before a clinically viable set of candidates emerges. Herein, a new organic compound, [porphyrin-diketopyrrolopyrrole (Por-DPP)] with the donor-acceptor structure was synthesized, where porphyrin was used as a donor unit while diketopyrrolopyrrole was used as an acceptor unit. Por-DPP exhibited efficient absorption extending from visible to NIR regions. After self-assembling into nanoparticles (NPs) (similar to 120 nm), the absorption spectrum of Por-DPP NPs broadened and red-shifted to some extent, relative to that of organic molecules. Furthermore, the architecture of NPs enhanced the acceptor-donor structure, leading to emission quenching and facilitating nonradiative thermal generation. The PCE of Por-DPP NPs was measured and calculated to be 62.5%, higher than most of ONPs. Under 808 nm laser irradiation, the Por-DPP NPs possessed a distinct photothermal therapy (PTT) effect in vitro and can damage cancer cells efficiently in vivo without significant side effects after phototherapy. Thus, the small-molecule porphyrin-based ONPs with high PCE demonstrated promising application in photoacoustic imaging-guided PTT.

Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.

Abstract:Carbon dots (CDs) are a member of fluorescent carbon nanomaterials that are widely applied in bioimaging, photothermal therapy (PTT), and biosensors for its tunable fluorescence, photothermal conversion property, and excellent biocompatibility. Surface passivation and doping especially the doping of N atoms are critical factors to enhance the fluorescent intensity of CDs. Until now, a variety of nitrogen-rich molecules has been applied for the surface passivation of CDs such as L-Dopa, amino acids, and polyethylenimine (PEI). Herein, we report the synthesis of fluorescent polydopamine (PDA)-passivated carbon dots (CD-PDA) via one-pot microwave-assisted pyrolysis within 5 min, dramatically simplifying the reaction process compared with the hydrothermal treatment reported before. DLS, FT-IR, UV-Vis, and fluorescence spectroscopy were used to confirm the components of CD-PDA and to illuminate the mechanism of its tunable photoluminescence (PL). Due to the doping of N atoms by PDA, quantum yield (QY) of the CD-PDA was measured at 5%, which was nearly triple the original CDs without adding PDA. Yield of CD-PDA was about 1.5 times of the CDs on account of the enhancement of nucleation site for the carbon dot formation with the phenolic group provided by PDA. Meanwhile, photothermal conversion efficiency of the CD-PDA was determined to be 35% because of the excellent NIR light-thermal conversion property of PDA. Overall, we provided an extremely efficient approach to fabricate the fluorescent N-doped CD-PDA with stable photothermal conversion efficiency and excellent biocompatibility. More importantly, the passivation of PDA enabled the CD-PDA synthesized in our research compatible for further modification through Michael addition or Schiff base reaction.

Abstract:Carbon dots ( CDs) with unique optical properties are a stable and highly biocompatible fluorescent material offering enormous application potential in optical imaging and photothermal cancer therapy (PTT). However, the low photothermal conversion efficiency and the high power density still impede the development of CDs in PTT. Herein, the nitrogen and oxygen co- doped CDs (N-O-CDs) with strong absorbance in the near infrared (NIR) region were prepared from 1,3,6-trinitropyrene (TNP) and N-containing polymer of branched polyethylenimine (BPEI) via a one-step molecular fusion route. The asprepared black N-O-CDs with high photostability and superb biocompatibility can be utilized not only as a new fluorescence imaging agent but also as an excellent PTT agent in vivo. The structure, optical properties, bioimaging, photothermal effect in vitro, and therapeutic efficiency in vivo of the N-O-CDs are investigated. The low power density (0.8 Wcm(-2)) and high photothermal conversion efficiency (eta = 38.3%) facilitate N-O-CDs to act as an ideal theranostic agent for fluorescence imaging and photothermal therapy in vitro and in vivo. Abstract:Photothermal conversion (PTC) nanostructures have great potential for applications in many fields, and therefore, they have attracted tremendous attention. However, the construction of a PTC nanoreactor with multi-compartment structure to achieve the combination of unique chemical properties and structural feature is still challenging due to the synthetic difficulties. Herein, we designed and synthesized a catalytically active, PTC gold (Au)@polydopamine (PDA) nanoreactor driven by infrared irradiation using assembled PS-b-P2VP nanosphere as soft template. The particles exhibit multi-compartment structure which is revealed by 3D electron tomography characterization technique. They feature permeable shells with tunable shell thickness. Full kinetics for the reduction reaction of 4-nitrophenol has been investigated using these particles as nanoreactors and compared with other reported systems. Notably, a remarkable acceleration of the catalytic reaction upon near-infrared irradiation is demonstrated, which reveals for the first time the importance of the synergistic effect of photothermal conversion and complex inner structure to the kinetics of the catalytic reduction. The ease of synthesis and fresh insights into catalysis will promote a new platform for novel nanoreactor studies.

Abstract:Due to the advantages of long wavelength emission, near‐infrared (NIR) fluorescent Carbon Dots (CDs) attract broad interest. However, strategies for manipulating NIR fluorescence of CDs are urgently needed. Here, the authors reveal an effective pathway to activate one‐photon and two‐photon NIR emission on CDs, providing a new avenue for developing CDs with ideal optical properties. Due to the complicated nature of carbon dots (CDs), fluorescence mechanism of near‐infrared (NIR) fluorescent CDs is still unrevealed and features highly controversial. Reliable and effective strategies for manipulating the NIR fluorescence of CDs are urgently needed. Herein, CDs with one‐photon (622 nm, QYs ≈ 17%) and two‐photon (629 nm) NIR fluorescence are prepared by acidifying o‐phenylenediamine‐based reaction sediments. Systematic analysis reveals that the protonation of amino groups increases the particle surface potential, disperse the bulk sediments into nano‐scale CDs. In the meanwhile, amino protonation of pyridinic nitrogen (–N=) structure inserts numerous n orbital energy levels between the π → π* transition, narrows the gap distance for photon transition, and induces NIR fluorescence emission on CDs. Present research reveals an effective pathway to activate CDs reaction sediments and trigger NIR emission, thus may open a new avenue for developing CDs with ideal optical properties and promising application prospects.

Abstract:Development of special organic materials that are able to absorb light energy in the second near-infrared window (NIR-II) is significantly important for treating deep-tissue-buried diseases or supplying power to implantable electronic devices. Herein, a narrow bandgap donor–acceptor (D-A) conjugated polymer with thiophene-fused benzodifurandione-based oligo(p-phenylenevinylene) (TBDOPV) as acceptor part and 2,2′-bithiophene (DT) as donor part was developed and exploited as a photothermal conversion material with high extinction coefficient and robust photostability in the NIR-II window. According to transient absorption analysis results, the photothermal conversion ability of this polymer is attributed to the fast internal conversion (IC) process. The high photothermal conversion efficiency makes this polymer a promising NIR-II adsorbing antenna to remotely actuate thermo-dependent devices, e.g., high-performance photothermal–electrical and photothermal–mechanical converters.

Rogach, and co-workers, in article number 1705913, through surface engineering of carbon dots, exploiting electron-acceptor groups, namely molecules or polymers rich in sulfoxide/carbonyl groups. Twophoton-induced NIR emission and three-photon-induced red emission are simultaneously observed for carbon dots in dimethyl sulfoxide under excitation of a 1400 nm femtosecond pulse laser in the NIR-II window.
Enhanced Imaging Brightness of NIR-II Fluorescent Conjugated Polymer Nanoparticles Prepared by Ternary Copolymerization Method and Its NIR-II Photothermal Application

Abstract:In order to simultaneously achieve excellent solubility , high brightness of second near-infrared (NIR-II, 1000-1700 nm) fluorescence, and strong NIR-II photothermal transduction capability of conjugated polymers, we employed a ternary copolymerization strategy to construct the NIR-II emissive conjugated polymers with strong electron acceptor and electron donor in our previous work. In this work, we further adjusted the ratio between the two electron donors, BDT and 2TC, to obtain a series of conjugated polymers, named as BDT-2TC(12), BDT-2TC(11), and BDT-2TC(21). These polymers have strong absorption in the NIR-II region and excellent solubility in organic phase. All of them show strong NIR absorption in the wavelength of 700-1200 nm extended to the NIR-II region and exhibit strong fluorescence emission in the NIR-II region of 1000-1400 nm, which was excited by an 808 nm laser. Thereafter, water-soluble and biocompatible nanoparticles (BDT-2TC(12) NPs) were facilely prepared by encapsulating the target conjugated polymer BDT-2TC(12) into the amphiphilic distearyl phosphatidylacetamide-methoxy polyethylene glycol (DSPE-mPEG) through nanoprecipitation method. These prepared nanoparticles have good size-stability and photostability, and exhibit strong absorption at both 808 nm and 1064 nm. Thus, under the irradiation of a 1064 nm laser, BDT-2TC(12) NPs showed an excellent NIR-II photothermal conversion effect (the photothermal conversion efficiency of the BDT-2TC(12) NPs aqueous solution was calculated to be 56.3%), which could realize the photothermal therapy (PTT) of tumor cells. In addition, under the 808 nm laser excitation, the nanoparticles can also achieve high-resolution NIR-II fluorescence imaging (FI) of the mouse blood vessels and other biological tissues.

Abstract:Carbonized polymer dots (CPDs) are remarkable optical materials with low preparation cost, high biocompatibility, and excellent optical properties. The emission wavelengths of current CPDs are limited in the visible to NIR‐I range, impeding their applications in the transparent NIR‐II window. A new series of NIR‐CPDs is reported through a two‐step hydrothermal/Knoevenagel reaction with adjustable absorption/emission wavelength for NIR‐II bioimaging. Carbon dots (CDs) or carbonized polymer dots (CPDs) are an emerging class of optical materials that have exceptional applications in optoelectronic devices, catalysis, detection, and bioimaging. Although cell studies of CPDs have produced impressive results, in vivo imaging requires available CPDs to fluoresce in the near‐infrared‐II (NIR‐II) window (1000−1700 nm). Here, a two‐step bottom‐up strategy is developed to synthesize NIR‐CPDs that provide bright emissions in both NIR‐I and NIR‐II transparent imaging windows. The designed strategy includes a hydrothermal reaction to form a stable carbon core with aldehyde groups, followed by the Knoevenagel reaction to tether the molecular emission centers. This procedure is labor‐saving, cost‐efficient, and produces a high yield. The NIR‐CPDs enable high‐performance NIR‐II angiography and real‐time imaging of the disease degree of colitis noninvasively. This technology may therefore provide a next‐generation synthesis strategy for CPDs with rational molecular engineering that can accurately tune the absorption/emission properties of NIR‐emissive CPDs.

Abstract:Carbon dots (CDs) with absorption and emission properties in near-infrared (NIR) bio-windows highlight their application in the integration of diagnosis and treatment. In this work, CDs with NIR-I emission and NIR-II absorption have been demonstrated, by constructing localized excited state and charge-transfer state. NIR fluorescence together with photothermal effect was observed in the CDs under NIR light illumination. The electron donors (D) and acceptors (A) groups around the carbon core of the CDs lead to D- π -A charge-transfer state, producing a strong electronic push–pull effect and yielding the desired NIR-II absorption with a maximum absorption peak at 1033 nm. Localized excited state from large conjugated rigid sp 2 domain leads to NIR-I emission of 670 nm. Multiphoton-induced NIR-I emission (quantum yield is 29%) and NIR-II induced heat with photothermal conversion efficiency of 41.19% have been realized. In addition, multiphoton cellular imaging and cancer photothermal therapy using the CDs as imaging and photothermal agents have been demonstrated. The results provide a route to CDs with NIR absorption/emission characters and prospect their potential application in multiple-photon bioimaging and photothermal therapy of cancers.

Abstract:An organic-inorganic hybrid core-shell nanostructure, based on mesoporous silica coated upconversion core-shell nanoparticles (NaGdF4:Yb,Er@NaGdF4:Yb@mSiO2-Dopa abbreviated here as UCNP@mSiO2-Dopa) that stably incorporates dopamine (Dopa) in the silica layer was introduced as a theranostic nanoplatform for optical imaging guided photothermal therapy (PTT) using NIR excitation. Silica-attaching polyethylenimine make the Dopa transforms into an active form (transferred Dopa) that strongly absorbs light under single 980 nm irradiation. We show that the activated UCNP@mSiO2-Dopa nanoplatform is able to produce a pronounced photothermal effect, that elevates water temperature from room temperature to 41.8 °C within 2 minutes, while concurrently emitting strong upconverted luminescence (UCL) for visualized guidance under 980 nm laser. In addition, we demonstrate the application of the same UCNP@mSiO2-Dopa nanoplatform for magnetic resonance imaging (MRI) and x-ray computed tomography (CT) enabled by the gadolinium (Gd) element contained in the UCNP. Importantly, the in vitro and in vivo anti-cancer therapeutic effects have been shown efficacious, implying the use of the described nanoplatform as an effective multi-modal imaging enabled PTT agent. Results from the in vivo biodistribution of UCNPs@mSiO2, cellular live/dead assay, and histologic analysis of main organs of treated mice, reveal that the UCNP@mSiO2-Dopa agents are bio-compatible with low toxicity.

Abstract:Fluorescence imaging (FI) and photoacoustic imaging (PA) play important roles in the real‐time assessment of cell‐based therapies. However, the limitations of conventional organic fluorescence contrast agents and the narrow range of the emission wavelength in the first near‐infrared (NIR‐I) window (750–900 nm) hamper applications of fluorescence imaging in living subjects. Abstract:Carbon dots (CDs) have significant potential for use in various fields including biomedicine, bioimaging, and optoelectronics. However, inefficient excitation and emission of CDs in both near-infrared (NIR-I and NIR-II) windows remains an issue. Solving this problem would yield significant improvement in the tissue-penetration depth for in vivo bioimaging with CDs. Here, an NIR absorption band and enhanced NIR fluorescence are both realized through the surface engineering of CDs, exploiting electron-acceptor groups, namely molecules or polymers rich in sulfoxide/carbonyl groups. These groups, which are bound to the outer layers and the edges of the CDs, influence the optical bandgap and promote electron transitions under NIR excitation. NIR-imaging information encryption and in vivo NIR fluorescence imaging of the stomach of a living mouse using CDs modified with poly(vinylpyrrolidone) in aqueous solution are demonstrated. In addition, excitation by a 1400 nm femtosecond laser yields simultaneous two-photon-induced NIR emission and three-photon-induced red emission of CDs in dimethyl sulfoxide. This study represents the realization of both NIR-I excitation and emission as well as two-photon- and three-photon-induced fluorescence of CDs excited in an NIR-II window, and provides a rational design approach for construction and clinical applications of CD-based NIR imaging agents.

Abstract:A novel concept and approach to engineering carbon nanodots (CNDs) were explored to overcome the limited light absorption of CNDs in low-energy spectral regions. In this work, we constructed a novel type of supra-CND by the assembly of surface charge-confined CNDs through possible electrostatic interactions and hydrogen bonding. The resulting supra-CNDs are the first to feature a strong, well-defined absorption band in the visible to near-infrared (NIR) range and to exhibit effective NIR photothermal conversion performance with high photothermal conversion efficiency in excess of 50%.

Abstract:Photothermal conversion in the second near-infrared (NIR-II) window allows deeper penetration and higher exposure to lasers, but examples of NIR-II photothermal agents are mainly formulated by inorganic compounds. In view of the underlying influence of inorganic materials, a novel NIR-II photothermal nanoagent based on a narrow band gap D-A conjugated polymer (TBDOPV-DT) with 2,2-bithiophene as the donor and thiophene-fused benzodifurandione-based oligo( p-phenylenevinylene) as the acceptor has been developed. More importantly, TBDOPV-DT nanoparticles (TBDOPV-DT NPs) are demonstrated to combine excellent photoacoustic imaging (PAI) and photothermal therapy (PTT) ability. TBDOPV-DT NPs exhibit dramatic photostability and heating reproducibility with a photothermal conversion efficiency of 50%. Especially, the NPs possess a remarkable PTT effect toward cancer cells in vitro and can eliminate tumor cells completely in vivo under 1064 nm laser irradiation, while no appreciable side effects have been observed. This study achieves PAI-guided cancer therapy and sheds light on the future of using organic polymer NPs for the NIR-II PTT of cancer.

Dye adsorption and superb NIR photothermal conversion property of carbon spheres

Abstract:Carbon spheres are often used as template for the fabrication of hollow nanostructures, but less themselves have been studied as high-performance materials. In this work, carbon spheres (CNSs) were synthesized by hydrothermal method and their dye adsorption and photothermal conversion efficiencies were investigated. Samples were characterized using SEM, EDX, XRD, FTIR, UV–vis and TGA measurements. The adsorption of methylene blue (MB) at different pH was investigated by carbon spheres and compared with the absorption of Congo red (CR) dye at pH = 7. Excellent adsorption of MB at neutral and basic pH was obtained by the samples. The photothermal properties of carbon spheres were investigated using a 3 W diode laser with a wavelength of 808 nm for different sample concentrations and different laser powers. The results showed that the photothermal properties was improved by increasing sample concentration and laser power. Maximum temperature increase of 70 °C in 6 min was obtained for concentration of 1 g/l and laser power of 3 W/cm 2 .

Orbicular-Donor-Acceptor System in N-doped Nanographene for Highly Efficient NIR-II Photothermal Therapy

Abstract:Developing stable and efficient photothermal agents (PTAs) for the second near-infrared window (NIR-II, 1 000-1700 nm) photothermal therapy (PTT) is highly desirable but remains challenging. Herein, a facile strategy to prepare NIR-II nano-PTA based on the ionic N-doped nanographene hexapyrrolohexaazacoronene (HPHAC) is reported featuring a specific orbicular-donor-acceptor (O-D-A) structure. Oxidizing HPHAC 1 to dication 12+ causes a substantial decrease in its band gap, leading to a shift in absorption from the confined UV region to a broad absorption range that reaches up to 1400 nm. The dication 12+ exhibits global aromaticity and excellent stability. Theoretical investigation demonstrates that the strong NIR-II absorption of 12+ is attributed to a distinct inner-to-outer intramolecular charge transfer. Encapsulating 12+ with amphiphilic polymers results in water-soluble 12+ NPs with retained optical characteristics. The 12+ NPs exhibit exceptional biocompatibility, intense photoacoustic responses, and a high photothermal conversion efficiency of 72% under the 1064 nm laser irradiation, enabling efficient PTT of cancer cells. The "O-D-A" system on HPHAC, which is created by a simple redox approach, provides a novel strategy to construct efficient NIR-II photothermal materials through molecular engineering of nanographenes.

Abstract:On account of the scarcity of molecules with satisfactory second near-infrared (NIR-II) response, the design of high-performance organic NIR photothermal materials has been limited. Herein, we investigate a cocrystal incorporating tetrathiafulvalene (TTF) and tetrachloroperylene dianhydride (TCPDA) components. A stable radical was generated through charge transfer from TTF to TCPDA, which exhibits strong and wide-ranging NIR-II absorption. The metal-free TTF–TCPDA cocrystal in this research shows high photothermal conversion capability under the 1064 nm laser irradiation and clear photothermal imaging. The remarkable conversion property — which is a result of twisted components in the cocrystal — has been demonstrated by analyses of single crystal X-ray diffraction, photoluminescence and femtosecond transient absorption spectroscopy as well as theoretical calculations. We have discovered that space charge separation and ordered lattice in the TTF–TCPDA cocrystal suppress the radiative decay, while simultaneously strong intermolecular charge transfer enhances the non-radiative decay. The twisted TCPDA component induces rapid internal conversion, while the distorted configuration in TTF–TCPDA favors an internal non-radiative pathway. This research has provided a comprehensive understanding of the photothermal conversion mechanism and opened a new way for the design of advanced organic NIR-II photothermal materials.
