Abstract:Near-infrared (NIR)-absorbing organic nanoparticles (ONPs) are emerging candidates for "one-for-all" theranostic nanomaterials with considerations of safety and formulation in mind. However, facile fabrication methods and improvements in the photothermal conversion efficiency (PCE) and photostability are likely needed before a clinically viable set of candidates emerges. Herein, a new organic compound, [porphyrin-diketopyrrolopyrrole (Por-DPP)] with the donor-acceptor structure was synthesized, where porphyrin was used as a donor unit while diketopyrrolopyrrole was used as an acceptor unit. Por-DPP exhibited efficient absorption extending from visible to NIR regions. After self-assembling into nanoparticles (NPs) (similar to 120 nm), the absorption spectrum of Por-DPP NPs broadened and red-shifted to some extent, relative to that of organic molecules. Furthermore, the architecture of NPs enhanced the acceptor-donor structure, leading to emission quenching and facilitating nonradiative thermal generation. The PCE of Por-DPP NPs was measured and calculated to be 62.5%, higher than most of ONPs. Under 808 nm laser irradiation, the Por-DPP NPs possessed a distinct photothermal therapy (PTT) effect in vitro and can damage cancer cells efficiently in vivo without significant side effects after phototherapy. Thus, the small-molecule porphyrin-based ONPs with high PCE demonstrated promising application in photoacoustic imaging-guided PTT.

Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.

Abstract:Water‐dispersible nanoparticles containing two nanographene‐porphyrin hybrids (NGP‐1‐NPs and NGP‐2‐NPs) are fabricated, displaying intense absorption in NIR‐I and NIR‐II windows and achieving high photothermal conversion efficiency of 60% and 69%, respectively. NIR‐II‐active NGP‐2‐NPs demonstrate promising photoacoustic responses along with high photostability and biocompatibility, enabling the photoacoustic imaging and efficient NIR‐II photothermal therapy of cancer in vivo. Photoacoustic imaging (PAI)‐guided photothermal therapy (PTT) in the second near‐infrared (NIR‐II, 1000–1700 nm) window has been attracting attention as a promising cancer theranostic platform. Here, it is reported that the π‐extended porphyrins fused with one or two nanographene units (NGP‐1 and NGP‐2) can serve as a new class of NIR‐responsive organic agents, displaying absorption extending to ≈1000 and ≈1400 nm in the NIR‐I and NIR‐II windows, respectively. NGP‐1 and NGP‐2 are dispersed in water through encapsulation into self‐assembled nanoparticles (NPs), achieving high photothermal conversion efficiency of 60% and 69%, respectively, under 808 and 1064 nm laser irradiation. Moreover, the NIR‐II‐active NGP‐2‐NPs demonstrated promising photoacoustic responses, along with high photostability and biocompatibility, enabling PAI and efficient NIR‐II PTT of cancer in vivo.

Abstract:Recently, extensive efforts have been devoted to the development of the second near-infrared bio-window (NIR-II, 1000-1700 nm) theranostic agents owing to the excellent tissue-penetration capability of NIR-II light. The exploration of organic NIR-II photothermal therapy materials, especially those with absorption peak over 1000 nm, is an appealing yet significantly challenging task. Herein, we have designed conjugated polymer nanoparticles (PIS NPs) with NIR-II absorption peak at 1026 nm through a combined strategy of introducing quinoid donor-acceptor (D-A) structures, constructing intramolecular "conformational locks" and extending the conjugation area to narrow the band gap. Irradiated at 1064 nm, PIS NPs showed remarkable photothermal conversion performance for efficient photothermal ablation of tumor cells in vitro and in vivo. This study provides useful insights into the rational design of organic NIR-II photothermal agents based on multiple strategies.

Abstract:Photothermal conversion in the second near-infrared (NIR-II) window allows deeper penetration and higher exposure to lasers, but examples of NIR-II photothermal agents are mainly formulated by inorganic compounds. In view of the underlying influence of inorganic materials, a novel NIR-II photothermal nanoagent based on a narrow band gap D-A conjugated polymer (TBDOPV-DT) with 2,2-bithiophene as the donor and thiophene-fused benzodifurandione-based oligo( p-phenylenevinylene) as the acceptor has been developed. More importantly, TBDOPV-DT nanoparticles (TBDOPV-DT NPs) are demonstrated to combine excellent photoacoustic imaging (PAI) and photothermal therapy (PTT) ability. TBDOPV-DT NPs exhibit dramatic photostability and heating reproducibility with a photothermal conversion efficiency of 50%. Especially, the NPs possess a remarkable PTT effect toward cancer cells in vitro and can eliminate tumor cells completely in vivo under 1064 nm laser irradiation, while no appreciable side effects have been observed. This study achieves PAI-guided cancer therapy and sheds light on the future of using organic polymer NPs for the NIR-II PTT of cancer.
Molecular engineering of D-A-D conjugated small molecule nanoparticles for high performance NIR-II photothermal therapy

Abstract:Chemically defined conjugated small molecules (CSMs) and their nanoparticles (CSMNs) have been widely used in phototheranostics. However, CSMNs have rarely been studied for second near-infrared (NIR-II) deep-tissue photothermal therapy (PTT) due to the lack of NIR-II-absorbing CSMNs. Herein, through a molecular engineering strategy of "increasing the number of thiophene bridges", three donor-acceptor-donor (D-A-D) CSMs, namely CSM0-2 in which the number represents the number of thiophene bridges, have been synthesized. The optical bandgap decreases with the increase of the number of thiophene bridges. Among them, CSM2 has strong absorption in both the first near-infrared (NIR-I) and NIR-II regions. CSMN2 exhibits high photothermal conversion efficiency (PTCE = 31.6%) in NIR-II and excellent biocompatibility. By using CSMN2, photoacoustic imaging (PAI)-guided in vivo tumor NIR-II PTT is achieved under low laser power density (0.5 W cm(-2)). The molecular engineering strategy reported here can potentially be applied to other D-A systems for the development of new materials in phototheranostics.

Abstract:Development of special organic materials that are able to absorb light energy in the second near-infrared window (NIR-II) is significantly important for treating deep-tissue-buried diseases or supplying power to implantable electronic devices. Herein, a narrow bandgap donor–acceptor (D-A) conjugated polymer with thiophene-fused benzodifurandione-based oligo(p-phenylenevinylene) (TBDOPV) as acceptor part and 2,2′-bithiophene (DT) as donor part was developed and exploited as a photothermal conversion material with high extinction coefficient and robust photostability in the NIR-II window. According to transient absorption analysis results, the photothermal conversion ability of this polymer is attributed to the fast internal conversion (IC) process. The high photothermal conversion efficiency makes this polymer a promising NIR-II adsorbing antenna to remotely actuate thermo-dependent devices, e.g., high-performance photothermal–electrical and photothermal–mechanical converters.

Abstract:1T-phase transition-metal dichalcogenides (TMDs) nanomaterials are one type of emerging and promising near-infrared II (NIR-II) photothermal agents (PTAs) derived from their distinct metallic electronic structure, but it is still challenging to synthesize these nanomaterials. Herein, PdTe2 nanoparticles (PTNs) with a 1T crystal symmetry and around 50 nm in size are prepared by an electrochemical exfoliation method, and the corresponding photothermal performances irradiated under a NIR-II laser have been explored. The encapsulation of 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-poly(ethylene glycol) (DSPE-PEG) endows PTNs with water solubility, enhanced photothermal stability, and high biocompatibility. Notably, PTN/DSPE-PEG displays a potent absorbance through the NIR-II zone and considerable photothermal conversion efficiency, which is up to 68% when irradiated with a 1060 nm laser. Ultrastable Near‐Infrared Conjugated‐Polymer Nanoparticles for Dually Photoactive Tumor Inhibition

Abstract:It is highly desired that satisfactory photoactive agents with ideal photophysical characteristics are explored for potent cancer phototherapeutics. Herein, bifunctional nanoparticles of low-bandgap donor-acceptor (D-A)-type conjugated-polymer nanoparticles (CP-NPs) are developed to afford a highly efficient singlet-to-triplet transition and photothermal conversion for near-infrared (NIR) light-induced photodynamic (PDT)/photothermal (PTT) treatment. CP-NPs display remarkable NIR absorption with the peak at 782 nm, and perfect resistance to photobleaching. Photoexcited CP-NPs undergo singlet-to-triplet intersystem crossing through charge transfer in the excited D-A system and simultaneous nonradiative decay from the electron-deficient electron acceptor isoindigo derivative under single-wavelength NIR light irradiation, leading to distinct singlet oxygen quantum yield and high photothermal conversion efficiency. Moreover, the CP-NPs display effective cellular uptake and cytoplasmic translocation from lysosomes, as well as effective tumor accumulation, thus promoting severe light-triggered damage caused by favorable reactive oxygen species (ROS) generation and potent hyperthermia. Thus, CP-NPs achieve photoactive cell damage through their photoconversion ability for synergistic PDT/PTT treatment with tumor ablation. The proof-of-concept design of D-A-type conjugated-polymer nanoparticles with ideal photophysical characteristics provides a general approach to afford potent photoactive cancer therapy.

Abstract:Photothermal therapy (PTT), a promising spatiotemporally controllable cancer treatment method, needs effective depth of laser irradiation to ensure the anticancer efficiency. Second near-infrared window (NIR-II) absorbing conjugated polymer (CP) nanomaterials are emerging candidates for a non-invasive treatment of cancer with considerations of safety. Herein, a new organic CP compound (Ppordpp) with the donor-acceptor structure of porphyrin and diketopyrrolopyrrole was synthesized, which showed efficient absorption extending in NIR-II and spontaneously self-assembled into NPs (∼190 nm), leading to emission quenching and facilitating nonradiative thermal generation. Photothermal conversion efficiency (PCE) of our Ppordpp NPs was up to 86.21 %, higher than most of reported PTT reagents. Under 940 nm (0.75 W/cm 2 ) laser irradiation for 10 min, the Ppordpp NPs showed dramatical PTT effects in vitro to kill 78% 4T1 cancer cells with 40 μg/mL incubation, and in vivo to almost completely inhibition tumor growth by 200 μg/mL injection without obvious side effects. Our CP nanomaterials Ppordpp NPs with a D-A structure provide new alternatives for precise cancer through photothermal therapy with high PCE.

Achieving Efficient NIR‐II Type‐I Photosensitizers for Photodynamic/Photothermal Therapy upon Regulating Chalcogen Elements

Abstract:Second near-infrared (NIR-II) window type-I photosensitizers have intrinsic advantages in photodynamic/photothermal therapy (PDT/PTT) of some malignant tumors with deep infiltration, large size, complicated location, and low possibility of surgery/radiotherapy. Herein, three chalcogen-element-based donor-acceptor-type semiconducting polymers (poly[2,2″-((E)-4,4″-bis(2-octyldodecyl)-[6,6″-bithieno[3,2-b]pyrrolylidene]-5,5″(4H,4″H)-dione)-alt-2,5-(thiophene)] (PTS), poly[2,2″-((E)-4,4″-bis(2-octyldodecyl)-[6,6″-bithieno[3,2-b]pyrrolylidene]-5,5″(4H,4″H)-dione)-alt-2,5-(selenophene)] (PTSe), and poly[2,2″-((E)-4,4″-bis(2-octyldodecyl)-[6,6″-bithieno[3,2-b]pyrrolylidene]-5,5″(4H,4'H)-dione)-alt-2,5-(tellurophene)] (PTTe)) are synthesized and fully characterized, demonstrating strong absorption in the NIR-II region. Upon adjusting the chalcogen elements, the intramolecular charge-transfer characteristics and the heavy-atom effect are tuned to enhance the intersystem crossing rate, improving the photodynamic effect. Moreover, the energy levels and Gibbs free energies are tuned to facilitate the type-I photodynamic process. As a result, PTTe nanoparticles (NPs) produce superoxide anion radicals (O2 •- ) more efficiently and demonstrate higher photothermal conversion efficiency than PTS and PTSe NPs upon NIR-II (1064 nm) laser irradiation, exhibiting unprecedented NIR-II type-I PDT/PTT performance in vitro and in vivo. This work provides ideas for achieving high-performance NIR-II type-I PDT/PTT semiconducting polymers for hypoxic oncotherapy.

Abstract:A novel low‐molecular‐weight dibenzofulvene‐based D‐π‐A phototheranostic agent (PTA) with NIR‐II emission and high PCE is developed by the strategies of enhancing D‐A strength, introducing molecular rotors/vibrators, and facilitating intermolecular π–π stacking. The PTA is successfully applied in the multi‐modal imaging (NIR‐II fluorescence/photoacoustic/photothermal imaging)‐guided photothermal therapy. Organic molecules with second near‐infrared (NIR‐II, 1000–1700 nm) emission have significant advantages over NIR‐I (600–900 nm) dyes, and have been widely used as phototheranostic agents (PTAs) and fluorescent probes. However, the two biggest challenges for such reagents are improving photothermal conversion efficiency (PCE), and simplifying tedious synthetic procedures. Herein, different from the traditional NIR skeleton, for the first time, it develops two novel dibenzofulvene‐based NIR‐II emission PTAs via D‐π‐A concept with high PCEs through two simple synthetic steps. The increased intramolecular charge transfer effect and extended π‐conjugation effectively red‐shift their emissions to the NIR‐II region. In addition, the introduction of molecular rotors/vibrators and the formation of π–π stacking prompt the two PTAs to exhibit high PCEs. Notably, FE‐IDMN nanoparticles (FE‐IDMN NPs) achieve an exciting PCE of 82.6%, higher than most reported photothermal agents with NIR‐II emission. FE‐IDMN NPs also possess good colloidal, pH, and photothermal stabilities, as well as excellent photoacoustic response, which is further successfully applied for NIR‐II fluorescence/photoacoustic/photothermal imaging‐guided photothermal therapy. This work provides a simple strategy for constructing new D‐π‐A PTAs with low molecular weight, NIR‐II emission, and high PCE for cancer therapy.

Abstract:The emergence of near-infrared-II (NIR-II) activated photomedicines has extended the penetration depth for noninvasive theranostics, especially for photothermal nanomedicines. The current early development stage for NIR-II activated photomedicines has focused on creating a greater variety of photothermal agents (PTAs) with superior photothermal conversion ability. However, there is no thorough review for NIR-II inorganic PTAs and most comparisons of the photothermal performances of NIR-II inorganic PTAs are made with NIR-I PTAs. This review will first discuss about the key mechanisms of NIR-II absorption and photothermal conversion. Subsequently, this review will summarize recently developed advanced NIR-II inorganic PTAs based on the dominant inorganic elements and provide a comparison of their NIR-II photothermal performances. The nanostructure design, enhancement strategies and potential biomedical applications will be listed and discussed. We hope this review will further inspire active development and study of NIR-II activated inorganic PTAs with good photothermal conversion ability, multifunctionality, biocompatibility or biodegradability, and disease targeting ability.

Acceptor-donor-acceptor-type Molecules with Large Electrostatic Potential Difference for Effective NIR Photothermal Therapy

Abstract:Although acceptor-donor-acceptor (A-D-A)-type molecules offer advantages in constructing NIR absorbing photothermal agents (PTAs) due to their strong intramolecular charge transfer and molecular planarity, their applications in photothermal therapy (PTT) of tumors remain insufficiently explored. In particular, the influence of ESP distribution on the optical properties of A-D-A photosensitizers has not been investigated. Herein, we analyze and compare the difference in ESP distribution between A-D-A-type small molecules and polymers to construct NIR absorbing PTAs with a high extinction coefficient (epsilon) and high photothermal conversion efficiency (PCE). The calculation results of density functional theory (DFT) indicate that the large ESP difference makes A-D-A-type small molecules superior to their polymer counterparts in realizing tight molecular packing and strong NIR absorbance. Among the as-prepared nanoparticles (NPs), Y6 NPs exhibited an obvious bathochromic shift of absorption peak from 711 nm to 822 nm, with the NIR-II emission extended to 1400 nm. Moreover, a high epsilon value of 5.69 L g-1 cm-1 and a PCE of 66.3% were attained, making Y6 NPs suitable for PTT. With a concentration of 100 mu g mL-1, Y6 NPs in aqueous dispersion yielded a death rate of 93.4% for 4T1 cells upon 808 nm laser irradiation (1 W cm-2) for 10 min, which is comparable with the best results of recently reported PTT agents. We explored a new way of utilizing the ESP difference to improve the molecular packing of (A-D-A) type molecules, and obtained a series of NIR PTAs with good PTT performance. The in vivo antitumor study demonstrates effective suppression of tumors.

Abstract:A stable alkyne‐bridged conjugated polymer, BBT‐BTE, is designed with strong NIR‐II absorption, and its nanoparticles achieve a high photothermal conversion efficiency of 71%. BBT‐BTE NPs achieve complete tumor ablation without any recurrence, and the biosafety of these NPs has also been further proven. This study offers feasible insight into the design of efficient photothermal materials in the NIR‐II window. Organic photothermal materials (PTMs) in the near‐infrared (NIR) range (1000–1700 nm) are more favorable for photothermal therapy (PTT) therapeutic applications because of their greater depth of tissue penetration and better biosafety. However, the photothermal conversion efficiency (PCE) of the few NIR‐II‐responsive organic PTMs that have been investigated is still slightly low. Here, a stable alkyne‐bridged conjugated polymer, BBT‐BTE, are explored with a high PCE for tumor ablation within the NIR‐II window. The BBT‐BTE synthesized by the Stille coupling reaction has strong NIR‐II absorption, and their nanoparticles (NPs) achieved a 71% PCE under 1064 nm laser excitation, with good photothermal stability. BBT‐BTE NPs are shown to be effective in completely ablating tumors without any recurrence in both in vitro and in vivo experiments. Additionally, the biosafety of these NPs is further proven by biochemical analysis and tissue biopsy. This study developed a high‐performance NIR‐II PTM and offered practical and feasible insights into the design of efficient photothermal materials specifically for the NIR‐II window.

Abstract:The contradiction of near-infrared II (NIR-II) emission and photothermal effects limits the development of phototheranostic agents (PTAs) in many emerging cutting-edge applications. Organic aggregates present a promising opportunity for the balance of competitive relaxation processes through the manipulation of molecular structure and packing. Herein, side chain phenyl isomerization-induced spatial conjugation was proposed for constructing A-D-A type NIR-II PTAs with simultaneous enhancement of fluorescence brightness and photothermal properties. Three pairs of mutually isomeric fluorophores, whose phenyls respectively located at the outside (o-series) and inside (i-series) of the side chain, were designed and synthesized. The positional isomerization of the phenyl endows the o-series crystals with strong spatial conjugation between the phenyl group on the side chain and the backbone, as well as interlocked planar network, which is different to that observed in the i-series. Thus, all o-series nanoparticles (NPs) exhibit red-shifted absorption, enhanced NIR-II emission, and superior photothermal properties than their i-series counterparts. A prominent member of the o-series, o-ITNP NPs, demonstrated efficacy in facilitating NIR-II angiography, tumor localization, and NIR-II imaging-guided tumor photothermal therapy. The success of this side chain phenyl isomerization strategy paves the way for precise control of the aggregation behavior and for further development of efficient NIR-II PTAs.

Abstract:Non-invasive cancer photothermal therapy (PTT) is a promising replacement for traditional cancer treatments. The second near-infrared region induced PTT (NIR-II PTT, 1000–1500 nm) with less energy dissipation has been developed for deeper-seated tumor treatment in recent years compared with the traditional first near-infrared light (750–1000 nm). In addition, the use of emerging inorganic 2D nanomaterials as photothermal agents (PTAs) further enhanced PTT efficiency due to their intrinsic photothermal properties. NIR-II light stimulated inorganic 2D nanomaterials for PTT is becoming a hot topic in both academic and clinical fields. This review summarizes the categories, structures, and photothermal conversion properties of inorganic 2D nanomaterials for the first time. The recent synergistic strategies of NIR-II responsive PTT combined with other treatment approaches including chemotherapy, chemodynamic therapy, photodynamic therapy, radiotherapy are summarized. The future challenges and perspectives on these 2D nanomaterials for NIR-II responsive PTT systems construction are further discussed.

A-DA′D-A Structured Organic Phototheranostics for NIR-II Fluorescence/Photoacoustic Imaging-Guided Photothermal and Photodynamic Synergistic Therapy

Abstract:Multimodal imaging-guided combinational phototherapies triggered by a single near-infrared (NIR) laser are highly desirable. However, their development is still a big challenge. Herein, we have developed an "acceptor–donor–acceptor′–donor–acceptor" structured organic phototheranostics (Y16-Pr) with strong light-harvesting ability in the NIR region. After being modified with polyethylene glycol (PEG), the obtained biocompatible nanoparticles (Y16-Pr-PEG NPs) could conduct NIR-II fluorescence imaging (FLI) and photoacoustic imaging (PAI) and perform photothermal therapy (PTT) and photodynamic therapy (PDT) simultaneously. Notably, Y16-Pr-PEG NPs showed an impressive photothermal conversion efficiency (PCE) of 82.4% under 808 nm laser irradiation. The irradiated NPs could also produce hydroxyl radicals (•OH) and singlet oxygen (1O2) for type I and type II PDT, respectively. In vivo and in vitro experiments revealed that the Y16-Pr-PEG NPs significantly inhibit tumor cell growth without apparent toxic side effects under laser irradiation. Abstract:Numerous photothermal agents (PTAs) require high-intensity and long-duration laser excitation for photothermal therapy (PTT), resulting in light damage to healthy skin and tissue as well as limiting their biomedical applications. Integrating desirable near-infrared (NIR) absorption and high photothermal conversion efficiency (PCE) into a single small-molecule PTA is an important prerequisite for realizing efficient PTT, but is a serious challenge. Herein, through molecular engineering strategy, an acceptor-donor-acceptor (A-D-A) type PTA (ADA3) was readily developed for 808 nm laser-driving photothermal imaging and PTT of tumor. Theoretical calculations and experiment results show molecular engineering strategy is significant in regulating the structure and energy gap of PTAs, so as to effectively induce a narrow band gap for NIR absorption and further optimize photothermal properties. ADA3 possesses molar extinction coefficient of 3.1 × 104 M-1 cm-1 at 808 nm, followed being assembled into nanoparticles, ADA3-NPs show high PCE of 80.3%. In vivo experiments indicate that ADA3-NPs have excellent antitumor capability under one-time, low-intensity and short-duration (808 nm, 330 mW/cm2, 3 min) laser irradiation. Therefore, this work definitely exemplifies the enormous potential of molecular engineering strategy and provides an effective method for developing small-molecule PTAs.
