Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:A novel seminaphthorhodafluor-based fluorescent probe was designed and prepared. It exhibited an outstanding off-on fluorescence response (about 45-fold enhancement) to Cu2+ because the structure changed from a spirocyclic form to a ring-open state. It showed satisfactory detection performances for Cu2+ with fast response speed, high selectivity and anti-interference, and a low detection limit (22.9 nM) due to its long emission wavelength and off-on fluorescence response mode. This probe was also satisfactorily applied for the monitoring of Cu2+ in living cells and zebrafish.

Abstract:Background: In recent years, environmental pollution has been increasing due to the excessive emission of toxic ions, which has caused serious harm to human health and ecological environment. There are various methods for detecting Cu2+, S2- and Zn2+, but the traditional ion detection methods have obvious disadvantages, such as poor selectivity and long detection time. Therefore, it is still crucial to develop simple, efficient and rapid detection methods. Results: A fluorescent probe based on benzothiazole, (E)-N'-(3-(benzo[d]thiazol-2-yl)-2-hydroxy-5-methylbenzylidene)-3,4,5-tris(benzyloxy)benzohydrazide (BT), was designed and synthesized. It was characterized using ESI-MS, 1H NMR, and 13C NMR. BT can be used as a chemosensor to detect Cu2+, S2- and Zn2+ in CH3CN/H2O (7:3, v/v, pH = 7.4, HEPES buffer: 0.1 M), with detection limits of 0.301 μM, 0.017 μM, and 0.535 μM, respectively. At an excitation wavelength of 320 nm, BT exhibits an "on-off-on" response to Cu2+/S2- and enhanced fluorescence response to Zn2+, with a change in fluorescence color from orange to green. The coordination ratio of ions to the probe was determined to be 1:1 through Job's plot and hydrogen spectral titration. The recognition mechanism was discussed in conjunction with theoretical calculations. Furthermore, the probe has been successfully used in test strips and medical swabs colorimetry, as well as live cell imaging. Significance: The probe BT lays the foundation for the design and synthesis of multifunctional fluorescent probes. As a portable detection method, probe BT was used to detect Cu2+, S2- and Zn2+ on strips. Furthermore, the probe was applied to biological cells to detect target ions with low cytotoxicity and excellent cell permeability. This indicating that it can be used as a potential candidate for tracking Cu2+ and S2- in clinical diagnostics and biological systems.

Abstract:Constructed on the benzothiazole-oxanthracene structure, a fluorescent probe RBg for Cu+ was designed under the ESIPT mechanism and synthesized by incorporating amide bonds as the connecting group and glyoxal as the identifying group. Optical properties revealed a good sensitivity and a good linear relationship of the probe RBg with Cu+ in the concentration range of [Cu+] = 0–5.0 μmol L−1. Ion competition and fluorescence-pH/time stability experiments offered further possibilities for dynamic Cu+ detection in an aqueous environment. HRMS analysis revealed a possible 1:1 combination of RBg and Cu+. In addition, colorimetric Cu+ detection and lysosome-targeted properties of the probe RBg were analyzed through RBg-doped PVDF nanofiber/test strips and RBg-Mito/Lyso trackers that were co-stained in living HeLa cells, enabling the probe's future applications as real-time detection methods for dynamic Cu+ tracking in the lysosomes and Cu+ detection under diversified conditions.

Abstract:Cu2+ in lysosomes are closely related to some diseases such as Menke's syndrome, Wilson's diseases and Alzheimer's diseases. Although many fluorescent probes have been constructed for the detection of Cu2+ in lysosomes, the poor water solubility and short wavelength emission (λem < 600 nm) limit the application of these probes. To solve this problem, a fluorescent probe DHUCu-1 with near-infrared emission was designed and synthesized in this work. DHUCu-1 could detect Cu2+ in PBS solution (1% alcohol as co-solvent) with high sensitivity and good selectivity. Moreover, DHUCu-1 could target lysosomes and respond to Cu2+ in lysosomes. This probe thus can be used as a tool for the exploration of physiological functions of Cu2+ and the related diseases.

Abstract:A near-infrared fluorescent probe (NUST-Cy-1) was disclosed here, which displays ratiometric and dual-channel response for Cu2+ (λex1 = 450 nm, λex2 = 750 nm) with large Stokes shifts (143 nm, 375 nm, 75 nm respectively). This probe demonstrates high sensitivity with low detection limit (1.4 μM) and selectivity for Cu2+ detection. Furthermore, fluorescent imaging of Cu2+ in vitro and vivo were successfully achieved.

Abstract:In this study, a new pyrazoline derivate (1-acetyl-3- (2-hydroxy-1-naphthyl)-5-(2-phenyl-2H-chromene)-2-pyrazoline, N) is described as fluorescent probe for fluorometric detection of Cu2+. This fluorescence of probe has a high selectivity only toward Cu2+ ion in the other metal ions. It is remarkably observed that the fluorescence intensity of N decreased in the presence of Cu2+ ions with a strong capability to resist perturbations.The Limit of detection (LOD) value is calculated as 8.85 × 10-7 M for Cu2+ while the complexation constant is calculated as 3.14 × 105 M−2 via fluorescence measurements. The interaction between of N and Cu2+ is reversible after adding PPi. The optimized calculation of the probe for Cu2+ is confirmed by TD-DFT calculations. N-Cu2+ is successfully applied in the intracellular imaging in living PK-21cells that will have a great chance for wide application.

Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:A novel highly selective and sensitive fluorescent probe (NJ1) had been designed and synthesized for Cu2+ detection by fluorescence quenching mechanism, and then the enhancement of fluorescence intensity with the addition of hydrogen sulfide in complex NJ1Cu aqueous solution at physiological conditions were described. This "on-off-on" type fluorescence recognition system was a reversible process, which could be utilized to monitor copper ion and hydrogen sulfide based on a complex Nj1Cu formation-Cu2+ displacement approach. Importantly, this real-time recognition process of Cu2+ and hydrogen sulfide exhibited excellent anti-interference ability. In addition, this new fluorescent probe also has potential utility for Cu2+ and hydrogen sulfide detection in living cells, providing a potential tool for investigating copper ion and hydrogen sulfide in living systems or environment. Abstract:Tracing copper ions levels in the environment and subcellular microenvironment is crucial due to the key role copper ions play in physiological and pathological processes. Herein, a novel naphthalimide-fused rhodamine probe Rh-Naph-Cu was prepared through modification with phenylhydrazine to produce a closed and non-fluorescent spirolactam. Based on the copper-induced spirolactam ring-opening and hydrolysis process, Rh-Naph-Cu can be employed as a fluorescence off-on probe for copper ions with high selectivity, high sensitivity (limit of detection: 33.0 nM), broad pH-response range (pH: 5.0-10.0), and color change visible with the naked eye. Rh-Nap-Cu could be made into test strips for the in-situ chromogenic detection of Cu2+. Significantly, Rh-Naph-Cu can be utilized for the detection of copper ions in living HeLa cells and zebrafish, and exhibits excellent lysosomal-targeting ability with high Pearson's correlation coefficient (PCC) of 0.96.

Abstract:A novel fluorescent probe DCM-CuCl was developed for the detection of Cu 2+ . With the intramolecular charge transfer effect recovered by a Cu-catalyzed hydrolysis reaction, the probe DCM-CuCl can detect the Cu 2+ faster through the molecular modification of chloride ions. In addition, the probe DCM-CuCl has a large Stokes shift (148 nm), NIR region emission (668 nm), and low detection limitation (0.13 μM) . Further, the probe could be used to detect the Cu 2+ in living cells and actual water samples.

Abstract:A new benzothiazole-based fluorescent probe 2-(benzo[d]thiazol-2-yl)-4-(1,3- dithian-2-yl)phenol (BT) with two different reaction sites, a thioacetal group (site 1 for Hg2+), and O and N atoms of the benzothiazole dye (site 2 for Cu2+), was designed and synthesized. The probe BT showed ratiometric fluorescent response to Hg2+ and fluorescence quenching behavior to Cu2+, which induces naked-eye fluorescent color changes from green to blue and colorless, respectively. Moreover, it displayed highly sensitivity and selectivity toward Hg2+ and Cu2+ without interference from other metal ions. The sensing mechanisms were also confirmed by 1H NMR titration, mass spectrum and Job's plot analyses. Finally, probe BT was successfully used for fluorescent imaging of Hg2+ and Cu2+ in living cells, demonstrating its potential applications in biological science.

Abstract:A novel ferrocenyl-naphthalimide multichannel probe 1 was designed and synthesized using a facile method. The color of the solution containing probe 1 changed from yellow to colorless upon the addition of Cu2+ or Hg2+. Interestingly, probe 1 exhibited highly selective fluorescent turn-on for Cu2+ and turn-off for Hg2+ in aqueous solution. Probe 1 was an electrochemical Cu2+ and Hg2+ ion sensor, in which the Fc/Fc+ redox couple was significantly shifted (ΔE1/2 = 178 mV and ΔE1/2 = 53 mV, respectively) upon complexation. Therefore, probe 1 can act as a naked-eye chemosensor, as well as an electrochemical and a fluorescent probe for Cu2+ and Hg2+. Furthermore, this is the first reported probe that can be used for the bifunctional fluorescent detection of intracellular Cu2+ and Hg2+ by fluorescent imaging studies. These characteristics give this probe considerable potential in the study and analysis of Cu2+ and Hg2+ in complex biosystems.

Abstract:Cu2+ and pH play a key role in life systems and it is necessary to develop an effective detection method. In view of the significant advantages of the fluorescent probe, we have synthesized a dual functional fluorescent probe NCNB based on naphthalimide for detecting Cu2+ and pH. The probe NCNB is sensitive (LOD = 44.8 nM) and fast for Cu2+ (within 150 s). It can be used not only for qualitative and quantitative detection of Cu2+ in actual water samples, but also for making pH fluorescent test paper with good effect. In addition, the probe NCNB has been successfully used for imaging detection of exogenous Cu2+ in Caco-2 cells with low toxicity.

Abstract:Developing fluoran dyes as fluorescent probes is interesting because it is of great significance to extend their applications from the printing industry to other fields such as fluorescence analysis and medicine. We here present a new quinoline-fluoran hybrid dye QF, which delivers fluorescent signals in water (pH: 4.0 - 9.0) with the maximum absorption and emission centered at 514 nm and 597 nm, respectively. In addition, a large Stokes shift (83 nm) can be observed when compared to that of rhodamine B. QF is further employed for the preparation of a new fluorescent probe QFH, which exhibits high selectivity towards copper ions with the limit of detection down to 1.54 nM via a Cu2+-triggered spirolactam ring opening and hydrolysis process. Significantly, QFH can be applied for the detection of copper ions at physiological pH conditions (pH: 6.0 - 9.0) and shows low cell cytotoxicity, which allows it to be a candidate for tracing Cu2+ by cell imaging.
