Abstract:1593 Objectives Copper is an essential element for the activity of a number of physiologically important enzymes and its disorders are related to many neurological symptoms such as Alzheimer9s, Mendes, and Wilson diseases. The question is whether copper ions (non-radio or radio-active) can be detected by molecular imaging. Herein we report a novel dansyl-based fluorescent imaging probe for detecting Cu2+. Methods Probe 1 is designed based on the intramolecular charge transfer (ICT) mechanism, and synthesized as shown in Scheme 1. In neutral MeOH-H2O solution conditions (HEPPS 10 μM, PH = 6.8, MeOH/H2O=1:1, v/v), the sensitivity and selectivity of probe 1 toward copper () ions, including the linear relationship between the concentration of copper ions and fluorescence intensity, are investigated. Results Probe 1 was synthesized in a total yield of 33% in 5 steps. Fluorescence experiments show that Cu2+ can significantly quench probe 1 from yellow to colorless at 20 μM while other chemically similar ions have little effect, and a good linear relationship between the concentration of copper ions and fluorescence intensity was observed. Even when other metal ions exist, probe 1 still shows excellent selectivity toward Cu2+ (Scheme 2). Conclusions Probe 1 was synthesized in a good yield and can detect copper () ions with high sensitive and selective. Study to determine if copper ions in neurological cells or tissue could be imaging effectively is underway.
