Abstract:In this work, novel cuprous oxide-demethyleneberberine (Cu2O-DMB) nanomaterials are successfully synthesized for photoresponsive-enhanced enzymatic synergistic antibacterial therapy under near-infrared (NIR) irradiation (808 nm). Cu2O-DMB has a spherical morphology with a smaller nanosize and positive ζ potential, can trap bacteria through electrostatic interactions resulting in a targeting function. Cu2O-DMB nanospheres show both oxidase-like and peroxidase-like activities, and serve as a self-cascade platform, which can deplete high concentrations of GSH to produce O2˙- and H2O2, then H2O2 is transformed into ˙OH, without introducing exogenous H2O2. At the same time, Cu2O-DMB nanospheres become photoresponsive, producing 1O2 and having an efficient photothermal conversion effect upon NIR irradiation. The proposed mechanism is that the generated ROS (O2˙-, ˙OH and 1O2) and hyperthermia can have synergetic effects for killing bacteria. Moreover, hyperthermia is not only beneficial for destroying bacteria, but also effectively enhances the efficiency of ˙OH production and accelerates GSH oxidation. Upon NIR irradiation, Cu2O-DMB nanospheres exhibit excellent antibacterial ability against methicillin-resistant Staphylococcus aureus (MRSA) and ampicillin-resistant Escherichia coli (AREC) with low cytotoxicity and bare bacterial resistance, destroy the bacterial membrane causing an efflux of proteins and disrupt the bacterial biofilm formation. Animal experiments show that the Cu2O-DMB + NIR group can efficiently treat MRSA infection and promote wound healing. These results suggest that Cu2O-DMB nanospheres are effective materials for combating bacterial infections highly efficiently and to aid the development of photoresponsive enzymatic synergistic antibacterial therapy.

Abstract:TiO 2 /TiOF 2 Z-scheme nanosheets have been successfully synthesized for photocatalytic antibacterial. The antibacterial efficiency of TiO 2 /TiOF 2 against E. coli and S. aureus were 99.90 % and 81.89 % at low material concentration (110 μg/mL), respectively, which are higher than those of pure TiO 2 , TiOF 2 , and Degussa P25. In situ molecular spectroscopy results demonstrate that the microstructure of the synthesized material can be reconstructed and optimized to enhance the exposure of the active sites·H 2 O and O 2 are effectively adsorbed on the catalyst surface and activated to form OH...Ti and O...Ti surface active species. Furthermore, the dense interface formed in TiO 2 /TiOF 2 acts as an efficient transport path for photoexcited electrons from TiO 2 to TiOF 2 , and thus accelerates the formation of reactive oxygen species. Finally, the mechanism of bacterial inactivation is systematically discussed considering the main active substances, cell morphological changes, and activity of antioxidant enzymes.

The Enhanced Near-Infrared Photocatalytic and Photothermal Effects of MXene-based Heterojunction for Rapid Bacteria-Killing

Abstract:Bacterial resistance threatens the health of human beings when they combat pathogenic diseases with antibiotics. Herein, an ecofriendly and antibiotics-free 0D/2D Schottky heterojunction (Ag2S/Ti3C2) was synthesized for the first time to treat bacterial infection with a high efficacy of 99.99% within 20 min under 808 nm NIR light irradiation, which was ascribed to the synergy of enhanced photocatalytic and photothermal effects. The density functional theory calculations show that when a redistribution of charge occurs at the interface between Ag2S and Ti3C2 due to the difference in their Fermi energy levels (EF), it results in an upward bend of the Ag2S energy band. Additionally, in the case of Ag2S/Ti3C2, the higher conduction band energy of Ag2S compared to the EF of Ti3C2 and the excellent electrical conductivity of the latter made the photoexcited electrons in Ag2S, including those hot electrons produced by surface plasma resonance (SPR), rapidly flow to Ti3C2, thus greatly increasing both photocatalytic and photothermal performance of Ti3C2. In vitro and in vivo tests showed that this photoresponsive system possessed not only highly effective antibacterial efficacy against Staphylococcus aureus infection but also excellent biocompatibility simultaneously. Therefore, this hybrid is expected to be a promising ecofriendly platform for the rapid treatment of bacterial infections using NIR light.

Abstract:Driven by market demand and resourcefulness of low-value minerals, the preparation of low-cost visible photocatalytic bactericidal materials has become a research hotspot. Here, a simple solvothermal method was constructed FeS 2 photosensitized TiO 2 , which is a promising antibacterial and photocatalytic sterilization strategy. The disinfection efficiency of TiO 2 /FeS 2 for Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) was 95.7 and 92.4%, respectively. In the darkness, TiO 2 /FeS 2 could wrap around the surface of bacterial cells and significantly decrease their metabolic activity of bacterial cells of E. coli and S. aureus to 21.4% and 30.1%, respectively. Under the light irradiation, TiO 2 /FeS 2 exhibited the light absorption feature of both TiO 2 and FeS 2 . Meanwhile, an internal electric field from FeS 2 to TiO 2 was established to realize electron transmission, which as dominant role cooperating with the surface redox reaction, allowed TiO 2 /FeS 2 produced obviously higher •OH, •O 2 – , and 1 O 2 to perform an ideal disinfection effect.

Abstract:In this study, zincperoxide (ZnO2) with broad energy gap was firstly used for visible-light-induced photocatalytic degradation of tetracycline (TC) and inactivation of Escherichia coli (E. coli). A small amount of ZnO2 (10 mg) could efficiently degrade 100 mL of 50 mg/L TC in a wide pH range (4-12), and the degradation performance was rarely suppressed by common matrix species and natural water sources. Also, 100 mg/L ZnO2 could inactivate around 7-log E. coli cells within 60 min under visible-light irradiation. Quenching experiments and electron paramagnetic resonance (EPR) results confirmed that superoxide radical (•O2-) and singlet oxygen (1O2) were the main reactive oxygen species (ROS), which were attributed to the self-sensitization of TC and the photoexcitation of released H2O2 under the catalysis of Zn(OH)2 from the hydrolysis of partial ZnO2, respectively. The pathways of TC degradation and processes of visible-light-induced TC degradation and E. coli inactivation were proposed and deduced in detail. This work presented the enhanced visible-light photocatalytic activities of ZnO2 for antibiotic degradation and bacterial inactivation, and provided a deep insight into the mechanisms of visible-light-induced TC degradation andE. coli inactivation over ZnO2.

Abstract:Abstract In view of increasing drug resistance, ecofriendly photoelectrical materials are promising alternatives to antibiotics. Here we design an interfacial Schottky junction of Bi 2 S 3 /Ti 3 C 2 T x resulting from the contact potential difference between Ti 3 C 2 T x and Bi 2 S 3 . The different work functions induce the formation of a local electrophilic/nucleophilic region. The self-driven charge transfer across the interface increases the local electron density on Ti 3 C 2 T x . The formed Schottky barrier inhibits the backflow of electrons and boosts the charge transfer and separation. The photocatalytic activity of Bi 2 S 3 /Ti 3 C 2 T x intensively improved the amount of reactive oxygen species under 808 nm near-infrared radiation. They kill 99.86% of Staphylococcus aureus and 99.92% of Escherichia coli with the assistance of hyperthermia within 10 min. We propose the theory of interfacial engineering based on work function and accordingly design the ecofriendly photoresponsive Schottky junction using two kinds of components with different work functions to effectively eradicate bacterial infection.

Ce and Er Co-doped TiO2 for Rapid Bacteria- Killing Using Visible Light.

Abstract:Bacterial infection and related diseases are threatening the health of human beings. Photocatalytic disinfection as a simple and low-cost disinfection strategy is attracting more and more attention. In this work, TiO2 nanoparticles (NPs) were modified by co-doping of Ce and Er using the sol–gel method, which endowed TiO2 NPs with enhanced visible light photocatalytic performance but not pure ultraviolet photocatalytic properties compared the untreated TiO2. Our results disclosed that as the doping content of Er increased, the photocatalytic activity of modified TiO2 NPs initially increased and subsequently decreased. The same trend occurred for Ce doping. When the doping dose of Er and Ce is 0.5 mol% and 0.2 mol%, the 0.5Ce0.2Ti–O calcined at 800 °C presented the best antibacterial properties, with the antibacterial efficiency of 91.23% and 92.8% for Staphylococcus aureus and Escherichia coli, respectively. The existence of Er ions is thought to successfully turn the near-infrared radiation into visible region, which is easier to be absorbed by TiO2 NPs. Meanwhile, the addition of Ce ions can effectively extend spectral response range and inhibit the recombination of electrons and holes, enhancing the photocatalytic disinfection activity of co-doped TiO2.

Abstract:When illuminated by near-UV light, titanium dioxide (TiO2) exhibits excellent bactericidal activity. However, there exist some different mechanisms for cell killing via photocatalysis. In the present study, the photocatalytically bactericidal mechanism of TiO2 thin films was investigated by atomic force microscopy (AFM) in conjugation with some other techniques. The decomposition process of the cell wall and the cell membrane was directly observed by AFM for the first time. The resultant change in cell permeability was confirmed by potassium ion (K+) leakage. Quantum dots (QDs) were designed originally as a probe to examine the cell permeability for macromolecules. The corresponding bactericidal activity of TiO2 thin films was examined by cell viability assay. These results suggested that the cell death was caused by the decomposition of the cell wall and the cell membrane and the resultant leakage of intracellular molecules.

Abstract:Ti doped BiOI microspheres were successfully synthesized through a solvothermal method. The photocatalysts were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), high-resolution transmission electron microscopy (HRTEM), X-ray photoelectron spectroscopy (XPS) and UV-vis diffuse reflectance spectra (DRS) spectroscopy, respectively. The as-synthesized microspheres had 3D hierarchical structures, and the morphologies and visible-light-driven (VLD) disinfection performances were found to be determined by the amount of loaded Ti. The incorporation of Ti in the lattice of BiOI broadened the band gap of BiOI and enhanced the VLD disinfection activity. Ti doped BiOI microspheres with the optimal Ti content exhibited excellent antibacterial performances against both representative Gram-negative and Gram-positive strains, which completely inactivated 3.0×10(7)CFUmL(-1)E. coli in 24min and 3.0×10(6)CFU mL(-1)S. aureus in 45min, respectively. Active species including h(+), e(-), O2(-) and H2O2 were found to play important roles in disinfection system. Moreover, the damage of cell membrane and emission of cytoplasm directly led to the inactivation.
Mechanism Insight into Rapid Photodriven Sterilization Based on Silver Bismuth Sulfide Quantum Dots

Abstract:Microbial contamination and the prevalence of resistant bacteria is considered a worldwide public health problem. Therefore, recently, great efforts have been made to develop photoresponsive platforms for the simultaneous photodynamic antibacterial (PDA) and photothermal antibacterial (PTA) therapy processes as mediated by specific light. However, owing to the absorption mismatches of the photothermal agents and photodynamic photosensitizers, it has been discovered that many synergistic photoresponsive antibacterial platforms cannot be excited by a single-wavelength light. In this study, silver bismuth sulfide quantum dots (AgBiS2 QDs) identified from the literature as a near-infrared light (NIR) that triggers bifunctional materials with simultaneous photodynamic and photothermal effects for photoresponsive bacterial killing were used. Specifically, AgBiS2 QDs were successfully synthesized via a bottom-up approach, using polyethylenimine (PEI) as an assistant molecule. With PEI wrapping, the attachment between the negatively charged membrane surfaces of the bacterial cells and AgBiS2 QDs was enhanced via the electrostatic interactions. The photodriven antibacterial activity of AgBiS2 QDs was then investigated against both S. aureus and E. coli. The results revealed a significant reduction in bacterial survival. The killing effect was found to be independent of the AgBiS2 QDs, and redox potentials controlled the photogenerated electrons that thermodynamically favored the formation of multiple reactive oxygen species (ROS). A possible phototriggered antibacterial mechanism was then proposed in which the AgBiS2 QDs are anchored first to the bacterial surface and then induce breaking on its outer membrane by high local heat and ROS under single 808 nm NIR laser illumination to finally induce bacterial death.
Self-activating electron transfer antibacterial strategy: Co 3 O 4 /TiO 2 P-N heterojunction combined with photothermal therapy

Abstract:Implant-associated infections are significant impediment to successful surgical outcomes, often resulting from persistent bacterial contamination. It has been hypothesized that bacteria can transfer electrons to semiconductors with comparable potential to biological redox potential (BRP). Building on this concept, we developed an antibiotic-free bactericidal system, Co3O4/TiO2-Ti, capable of achieving real-time and sustainable bactericidal effects. Our study demonstrated that Co3O4/TiO2-Ti, possessing an appropriately set valence band, initiated charge transfer, ROS production, and membrane damage in adherent Staphylococcus aureus (S. aureus). Notably, in vivo experiments illustrated the remarkable antibacterial activity of Co3O4/TiO2-Ti, while promoting soft-tissue reconstruction and demonstrating excellent cytocompatibility. Transcriptomic analysis further revealed a down-regulation of aerobic respiration-associated genes and an up-regulation of ROS-associated genes in S. aureus in the presence of Co3O4/TiO2-Ti compared to Ti. Kyoto encyclopedia of genes and genomes (KEGG) analysis and gene set enrichment analysis (GSEA) identified alterations in respiratory metabolism, oxidative phosphorylation, and the synthesis of amid acid signaling pathway in S. aureus cultured on Co3O4/TiO2-Ti. Furthermore, when combined with near-infrared (NIR) irradiation and photothermal therapy (PTT), Co3O4/TiO2-Ti eliminated 95.71% of floating and adherent S. aureus in vitro. The findings suggest that this antibiotic-free strategy holds substantial promise in enhancing implant sterilization capabilities, thereby contributing to the prevention and treatment of bacterial infections through bandgap engineering for implants and NIR irradiation.

Abstract:Construction of multiple electronic transmission channels in a photocatalytic system is in favor of boosting the charge separation to realize a high photocatalytic antibacterial property. Herein, a novel hierarchical ternary photocatalyst consisting of TiO2/W18O49 Z-scheme heterojunction and Ti3C2Tx-MXene cocatalyst was synthesized via a simple two-step solvothermal method. Dual charge transfer routes were realized in this TiO2/W18O49/Ti3C2Tx-MXene composite. The electrons on TiO2 can transfer either through the Z-scheme mode to combine with the holes of W18O49, or through the Schottky-junction to Ti3C2Tx-MXene cocatalyst. The TiO2/W18O49/Ti3C2Tx-MXene composite maintained high photooxidation-reduction capacity and possessed fast charge separation behavior. As a result, the TiO2/W18O49/Ti3C2Tx-MXene heterojunction had a high photocatalytic bactericidal rate of 93.7% for Escherichia coli (E. coil) under light (λ ≥ 365 nm, 6 h), which was 3.3, 5.5, 3.1 and 2.9 times that of MXene (28.6%), TiO2 (17.0%), W18O49 (30.0%) and TiO2/MXene (32.8%), respectively. In addition, the ternary heterojunction illustrated good biocompatibility. This work provides a simple method of constructing multiple electronic transmission channels for high bactericidal performance and proves the effectiveness of photocatalytic materials in the biological field.

Abstract:Harmful bacterial flourish with the increase in environmental pollution and pose a great threat to human health. Thus, developing new and efficient antibacterial materials is imperative to reduce the pollution caused by traditional sterilization materials and improve sterilization efficiency. In this study, a new photocatalytic antibacterial material was developed to achieve an efficient antibacterial effect. Ti3C2Tx@CuS composites were synthesized by simple hydrothermal method, by which copper sulfide (CuS) nanoparticles were anchored on the surface of Ti3C2Tx to sharply improve the photocatalytic its antibacterial ability. Ti3C2Tx@CuS exhibits excellent antibacterial activity against Escherichia coli and Staphylococcus aureus with bactericidal rates of 99.6% and 99.1%, respectively. Photoluminescence spectroscopy (PL), decay time PL, photocurrent test, electrochemical impedance spectroscopy and finite element method showed that the formation of Ti3C2Tx@CuS heterojunction promoted the separation of electrons and holes, improved the electron transport efficiency, and elevated the generation of reactive oxygen species. Moreover, Ti3C2Tx@CuS has a stronger photothermal effect and causes more heat release than CuS to improve antibacterial performance. The Ti3C2Tx@CuS heterojunction has a broad application prospect in the disinfection and antibacterial fields.

Visible-light-induced bactericidal properties of a novel thiophene-based linear conjugated polymer/TiO2 heterojunction

Abstract:The low utilization of visible light and the fast recombination of photogenerated electron-hole pairs are the two intrinsic defects that have hindered the antibacterial applications of TiO2. The addition of organic photocatalytic agents to form heterojunctions with TiO2 powder can effectively address these problems. A novel linear conjugated polymer poly[(thiophene-ethylene-thiophene)-thiophene-3-carboxylic acid decyl ester] (PTCD) was successfully synthesized via Stille coupling polymerization. PTCD and TiO2 can form a heterojunction photocatalyst (PTCD/TiO2), and this product was characterized using NMR and XRD. The UV-vis spectra showed that the absorption edge of the PTCD/TiO2 heterojunction extends to approximately 700 nm, which indicates that the visible-light utilization is greatly improved. Staphylococcus aureus (S. aureus) was selected as the target organism to test the photocatalytic antimicrobial activity of the material. Photogenerated electrons can undergo directional transmission of the PTCD polymer to TiO2 on the PTCD/TiO2 heterojunction to realize excellent antibacterial properties. With an optimized PTCD loading ratio of 30%, PTCD/TiO2 showed an antibacterial rate that was 14.5 times greater than that of pure TiO2 in 4 h under visible-light irradiation.

Abstract:Photothermal therapy is an ideal non-invasive treatment for bacterial infections. However, if the photothermal agents are unable to target bacteria, it can also cause thermal damage to healthy tissue. This study describes the fabrication of a Ti3C2Tx MXene-based photothermal nanobactericide (denoted as MPP) which targets bacteria by modifying MXene nanosheet with polydopamine and bacterial recognition peptides CAEKA. The polydopamine layer blunts the sharp edges of MXene nanosheets, preventing their damage to normal tissue cells. Furthermore, as a constituent of peptidoglycan, CAEKA can recognize and penetrate the bacterial cell membrane based on similar compatibility. The obtained MPP exhibits superior antibacterial activity and high cytocompatibility compared to the pristine MXene nanosheets. In vivo studies showed that MPP colloidal solution under 808 nm NIR light can effectively treat a subcutaneous abscess caused by multi-drug resistant bacterial infection without adverse effects.

Abstract:The photocatalytic disinfection of pathogenic bacteria in water was investigated systematically with AgI/TiO2 under visible light (lambda > 420 nm) irradiation. The catalyst was found to be highly effective in killing Escherichia coli and Staphylococcus aureus. The adsorbed *OH and hVB+ on the surface of the catalyst were proposed to be the main active oxygen species by study of electron spin resonance and the effect of radical scavengers. The process of destruction of the cell wall and the cell membrane was verified by TEM, potassium ion leakage, lipid peroxidation, and FT-IR measurements. Some products from photocatalytic degradation of bacteria such as aldehydes, ketones, and carboxylic acids were identified by FT-IR spectroscopy. These results suggested that the photocatalytic degradation of the cell structure caused the cell death. The electrostatic force interaction of the bacteria-catalyst significantly affected the efficiency of disinfection on the basis of the E. coli inactivation under different conditions.

Abstract:Carbon–semiconductor hybrid quantum dots are classical carbon dots with core carbon nanoparticles doped with a selected nanoscale semiconductor. Specifically, on those with the nanoscale TiO2 doping, denoted as CTiO2-Dots, their synthesis and thorough characterization were reported previously. In this work, the CTiO2-Dots were evaluated for their visible light-activated antibacterial function, with the results showing the effective killing of not only Gram-positive but also the generally more resistant Gram-negative bacteria. The hybrid dots are clearly more potent antibacterial agents than their neat carbon dot counterparts. Mechanistically, the higher antibacterial performance of the CTiO2-Dots is attributed to their superior photoexcited state properties, which are reflected by the observed much brighter fluorescence emissions. Also considered and discussed is the possibility of additional contributions to the antibacterial activities due to the photosensitization of the nanoscale TiO2 by its doped core carbon nanoparticles.

A Self-Activating Electron Transfer Antibacterial Strategy: Co3O4/TiO2 P-N Heterojunctions Combined with Photothermal Therapy.

Abstract:Implant-associated infections are significant impediments to successful surgical outcomes, often resulting from persistent bacterial contamination. It has been hypothesized that bacteria can transfer electrons to semiconductors with comparable potential to the biological redox potential (BRP). Building on this concept, we developed an antibiotic-free bactericidal system, Co3O4/TiO2-Ti, capable of achieving real-time and sustainable bactericidal effects. Our study demonstrated that Co3O4/TiO2-Ti, possessing an appropriately set valence band, initiated charge transfer, reactive oxygen species (ROS) production, and membrane damage in adherent Staphylococcus aureus (S. aureus). Notably, in vivo experiments illustrated the remarkable antibacterial activity of Co3O4/TiO2-Ti, while promoting soft-tissue reconstruction and demonstrating excellent cytocompatibility. Transcriptomic analysis further revealed a down-regulation of aerobic respiration-associated genes and an up-regulation of ROS-associated genes in S. aureus in the presence of Co3O4/TiO2-Ti compared to Ti. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis and gene set enrichment analysis (GSEA) identified alterations in respiratory metabolism, oxidative phosphorylation, and the synthesis of amino acid in S. aureus cultured on Co3O4/TiO2-Ti. Furthermore, when combined with near-infrared (NIR) irradiation and photothermal therapy (PTT), Co3O4/TiO2-Ti eliminated 95.71% of floating and adherent S. aureus in vitro. The findings suggest that this antibiotic-free strategy holds substantial promise in enhancing implant sterilization capabilities, thereby contributing to the prevention and treatment of bacterial infections through bandgap engineering of implants and NIR irradiation.
Local Photothermal/Photodynamic Synergistic Therapy by Disrupting Bacterial Membrane to Accelerate Reactive Oxygen Species Permeation and Protein Leakage

Abstract:Bacterial infection is still a ticklish clinical challenge even though some advanced antibacterial materials and techniques have been put forward. This work reports that rapid and effective antibacterial performance is achieved by the synergistic local photothermal and photodynamic therapy (PTDT). Within 10 min of light irradiation, both Escherichia coli and Staphylococcus aureus are almost completely eliminated by the action of photothermy (52.1 °C) and limited reactive oxygen species (ROS), the corresponding bacterial killing efficiencies are 99.91 and 99.97%, respectively, which are far higher than single modal therapy, i.e., photothermal therapy or photodynamic therapy with antibacterial efficacy of 50 or 70%, respectively. The mechanism is that bacterial membrane permeation is increased by PTDT because photothermy shows more severe impact only on E. coli by destroying the outmost bacterial panniculus, whereas the inner panniculus of the two kinds of bacteria is more sensitive to ROS. Hence, ROS penetrates the bacterial membrane more easily, and meanwhile, the proteins in the bacteria are severely lost after the bacterial membrane disruption, which leads to bacterial death. In vivo results reveal that rapid and effective sterilization is an important process to accelerate wound healing, and the traumas on the rats' backbones heal well within 12 days by PTDT. Furthermore, the PTDT is friendly to major organs of rats during the therapeutic process. Therefore, the synergistic therapy system can be a safe therapeutic system for clinical sterilization with great potential. More importantly, the antibacterial mechanism presented in this work has great guiding significance for the design of other advanced antibacterial systems and techniques.
