Abstract:The need for an excellent antibacterial material that is sufficiently powerful to never develop bacterial resistance is urgent. In this study, a series of novel polyacrylonitrile-based fibres with chelated Ag ions (referred to as Ag-SH-PANF) were prepared by a two-step chemical modification process: grafting and chelating. The properties of the as-prepared Ag-SH-PANF were characterized by Fourier transform infrared (FT-IR) spectroscopy, thermogravimetric analysis (TGA), X-ray diffraction (XRD), scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). The antibacterial activities of Ag-SH-PANF were examined against pathogenic bacteria, and an antibacterial mechanism was explicated based on the release of Ag ions from the fibres' surfaces. The results showed that, although chelation occurred between the Ag ions and the grafted amino, sulfhydryl and disulfide groups, Ag-SH-PANF retained its fine microstructure and thermal stability. Moreover, Ag-SH-PANF displayed excellent antibacterial ability against pathogenic bacteria as well as good washing durability. In terms of the antibacterial mechanism, Ag ions are the main bactericidal agents in the role of catalysts and are not consumed in the antibacterial process. Nonetheless, a relatively higher concentration of Ag ions can accelerate the bactericidal process.

Abstract:The abuse of antibiotics has led to bacterial drug resistance, which hinders traditional treatments for infection. It is a vitally important task to develop new methods for combating bacterial infection. In this work, we design a multifunctional peptide (BrEK) that consists of three functional building blocks: an antimicrobial sequence (BF2b), a gelatinase B responsive linker, and a zwitterionic antifouling block. The BrEK peptide is conjugated onto gold nanorods (AuNRs) via a cysteine on one end of the peptide sequence, with excellent photothermal conversion ability under near-infrared (NIR) light. The obtained AuNR@BrEK are tested in vitro and in vivo for antibiotic capacity against methicillin-resistant Staphylococcus aureus (MRSA). The multifunctional peptide coating provides the nanocomposite excellent biocompatibility, relative long circulation in blood, infection microenvironment responsive targeting ability, and unique antibacterial performance. Ultimately, we aim to eliminate drug resistance bacteria by (1) integrating infection microenvironment responsiveness for efficient targeting, (2) capitalizing on antibacterial capacity of BF2b peptides, and (3) developing photothermal sterilization. Transmittance electron microscopy, dynamic light scatter, and zeta potential measurements are introduced to characterize the synthesis of AuNR@BrEK. The evaluation of particle stability is conducted by Fouriertransform infrared spectroscopy and UV?vis spectroscopy. Photothermal conversion capacity of AuNR@BrEK is examined via photothermal effect measurement. Cell viability and hemolysis assay are conducted to further assess the biocompatibility of AuNR@BrEK (200 mu g/mL). Lastly, results from the in vitro and in vivo experiments indicate the outstanding antibacterial activity of AuNR@BrEK (10 mg/kg). Such multifunctional gold nanorods represent an important step forward in the goal to combat bacterial resistance.

Abstract:Multivalent peptide nanofibers have attracted intense attention as promising platforms, but the fabrication of those nanofibers is mainly dependent on the spontaneous assembly of β-sheet peptides. Herein we report an alternative approach to the creation of nanofibers: the polyoxometalate-driven self-assembly of short peptides. The resultant nanofibers with concentrated positive charges are excellent multivalent ligands for binding with bacterial cells and thus lead to a salient improvement in bioactivity.

Abstract:The pervasive employment of antibiotics has engendered the advent of drug-resistant bacteria, imperiling the well-being and health of both humans and animals. Infections precipitated by such multi-resistant bacteria, especially those induced by methicillin-resistant Staphylococcus aureus (MRSA), pervade hospital settings, constituting a grave menace to patient vitality. Antimicrobial peptides (AMPs) have garnered considerable attention as a potent countermeasure against multidrug resistant bacteria. In preceding research endeavors, an insect-derived antimicrobial peptide is identified that, while possessing antimicrobial attributes, manifested suboptimal efficacy against drug-resistant Gram-positive bacteria. To ameliorate this issue, this work enhances the antimicrobial capabilities of the initial beta-hairpin AMPs by substituting the structural sequence of the original AMPs with variant lengths of hydrophobic amino acid-hydrophilic amino acid repeat units. Throughout this endeavor, this work has identified a number of peptides that possess highly effective antibacterial characteristics against a wide range of bacteria. Additionally, some of these peptides have the ability to self-assemble into nanofibers, which then build networks in a distinctive manner to capture bacteria. Consequently, they represent prospective antibiotic alternatives for addressing wound infections engendered by drug-resistant bacteria. Several peptides have been identified by comprehensive examination of antimicrobial peptides (AMPs) derived from insects. These peptides exhibit both a wide range of resistance against drug-resistant bacteria and the ability to spontaneously form nanofibrous networks for trapping bacteria. Peptide‐AIE Nanofibers Functionalized Sutures with Antimicrobial Activity and Subcutaneous Traceability

Abstract:As one of the most widely used medical device, sutures face challenges related to surgical site infections (SSIs) and lack of subcutaneous traceability. In the present study, we have developed a facile and effective approach using peptide‐AIE nanofibers (NFs‐K18) to create fluorescent‐traceable antimicrobial sutures, which have been applied to four commercially available sutures. The functionalized sutures of PGAS‐NFs‐K18 and PGLAS‐NFs‐K18 exhibited fluorescence with excellent penetration from 4 mm chicken breasts. They also demonstrated remarkable stability after 24 h of white light illumination and threading through chicken breasts 10 times. These sutures efficiently generated ROS, resulting in a significant suppression of four clinical bacteria, with the highest antimicrobial rate of nearly 100%. Moreover, the sutures exhibited favorable hemocompatibility and biocompatibility. In vivo experiments demonstrated that the optimized PGLAS‐NFs‐K18 suture displayed potent antimicrobial activity against MRSA, effectively inhibiting inflammation and promoting tissue healing in both skin wound and abdominal wall wound models, outperforming the commercially available Coated VICRYL® Plus Antibacterial suture. Importantly, PGLAS‐NFs‐K18 exhibited sensitive subcutaneous traceability, allowing for accurate in situ monitoring of its degradation. Abstract:Objective: In order to investigate the potential biomedical applications of silver nanoparticle (Ag NP)-immobilized electrospun nanofibers with different surface functionalities. Methods: Silver nanoparticles were immobilized within water-stable electrospun polyethylenimine (PEI)/polyvinyl alcohol (PVA) nanofibers by an in-situ reduction method after complexing Ag + . ions with the free PEI amine groups. The obtained Ag NP-incorporated PEI/PVA nanofibers were then hydroxylated, carboxylated, and acetylated to generate different surface functionalities. Different techniques were employed to characterize the Ag NP-containing nanofibers with different surface functionalities. Results: In vitro antibacterial activity tests show that Ag NP-containing nanofibrous mats have high antibacterial activity and are capable of inhibiting the growth of both S. aureus and E. coli bacteria. Cell viability assay data show that the Ag NP-containing nanofibers are cytocompatible, and those treated by hydroxylation and acetylation display better cytocompatibility than those treated by carboxylation and the pristine non-modified fibers to promote cell adhesion and proliferation. Conclusion: Therefore, the hydroxylated or acetylated Ag NP-containing PEI/PVA nanofibers have a great potential for wound dressing, biological protection and tissue engineering applications.

Rapid Antibacterial Effects of Silk Fabric Constructed through Enzymatic Grafting of Modified PEI and AgNP Deposition

Abstract:Enzymatic antibacterial finishing is an eco-friendly alternative to develop functional silk-based materials. However, the low accessibility of tyrosine residues distributed in fibroin chains restricts the laccase-mediated functionalization of silk fibers (SF). To address this issue, a highly reactive p-hydroxyphenylacetic acid-modified polyethyleneimine (mPEI) was enzymatically grafted onto fibroin using laccase, aiming at constructing an antibacterial matrix of mPEI on the fiber surface. Subsequently, in situ deposition of silver nanoparticles (i.e., AgNPs) into the newly built mPEI network was performed to form a rapid antibacterial layer. The results indicated that laccase efficiently catalyzes the mPEI coupling, the zeta potential of SF-g-mPEI increases from −32 to 21.70 mV, and the silver content reaches 1.81% after AgNP embedment. Based on the combined two-step treatments, the obtained silk fabric exhibited excellent antibacterial abilities against two bacteria, including Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). The antibacterial rates of both bacteria reached 99.9% within 30 min of contact, remaining over 99.9% within 18 h of contact even after washing 10 times. Abstract:Facing the global health crisis caused by drug-resistant bacteria, antimicrobial peptides and their analogues offer exciting solutions to this widespread problem. Without additionally introducing a fluorescent probe, novel nanoengineered peptide-grafted hyperbranched polymers (NPGHPs) are constructed for their combined outstanding antimicrobial activity and sensitive bacterial detection in real time. Hyperbranched polyamide amine (H-PAMAM) that exhibits aggregation-induced emission (AIE) effects is synthesized. Then, NPGHPs are prepared by ring-opening polymerization of α-amino acid N-carboxyanhydrides on the periphery of the H-PAMAM. The NPGHPs exhibit high-efficiency antibacterial properties against a wide spectrum of bacteria, especially against Gram-negative bacteria. On the basis of the AIE effect of NPGHPs, the interaction between NPGHPs and Escherichia coli is explored and the fluorescence intensity of NPGHPs is dependent on the number of E. coli present. Thus, a method for monitoring E. coli concentration is developed, and the detection limit is 1 × 104 CFU mL-1. Furthermore, NPGHPs are used as fluorescent probes to visualize antibacterial process via lighting-up bacteria. NPGHPs can penetrate the membrane of bacteria and cause cell rupture and apoptosis. In addition, the excellent selectivity of NPGHPs toward bacteria over mammalian cells makes them bright prospects for clinical applications.

Abstract:Supramolecular gels have promising applications in the synthesis of inorganic–organic composite nanomaterials, and gels with excellent mechanical strength can be used to synthesize scaffolds for metal nanoparticles. However, external reducing agents usually need to be added in this process. In this work, a novel peptide gel containing reduced sulfhydryl groups in the side chains was constructed and silver nanoparticles formed in situ in the gel fibers, while eco-friendly and natural light could accelerate the formation of AgNPs without addition of any reducing agent. The coordination of Ag+ led to improvement in the mechanical properties, self-recovery ability and gel formation time of the FFCWR. The AgNPs/FFCWR exhibited excellent catalytic ability in degradation of the methylene blue and showed the antibacterial properties to both gram-negative (E. coli) and gram-positive (S. aureus) bacteria. The composite system may have promising applications in the field of biomedical and dye degradation.

Abstract:Silk fibroin (SF) has excellent biocompatibility and is one of the most commonly used polymer materials. However, SF fibers have serious drawbacks as antibacterial materials due to their lack of stability and bacterial resistance. Therefore, it is of paramount significance to enhance the stability and bolster the bacterial resistance of SF fibers. In this study, SF fibers were fabricated and loaded with Ag nanoparticles (AgNPs) to improve the antimicrobial properties of the fibers. The impact of reduction conditions on the size of AgNPs was also investigated. In an antibacterial test, the fibers that were prepared exhibited over 98% bacterial resistance against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Therefore, as an efficient antibacterial material, these fibers are expected to become a candidate material in medical and textile fields. This study offers a novel approach for the utilization of SF fibers in the realm of antibacterial applications.

Abstract:Antimicrobial peptides (AMPs) are a new class of promising antibacterial agents. We prepared electrospinning chitosan (CS)-polyethylene oxide (PEO) nanofiber membranes containing different concentrations of an antibacterial peptide NP10. The average diameter of nanofibers increased with the total concentration of NP10. The FTIR shows that all the peaks of CS-PEO nanofiber membranes with different concentrations of NP10 were almost the same as those of pure CS-PEO nanofiber membranes, and only the peak intensity changes. Adding NP10 can improve the thermal stability of CS-PEO nanofiber membranes. In the in vitro release experiment, NP10 was released from the CS-PEO-0.5%NP10 nanofiber membrane in a burst first and then slowly and continuously. Simultaneously, the CS-PEO-NP10 nanofiber membrane had good antibacterial activity against Escherichia coli and Staphylococcus aureus and good biocompatibility. In animal wound healing experiments, CS-PEO-0.5%NP10 nanofiber membrane had advantages over gauze and CS-PEO nanofiber membrane in wound healing. These properties may provide a choice for the clinical application of AMPs and treatment of wound infections.

Abstract:Protein and peptide can assemble into functional amyloid fibrils with distinct architectures which fulfill a variety of different biological functions in living organism, then can be degraded. In this study, inspired from living cell, by incorporating an amyloidogenic segment and an enzyme-cleavage segment together, we design a peptide (enzyme-cleavage amyloid peptides, termed as EAP)-based functional fibril which can be specifically degraded by gelatinase. To further gain molecular insight of EAP fibril assembly and degradation, we determined the atomic structure of the EAP fibril by using cryo-EM. Notably, the amyloidogenic segment of EAP adopts β-strand conformation and mediates EAP fibril formation mainly via steric zipper-like interaction. While, the enzyme-cleavage segment partially involves in self-assembling, but also exhibits high structural flexibility in the fibril structure with accessibility to gelatinase binding and degradation. Moreover, we applied the EAP fibril as a tunable scaffold for developing degradable self-assembled antimicrobial fibrils (SANs) by integrating Melittin and EAP together. The SANs exhibit superior antimicrobial activity for killing bacteria, and significantly improved the stability and biocompatibility of Melittin. Meanwhile SANs are automatically eliminated by gelatinase secreted from the targeted bacteria. Our work provides a new strategy for rational design of functional fibrils with a feed-back regulation loop for optimizing the biocompatibility and biosafety of the designed fibrils, which may shed light on further development of smart peptide-based biomaterials for biomedical applications.

Abstract:Bacterial infections have been a serious threat to mankind throughout history. Natural antimicrobial peptides (AMPs) and their membrane disruption mechanism have generated immense interest in the design and development of synthetic mimetics that could overcome the intrinsic drawbacks of AMPs, such as their susceptibility to proteolytic degradation and low bioavailability. Herein, by exploiting the self-assembly and pore-forming capabilities of sequence-defined peptoids, we discovered a family of low-molecular weight peptoid antibiotics that exhibit excellent broad-spectrum activity and high selectivity toward a panel of clinically significant Gram-positive and Gram-negative bacterial strains, including vancomycin-resistant Enterococcus faecalis (VREF), methicillin-resistant Staphylococcus aureus (MRSA), methicillin-resistant Staphylococcus epidermidis (MRSE), Escherichia coli, Pseudomonas aeruginosa, and Klebsiella pneumoniae. Tuning the peptoid side chain chemistry and structure enabled us to tune the efficacy of antimicrobial activity. Mechanistic studies using transmission electron microscopy (TEM), bacterial membrane depolarization and lysis, and time-kill kinetics assays along with molecular dynamics simulations reveal that these peptoids kill both Gram-positive and Gram-negative bacteria through a membrane disruption mechanism. These robust and biocompatible peptoid-based antibiotics can provide a valuable tool for combating emerging drug resistance.

Abstract:Over the past few decades, the overuse of antibiotics has led to the emergence of resistant bacteria and environmental issues. Both silver nanoparticles (AgNPs) and antimicrobial peptides (AMPs) hold potential to replace antibiotics. Combining both AMPs and AgNPs into a composite material may create novel properties such as enhanced antibacterial activity, lower cytotoxicity and favorable stability in aqueous solution. We designed a 13 amino acid peptide (in short, P-13) with two functional regions: one is for antibacterial activity, and the other for reducing and stabilizing AgNPs with containing cysteine (C) residues in its C-terminus. With a single step reaction, we have successfully synthesized P-13 protected AgNPs (P-13@AgNPs) with a hydrodynamic diameter of about 11 nm. In the preliminary antibacterial activity assay, the minimum inhibitory concentrations (MICs) of P-13@AgNPs were up to 7.8 mu g/mL against E. colt, S. aureus and B. pumilus, and 15.6 mu g/mL against P. aeruginosa. Moreover, Flow cytometry analysis of E. coll., S. aureus, P. aeruginosa and B. pumilus show that the mortality of the strains reached 96 %, 96 %, 91 % and 90 %, respectively. The cytotoxicity of AgNPs was reduced dramatically after protected by P-13, and P-13 was favorable for the stability of the AgNPs solution. We believe this work could set up an example to make the best use of the individual material's properties to produce novel nanocomposites with better antibacterial activity.

Abstract:Electrospinning is an economical and relatively simple method to produce continuous and uniform nanofibers from almost any synthetic and many natural polymers. Because of the high specific surface area, tunable pore size, and flexibility, the nanofibrous membranes are finding an increasingly wide range of applications. Some particular attention has been devoted to antibacterial nanofibers for applications such as wound dressings. A variety of biocides, e. g., antibiotics, quaternary ammonium salts, triclosan, biguanides, (silver, titanium dioxide, and zinc oxide) nanoparticles and chitosan have been incorporated by various techniques into nanofibers that exhibit strong antibacterial activity in standard assays. However, the small diameters of the nanofibers also mean that the incorporated biocides are often burst released once the materials are submerged in an aqueous solution. Nevertheless, several strategies, such as core-sheath structure of the nanofiber, covalent bonding of the biocide on the fiber surface and adsorption of the biocide in nanostructures, can be utilized to sustain the release over several days. This review summarizes recent development in the fabrication of antibacterial nanofibers, the release profiles of the biocides and their applications in in vivo systems. Optimization of Antibacterial Activity and Biosafety through Ultrashort Peptide/Cyclodextrin Inclusion Complexes

Abstract:Engineered ultrashort peptides, serving as an alternative to natural antimicrobial peptides, offer benefits of simple and modifiable structures, as well as ease of assembly. Achieving excellent antibacterial performance and favorable biocompatibility through structural optimization remains essential for further applications. In this study, we assembled lipoic acid (LA)–modified tripeptide RWR (LA–RWR) with β–cyclodextrin (β–CD) to form nano–inclusion complexes. The free cationic tripeptide region in the nano–inclusion complex provided high antibacterial activity, while β–CD enhanced its biocompatibility. Compared with peptides (LA–RWR, LA–RWR–phenethylamine) alone, inclusion complexes exhibited lower minimum inhibitory concentrations/minimum bactericidal concentrations (MICs/MBCs) against typical Gram–negative/Gram–positive bacteria and fungi, along with improved planktonic killing kinetics and antibiofilm efficiency. The antibacterial mechanism of the nano–inclusion complexes was confirmed through depolarization experiments, outer membrane permeability experiments, and confocal laser scanning microscopy observations. Furthermore, biological evaluations indicated that the hemolysis rate of the inclusion complexes decreased to half or even lower at high concentrations, and cell viability was superior to that of the non–included peptides. Preliminary in vivo studies suggested that the inclusion complexes, optimized for antibacterial activity and biosafety, could be used as promising antibacterial agents for potential applications.

Abstract:Materials with a hierarchical structure often demonstrate superior properties with combined and even synergistic effects of multiple functions. Herein, we report the design of a new class of material with a multicompartment nanofibrous structure as a promising candidate for antibacterial wound dressing and functional textile applications. The design consists in first synthesizing nanocapsules loaded with functional payloads and subsequently embedding the nanocapsules into polymer nanofibers by using the colloid-electrospinning technique. The nanocontainer-in-nanofiber structure allows for a selective and separate loading of different functional agents with different polarities, and it offers a flexible combination of the properties of nanocontainers and nanofibers. An example of the potential for these multicompartment materials is demonstrated here, in which the synergistic antibacterial effect against E. coli K-12 and B. Subtilis combined with anti-UV property is shown.

Abstract:Drug‐resistant bacteria and biofilm‐associated infections are prominent problems in the field of antibacterial medicine, seriously affecting human and animal health. Despite the great potential of nanomaterials in the antibacterial field, overcoming the paradox of size and charge, efficient penetration, and retention within biofilms remain a formidable challenge. Here, we designed self‐assembling chimeric peptide nano‐assemblies composed of multiple functional fragments for the treatment of drug‐resistant bacteria and biofilm‐associated infections. Notably, the chimeric peptide self‐assembling into nanofibers at pH 7.4, transformable into nanoparticles in the acidic biofilm‐infected microenvironment at pH 5.0, thus achieving a size reduction and charge increase, improving the penetration into the bacterial biofilms, and killing drug‐resistant bacteria by a mechanism dominated by membrane cleavage. In vivo mouse and piglet infection models confirm the ability of chimeric peptide nano‐assemblies to reduce bacterial load within biofilms. Collectively, this research on pathological environment‐driven nano‐structural transformations may provide a theoretical basis for designing high‐performance antibacterial nanomaterials and advance the application of peptide‐based nanomaterials in medicine and animal husbandry. A Bioinspired Atomically Thin Nanodot Supported Single‐Atom Nanozyme for Antibacterial Textile Coating

Abstract:Atomic copper on atomically thin C3N4 nanodots with high peroxidase‐like activity is first synthesized by a "self‐tailoring" strategy for antimicrobial textile coating. Due to the size effect of the nanodots optimizing the coordination structure, the unique local coordination structure of Cu–N3 moiety as the active site is conducive to activating H2O2 into •OH, thus endowing them with superior antibacterial performance. Surface antibacterial coatings with outstanding antibacterial efficiency have attracted increasing attention in medical protective clothing and cotton surgical clothing. Although nanozymes, as a new generation of antibiotics, are used to combat bacteria, their catalytic performance remains far from satisfactory as alternatives to natural enzymes. Single‐atom nanodots provide a solution to the low catalytic activity bottleneck of nanozymes. Here, atomically thin C3N4 nanodots supported single Cu atom nanozymes (Cu‐CNNDs) are developed by a self‐tailoring approach, which exhibits catalytic efficiency of 8.09 × 105 M−1 s−1, similar to that of natural enzyme. Experimental and theoretical calculations show that excellent peroxidase‐like activity stems from the size effect of carrier optimizing the coordination structure, leading to full exposure of Cu‐N3 active site, which improves the ability of H2O2 to generate hydroxyl radicals (•OH). Notably, Cu‐CNNDs exhibit over 99% superior antibacterial efficacy and are successfully grafted onto cotton fabrics. Thus, Cu‐CNNDs blaze an avenue for exquisite biomimetic nanozyme design and have great potential applications in antibacterial textiles.
