Abstract:A simple and highly efficient one-pot method for the construction of α-hydroxyketone phosphates from terminal alkynes and H-phosphine oxides has been developed in the presence of PhI（OAc） 2 and H 2 O.The present protocol provides an attractive approach to α-hydroxyketone phosphates in good to high yields,with the advantages of operation simplicity,the use of commercially available materials,broad substrate scope,high atom efficiency and good tolerance to scale-up synthesis.

Abstract:On account of the broad utilities of organophosphorus compounds, the development of highly efficient and concise phosphination methods is significantly important and urgent. Herein, we disclose a novel method for the synthesis of phosphorylated heterocycles: versatile intermediate propargylamines serving as a new type of radical acceptors incorporated in P-radicals via a photocatalytic strategy. This reaction proceeds through a cascade phosphinoylation/ cyclization/oxidation/aromatization pathway using readily available starting materials under mild conditions of light with excellent atom economy, catalyzed by AgOAc or fac-Ir(ppy)(3). One of the phosphorylated quinolines was selected, as an example, as an electron-transporting material for fabricating phosphorescence organic light-emitting diodes displaying excellent electroluminescence performances with a maximum external quantum efficiency of 21.9% with negligible efficiency roll-off ratios.

Abstract:We report a straightforward, catalyst-free method for the cathodic benzylic C–H hydroxyalkylation/arylation of N-(alkyloxy)phthalimides with ketones or cyanopyridines. The reaction is driven by an electrochemically induced 1,2-hydrogen atom transfer involving alkoxy radicals generated by O–N bond cleavage. This protocol provides an efficient approach for the synthesis of a wide range of valuable unsymmetrical 1,2-diols and diarylmethanols.

Abstract:A new method has been established for formation of ethers from aldehydes and alcohols – a net reductive etherification. Reactions of these entities with phosphines in the presence of acid enable formation of α-(alkoxyalkyl)phosphonium salts, which, upon hydrolysis, result in formation of ether products in isolated yields of 63–92%. Formation and hydrolysis of the α-(alkoxyalkyl)phosphonium salts were done in an efficient telescoped two-step, one-pot process. Formation of the key phosphonium salt intermediates was found to occur in preference to acetal formation, and is proposed to involve interception of oxocarbenium ions formed by reaction of the aldehyde, alcohol and acid by phosphine. This method represents the first instance in which net reductive etherifications have been achieved without the requirement for use of hydrides or hydrogen as reductants, and exhibits excellent functional group tolerance, thus enabling facile hydride-free synthesis of ethers. These are amongst the most important functional groups in organic synthesis and the most frequently encountered functional groups in the structures of pharmaceutical agents.

Abstract:We herein report a direct electrochemical dehydrogenative CH phosphonylation of thiazoles derivatives with H2 evolution. Employing electricity as the green and sole oxidant, cheap metal as electrode, the anodic oxidation together with cathodic hydrogen evolution process provides a green and efficient strategy for CH phosphonylation. A diverse range of phosphorus products were constructed under external metal and oxidant-free conditions at ambient temperature, featuring atom economy, simple operation and wide reaction scope.

Abstract:Various alpha,beta-unsaturated (E)-alkenylphosphonamidates were prepared in excellent stereo- and regioselectivities via the electrophilic hydrophosphonylation of cyclic and acyclic aldimines with alkylphosphonochloridates in the presence of base. For unsaturated aldimines, only their respective imine bonds were hydrophosphonylated regiospecifically. The reaction provided a new strategy for the synthesis of (E)-alk-1-enylphosphonamidates via a tandem phosphorylation, deprotonation, and intramolecular 1,5-hydride transfer sequence process from readily available aldimine and alkylphosphonochloridate starting materials. The strategy established the first one-pot electrophilic rather than nucleophilic hydrophosphonylation of aldimines without external hydride donors.

Abstract:Catalytic difunctionalization with the direct activation of (O)P-H bonds has been recently established as a potentially robust platform to generate valuable organophosphorus compounds. In terms of 1,3-enynes, despite of the various catalytic methods developed for hydrophosphorylation, the radical-mediated hetero-functionalization of two different atoms has been less explored. In this study, we disclosed an electrochemically induced hydroxyphosphorylation of 1,3-enynes for the construction of phosphinyl-substituted propargyl alcohols. The system involves the direct activation of both arylphosphine oxides and oxygen in ambient air with no external metal or additive needed. The use of electrochemistry ensures the regioselective, atom-economic and eco-friendly for the difunctionalization process. This strategy highlights the advantages of mild reaction conditions, readily available starting materials and broad substrate scope, showing its practical synthetic value in organic synthesis.

Abstract:An unprecedented electrochemical selenium-π-acid promoted hydration of internal alkynes bearing a phosphonate auxiliary was described. Thus, valuable (hetero)aryl and alkyl ketones could be accessed under mild, metal- and external oxidant-free conditions. This protocol features high atom-economy, good chemo- and regio-selectivity, excellent functional group tolerance and easily transformable products. Control experiments demonstrate that phosphonate assistance is essential for this transformation.

Abstract:It was found that nucleoside H-phosphonothioate, H-phosphonodithioate, and H-phosphonoselenoate monoesters in the presence of iodine underwent rapid oxidative esterification with various hydroxylic components to produce in "one pot" reactions the corresponding nucleoside phosphorothioate, nucleoside phosphorodithioate and nucleoside phosphoroselenoate diesters in good yields. The transformation represents a novel alternative method for the synthesis of internucleotide phosphate bridge, bearing sulfur or selenium atoms. Mechanistic studies have revealed metaphosphate analogues as the probable intermediates, involved in the reactions.

Abstract:Addition of P-H species to carbonyl groups, namely the Pudovik reaction, normally delivers hydroxyl phosphorus compounds, along with phosphate byproducts in some cases. A few controllable systems starting from phosphites were set up to mainly provide the phosphates. Herein, we present a highly selective protocol starting from phosphonate precursors leading to phosphinate derivatives. Enantioenriched phosphinates were successfully achieved from chiral phosphine oxide precursors. Experimental and theoretical investigations were conducted to understand the mechanistic details.
P-Chloride-Free Synthesis of Phosphoric Esters: Microwave-Assisted Esterification of Alkyl- and Dialkyl Phosphoric Ester-acids obtained from phosphorus pentoxide

Abstract:It is a reasonable endeavour to replace P-chloride starting materials (e.g. POCl3) by greener and cheaper reagents. Our purpose was to start from phosphorus pentoxide, i.e. to utilize its reaction with alcohols in the preparation of (HO)2P(O)(OR) and HOP(O)(OR)2, and to convert the mixtures of the corresponding monoester and diester so obtained to the target trialkyl esters. Separate experiments showed that the monobutylphosphate undergo microwave (MW)-assisted esterification with butanol in the presence of [bmim][BF4] catalyst at 200 °C to afford dibutylphosphate in a selective manner (~95%) that, in turn, may be converted to tributylphosphate by alkylation under MW irradiation. In this way, the mixtures of (HO)2P(O)(OR) and HOP(O)(OR)2 obtained by the practical reaction of phosphorus pentoxide and alcohol (ROH) could also be converted in two additional steps to the corresponding trialkyl esters. The three-step synthesis of trialkylphosphates starting from phosphorus pentoxide was also transformed in a one-pot (step 1: preparation of the monoester diester mixture, step 2: diesterification) and telescoping (step 3: triesterification) variation avoiding the isolation and purification of the intermediates, and affording the triesters in 86-93% yields. The three- and two-step P-chloride-free methods developed are “green” and of more general value.

Abstract:Here an unprecedented formal deformylative phosphonylation is disclosed. Under the catalysis of 1 mol % 1,2,3,5-tetrakis(diphenylamino)-4,6-dicyanobenzene (4DPAIPN) and the presence of 200 mol % triethylamine as additive, 4-alkyl-1,4-dihydropyridines (DHPs), derived from aldehydes, underwent phosphonylation smoothly with 9-fluorenyl o-phenylene phosphite, allowing facile synthesis of alkylphosphonates. This strategy is applicable to primary, secondary, and even tertiary DHPs, and exhibits a broad substrate scope and excellent functional group tolerance, thereby enabling the late-stage phosphonylation of a series of naturally-ocurring bioactive and biorelevant molecules.

Abstract:A highly efficient and environmentally-friendly base-mediated transition metal-free direct thiophlic catalytic approach has been reported for the synthesis of S-benzhydryl-phosphorothioates by reacting phosphite nucleophiles with diarylmethanethione. A wide variety of thioketones were coupled with different phosphite derivatives to provide the corresponding phosphorothioates in good to excellent yields. The control experiments, and density functional theory (DFT) calculations relies on the regio-selective thiophilic addition of phosphite nucleophile via umpolung protocols.

Abstract:Two Mo(0) phosphenium complexes containing ancillary secondary phosphine ligands have been investigated with respect to their ability to participate in electrophilic addition at unsaturated substrates and subsequent P‐H hydride transfer to "quench" the resulting carbocations. These studies provide stoichiometric "proof of concept" for a proposed new metal‐catalyzed electrophilic hydrophosphination mechanism. The more strongly Lewis acidic phosphenium complex, [Mo(CO)4(PR2H)(PR2)]+ (R = Ph, Tolp), cleanly hydrophosphinates 1,1‐diphenylethylene, benzophenone, and ethylene, while other substrates react rapidly to give products resulting from competing electrophilic processes. A less Lewis acidic complex, [Mo(CO)3(PR2H)2(PR2)]+, generally reacts more slowly but participates in clean hydrophosphination of a wider range of unsaturated substrates, including styrene, indene, 1‐hexene, and cyclohexanone, in addition to 1,1‐diphenylethylene, benzophenone, and ethylene. Mechanistic studies are described, including stoichiometric control reactions and computational and kinetic analyses, which probe whether the observed P‐H addition actually does occur by the proposed electrophilic mechanism, and whether hydridic P‐H transfer in this system is intra‐ or intermolecular. Preliminary reactivity studies indicate challenges that must be addressed to exploit these promising results in catalysis.

Abstract:An efficient, LiN(SiMe3)2‐mediated phosphorylation of terminal alkynes with chlorodiphenylphosphine oxide has been developed to synthesize alkynylphosphine oxides. This transition‐metal‐free protocol was featured with simple operation, mild reaction conditions, and broad substrate scope. The tolerance of halogen and ester substituents displayed the advantages of this new method over the traditional ones. In addition, the application potential was demonstrated by the gram‐scale reaction. When using diisopropyl chlorophosphate and chlorodiphenylphosphine, the corresponding alkynylphosphate and alkynylphosphine could also been produced.

Abstract:Direct α-phosphonylation of an unprotected secondary amine in a single step is of practical importance to amino phophophates. However, this protocol is limited due to the high redox barrier of unprotected amine. In this paper, we report C-H phosphonylation of an unprotected secondary amine via an electrochemical approach in the presence of catalytic carboxylate salt. This metal-free and exogenous oxidant-free method furnishes diverse target molecules with satisfactory yield under mild reaction conditions. Successful application of the protocol in a gram-scale experiment demonstrates the potential utility for further functionalization.
