Abstract:A novel method was developed to separate the tea polyphenols epigallocatechin gallate (EGCG) and epicatechin gallate (ECG) by two-step chromatography. In the first-step, four different macroporous absorption resins were compared by dynamic desorption, of which only AB-8 resin resulted in enriched fractions for EGCG and ECG. Their purity was 53.69 %, and 65.77 %, with recovery rates of 88.05 %, and 83.46 %, respectively. For the second-step, The recovery rates of 85.36 % and 86.09 % for EGCG and ECG were obtained respectively by a novel stationary phase of beta-CD bonded agar gel. The overall recovery efficiency of the complete procedure was 75.16 % for EGCG and 71.85 % for ECG, respectively, and the purity of the obtained preparations reached 97.63 % for EGCG and 98.09 % for ECG. The chromatographic behavior of beta-cyclodextrin bonded agar gel was studied with elution by acetonitrile, which indicated that both hydrophilic and hydrophobic interactions between the polyphenols and the solid phase occurred.

Abstract:A dual-task method for the simultaneous separation and purification of (-)-epigallocatechin gallate (EGCG) and caffeine (CAF) from crude extract of green tea was established by size exclusion effect onto hydroquinone modified porous adsorbents. The results showed that hydroquinone modified porous adsorbents P4 provided the best separation power due to it has more porous structure and phenolic hydroxyl group. The adsorption-desorption behaviors of EGCG and CAF onto P4 adsorbents were investigated. Adsorption kinetics of EGCG and CAF results showed that the adsorption followed the pseudo-second-order kinetic model. The results also indicated that the equilibrium adsorption data best fit the Langmuir model. Meanwhile, EGCG and CAF were separated successfully onto P4 adsorbents packed columns in a gradient eluent process, and P4 adsorbents exhibited the size exclusion effect for small molecules CAF. Based on the phenolic hydroxyl group and size exclusion effect of P4 adsorbents, the high purity EGCG and CAF were obtained with 40% (v/v) ethanol eluent successively. The process fulfilled the task of simultaneous separation and purification of EGCG and CAF, and proved to be a promising basis for preparations of difficult to obtain active components that have similar polarity and different sizes of molecules and derived from the same natural products.
Separation and Purification Process of Total Flavones from Radix Tetrastigmae with Macroporons Resins

Abstract:OBJECTIVE To screen the suitable resin for separation and purification of total flavones from Radix tetrastigmae,and to establish the optimum parameters of separation and purification process.METHODS The adsorption capacities and desorption ratios of ten selected macroporous resins were evaluated with UV method.Suitable resins were chosen to study the performance of static-dynamic adsorption and desorption,and to optimize the process parameters.RESULTS HPD826 resin showed advantageous adsorption and desorption properties with thermodynamic parameter according with the Freundlich equation.The optimal conditions were determined as follows: for adsorption,the flow speed was 2 BV·h-1;for desorption,wash the adsorbate-laden column first with 2 BV water,then with 3 BV 70% ethanol-water.Under this condition,the content of total flavones increased by 2.44-fold.CONCLUSION It is feasible to separate and purify Radix tetrastigmae total flavones with HPD826 resin.

Abstract:(-)-Epigallocatechin gallate (EGCg)-rich extract (EGCg > 700 mg g(-1)) was prepared from green tea leaves through a three-stage process consisting of liquid-liquid extraction and silica column purification. Crude tea extract was dissolved in ethyl acetate. After filtration, the solution was extracted by 10 g L-1 citric acid solution twice, and then passed through silica column. The catechins compounds in the ethyl acetate eluate were back extracted to the aqueous phase, then extracted with a mixed solution of n-hexane/ethyl acetate (2/5, v/v) 3 times, concentrated, and freeze dried. 12.8g EGCg-rich extract containing 709 mg g(-1) EGCg and 965 mg g(-1) total catechins was obtained from 300g green tea leaves, with an EGCg recovery of 26.1% and a yield of 4.3%. Abstract:An adsorption separation method using Polyamide-6 (PA) as an adsorbent was developed to separate catechins from green tea extract. The adsorption capacity of total catechins for PA was 193.128 mg g⁻¹ with an adsorption selectivity coefficient K(A)(B) of total catechins over caffeine 21.717, which was better than macroporous resin model HPD 600. The Langmuir model and the pseudo-second order mode were primely fitted to describe its equilibrium data and adsorption kinetics, respectively. PA column separation by two-step elution using water and 80% (v/v) aqueous ethanol was established to prepare catechins complex which contained 670.808 mg g⁻¹ total catechins and 1.828 mg g⁻¹ caffeine. It is considered that PA was a promising adsorbent for selective isolation of catechins.

Abstract:A method is described for successive purification of epigallocatechin gallate(EGCG)and epicatechin gallate(ECG)by medium-pressure silica gel column chromatography.The raw material was 98 % tea polyphenols.The operating conditions of the chromatographic steps were investigated and optimized,using 1 200 mm×80 mm stainless steel column as chromatographic column.The optimum condition was as followed:160-280 μm silica gel as filling material,ethyl acetate-petroleum ether-formic acid 6 ∶4 ∶1 as eluting solvent,flowing rate 30 mL/min,loading amount 35 g/kg silica gel.The average yields of 98 % EGCG and 98 % ECG were 85.5 % and 80.3 %,respectively.A reclaimed eluting solvent can be used repeatedly,it's pH value is first adjusted,and concentration ratio is then adjusted by TLC method.The chromatographic column which was regenerated by ethyl acetate,and equilibrated with petroleum,can be used repeatedly.

Abstract:Using low grade green tea as raw material, the process conditions of extracting, separating, purifying tea-polyphenols, caffeine and tea-polysaccharide were studied. A new comprehensive extraction process was developed. The results demonstrate that, by adding multiple enzyme liquor of cellulase and pectase, the leaching rate of active constituents of green tea is improved and the loss of activity is reduced; enzyme extraction liquor is refined through ultrafiltration membrane, and tea-polysaccharide is effectively produced from the membrane concentration liquor. By using H-600 resin adsorption and stepwise elution chromatography process for tea-polyphenols and caffeine separation, this process can solve the problem in conventional process in which caffeine must be eliminated with poisonous solvent. The calculation of height of mass transfer zone and total mass transfer coefficient of adsorption column provides a basis for the design of size-enlarged plant.

Abstract:AbstractTo separate and purify tea polyphenols and caffeine from discarded green tea crude extract with macroporous adsorption resins (MARs), the adsorption properties of twenty kinds of MARs are systematically evaluated. The results show that GS-18 has higher separation efficiency than other MARs, and the experimental data fit best to the pseudo-second-order kinetic model. And adsorption isotherms and thermodynamics of tea polyphenols and caffeine on GS-18 are investigated. The separation parameters of tea polyphenols and caffeine from discarded green tea crude extract are optimized according to dynamic adsorption/desorption experiments with the column packed with GS-18. Through only one cycle treatment, the content of tea polyphenols increases 3.0-fold from 20 to 60 % with a recovery yield of 91 % and that of caffeine increases 3.1-fold from 7 to 22 % with a recovery yield of 98 %. The results demonstrate that the method of MARs will provide useful help for the development of large-scale separation of tea polyphenols and caffeine from discarded green tea in industry.

Abstract:A highly efficient column-chromatographic extraction (CCE) followed by sequential adsorption to extract and separate bioactive compounds from green tea was developed. Tea powder was loaded into columns with 4-fold solvents and eluted through a cyclic CCE. High-quality tea extracts with greater than 90% extraction efficiencies of polyphenols, epigallocatechin-3-gallate, caffeine, theanine and polysaccharides were obtained with 4-fold water circulated five times among different columns at 70°C. Similar results, except for low polysaccharide extraction (35.5%), were obtained with 4-fold 30% ethanol circulated three times at room temperature. The highly concentrated water extraction was directly passed through columns of polyamide, DM130 macroporous and 732 ion exchange resins, resulting in high-purity polyphenols (99%), caffeine (98%) and theanine (98%) after simple purification of the eluates from each column. This method uses simple equipment, minimum solvents and can be used for both quantitative analysis and continuous preparation of high-quality tea extracts and bioactive compounds.

Abstract:Epigallocatechin-3-O-gallate (EGCG) was modified by catalytic esterification with hexadecanoyl chloride. A long-chain fatty acid ester derivative was obtained from the purification of this reaction product by high-speedcounter-current chromatography using a solvent system composed of n-hexane-ethyl acetate-methanol-water (1:1:1:1, v/v). The structure of the derivative, epigallocatechin-3-O-gallate-4'-hexadecanate, was elucidated by IR, MS and 1H NMR.

Abstract:Adsorbent is one of the most important factors for separation efficiency in fixed-bed purification techniques. The adsorption behavior of catechins and caffeine onto polyvinylpolypyrrolidone (PVPP) was investigated by static adsorption tests. The results showed that catechins rather than caffeine were preferred to adsorb onto PVPP since the adsorption selectivity coefficient of total catechins vs caffeine was around 22.5, and that adsorption of catechins could be described by the pseudo-second-order model. Adsorption amount of caffeine onto PVPP in green tea extracts solution was much higher than that in purified caffeine solution although the initial concentration of caffeine was similar in the two solutions, indicating the caffeine might be attached with catechins which were adsorbed by PVPP instead of being adsorbed by PVPP directly. The results also showed that the adsorption capacity of catechins and caffeine decreased with an increase in temperature, and that Freundlich and Langmuir models were both suitable for describing the isothermal adsorption of catechins, but not suitable for caffeine. The predicted maximum monolayer adsorption capacity of total catechins by PVPP was 671.77 mg g(-1) at 20 °C, which was significantly higher than that by other reported adsorbents. The thermodynamics analyses indicated that the adsorption of catechins onto PVPP was a spontaneous and exothermic physisorption process, revealing lower temperature was favorable for the adsorption of catechins. Elution tests showed that the desorption rates of catechins and caffeine were higher than 91% and 99% after two elution stages; in detail, almost all of the caffeine could be washed down at the water eluting stage, while catechins could be recovered at the dimethyl sulfoxide/ethanol solution eluting stage. Thus, the PVPP could be used as an excellent alternative adsorbent candidate for separating catechins from crude tea extracts, although some investigations, such as exploring the new eluants with low boiling point and high desorption efficiency, should be conducted furthermore.

Abstract:The process of purification of crude tea polyphenols by column chromatography was investigated.The method was carried out with silica gel as adsorbent and ethyl acetate as elution.Thus,tea polyphenols was obtained which the content of catethin was over 90% and the total content of EGCG,GCG and ECG was over 75%,the content of caffeine was unchecked by HPLC.The results showed that this method was effective and simple.

Abstract:A novel method for the extraction of (−)epigallocatechin gallate of high purity from green tea leaves is proposed in this study. The method comprised a two-stage water-based extraction followed by successive use of microfiltration and ultrafiltration. Microfiltration was used as a pretreatment to ultrafiltration. The best process conditions of each unit operation were estimated by performing well-planned experimentations. The clarified liquor was dried to powder by freeze drying. Chemical analyses revealed that the tea powder contained about 90% of polyphenols. The purity of (−)epigallocatechin gallate was found to be about 80%, while its average yield was 1.22 g/l. The method outlined in this study may have remarkable importance for the bulk production of high-purity (−)epigallocatechin gallate with potential application in pharmaceutical, cosmetic, and food-processing industries. Besides being a green process, this method can be easily scaled up for the commercial production of (−)epigallocatechin gallate.

Abstract:Tea catechin oxy-polymers were obtained by oxidizing tea catechinwith H2O2 and their isolation conditions of HPLC were studied. MS showed that their molecular weight range from 906.4 to 1962.4, which suggested that they might be dimer to pentamer of EGCG; Isocratic and linear gradient elution could not separate them from each other, but a method of combining linear and step gradients elution could separate them from each other. This HPLC method might provide a new way to separate oxy-polymers of catechin.

Abstract:A novel method to separate and purify tea seed polysaccharide and tea seed saponin from camellia cake extract by macroporous resin was developed. Among four kinds of resins (AB-8, NKA-9, XDA-6, and D4020) tested, AB-8 macroporous resin possessed optimal separating capacity for the two substances and thus was selected for the separation, in which deionized water was used to elute tea seed polysaccharide, 0.25% NaOH solution to remove the undesired pigments, and 90% ethanol to elute tea seed saponin. Further dynamic adsorption/desorption experiments on AB-8 resin-based column chromatography were conducted to obtain the optimal parameters. Under optimal dynamic adsorption and desorption conditions, 18.7 and 11.8% yield of tea seed polysaccharide and tea seed saponin were obtained with purities of 89.2 and 96.0%, respectively. The developed method provides a potential approach for the large-scale production of tea seed polysaccharide and tea seed saponin from camellia cake.

Adsorption and Separation of Tea Polyphenols and Caffeine by Natural Adsorbents

Abstract:To obtain natural and safe absorbents with low costs, two plant residues, Rice Grain Hull (RGH) and Retinervus Luffae Fructus (RLF), one burnt red brick powder, one mineral stone powder and sea sands were tested for their absorption and separation on polyphenols and caffeine in green tea extracts, using HPLC and spectrophotometric methods for determination of EGCG (Epigallocatechin-3-gallate), caffeine and total polyphenols. RGH and RLF had absorption rates of polyphenols of 65% and 62% , respectively, which are slightly lower than those of the best absorbents of macroporous resin DM130 (73%) and polyamide resin (81%). The absorption rates of caffeine by RGH and RLF were 24% and 18% , respectively, in between those of DM130 (68% ) and polyamide (15%). Red brick powder, stone power and sea sand had low absorption to polyphenols (38%, 29%, 3%, respectively) and caffeine (24%, 15%, 15%, respectively). In column chromatographic separation with gradient elution of 1 BV of H2O, 20% and 40% ethanol, and 3 BVs of 80% ethanol, polyamide can be used to effectively separate caffeine (90% in the H2O and 20% ethanol fractions) and EGCG (90% in the 80% ethanol fraction). DM130 can not be used to effectively separate caffeine and EGCG, 94%~96% of both compounds were in the 40% and 80% ethanol fractions. The performances for RGH and RLF are in between the two resins. Caffeine was 92% for RGH and 87% for RLF in H2O, 20% and 40% ethanol fractions, while EGCG were 84% for RGH and 76% for RLF in 80% ethanol fractions. RGH and RLF can be used to separate caffeine and EGCG effectively. Results indicate that RGH and RLF can be used as adsorbent materials for adsorption and separation of EGCG and caffeine in tea extracts.

Abstract:In the present study, feasibility of producing high quality of tea polyphenols, theanine and caffeine from the extract of summer green tea by column chromatography was investigated. Crude extract of summer green tea obtained by hot water extraction was passed through columns of polyamide and NKA-II macroporous resin respectively, resulting in tea polyphenols-enriched fraction (96.27 +/- 1.78%), theanine-enriched fraction (99.02 +/- 0.43%) and caffeine-enriched fraction (99.25 +/- 0.36%). The recovery rates for tea polyphenols, theanine and caffeine were 72.42 +/- 1.41%, 66.12 +/- 1.66% and 62.07 +/- 2.17%, respectively. The separation procedure allowed the production of such products by decreasing impurities and avoiding the use of poisonous organic solvent. The results suggested that it was possible to produce high-quality products of rich-in tea polyphenols, theanine and caffeine by a simple way. From industrial point of view, this novel method has many merits such as high efficiency of separation, low consumption of energy and environment-friendly procedure.

Abstract:SummaryDesorption of catechins and caffeine from polyvinylpolypyrrolidone (PVPP) was comprehensively investigated. The result showed that caffeine could easily be desorbed from PVPP by the tested solvents except n‐hexane, while catechins could only be thoroughly done by dimethyl sulphoxide (DMSO) and N, N‐dimethylformamide (DMF). Excellent desorption efficiency of DMSO and DMF might be attributed to their high dipole moments and H‐bond potentials. Addition of ethanol was recommended considering the elution efficiency and fluidity, but ethanol volume should be <40% (v/v) for DMSO or 20% (v/v) for DMF. Desorption would get to equilibrium within 1 h and followed the pseudo‐second‐order model. Caffeine and catechins could be separately desorbed through two‐stage elution procedure, that is, water or 20% aqueous ethanol for desorbing caffeine and part of nongalloylated catechins and DMSO/ethanol (8/2, v/v) for eluting the remaining catechins. Highly purified catechins (~95%) with high level (~70%) galloylated catechins would be achieved when the desorption procedure was applied in column chromatograph.

Abstract:A simple, clean and economical way of separation and preparation noncaffeine tea polyphenols from green tea waste in one-step by Sephadex LH-20 column chromatography was developed. 20% (v/v) ethanol solution was used for removing caffeine and other components in the first stepwise elution, and tea polyphenols was eluted with 60% (v/v) ethanol solution by using reversed flow in the second stepwise elution. The yield of tea polyphenols above 11% with the purity about 98% and the content of catechins more than 91%, and the content of caffeine in tea polyphenols have not been detected by high-performance liquid chromatography. The recovery of tea polyphenols by column chromatography were about 86%, and the stability of preparative way is very good. Compared with conventional extraction and decaffeination ways, this way has more excellent advantages, such as cheaper raw material, no toxic residues, higher yield, commercially feasible and environment friendly.
