Abstract:To obtain natural and safe absorbents with low costs, two plant residues, Rice Grain Hull (RGH) and Retinervus Luffae Fructus (RLF), one burnt red brick powder, one mineral stone powder and sea sands were tested for their absorption and separation on polyphenols and caffeine in green tea extracts, using HPLC and spectrophotometric methods for determination of EGCG (Epigallocatechin-3-gallate), caffeine and total polyphenols. RGH and RLF had absorption rates of polyphenols of 65% and 62% , respectively, which are slightly lower than those of the best absorbents of macroporous resin DM130 (73%) and polyamide resin (81%). The absorption rates of caffeine by RGH and RLF were 24% and 18% , respectively, in between those of DM130 (68% ) and polyamide (15%). Red brick powder, stone power and sea sand had low absorption to polyphenols (38%, 29%, 3%, respectively) and caffeine (24%, 15%, 15%, respectively). In column chromatographic separation with gradient elution of 1 BV of H2O, 20% and 40% ethanol, and 3 BVs of 80% ethanol, polyamide can be used to effectively separate caffeine (90% in the H2O and 20% ethanol fractions) and EGCG (90% in the 80% ethanol fraction). DM130 can not be used to effectively separate caffeine and EGCG, 94%~96% of both compounds were in the 40% and 80% ethanol fractions. The performances for RGH and RLF are in between the two resins. Caffeine was 92% for RGH and 87% for RLF in H2O, 20% and 40% ethanol fractions, while EGCG were 84% for RGH and 76% for RLF in 80% ethanol fractions. RGH and RLF can be used to separate caffeine and EGCG effectively. Results indicate that RGH and RLF can be used as adsorbent materials for adsorption and separation of EGCG and caffeine in tea extracts.
