Abstract:A kind of analytical method by high performance liquidchromatography( HPLC) for tea catechins with ingradient elution system was put forward. In the meantime，the method was investigated in the methodological aspect through separa ting and determining the main four catechins (EC，ECG，EGC，EGCG) of tea polyphe nols. The optimum chromatographic conditions were as follows：ODS column (5 μm ， 4.6×250 mm)，water/acetonitrile/ethylacetate (v/v/v，8 6∶12∶2) as mobile phase ( add H2SO4 to adjust pH 3.0～4.0)，UV detector (280 nm)，flow rate:1.0 ml/ min. Moreover，the precision and stability of the method was discussed.

Abstract:Tea polyphenols is an ideal natural anti-oxidant,which has being widely applied in many fields.The methods of extraction of tea polyphenols from green tea have been become one of hot points of tea research.In the present paper,methods of extraction of tea polyphonols from green tea were reviewed,including solvent extraction,ionic precipitation,supercritical CO2 extraction,resin adsorption,ultrasonic assistant extraction,microwave assistant extraction and so on.We also discussed basic principle,producing routine and advantage or disadvantage of each method at the same time.

Abstract:In this paper, theaflavins extracts from black tea and tea polyphenols were used as control, composition of theaflavins in enzymatic oxidation products was analyzed by HPLC, and the antioxidant activity was investigated by the technique of chemiluminescence. Results showed that, TF2A was the main composition in enzymatic oxidation products. In theaflavins extracts from black tea, there mainly existed four kinds of theaflavins and their contents were not greatly fluctuated. By employing CuSO4-Vc-H2O2-yeast and Fe2+-H2O2-yeast systems, antioxidant activity of enzymatic oxidation products was better than theaflavins extracts from black tea and tea polyphenols.

Abstract:Tea polyphenol product is a kind of complex product extracted from tea.Applicability of ferrous tartrate method and Folin-Ciocalteu method in determining total polyphenols' content of tea polyphenol products were discussed in this paper.The results showed that both of them were stable with RSD <5.1.Two methods had different results,because the measuring principles of them were different.As the purity of tea polyphenol products improving,differences between two methods' results were bigger.Status of Folin-Ciocalteu-standard curve method ranged from 30.08% to 88.76% while several data of ferrous tartrate method exceeded 100%.It was summarized that Folin-Ciocalteu methods including Folin-Ciocalteu-standard curve method and Folin-Ciocalteu-conversion ratio method were better than ferrous tartrate method in determining total polyphenols' content of tea polyphenol products and it was calculated that the conversion coefficient was 88.13μg/mL.

Abstract:Polyphenols, the most abundant components in tea, determine the quality and health function of tea. The analysis of polyphenols in tea is a topic of increasing interest. However, the complexity of the tea matrix, the wide variety of teas, and the difference in determination purposes puts forward higher requirements for the detection of tea polyphenols. Many efforts have been made to provide a highly sensitive and selective analytical method for the determination and characterization of tea polyphenols. In order to provide new insight for the further development of polyphenols in tea, in the present review we summarize the recent literature for the detection of tea polyphenols from the perspectives of determining total polyphenols and individual polyphenols in tea. There are a variety of methods for the analysis of total tea polyphenols, which range from the traditional titration method, to the widely used spectrophotometry based on the color reaction of Folin–Ciocalteu, and then to the current electrochemical sensor for rapid on-site detection. Additionally, the application of improved liquid chromatography (LC) and high-resolution mass spectrometry (HRMS) were emphasized for the simultaneous determination of multiple polyphenols and the identification of novel polyphenols. Finally, a brief outline of future development trends are discussed.

Abstract:Tea and coffee, the most popular beverages in the world, have been consumed for thousands of years for their alluring flavors and health benefits. Polyphenols, particularly flavonoids and phenolic acids, are of great abundance in tea and coffee and contribute a lot to their flavor and health properties. This paper reviews the polyphenol chemistry of tea and coffee, specifically their stability, and scavenging ability of reactive oxygen species (ROS) and reactive carbonyl species (RCS). During the manufacturing and brewing process, green tea and black tea polyphenols undergo epimerization and oxidation, respectively. Meanwhile, the lactonization and the polymerization of chlorogenic acid are the major causes for the degradation of polyphenols in coffee. Tea catechins, besides having antioxidant properties, have the novel characteristic of trapping reactive carbonyl species. The A ring of the catechins is the binding site for RCS trapping, whereas the B ring is the preferred site for antioxidation.

Abstract:In this study, an effective ultra-high performance liquid chromatography (UHPLC) method for simultaneous determination of 15 constituents of catechins, caffeine, gallic acid and theaflavins was developed. Employing the developed UHPLC method, the contents of the 15 constituents were analyzed in 245 samples of 6 types of Chinese teas collected from different places. The catechin contents in these samples ranged from 16.50 to 94.35 mg/g. Theaflavins were not detected in green tea and white tea, and caffeine did not strongly relate to germplasm resource or fermentation degree. Principal component analysis (PCA) of UHPLC data revealed that white, oolong and green tea samples could be clustered closely, demonstrating notable difference of functional components between the three types of teas resulting from preparation process and varieties. The constituent profile could be primarily attributed to the structural transformation of catechins and pigments during tea processing. This study would advance our understanding of chemical profile of Chinese teas afforded by different processing technologies.
A HPLC Method for Detection of 17 Characteristic Components in Tea Extract

Abstract:A HPLC method has been developed for the determination of tea for seven catechins ((+)-catechin, (−)-epicatechin, (−)-epigallocatechin, (−)-epigallocatechin gallate, (−)-epicatechin gallate, (−)-gallocatechins and (−)-gallocatechin gallate), two flavonols (quercetin, kaempferol), seven organic acids (Gallic acid, ferulic acid, protocatechuic acid, p-hydroxybenzoic acid,p-coumaric acid, erucic acid,vanillic acid) and caffeine. Using HPLC with gradient elution and photodiode array detection, seventeen compounds were separated and detected by optimizing the column temperature, mobile phase ratio and detection wavelength. The validity of this method was confirmed by the quantitative measurement of seventeen characteristic compounds. The correlation coefficient of all standards curve was 0.9967~1.0000 and the detection limits (s/N = 3) were 0.010~1.2 mg/L. The average recoveries of 17 characteristic components were between 81.50 and 124.80%. 59 green tea samples (origin: Hangzhou, China) were analyzed and 11 characteristic components were found in all samples and their content of different samples varied greatly. The catechins were the most abundant constituents in teas with up to 79.5~156.6 g/kg and the contents of (−)-epigallocatechin gallate were the highest. Quercetin and kaempferol were only found in some tea samples. Protocatechuic acid, p-hydroxybenzoic acid, ferulic acid and erucic acid were not detected in the tea samples. This method is ideally suited for routine analysis for the determination of bioactive components in green tea with good repeatability and accuracy of results. Furthermore, this method can be applied to all kinds of tea and tea products and provided a detection method for establishing the HPLC fingerprint of a certain tea.

Determination of Flavonol Glycosides in Tea Samples by Ultra-High Performance Liquid Chromatography-Photodiode Array Detection-Tandem Mass Spectrometry

Abstract:An ultra?high performance liquid chromatography?photodiode array detection?tandem mass spectrometry ( UPLC?PDA?MS/MS) method was developed for the determination of flavo?nol glycosides in tea samples. The chromatographic separation was performed on an UPLC HSS T3 column by gradient elution with the mobile phases of acetonitrile and water both containing 0?1%( v/v) formic acid. A total of 15 flavonol glycosides which include 3 myricetin glycosides, 6 quercetin glycosides and 6 kaempferol glycosides were positively identified in green and black tea samples by comparing the retention times and mass spectra of the samples with standards and publications. The quantities of flavonol glycosides were relatively calculated with the stand?ard quercetin?3?rhamnosylglucoside ( Q?GRh) which was calibrated with external quantification method using multi?reaction monitoring ( MRM ) mode. The results showed that there were different flavonol glycoside distributions in green tea and black tea. The total amount of flavonol glycosides in green tea was 1?7 times of that in black tea. The major flavonol glycosides in green tea were myricetin?3?galactoside ( M?Ga) , myricetin?3?glucoside ( M?G) , quercetin?3?glucosyl?rhamnosyl?galactoside ( Q?GaRhG ) , quercetin?3?glucosyl?rhamnosyl?glucoside ( Q?GRhG) , kaempferol?3?glucosyl?rhamnosyl?galactoside ( K?GaRhG ) and kaempferol?3?glucosyl?rhamnosyl?glucoside ( K?GRhG) , but for black tea, the major flavonol glycosides were querce?tin?3?rhamnosylglucoside ( Q?GRh) , quercetin?3?glucoside ( Q?G) , kaempferol?3?rhamnosylglu?coside (K?GRh) and kaempferol?3?galactoside (K?Ga). The present method is accurate, con?venient for the rapid identification of flavonol glycosides and analysis of constituent distribution for green and black teas.

Abstract:Despite the fact that mankind has been drinking tea for more than 5000 years, its chemical composition has been studied only in recent decades. These studies are primarily carried out using chromatographic methods. This review summarizes the latest information regarding the chemical composition of different tea grades by different chromatographic methods, which has not previously been reviewed in the same scope. Over the last 40 years, the qualitative and quantitative analyses of high volatile compounds were determined by GC and GC/MS. The main components responsible for aroma of green and black tea were revealed, and the low volatile compounds basically were determined by HPLC and LC/MS methods. Most studies focusing on the determination of catechins and caffeine in various teas (green, oolong, black and pu-erh) involved HPLC analysis. Knowledge of tea chemical composition helps in assessing its quality on the one hand, and helps to monitor and manage its growing, processing, and storage conditions on the other. In particular, this knowledge has enabled to establish the relationships between the chemical composition of tea and its properties by identifying the tea constituents which determine its aroma and taste. Therefore, assessment of tea quality does not only rely on subjective organoleptic evaluation, but also on objective physical and chemical methods, with extra determination of tea components most beneficial to human health. With this knowledge, the nutritional value of tea may be increased, and tea quality improved by providing via optimization of the growing, processing, and storage conditions.

Abstract:Tea, as a beverage, has been reputed for its health benefits and gained worldwide popularity. Tea polyphenols, especially catechins, as the main bioactive compounds in tea, exhibit diverse health benefits and have wide applications in the food industry. The development of tea polyphenol-incorporated products is dependent on the extraction, purification, and identification of tea polyphenols. Recent years, many green and novel extraction, purification, and identification techniques have been developed for the preparation of tea polyphenols. This review, therefore, introduces the classification of tea and summarizes the main conventional and novel techniques for the extraction of polyphenols from various tea products. The advantages and disadvantages of these techniques are also intensively discussed and compared. In addition, the purification and identification techniques are summarized. It is hoped that this updated review can provide a research basis for the green and efficient extraction, purification, and identification of tea polyphenols, which can facilitate their utilization in the production of various functional food products and nutraceuticals.

Abstract:Pu-erh tea is a unique microbial fermented tea produced from the sun-dried leaves of large-leaf tea species (Camellia sinensis (Linn.) var. assamica (Masters) Kitamura) in the Yunnan province of China. Pu-erh tea has become increasingly popular in Southeast Asia may be due to its multiple health benefits. The special sensory characteristics of Pu-erh tea arise from the multitudinous chemical changes and transformations of the chemical constituents of the sun-dried green tea leaves that occur during the post-fermentation process. Many functional components have been isolated from Pu-erh tea and identified. In this paper, modern processing techniques and their effects on the transformation of the chemical constituents and the major chemical components of Pu-erh tea are reviewed and discussed.

Abstract:Qualitative and quantitative analysis of tea polyphenols in green tea, jasmine tea, Kuding tea, and ginseng tea was carried out by HPLC-UV/MS. There were 5 catechins in green tea and 7 catechins in jasmine tea identified, but no catechins were identified in Kuding tea and ginseng tea. The total content of catechins was 2% in green tea and 4% in jasmine tea, respectively.

Abstract:The classification of macrofeticulaf resin and their principles of adsorption and desorption in isolation and purification of tea polyphenols were reviewed in the present paper. The application of macrofeticulaf resin to isolation and purification of tea polyphenols included preparation of decaffeinated tea polyphenols, continuous extraction and isolation of functional components from tea and purification of tea polyphenols. The aspects of food safety and organic solvent residues in application of macrofeticulaf resin were also discussed.

Abstract:Polyphenols, a ubiquitous group of secondary plant metabolites sharing at least one aromatic ring structure with one or more hydroxyl groups, represent a large group of natural antioxidants abundant in fruits, vegetables, and beverages, such as grape juice, wine, and tea, and are widely considered to contribute to health benefits in humans. However, little is yet known concerning their bioactive forms in vivo and the mechanisms by which they may alter our metabolome, which ultimately contribute toward disease prevention. Here we report a study to determine the metabolic fate of polyphenolic components in a Chinese tea (Pu-erh) in human subjects using a metabonomic profiling approach coupled with multivariate and univariate statistical analysis. Urine samples were collected at 0 h, 1 h, 3 h, 6 h, 9 h, 12 h, and 24 h within the first 24 h and once a day during a 6 week period including a 2 week baseline phase, a 2 week daily Pu-erh tea ingestion phase, and a 2 week "wash-out" phase, and they were analyzed by gas chromatography mass spectrometry and liquid chromatography mass spectrometry. The dynamic concentration profile of bioavailable plant molecules (due to in vivo absorption and the hepatic and gut bacterial metabolism) and the human metabolic response profile were measured and correlated with each other. This study demonstrates that the metabonomic strategy will enable us to integrate the overwhelming amount of metabolic end points as a systems' response to the absorption, metabolism, and disposition of a multicomponent botanical intervention system, leading to a direct elucidation of their mechanisms of action.

Abstract:In this study, the chemical constituents of pu-erh tea, black tea, and green tea, as well as those of pu-erh tea products of different ages, were analyzed and compared using a chemical profiling approach. Differences in tea processing resulted in differences in the chemical constituents and the color of tea infusions. Human biological responses to pu-erh tea ingestion were also studied by using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS) in conjunction with multivariate statistical techniques. Metabolic alterations during and after pu-erh tea ingestion were characterized by increased urinary excretion of 5-hydroxytryptophan, inositol, and 4-methoxyphenylacetic acid, along with reduced excretion of 3-chlorotyrosine and creatinine. This study highlights the potential for metabonomic technology to assess nutritional interventions and is an important step toward a full understanding of pu-erh tea and its influence on human metabolism.

Abstract:Tea (Camellia sinensis) is the second most commonly consumed beverage in the world, and its consumption has been associated with several important health benefits due in part to its high concentration of polyphenolic compounds. Tea varieties, including white, green, oolong, pu-erh, and black tea, are all produced from the same plant but differ in postharvest processing. The level of oxidative processing from white to black tea (low to high) drives changes in polyphenol profiles from monomeric polyphenols (catechins) to oxidative products (theaflavins and thearubigins) that convert the subtle yellow color of green tea to the distinctive red/copper color of oolong and black tea. Polyphenol compounds from green teas have been well studied; however, oxidative products are more difficult to characterize and may require a combination of analytical techniques for identification and quantification. This technical summary introduces the major polyphenols in tea and discusses the main steps of tea processing and instrumentation used for polyphenol analysis.
