Abstract:Based on the nonlinear plasmonic scattering response to the modulated excitation in time, we realized a single-wavelength super-resolution imaging method on a custom-built system which is named as a scattering saturation STED (ssSTED) microscope. Abstract:Stimulated emission depletion microscopy (STED) has been proved to be a feasible and straightforward method of breaking the conventional diffraction barrier in the far-field. In this paper, we design and setup a home-built high speed STED microscope. Both the excitation beam (488nm) and the depletion beam (592nm) are provided by continuous wave lasers. By using a pair of galvo mirrors (TILL Yanus IV Digital Scan Head), we realize a pixel dwell time down to 2 μs in the experiment which enables an acquisition speed of 2 frames per second in an imaging field of 5*5 μm with an individual pixel size of 10nm. The image acquisition process is controlled by the software Imspector. In the manuscript, we give a clear description on how to build the microscope and also conduct several experiments to evaluate its performance in practice. A spatial resolution of <100nm, which is well beyond the diffraction barrier has been demonstrated in both nanoparticles and biomedical samples. Featuring a superresolution together with a high imaging speed, our STED microscope has big potential to be widely applied in related scientific researches.

Abstract:Super-resolution optical microscopy is a useful tool to investigate physical and biological characteristics on the sub-100-manometer scale. Here, we develop pulsed saturated absorption competition (pSAC) microscopy to break the diffraction limit in imaging of a series of nonbleaching nanoparticles. Based on the confocal scheme, the input laser beam is divided into doughnut-shaped saturated pulse and Gaussian time-modulated pulse. By adjusting the time sequential of the two pulses, the doughnut-shaped beam transiently saturates the excited-state absorption in the periphery of the focal volume, immediately followed by the Gaussian readout beam which mainly excites fluorophores at the focal center. Through the inhibition of the saturation pulse that confines the fluorescence emission to the central subregion, lock-in detection scheme is utilized to select the characteristic fluorescence signals. We numerically and experimentally demonstrate the resolving capacity of pSAC in imaging biological specimens and subdiffraction resolutions as high as lambda/13 and lambda/12 are reached in nonfluorescence and fluorescence observation, respectively. The experimental results also show that, compared with stimulated emission depletion (STED) microscopy, pSAC is beneficial to achieve background-suppressed imaging when considering lower-power and fluorescent dye selection.

Abstract:Far-field super-resolution imaging techniques such as stimulated emission depletion (STED), stochastic optical reconstruction microscopy (STORM), and photoactivation localization microscopy (PALM) etc., have enabled fluorescence labeled nanoscale imaging and analysis. However, it is necessary or preferable to develop label-free high resolution imaging techniques to avoid the problems of phototoxicity with live cells and moreover, for many non-biological applications where fluorescent labelling is simply not feasible. Recently, by capturing the exponentially decaying evanescent waves and propagating it to far field, novel imaging technique like hyperlens and microsphere contacting technologies, micro-fiber and nanowire illumination technology have brought new opportunities to label-free super resolution imaging, but still suffer from weak signal compared to background noise. On the other hand, there is lack of criteria reported to quantify the imaging quality of label free far-field nanoscopy, that will slow down the developing of label-free far-field nanoscopy. We introduce CNR first to label-free far-field nanoscopy to quantify the imaging performance and investigate the key influencing elements systematically. In our study, we investigated and analyzed the key elements to achieve high CNR label-free wide-field far-field sub-diffraction imaging. By optimizing the key factors including polarization, materials, as well as fabrication conditions, sub-diffraction imaging with resolution of 122 nm in a large FOV has been achieved experimentally. This work has provided an efficient and convenient way to realize high contrast wide-field far-field label-free super-resolution based on evanescent wave illumination.

Abstract:To investigate fine structure in biological samples, over recent decades, super-resolution methods in light microscopy have been honed. Here, we report a technique integrating nonlinearity arising from fluorophore saturation with a new version of I5S to produce quasi-isotropic three-dimensional (3D) wide-field (WF) super-resolution imaging. This extended technique—saturated I5S (SI5S)—promises lateral and axial resolutions below 60 nm when the illuminating light intensity approaches the saturation threshold in simulation. A linear processing algorithm is used to acquire SI5S image reconstructions, and comparisons with 3D structured illumination microscopy (3D-SIM) and I5S show that a substantial improvement in resolution can be realized using SI5S. The notable improvement in 3D resolution means that SI5S could facilitate volumetric visualization of intracellular fine structure within biological samples.

Abstract:Stimulated emission depletion (STED) microscopy is one of the breakthrough technologies that belong to far-field optical microscopy and can achieve nanoscale spatial resolution. We demonstrate a far-field optical nanoscopy based on continuous wave lasers with different wavelengths, i.e., violet and green lasers for excitation and STED, respectively. Fluorescent dyes Coumarin 102 and Atto 390 are used for validating the depletion efficiency. Fluorescent nanoparticles are selected for characterizing the spatial resolution of the STED system. Linear scanning of the laser beams of the STED system along one line of a microscope slide, which is coated with the nanoparticles, indicates that a spatial resolution of about 70 nm has so far been achieved. A two-dimensional image of the particle pattern of the STED system is constructed and compared with scanning confocal microscope. The present work has further extended the application of the STED microscopy into the blue regime.

Abstract:To overcome the diffraction barrier, super-resolution microscopy is contrived and has witnessed scientific developments in varying fields, especially in last few decades, such as stochastic optical reconstruction microscopy, stimulated emission depletion microscopy (STED), mirror-enhanced super-resolution microscopy (MEANS), and fluorescence emission difference microscopy (FED). Recently, saturated competition microscopy (SAC) was developed to realize high sub-diffraction resolution in either fluorescent or non-fluorescent imaging. Compared with STED, SAC features non-constraint in fluorescent dye selection. Nevertheless, the lateral resolution is limited in consideration of photobleaching side effects. Also, the axial resolution enhancement of SAC has not been demonstrated. In this study, a method, combining FED, MEANS, and SAC, is presented to improve the three-dimensional (3D) resolution. The numerical study reveals that the lateral resolution is close to 0.085 lambda. and axial resolution can be enhanced to 0.184 lambda. In addition, the SNR is improved simultaneously. The availability to improve 3D resolution of SAC is believed to be significant for biological imaging in the future. (C) 2020 Optical Society of America

Abstract:Stimulated emission depletion nanoscopy and its derivatives based on saturation induced competition effects have become an indispensable tool for studying cellular events and their dynamics in living conditions. The successful implementation of these techniques heavily relies on the competition between excitation induced spontaneous emission and stimulated emission from fluorescent dyes. The use of two laser beams at different wavelengths perplexes the optical system and the high intensity saturation beam inevitably introduces detrimental photobleaching effects. Harnessing the emerging saturation scattering of plasmonic nanoparticles, here, we demonstrate a novel fluorescence-free single-wavelength super-resolution imaging technique using gold nanoparticles. A lateral resolution of 101.2 nm (<λ/5) is achieved through introducing saturation scattering competition (SSC) of 60 nm gold nanospheres between dual beams at the same wavelength. In addition, the SSC drastically reduces the saturation intensity by three orders of magnitude than the conventional stimulated emission depletion process at comparable resolutions. As a proof of concept, we realized robust single-wavelength super-resolved imaging in dMG-63 cells with a simplified system. The current technique provides a new modality of biosample-friendly technology for optical super-resolution imaging.

Abstract:Abstract. We have designed and built a time-gated continuous wave stimulated emission depletion (CW-STED) nanoscopy to visualize microstructures beyond the diffraction limit. An off-line time-gating detection was performed with the help of time-correlated single-photon counting technique. Experimental results showed that before time-gating, the resolution of our system was about 75 nm with a depletion beam (592 nm) power of 200 mW. By using the off-line time-gating detection, the resolution was further improved to 38 nm. Biological samples were also used to test the performance of our time-gated CW-STED, and a resolution of 70 nm was achieved with a depletion beam (592 nm) power of 85 mW. Detailed principles of time-gated CW-STED were discussed in the text. The time-gated STED provides a better resolution with finite laser power.

Abstract:Abstract Stimulated emission depletion microscopy (STED) is a powerful tool for studying nanoscale cell structure and activity, but the difficulties it encounters in multicolor imaging limit its application in biological research. To overcome the disadvantages of limited number of channels and high cost of multicolor STED imaging based on spectral identity, we introduced lifetime into live-cell multicolor STED imaging by separating selected dyes of the same spectrum by phasor analysis. Experimental results show that our method enables live-cell STED imaging with at least 4 colors, enabling observation of cellular activity beyond the diffraction limit.

Abstract:Super-resolution microscopy, as a powerful tool of seeing abundant spatial details, typically can only distinguish a few distinct targets at a time due to the spectral crosstalk between fluorophores. Spaser (i.e., surface plasmon laser) nanoprobes, which confine lasing emission into nanoscale, offer an opportunity to eliminate such obstacle. Here, realized is narrow band stimulated emission depletion (STED) nanoscopy on spaser nanoparticles by collecting the coherent spasing signals. Demonstrated are the physics concept and feasibility of erasing spaser emission by using a depletion beam to suppress the population inversion, which lays the foundation of spaser-based STED super-resolution. Thanks to the small size (47 nm) and narrow spectral linewidth (3.8 nm) of the spaser nanoparticles, a 74 nm spatial resolution in STED imaging within an acquisition bandwidth of 10 nm is finally obtained. These spaser nanoparticles, if multiplexing with different wavelengths, in principle, allow for spectral-multiplexed imaging, sensing, cytometry, and light operation of a large number of targets all at once.

Abstract:Noble metal nanoparticles (NPs), owing to their unique optical and physicochemical properties, are routinely used for optical imaging and labeling of biological specimens. Even though they can provide vital information for studying multiple cellular events and their interplays at the same time, optically multiplexing and resolving specific NPs within a diffraction‐limited region labeled in complex biological specimens remains a fundamental challenge. By introducing and manipulating plasmonic resonance assisted saturable scattering effects, multiplexed fluorescence‐free super‐resolution imaging of gold NPs in tumor cells with remarkable subdiffraction resolution is demonstrated. The saturable scattering allows fluorescence‐free resolving single plasmonic nanoprobes with significantly improved resolution down to ≈100 nm. The revealed plasmonic resonance assisted saturation effect as well as the associated spectral flexibility to variant sizes provides access to multiplexing capability in complex bioenvironments. The demonstrated feasibility of two‐color super‐resolution cellular imaging is achieved at ultralow suppression powers ≈0.28 MW cm−2, corresponding to a two‐order of magnitude improvement compared to the state‐of‐the‐art of stimulated emission depletion (STED) nanoscopy.

Abstract:We propose an approach that uses a conventional optical microscope to achieve optical super-resolution. This approach will provide a direct, non-invasive, and far-field access in the observation of both metallic and non-metallic objects with a spatial resolution of tens-of-nanometres, achieved in a single snap shot. This method, combines near-field illumination by the micro-fiber and a passive spatial frequency shift by the sample itself, offering a promising approach to surface tomography imaging without localized field enhancement by surface plasmon polaritons. Our results present the potential of breaking the diffractive barrier using a conventional wide-field optical microscope within the visible spectrum.

Abstract:We propose a wide-field super-resolved optical microscopic imaging technique based on subwavelength slit arrays embedded in a thin silver film to generate surface plasmon (SP) standing wave interference patterns. These fringes carrying high spatial frequency information serve as excitation profiles to excite the nanoscale fluorescence objects. The super-resolved fluorescence density distribution is reconstructed from a weight sum of a series of fluorescence images with differently phase-shifted SP standing wave illumination. Simulation and experimental results show that the lateral resolution of the reconstructed fluorescence density image is enhanced by 0.28 λ SP in two dimensions, which is twofold better than that of conventional high numerical aperture fluorescence microscopy. This technique benefits from a grating coupler to offer a simple way for the generation and phase shift of SP standing wave excitation profiles in two dimensions. The flat configuration, wide field, and noninvasive nature make this approach suitable for real-time analyzing the fine details of bio-samples in biochip applications.

Abstract:We report a Gauss-Bessel STED microscope for super-resolution deep imaging. Improved lateral resolution at 95μm-depth in agarose phantom of brain tissue is about 110nm, which is nearly 2-fold improvement on resolution comparing to conventional STED imaging.

Abstract:Conventional far-field microscopy cannot directly resolve the sub-diffraction spatial distribution of localized surface plasmons in metal nanostructures. Using BaTiO 3 microspheres as far-field superlenses by collecting the near-field signal, we can map the origin of enhanced two-photon photoluminescence signal from the gap region of gold nanosphere dimers and gold nanorod dimers beyond the diffraction limit, on a conventional far-field microscope. As the angle θ between dimer’s structural axis and laser polarisation changes, photoluminescence intensity varies with a cos 4 θ function, which agrees quantitatively with numerical simulations. An optical resolution of about λ/7 (λ: two-photon luminescence central wavelength) is demonstrated at dimer’s gap region.
