Development of a Novel Second-Generation Polyester Dendritic Functionalized Graphene Oxide for Efficient Pipette Tip Micro Solid-Phase Extraction and Detection of Venlafaxine and Desvenlafaxine in Urine

Abstract:The monitoring therapeutic concentrations of venlafaxine and its metabolite desvenlafaxine is crucial for optimizing treatment efficacy and safety, guiding personalized treatment strategies for depression. In this study, a novel adsorbent, second-generation polyester dendritic functionalized graphene oxide (PDG-2), was developed for pipette tip micro solid-phase extraction of venlafaxine and desvenlafaxine from urine. Leveraging the facile modification properties of graphene oxide and the controlled synthesis capabilities of dendritic materials, PDG-2 exhibited a rough surface and numerous active groups with adsorption capacities of desvenlafaxine and venlafaxine up to 187 mg g−1 and 230 mg g−1. Optimal adsorption was achieved with just 1.5 mg of PDG-2, and the best elution was obtained using a trifluoroacetic acid-acetonitrile (1:99, v/v) solution. Adsorption kinetics followed a pseudo-second-order model, indicating chemical adsorption. Method validation confirmed excellent linearity (0.1−10 μg mL−1, r ≥ 0.9999), low detection limits (2.6−3.0 ng mL−1), high precision (relative standard deviation ≤ 4.6%), and accurate recovery rates (89.3−103.2%). Practical application was demonstrated by successfully detecting the target compounds in urine samples from volunteers. PDG-2 showed high reusability, maintaining over 90% recovery rates after multiple uses. Compared with commercial adsorbents, PDG-2 outperformed HLB and MCX and matched the performance of C18 with a much smaller amount. This research highlights the efficacy of dendritic polyester modification for enhancing adsorption performance, providing a reliable method for therapeutic drug monitoring of venlafaxine and its metabolites, and serving as a reference for future studies on multilayered branched modifications.
