Abstract:This volume provides a comprehensive overview of the major applications and potential of fungal biotechnology. The respective chapters report on the latest advances and opportunities in each topic area, proposing new and sustainable solutions to some of the major challenges faced by modern society. Aimed at researchers and biotechnologists in academia and industry, it represents essential reading for anyone interested in fungal biotechnology, as well as those working within the broader area of microbial biotechnology. Written in an accessible language, the book also offers a valuable reference resource for decision-makers in government and at non-governmental organizations who are involved in the development of cleaner technologies and the global bioeconomy. The 21st century is characterized by a number of critical challenges in terms of human health, developing a sustainable bioeconomy, facilitating agricultural production, and establishing practices that support a cleaner environment. While there are chemical solutions to some of these challenges, developing bio-based approaches is becoming increasingly important. Filamentous fungi, ‘the forgotten kingdom,’ are a group of unique organisms whose full potential has yet to be revealed. Some key properties, such as their exceptional capacity to secrete proteins into the external environment, have already been successfully harnessed for the production of industrial enzymes and cellulosic biofuels. Many further aspects discussed here –such as feeding the hungry with fungal protein, and the potential applications of the various small molecules produced by fungi –warrant further exploration. In turn, the book covers the use of fungal cell factories to produce foreign molecules, e.g. for therapeutics. Strategies including molecular approaches to strain improvement, and recent advances in high-throughput technologies, which are key to finding better products and producers, are also addressed. Lastly, the book discusses the advent of synthetic biology, which is destined to greatly expand the scope of fungal biotechnology. Abstract:Fungi have been used to better the lives of everyday people and unravel the mysteries of higher eukaryotic organisms for decades. However, comparing progress and development stemming from fungal research to that of human, plant, and bacterial research, fungi remain largely understudied and underutilized. Recent commercial ventures have begun to gain popularity in society, providing a new surge of interest in fungi, mycelia, and potential new applications of these organisms to various aspects of research. Biotechnological advancements in fungal research cannot occur without intensive amounts of time, investments, and research tool development. In this review, we highlight past breakthroughs in fungal biotechnology, discuss requirements to advance fungal biotechnology even further, and touch on the horizon of new breakthroughs with the highest potential to positively impact both research and society.

Abstract:In the future bio-based society, leaving the fossil-based era, a wide range of challenges will have to be solved by new biological solutions. Among the core technologies for this transformation are fungal (and bacterial) production of proteins, metabolites and chemicals and biological processing (using microbial enzymes, substituting for chemical processing). Currently, new biorefinery technologies are being developed, aimed at converting crop residues and industrial sidestreams and waste streams to higher-value products. The new biorefinery development started with conversion of wheat straw and corn stover to biofuel. Over the last 5–10 years, it has expanded rapidly into cascading biomass-conversion processes, unlocking the full potential of the biomass and producing high-value products, such as food and feed ingredients as well as specialty chemicals and materials. Fungi have taken centre-stage in this development. In this chapter, we provide a comprehensive overview of what comes next, depicting in which areas fungi, fungal enzymes and fungal metabolites can contribute significantly to addressing global challenges, first and foremost related to mitigating climate change and providing sufficient food for a rapidly growing global population. However, fungi can do much more! The chapter reviews a broad spectrum of such opportunities for creation of societal and environmental value: opening paths towards improved health for humans and animals, discovery of new drug candidates and reducing risk of developing lifestyle diseases and antibiotic resistance and substituting agricultural pesticides with biologicals. For all the areas reviewed where fungi can be of major importance for contributing to a more sustainable world, we pinpoint where new biotechnology challenges have to be overcome to unlock the opportunities.

Abstract:Fungi have been extensively studied for their capacity to biotransform a wide range of natural and xenobiotic compounds. This versatility is a reflection of the broad substrate specificity of fungal enzymes such as laccases, peroxidases and cytochromes P450, which are involved in these reactions. This review gives an account of recent advances in the understanding of fungal metabolism of drugs and pollutants such as dyes, agrochemicals and per- and poly-fluorinated alkyl substances (PFAS), and describes the key enzymes involved in xenobiotic biotransformation. The potential of fungi and their enzymes in the bioremediation of polluted environments and in the biocatalytic production of important compounds is also discussed.

Abstract:The environmental problems caused by the use of fossil fuels have increased the worldwide need for biomass use. Fungal technology provides a gentle, environmentally friendly, low-cost solution for biomass conversion and is widely used in the production of chemicals and biofuels because of its advantages in lignin degradation, saccharification, fermentation, and lipid accumulation. The causes and consequences of using fungal technology in biomass conversion must be determined, to help us to understand the proper application of fungal technologies in biomass conversion. For this purpose, we reviewed the fungal types involved in biomass conversion, the advantages and limitations of fungal technology, and strategies for improvement. We systematically reviewed the advances in how these advantages and improvement strategies can be effectively combined in the conversion of biomass into value-added products, such as alcohols, gas fuel, biodiesel, organic acids, spices, medicines, compost, and feed. The future directions based on fungi technology have also prospected. It is certain that strain screening, parameter optimization, coupling processes, and the introduction of genetic engineering throughout the application of fungal technology in biomass conversion. Fungal technology deserves more attention and further exploration as a promising sustainable biomass utilization strategy.

Abstract:Fumaric acid is an industrially important platform chemical, which has been widely used in food, chemicals, agriculture and pharmaceutical industries. Due to the rising price of raw materials in traditional petrochemical method and demand for sustainable development, fermentative fumaric acid production has attracted great attention. In this context, various strains with enhanced fumaric acid production or elimination of by-products were developed through mutagenesis or metabolic engineering. In addition, renewable organic wastes together with the fermentation process optimization have also been widely investigated. Accordingly, this review will comprehensively summarize the achievements of fermentative fumaric acid production in recent years and major obstacles occurring in industrialization to provide insights and perspective for future microbial fermentation of fumaric acid.

Abstract:Natural products play important roles not only in the environment but also as useful compounds in various applications like in medicine or plant protection. An enormous number of such compounds have derived from microorganisms colonizing various habitats. Traditionally, new isolates of bacteria or fungi have been screened for their potential to produce biologically active compounds. In the post genomic era, however, there is a growing number of novel methods based on genetic engineering to obtain new metabolites. In this review, we summarize the recent progress made in the development of novel promising approaches for natural product discovery in fungi using genome mining, activation of silent gene clusters, heterologous expression of biosynthesis genes, exchange of enzyme modules as well as redesign of metabolic flux.

Abstract:Fungi are an understudied resource possessing huge potential for developing products that can greatly improve human well-being. In the current paper, we highlight some important discoveries and developments in applied mycology and interdisciplinary Life Science research. These examples concern recently introduced drugs for the treatment of infections and neurological diseases; application of –OMICS techniques and genetic tools in medical mycology and the regulation of mycotoxin production; as well as some highlights of mushroom cultivaton in Asia. Examples for new diagnostic tools in medical mycology and the exploitation of new candidates for therapeutic drugs, are also given. In addition, two entries illustrating the latest developments in the use of fungi for biodegradation and fungal biomaterial production are provided. Some other areas where there have been and/or will be significant developments are also included. It is our hope that this paper will help realise the importance of fungi as a potential industrial resource and see the next two decades bring forward many new fungal and fungus-derived products.

Abstract:Our planet, which operates as a closed system, is facing increasing entropy due to human activities such as the overexploitation of natural resources and fossil fuel use. The COP28 in Dubai emphasized the urgency to abandon fossil fuels, recognizing them as the primary cause of human‐induced environmental changes, while highlighting the need to transition to renewable energies. We promote the crucial role of microbes for sustaining biogenic cycles to combat climate change and the economic potential of synthetic biology tools for producing diverse non‐fossil fuels and chemicals, thus contributing to emission reduction in transport and industry. The shift to 'green chemistry' encounters challenges, derived from the availability of non‐food residues and waste (mainly lignocellulosic) as raw material, the construction of cost‐effective bioprocessing plants, product recovery from fermentation broths and the utilization of leftover lignin residues for synthesizing new chemicals, aligning with circular economy and sustainable development goals. To meet the Paris Agreement goals, an urgent global shift to low‐carbon, renewable sources is imperative, ultimately leading to the cessation of our reliance on fossil fuels.

Abstract:Abstract Fungi have the ability to transform organic materials into a rich and diverse set of useful products and provide distinct opportunities for tackling the urgent challenges before all humans. Fungal biotechnology can advance the transition from our petroleum-based economy into a bio-based circular economy and has the ability to sustainably produce resilient sources of food, feed, chemicals, fuels, textiles, and materials for construction, automotive and transportation industries, for furniture and beyond. Fungal biotechnology offers solutions for securing, stabilizing and enhancing the food supply for a growing human population, while simultaneously lowering greenhouse gas emissions. Fungal biotechnology has, thus, the potential to make a significant contribution to climate change mitigation and meeting the United Nation’s sustainable development goals through the rational improvement of new and established fungal cell factories. The White Paper presented here is the result of the 2nd Think Tank meeting held by the EUROFUNG consortium in Berlin in October 2019. This paper highlights discussions on current opportunities and research challenges in fungal biotechnology and aims to inform scientists, educators, the general public, industrial stakeholders and policymakers about the current fungal biotech revolution.

Abstract:Applications of plant biotechnology play increasingly important role in solving food and energy crisis. As the world’s largest country by population, China pays special attention to the development of agricultural biotechnology and biomass energy technology. Several excellent review articles have introduced the research progress of China’s crop biotech and bioenergy. However, plant biotechnology in China has more diverse applications for non-food uses and environmental sustainability. This paper reviews the recent applications of plant biotechnology in bioenergy, biocontrol, traditional Chinese medicine (TCM), and phytoremediation of polluted environments. The emphasis is mainly focused on the bioconversion of biofuels especially bioethanol and the activated carbon, the survey of energy plants rich in hydrocarbons and carbohydrates, the biocontrol agents (plant epiphytic yeasts) and phytoremediation of heavy metal pollution and water eutrophication. Moreover, plant biotechnology shows very good efficacy in the identifications of confused medicinal plants, active constituents of TCM and the taxonomic identification of plant diseases and insects. The huge number of biomass residue and abundant plant resources provide vast application perspectives for China’s plant biotechnology. Abstract:Fungi are an understudied, biotechnologically valuable group of organisms. Due to the immense range of habitats that fungi inhabit, and the consequent need to compete against a diverse array of other fungi, bacteria, and animals, fungi have developed numerous survival mechanisms. The unique attributes of fungi thus herald great promise for their application in biotechnology and industry. Moreover, fungi can be grown with relative ease, making production at scale viable. The search for fungal biodiversity, and the construction of a living fungi collection, both have incredible economic potential in locating organisms with novel industrial uses that will lead to novel products. This manuscript reviews fifty ways in which fungi can potentially be utilized as biotechnology. We provide notes and examples for each potential exploitation and give examples from our own work and the work of other notable researchers. We also provide a flow chart that can be used to convince funding bodies of the importance of fungi for biotechnological research and as potential products. Fungi have provided the world with penicillin, lovastatin, and other globally significant medicines, and they remain an untapped resource with enormous industrial potential.

Abstract:Endophytes are microorganisms that colonize the interior of host plants without causing apparent disease. They have been widely studied for their ability to modulate relationships between plants and biotic/abiotic stresses, often producing valuable secondary metabolites that can affect host physiology. Owing to the advantages of microbial fermentation over plant/cell cultivation and chemical synthesis, endophytic fungi have received significant attention as a mean for secondary metabolite production. This article summarizes currently reported results on plant-endophyte interaction hypotheses and highlights the biotechnological applications of endophytic fungi and their metabolites in agriculture, environment, biomedicine, energy, and biocatalysts. Current bottlenecks in industrial development and commercial applications as well as possible solutions are also discussed.

Abstract:Pollution is a global menace that poses harmful effects on all the living ecosystems and to the Earth. As years pass by, the available and the looming rate of pollutants increases at a faster rate. Although many treatments and processing strategies are waged for treating such pollutants, the by-products and the wastes or drain off generated by these treatments further engages in the emission of hazardous waste. Innovative and long-lasting solutions are required to address the urgent global issue of hazardous pollutant remediation from contaminated environments. Myco-remediation is a top-down green and eco-friendly tool for pollution management. It is a cost-effective and safer practice of converting pernicious substances into non-toxic forms by the use of fungi. But these pollutants can be transformed into useable products along with multiple benefits for the environment such as sequestration of carbon emissions and also to generate high valuable bioactive materials that fits as a sustainable economic model. The current study has examined the possible applications of fungi in biorefineries and their critical role in the transformation and detoxification of pollutants. The paper offers important insights into using fungal bioremediation for both economically and environmentally sound solutions in the domain of biorefinery applications by combining recent research findings.

Abstract:<span>Current fossil-based commercial products pose a serious threat to global reserves of natural resources and the preservation of the natural environment. During recent decades, great efforts have been made to increase the availability of non-utilizable biomass as alternative feedstocks and reduce environmental pollution to achieve a more sustainable bioeconomy. Several bio-sectors have emerged for the production of bio-based products to replace fossil-based equivalents through bioprocesses using biomass feedstocks. Indeed, advanced microbial fermentation technologies encompassing metabolic engineering and genome-based systems biology approaches have enabled the design and development of new bio-based refineries using engineered platform cells. Herein, we focus on recent progress in the area of microbial fermentation-aided bioprocesses for the production of bio-based products derived from naturally occurring biomasses as feedstocks. Furthermore, we discuss the application of bio-based products and remaining technical barriers and assess possible biorefineries.<b>Graphical </b></span>

Abstract:Fusarium is a large genus of filamentous fungi and widely distributed in soil and associated with plants. Because of its great morphological variation, the classification of Fusarium has always been a thorny problem in the academic circle. With the PCR technology, the identification methods of molecular markers and rDNA analysis of Fusarium improved the accuracy of morphological identification, and laid a foundation for further research and application of Fusarium. Fusarium can produce a variety of key enzymes, including cellulase, pectinase and xylan hydrolase. The important drugs or drug intermediates can be obtained via the bio-transformation, so they have potential commercial value. Fusarium can degrade a variety of environmental pollutants and has important potential in environmental protection. In particular, biological ethanol can be produced through biological transformation to solve the energy crisis. In the paper, the identification and enzyme production of Fusarium species are described in detail. The properties of the main enzymes from Fusarium and the application in the production of bioethanol are reviewed in detail.

Abstract:The fungi-based technology provided encouraging scenarios in the transition from a conventionally based economic system to the potential security of sources closely associated with the agricultural sphere such as the agriculture. In recent years, the intensification of fungi-based processes has generated significant gains, additionally to the production of materials with significant benefits and strong environmental importance. Furthermore, the growing concern for human health, especially in the agriculture scenario, has fostered the investigation of organisms with high biological and beneficial potential for use in agricultural systems. Accordingly, this study offered a comprehensive review of the diversity of the soil fungal microbiome and its main applications in a biotechnological approach aimed at agriculture and food chain-related areas. Moreover, the spectrum of opportunities and the extensive optimization platform for obtaining fungi compounds and metabolites are discussed. Finally, future perspectives regarding the insurgency of innovations and challenges on the broad rise of visionary solutions applied to the biotechnology context are provided.

Abstract:The reliance of the transport sector on fossil fuels has led to increasing concerns related to sustainability and environmental impact. Growing petroleum shortages and global climate change caused by greenhouse gas emissions have given an impetus to the development of renewable and low-carbon energy sources. Fatty acidderived hydrocarbons are an attractive drop-in biofuel to substitute fossil fuels consumed by the transport sector, especially heavy vehicles. Compared to traditional biofuels such as ethanol and biodiesel, fatty acidderived hydrocarbons have higher energy density and better compatibility with existing storage and transport systems. As a common microbial platform for biofuel production, yeasts have been engineered to produce fatty acid-derived hydrocarbons. Here, we attempted to comprehensively review the latest advances in metabolic engineering strategies for the production of fatty acid-derived hydrocarbons in the yeasts Saccharomyces cerevisiae and Yarrowia lipolytica. The heterologous hydrocarbon synthesis pathways and combinational metabolic engineering strategies have been summarized, followed by a discussion of future research directions for the development of yeasts as industrial hydrocarbon producers.
