Abstract:Lipases are ubiquitous in animals, plants and microorganisms, but industrial lipases are mainly produced by microorganisms. The industrial application of lipases is constrained by its high cost and enzymatic characteristics such as temperature and pH tolerance, specific activity, substrate specificity and solvent resistance. Despite the abundance of information regarding microbial lipase genes and proteins, there are currently very few commercial lipase preparations that are suitable for application in the food, pharmaceutical and energy industries. Accordingly, tremendous research efforts have been made to mine novel alternative lipase resources. This article mainly presents a review of the approaches used to mine microbial lipase resources including environmental screening and metagenomics screening for mining new lipases with good properties, genetic improvement of lipase-producing strains, recombinant expression of lipases in genetically engineered strains, lipase modification by protein engineering, lipase immobilization and optimization of reaction conditions of immobilized lipase.

Abstract:Bioenergy is of increasing interest as a renewable, environmentally friendly alternative to energy derived from fossil fuels. Through a variety of processes, biomass can be converted to solid, liquid, or gaseous biofuels. However, various challenges persist to the maintenance and development of biomass-to-energy utilization. This special issue focuses on the most recent advances in the frontier research of bioenergy and biomass utilization and features six selected papers with high quality. The paper titled “Nanofibrillated Cellulose and Copper Nanoparticles Embedded in Polyvinyl Alcohol Films for Antimicrobial Applications” by T. Zhong et al. develops a method to produce hybrids of TEMPO nanofibrillated cellulose (TNFC) and copper nanoparticles. The hybrid material is embedded in polyvinyl alcohol thermoplastic resin and the films are produced using a solvent casting method. The authors evaluate the films in terms of its morphology and thermal and antimicrobial properties and prove that TNFC-copper nanoparticles as antimicrobial nanofillers are valuable for PVA applications. The paper titled “Response of Arbuscular Mycorrhizal Fungi to Hydrologic Gradients in the Rhizosphere of Phragmites australis (Cav.) Trin ex. Steudel Growing in the Sun Island Wetland” by L. Wang et al. assesses the variations of hydrologic gradients in the relationships among AM fungi, reed, and rhizospheric microorganisms. The authors discover water content in soil and reed growth parameters are both positively associated with AM fungi colonization, but only the positive correlations between reed biomass parameters and the colonization could be expected, or both the host plant biomass and the AM fungi could be beneficial. This study could shed light on the mechanisms inside AM fungi-reed symbioses and would be referred to for optimizing the combined phytorhizoremediation. The paper titled “Production by Tobacco Transplastomic Plants of Recombinant Fungal and Bacterial Cell-Wall Degrading Enzymes to Be Used for Cellulosic Biomass Saccharification” by L. Paolo et al. presents work on saccharification of plant biomass. The authors express in tobacco chloroplasts microbial genes encoding five cellulases and a polygalacturonase. Leaf extracts containing the recombinant enzymes show the ability to degrade various cell-wall components under different conditions. In addition, a thermostable xylanase is also tested in combination with a cellulase and a polygalacturonase to study the cumulative effect on the depolymerization of a complex plant substrate. Results demonstrate the feasibility of using transplastomic tobacco leaf extracts to convert cell-wall polysaccharides into reducing sugars, fulfilling a major prerequisite of large scale availability of a variety of cell-wall degrading enzymes for biofuel industry. The paper titled “Enzymatic Saccharification of Lignocellulosic Residues by Cellulases Obtained from Solid State Fermentation Using Trichoderma viride” by T. Sartori et al. presents an approach that uses cellulolytic complex produced by Trichoderma viride in solid state fermentation to hydrolyze lignocellulosic residues. This produced enzyme shows viability in comparison with the commercial cellulase enzyme. The synthesis of cellulases by microorganisms from lignocellulosic residues is a process of great interest, representing the search for renewable sources to replace the fossil energetic matrix. The paper titled “Improving Biomethane Production and Mass Bioconversion of Corn Stover Anaerobic Digestion by Adding NaOH Pretreatment and Trace Elements” by C. Liu et al. describes an approach to improve biomethane production from corn stover. The authors use NaOH pretreatment and trace elements supplementation as a strategy to enhance the biodegradability of corn stover for biomethane production. The approach is compared with only NaOH-pretreated and untreated corn stover, and the paper highlights the supplementation of trace elements. The paper titled “Biochemical Modulation of Lipid Pathway in Microalgae Dunaliella sp. for Biodiesel Production,” by A. F. Talebi et al., explores the use of myoinositol to modulate LP and biodiesel quality. Inclusion of myoinositol in the media indeed improves the total lipid accumulation and biodiesel quality parameters, and this work highlights the fact that the biochemical modulation strategies should be progressively considered in the hope of finding more efficient and economically feasible strategies leading to more viable production systems.

Abstract:MIP-1 alpha is a secreted chemokine which can inhibit hematopoietic stem cells and modulate inflammatory responses. It is also an inhibitor of HIV replication in CD8+ T-cells, MIP-1 alpha is expressed in transformed B cells and can also be induced during cellular activation of CD4+ T-cells and monocytes. We have previously identified a new transcription factor family (the MNP family) whose expression is crucial for the induction of MIP-1 alpha transcription during cellular activation. Monocytes and transformed B-cells normally express MNP-1 strongly and MNP-2 weakly, while T-cells strongly express only MNP-2. In this communication we show evidence identifying a new member of the MNP transcription family, MNP-3, in PMA differentiated HL60 cells.

Abstract:Polyunsaturated fatty acids (PUFAs) of the ω-3 and ω-6 class (e.g., α-linolenic acid, linoleic acid) are essential for maintaining biofunctions in mammalians like humans. Due to the fact that humans cannot synthesize these essential fatty acids, they must be taken up from different food sources. Classical sources for these fatty acids are porcine liver and fish oil. However, microbial lipids or single cell oils, produced by oleaginous microorganisms such as algae, fungi and bacteria, are a promising source as well. These single cell oils can be used for many valuable chemicals with applications not only for nutrition but also for fuels and are therefore an ideal basis for a bio-based economy. A crucial point for the establishment of microbial lipids utilization is the cost-effective production and purification of fuels or products of higher value. The fermentative production can be realized by submerged (SmF) or solid state fermentation (SSF). The yield and the composition of the obtained microbial lipids depend on the type of fermentation and the particular conditions (e.g., medium, pH-value, temperature, aeration, nitrogen source). From an economical point of view, waste or by-product streams can be used as cheap and renewable carbon and nitrogen sources. In general, downstream processing costs are one of the major obstacles to be solved for full economic efficiency of microbial lipids. For the extraction of lipids from microbial biomass cell disruption is most important, because efficiency of cell disruption directly influences subsequent downstream operations and overall extraction efficiencies. A multitude of cell disruption and lipid extraction methods are available, conventional as well as newly emerging methods, which will be described and discussed in terms of large scale applicability, their potential in a modern biorefinery and their influence on product quality. Furthermore, an overview is given about applications of microbial lipids or derived fatty acids with emphasis on food applications.

Abstract:Microalgae, with the merits of environmental adaptability, high photosynthetic efficiency and high lipid content, have become one of the most promising raw materials for bio-fuels production, especially for the biodiesel production. In this paper, the advances of technologies in microalgae harvesting and lipid extraction worldwide are reviewed. Firstly, several methods such as flocculation, centrifugation and filtration, as well as their mechanisms and applications for microalgae recovery are introduced. Then, the practical lipid extraction technologies including organic solvents, supercritical and subcritical fluids are evaluated based on their operation conditions and energy consumption. In particular, a novel method of in-situ lipid milking is introduced to allow synchronous culture and lipid extraction of microalgae. Finally, the main problems within the downstream processing of algal oil are summarized and the possible solutions to produce cost-efficient algal biodiesel are proposed.

Abstract:The Cunninghamella genus has been utilized for the production of PUFA-rich lipids. Therefore, we investigate the impact of plant oil supplementation in the culture medium (soybean oil, rice bran oil, and perilla oil), selected based on their different fatty acid predominant, on lipid production and fatty acid composition in C. elegans (TISTR 3370). All oils significantly boosted fungal growth, each influencing distinct patterns of lipid accumulation within the cells. The cells exhibited distinct patterns of lipid accumulation, forming intracellular lipid bodies, influenced by the different oils. Monounsaturated fatty acids (MUFAs) were found to be the most abundant, followed by polyunsaturated fatty acids (PUFAs) and saturated fatty acids (SFAs) in the fungal lipid cultures. Oleic acid was identified as the primary MUFA, while palmitic acid was the predominant SFA in perilla oil supplements. Remarkably, perilla oil supplement provided the highest total lipid production with arachidonic acid being exclusively detected. The percentage of PUFAs ranged from 12% in the control to 33% in soybean oil, 32% in rice bran oil, and 61% in perilla oil supplements. These findings offer valuable opportunities for advancing biotechnological applications in lipid production and customization, with implications for food and nutrition as well as pharmaceuticals and cosmetics.

Abstract:Abstract Microbial oils have gained massive attention because of their significant role in industrial applications. Currently plants and animals are the chief sources of medically and nutritionally important fatty acids. However, the ever-increasing global demand for polyunsaturated fatty acids (PUFAs) cannot be met by the existing sources. Therefore microbes, especially fungi, represent an important alternative source of microbial oils being investigated. Mucor circinelloides —an oleaginous filamentous fungus, came to the forefront because of its high efficiency in synthesizing and accumulating lipids, like γ-linolenic acid (GLA) in high quantity. Recently, mycelium of M. circinelloides has acquired substantial attraction towards it as it has been suggested as a convenient raw material source for the generation of biodiesel via lipid transformation. Although M. circinelloides accumulates lipids naturally, metabolic engineering is found to be important for substantial increase in their yields. Both modifications of existing pathways and re-formation of biosynthetic pathways in M. circinelloides have shown the potential to improve lipid levels. In this review, recent advances in various important metabolic aspects of M. circinelloides have been discussed. Furthermore, the potential applications of M. circinelloides in the fields of antioxidants, nutraceuticals, bioremediation, ethanol production, and carotenoids like beta carotene and astaxanthin having significant nutritional value are also deliberated.

Abstract:The oleaginous fungus Mortierella alpina has distinct advantages in long-chain PUFAs production, and it is the only source for dietary arachidonic acid (ARA) certificated by FDA and European Commission. This review provides an overall introduction to M. alpina, including its major research methods, key factors governing lipid biosynthesis, metabolic engineering and omics studies. The current research interests in M. alpina focus on improving lipid yield and fatty acid desaturation degree by enhancing fatty acid precursors and the reducing power NADPH, and genetic manipulation on PUFAs synthetic pathways is carried to optimise fatty acid composition. Besides, multi-omics studies have been applied to elucidate the global regulatory mechanism of lipogenesis in M. alpina. Current research challenges lie in strain breeding and cost control due to the coenocytic mycelium, long fermentation period and insufficient conversion rate from carbon to lipid. In this review, we also proposed future research goals based on a multidimensional engineering: obtaining ideal chassis by directional evolution and high-throughput screening; rewiring central carbon metabolism and inhibiting competitive pathways by multi-gene manipulation system to enhance carbon-lipid conversion rate; optimization of protein function based on post-translational modification; application of dynamic fermentation strategies suitable for different fermentation phases. By reviewing the comprehensive research progress of this oleaginous fungus, we aim to further comprehend the fungal lipid metabolism and provide reference information and guidelines for the exploration of microbial oils from the perspectives of fundamental research to industrial application.

Abstract:PURPOSE: Oleaginous yeasts, fatty acids biosynthesis and regulation in the oleaginous yeasts and the fatty acids from the oleaginous yeasts and their applications are reviewed in this article.RESULTS: Oleaginous yeasts such as Rhodosporidium toruloides, Yarrowia lipolytica, Rhodotorula mucilaginosa, and Aureobasidium melanogenum, which can accumulate over 50% lipid of their cell dry weight, have many advantages over other oleaginous microorganisms. The fatty acids from the oleaginous yeasts have many potential applications. Many oleaginous yeasts have now been genetically modified to over-produce fatty acids and their derivatives. The most important features of the oleaginous yeasts are that they have special enzymatic systems for enhanced biosynthesis and regulation of fatty acids in their lipid particles. Recently, some oleaginous yeasts such as R. toruloides have been found to have a unique fatty acids synthetase and other oleaginous yeasts such as A. melanogenum have a unique highly reducing polyketide synthase (HR-PKS) involved in the biosynthesis of hydroxyl fatty acids.CONCLUSIONS: It is necessary to further enhance lipid biosynthesis using metabolic engineering and explore new applications of fatty acids in biotechnology.

Abstract:Oleaginous fungi have attracted a great deal of interest for their potency to accumulate high amounts of lipids (more than 20% of biomass dry weight) and polyunsaturated fatty acids (PUFAs), which have a variety of industrial and biological applications. Lipids of plant and animal origin are related to some restrictions and thus lead to attention towards oleaginous microorganisms as reliable substitute resources. Lipids are traditionally biosynthesized intra-cellularly and involved in the building structure of a variety of cellular compartments. In oleaginous fungi, under certain conditions of elevated carbon ratio and decreased nitrogen in the growth medium, a change in metabolic pathway occurred by switching the whole central carbon metabolism to fatty acid anabolism, which subsequently resulted in high lipid accumulation. The present review illustrates the bio-lipid structure, fatty acid classes and biosynthesis within oleaginous fungi with certain key enzymes, and the advantages of oleaginous fungi over other lipid bio-sources. Qualitative and quantitative techniques for detecting the lipid accumulation capability of oleaginous microbes including visual, and analytical (convenient and non-convenient) were debated. Factors affecting lipid production, and different approaches followed to enhance the lipid content in oleaginous yeasts and fungi, including optimization, utilization of cost-effective wastes, co-culturing, as well as metabolic and genetic engineering, were discussed. A better understanding of the oleaginous fungi regarding screening, detection, and maximization of lipid content using different strategies could help to discover new potent oleaginous isolates, exploit and recycle low-cost wastes, and improve the efficiency of bio-lipids cumulation with biotechnological significance.

Abstract:Abstract The recent energy crisis has triggered significant attention on the microbial synthesis of lipids, which comprise the raw material for biodiesel production. Microbial oil accumulation with filamentous fungi has great potential because filamentous fungi can form pellets during cell growth, and these pellets are much easier to harvest from cell broth. This paper focuses on the cell pelletization process of the oleaginous Mucor circinelloides . We have studied the effect of various cultural conditions on pelletized cell growth and lipid accumulation. This study is the first to report that pH adjustment during cell growth plays a key role in pellet formation of M. circinelloides and describes a handy method by which to induce cell pelletization in submerged fungal cultivation. Our study reveals that cell growth and lipid production are not significantly affected by pelletization and that lipid accumulation is triggered at stressed conditions, such as a high carbon-to-nitrogen ratio and high temperature.

Abstract:With filamentous fungus as parent strain and corn hydrolysate as substrate for deep ferment cultivation,the influences of the parameters such as sugar concentration,nitrogen source,temperature,pH and fermentation time on the filamentous fungus oil production.The optimal fermentation conditions are as follows:corn hydrolysate concentration at 13°Bx;2.4%NaNO3 as carbon source;3% corn plasm;pH 5.5 and below 26 ℃ temperature.The cultivation is for 61 h to get the oil up to 13.69% on a cellular biomass basis with biomass yield of 19.45 g/L.The main components of the microbial lipids are 16-carbon and 18-carbon fatty acids which include palmitic acid(17.16%),palmitoleic acid(1.63%),stearic acid(1.87%),oleic acid(58.37%),γ-linolenic acid(12.44%),α-linolenic acid(6.22%),based on GC analysis.

Abstract:An overview of domestic and foreign recent status on bio-oil analysis and characterization was presented,which focused on the applications of some spectroscopy and chromatography techniques in chemical structure characterization of the small organic molecules,such as,Fourier transform infrared spectroscopy(FTIR),nuclear magnetic resonance(NMR),mass spectrometry(MS),gas chromatography(GC),high performance liquid chromatography(HPLC) techniques,as well as the one-and two-dimensional chromatography coupling technology.The evaluations on the role and value of these techniques in structure characterization were afforded,and the detailed descriptions of the quality standards and some typical analysis methods of bio-oil were involved,and then the effects of physical and chemical properties of bio-oil on its separation and analysis were discussed.Moreover,the structure characterizations of oligosaccharide and pyrolytic lignin remained in bio-oil were introduced.Finally,the main problems existed in bio-oil separation and analysis were summarized,and the prospect on the development trend for bio-oil separation and analysis was presented.

The properties and standard as well as research advances and application of biodiesel in the world were introduced, and the methods for production of biodiesel by transesterification including homogeneous chemical catalysts, heterogeneous chemical catalysts, biocatalyst reaction and supercritical technology were reviewed. Moreover, the future prospects of biodiesel in China were pointed out.

Elicitation is a strong tool to enhance the productivity in microbial cells to obtain more compounds of interest. In this study, in order to improve the fungus growth and its terpenoids production, various vegetable oils were added in the fermentation broth of A. cinnamomea. It was found that corn oil from a group of vegetable oils exhibited the best effect on the biomass and triterpenoid content. After optimization, the initial addition of 1% (v/v) corn oil plus the inoculation of 10% (v/v) mycelia led to a maximum triterpenoid yield (532.3 mg L−1), which was increased as much as fourfold compared to the blank control. Differential transcriptome analysis demonstrated that corn oil significantly enriched several metabolic pathways including glycolysis/gluconeogenesis, propanoate metabolism and transmembrane hydrophobins. The enriched pathways interacted with deferentially expressed genes (DEGs) induced by corn oil treatment. Our research provides a potential strategy for the large production of triterpenoids by the improved fermentation of A. cinnamomea.

Abstract:The development of liquid bioenergy bio-oil from microalgae has recently attracted much attention, while most of them focused on single feedstock. In this work, super/subcritical liquefaction was applied for bio-oil production from hybrid microalgae. The effects of liquefaction conditions on yield of products were investigated. The results show that the hybrid microalgae Chlorella and Spirulina with an appropriate mass ratio present synergistic effects for liquefaction. Lanthanum oxide as catalysts in hydrothermal liquefaction system can improve the quality of the bio-oil. Supercritical alcoholic liquefaction systems have much higher bio-oil yield, and the maximum yield of bio-oil reaches 74.71%. The solvent methanol and ethanol in supercritical conditions could upgrade the components of the bio-oils and enhance their combustion performances. The comprehensive properties of the bio-oil from hybrid microalgae under supercritical liquefaction conditions show a potential prospect for application.
