A Novel and Rapid Approach to Fluorescent Automated Lab-In-Syringe Sensing of Ammonium

Abstract:The determination of ammonium is vital to oceanography and marine biology, as it significantly impacts nitrogen cycling and nutrient balance in marine environments. The study aims to establish a rapid and highly sensitive online method for detecting ammonium at room temperature and without the need for solid phase extraction. An innovative automated Lab-In-Syringe rapid fluorescence approach for detecting ammonium in natural waters is introduced in this study. The methodology underlying this approach revolves around the fluorescence reaction of M2OPA with ammonium, offering a promising avenue for efficient and precise detection. At room temperature, the detection limit was determined to be 9.04 nmol/L, and the reaction was completed within 300 s. The evaluation of salinity and carry-over effects did not indicate any significant interference when utilizing the established method for determining ammonium. Additionally, the impact of the 21 types of organic nitrogen compounds on this method is essentially negligible. The method was effectively utilized to quantify the concentration of ammonium in both seawater and river water, affirming its applicability to ammonium determination in natural water. Rigorous evaluation demonstrated the stability of the M2OPA reagent for a minimum of 6 days, even when stored at room temperature. In conclusion, compared with previously reported fluorescence methods, the present method exhibits rapid reactivity at room temperature, making it suitable for practical implementation in challenging environments, including field settings and aboard maritime vessels. It also simplifies the design of the instrument. Furthermore, the implementation of Lab-In-Syringe not only conserves energy but also reduces reagent consumption, aligning with the principles of Green Analytical Chemistry.
