Abstract:An ion chromatograph single-pump column-switching system was developed for the determination of ammonium in high salt matrices. The simple system included a pump, a suppressor, two valves, two columns, a single eluent, and a conductivity detector. Using this method, both online collection and matrix elimination were achieved. Ammonium was eluted from the acceptor loop to the analytical column circularly. Under the optimized separation conditions, the method showed good linearity (r = 0.9997) in the range of 0.05-2.0 mg/L and satisfactory repeatability (RSD > 4%, n = 6). The limit of detection was 4 mu g/L (S/N = 3) with 25 mu l injection. The utilization of the method was demonstrated by the analysis of seawater, with satisfactory spiked recoveries of ammonium between 98.8% and 108.7%. The result showed that the single-pump column-switching system was convenient and practical for the determination of low-level ammonium in high-salinity samples.
Flow-injection analysis with fluorescence detection for the determination of trace levels of ammonium in seawater

Abstract:A method using flow-injection, gas-diffusion, derivatisation and then fluorescent detection has been established for ammonium ion determination in seawater. The fluorescent derivative formed by reacting ortho-phthaldialdehyde (OPA) and sulfite with ammonia gives high sensitivity while removing potential interferences. This is required to measure the low concentrations of ammonium often seen in the open ocean. The experimental conditions (flow-rate, reagent concentrations, membrane configurations, etc.) were manipulated to improve performance. For a sample throughput of 30 samples h(-1), the limit of detection was 7 nM, the coefficient of variation was 5.7% at 800 nM, and the calibration curve was linear to at least 4 micromol L(-1). Interferences were minimised by a gaseous diffusion step. Volatile small molecular-weight amines as interferents were discriminated against by this method. They neither passed through the membrane as efficiently as ammonia, nor reacted as readily with OPA when sulfite was the reductant. Contamination by ammonia from laboratory and shipboard sources complicates application of the method to natural waters, especially measurement of low concentrations (<100 nM) in open-ocean waters. Steps to overcome contamination are described in detail. Some results are presented for ammonium determination in Southern Ocean and Huon Estuary (Tasmania) waters.

Abstract:An improved method for the determination of ammonium in seawater was established using purge-and-trap pretreatment coupled with flow injection fluorescence. An appropriate volume of sodium hydroxide was added to seawater to convert ammonium to gaseous ammonia that was purged from the solution to the vapor phase by argon at 350 mL/min for 15 min at 80°C and trapped in hydrochloric acid. The solution was analyzed based on the fluorescence detection of the reaction product of o-phthaldialdehyde, sulfite, and ammonium. Parameters affecting the purge-and-trap efficiency and determination were optimized, and the matrix effects of seawater were investigated. The influence of primary amines and amino acids was tested and negligible effects were found. The detection limit was 7.4 nmol/L. The relative standard deviation was 4.4% for a seawater fortified with 200 nmol/L ammonium. Compared to the use of gas diffusion cell, this approach offers higher sensitivity, higher collection efficiency, and negligible effects from salinity. In addition, deionized water was used for standard preparation instead of matrix-matched standards.

Abstract:In order to study the ability of peroxydisulfate in degrading ammonia nitrogen and other nitrogen compounds in water, it is necessary to analyze the content of nitrogen compounds and peroxydisulfate in the water.Simultaneous determination of ammonia nitrogen and peroxydisulfate in lake water by dual system ion chromatography was proposed.Ammonia nitrogen could be decomposited by peroxydisulfate under the proper condition of light, heat and transition metal ions.KOH and MSA were used as eluent, and suppression conductivity detector was used for detection.The detection limits (LODs) of ammonia nitrogen and peroxydisulfate were 0.005 mg·L-1 and 0.009 mg·L-1, respectively, based on the signal-to-noise ratio of 3 (S/N=3) and the 25 μL injection volume.Relative standard deviations (RSDs) for retention time and peak area were all less than 1.78%.The easy and fast method was performed with satisfactory recoveries between 98.2% to 103.5% for all ions.It provides a convenient, rapid and effective analysis method for environmental monitoring.

Abstract:An analytical method that uses multichannel flow injection is reported to determine ammonia in seawater. The sample throughput was twenty-eight samples per hour. The linear dynamic range for ammonia was from 20.0–1000.0 μg N · L−1 with a limit of detection of 0.27 μg N · L−1, and recovery values between 98.1% and 104.6%. The relative standard deviation values for nitrogen as ammonia were 2.15–3.33% (n = 6). Statistical t-test analysis indicated that analysis by the flow injection method provided results that were statistically the same as standard procedures. The reported method was suitable for the determination of ammonia in seawater.

A flow injection-fluorometric method for the determination of ammonium in fresh and saline waters with a view to in situ analyses

Abstract:A version of the orthophthaldialdehyde-fluorescence ammonium determination for flow injection analysis (FIA) is presented here, with a view to its use for in situ, low-power consumption systems. Thus, the reaction temperature was limited to 30 degrees C and FIA was used in stop-flow mode (3 min stop). The calibration is linear up to 50 mumol l-1, but concentrations up to 100 mumol l-1 can be measured. Repeatability is around 1% in the range of 0.5-4 mumol l-1 and the detection limit is about 0.03 mumol l-1. Over the salinity range of 5-35 (seawater practical salinity scale) the salt effect is almost negligible (within +/- 2%); and below salinity of 5 it increases to a maximum of -9% in fresh water compared to seawater. Hydrogen carbonate, dissolved oxygen and turbidity (either suspended sediments or phytoplankton cells) have almost no adverse effect in a wide range of concentrations, covering most natural water conditions. Relative interference of primary amines is negligible and mercury (a common sample preservative) does not depress the signal up to 20 mg l-1 Hg II. Sulfide, that may be present in areas with anoxic waters, depresses the signal only slightly and linearly (-9% at 100 mumol l-1 S2-). The method appears to be convenient for the determination of ammonium in most coastal, estuarine and fresh waters. Sample throughput is 9 h-1. The performance of the method can be improved, either by increasing the reaction time (low throughput) or, if enough energy is available, by increasing the reaction temperature (non-stop-flow mode, high throughput). Combining FIA and fluorometry appears to be interesting for in situ determination (submersible devices) of dissolved compounds in environments with variable salinity and turbidity (especially coastal and estuarine waters).

Abstract:A multisyringe flow injection system (MSFIA) coupled to a gas-diffusion cell has been developed for the conductometric determination of ammonium in different water samples. Operation strategies, membrane, reagent concentrations, and flow rates have been studied to optimize the sensitivity of detection and to fit the required working range. The proposed MSFIA system has been compared with former FIA and SIA systems using gas diffusion. The system was applied to the determination of ammonium in water samples of different matrices in order to evaluate its performance. These samples were coastal waters, pond waters, and compost aqueous extracts. Good recoveries of 102 ± 13% were obtained and no significant differences with the reference methods were found. The system can be used for a wide concentration range of ammonia, from 0.075 to 360 mg L−1, without sample dilution and with a precision better than 2% of RSD. The throughput of the method was 32 injections per hour.

Abstract:<p>This work presents a lab-made automatic flow-batch system for headspace single-drop microextraction and on-drop conductometric sensing ammonium. Sample and NaOH solution are simultaneously pumped into a reaction chamber (RC), where ammonium is converted to ammonia by raising pH. The converted ammonia then diffuses into the headspace of the RC, and reacts with a 100 mM boric acid drop. The conductivity of the drop is measured by an on-drop conductivity probe, which is made by two stainless-steel contacting electrodes. The result shows that the increasing rate of conductivity has a linear relationship to the ammonium concentration in sample (R<sup>2</sup>=0.9945). This method has a linear range up to 400 μM, a limit of detection 2.8 μM, a relative standard deviation of 3.0% (200 μM, n=10) and carryover coefficient 0.028. Measurements of river waters, lake waters and wastewaters have been demonstrated. The recoveries have achieved from 99.0-114%. This method avoids using of harmful or odorous reagents and follows the concept of green chemistry.</p>

Abstract:Crude aqueous extract from the orchid ‘Dendrobium Sonia earsakul’ was utilised as a natural product reagent in flow injection analysis (FIA) incorporating a gas diffusion unit (GD) for the determination of ammonia nitrogen. Sample solution was injected into a NaOH donor stream to generate ammonia gas (NH3). In the GD unit, NH3 diffused across a PTFE gas-permeable membrane into the acceptor stream of the orchid extract. As the result, the aqueous orchid reagent became more alkaline and its colour changed from purple to green. The change in the colour of orchid acceptor correlated with the concentration of ammonia nitrogen in the sample and its absorbance monitored by a spectrophotometer at 600 nm. Ammonia nitrogen in chemical fertiliser samples and wastewater samples from agricultural fields were determined and reported as %N (w/w) and mg N L−1, respectively. For chemical fertilisers which contained high content of ammonia nitrogen, a flow rate of 1.0 mL min−1 and injection volume of 100 µL were used with a linear range of 5–40 mmol L−1 and detection limit of 2.12 mmol L−1. However, a higher sensitivity was required for wastewater samples having low ammonia nitrogen content. The flow rate was reduced to 0.3 mL min−1 and the injection volume increased to 1000 µL. As a result, detection limit of 0.76 mmol L−1 was achieved with linear range of 1–5 mmol L−1. The results of our method agreed well with that using the OPA method employing fluorescence detection.

Abstract:N-(1-Naphthyl)ethylenediamine-reversed phase flow injection-spectrophotometric determination of trace nitrite nitrogen in water was studied. N-(1-Naphthyl)ethylenediamine dihydrochloride solution was injected into the mixed flow of the water sample and sulanilamide solution. Red dye formed by the reactions was spectrophotometrically detected at λmax 540nm. The linear range is 0.004 to 0.18mg/L nitrite nitrogen with the detection limit of 0.0012 mg/L and the determination frequency of 50 times per hour. This method has high sensitivity, good selectivity and fast analytical speed.It has been applied to the determination of trace nitrite nitrogen in Xuanwu Lake water in Nanjing with relative standard deviation of 3.5% for the determination results and mean relative error of -4.2% for the determination results compared with that of N-(1-naphthyl)ethylenediamine spectrophotometry method (national standard method), indicating satisfactory precision and accuracy of the determination results.

Abstract:In the last few decades, various techniques, including spectroscopic, mass spectrometric, chemiluminescence and wet chemical methods, have been developed and applied for the detection of gaseous ammonia (NH3). We developed an online NH3 monitoring system – salicylic acid derivatization reaction and long-path absorption photometer (SAC-LOPAP) – based on a selective colorimetric reaction to form a highly absorbing reaction product and a LOPAP, which could run stably for a long time and be applied to the continuous online measurement of low concentrations of ambient NH3 by optimizing the reaction conditions, adding a constant-temperature module and liquid flow controller. The detection limit reached with this instrument was 40.5 parts per trillion (ppt) with a stripping liquid flow rate of 0.49 mL min−1 and a gas sample flow rate of 0.70 L min−1. An inter-comparison of our system with a commercial Picarro G2103 analyzer (Picarro, USA) in Beijing was presented, and the results showed that the two instruments had a good correlation with a slope of 1.00 and an R2 of 0.96, indicating that the SAC-LOPAP instrument involved in this study could be used for the accurate measurement of NH3.
A Reversible Spectrophotometric Method Based on a Coupled Microfluidic Chip for Highly Selective Ammonium Detection

Abstract:A coupled chip aiming at economical and highly selective ammonium detection was fabricated. It consisted of a reaction chip, a gas-diffusion chip, and a detection chip. Zinc tetraphenylporphyrin dyed on the cation-exchange resin microbeads was used as the indicating material to avoid excess consumption for its reversibility. PDMS was selected as the material of the gas-diffusion membrane. A portable spectrometer was applied for spectrum analysis. By analysis of spectrum change, the high selectivity was confirmed because no component had interference on detection effect. Good performance was shown for all the tested concentrations (0.2–50 mg·L−1). The stability and reversibility were also judged by the spectrum data obtained from the indicating process and the recovering process. Finally, real samples containing ammonium were tested and the results were compared to those came from a standard method to confirm the accuracy of our method.
Simple and sensitive determination of trace nitrite in water by zero-crossing first-derivative synchronous fluorescence spectrometry using 6-amino-1,3- naphthalenedisulfonic acid as a new fluorescent probe

Abstract:A new fluorescent probe, 6-amino-1,3-naphthalenedisulfonic acid (ANDSA), has been developed for the determination of trace nitrite in different waters. This probe is based on the selective reaction of nitrite with ANDSA in hydrochloric acid solution to form the corresponding diazonium acid in sodium hydroxide solution at room temperature. The diazonium acid produced has high fluorescence intensity at 488 nm (excitation at 367 nm), whereas ANDSA has high fluorescence intensity at 465 nm (excitation at 354 nm). The synchronous fluorescence (Δλ = 121 nm) spectrum and the first-derivative synchronous fluorescence spectrum of diazonium acid greatly overlapped with those of ANDSA. The zero-crossing method was used to measure the first-derivative value (dF/dλ) of the first-derivative spectra, in which physical separation of excess ANDSA was unnecessary. The zero-crossing point was located at 351.2 nm for ANDSA, at which dF/dλ of diazonium acid was proportional to the nitrite concentration. Therefore, dF/dλ at 351.2 nm was selected as the analytical signal. Under the optimized experimental conditions, the signal intensity was linear over a nitrite concentration range of 0.006–0.075 mg L−1, with a correlation coefficient better than 0.9994. The limit of detection was 2.1 μg L−1 for the determination of nitrite by the proposed method. The relative standard deviation of the method for the determination of nitrite in real water samples was below 2.45%, and the corresponding recoveries were between 95.7% and 104.1%. The validity of the proposed method was further confirmed by comparison with the reference method with use of the t test.Graphical abstractANDSA reacts with nitrite to form diazonium acid with higher fluorescence intensity. Abstract:The ultraweak chemiluminescence (CL) from the reaction of hydrogen peroxide and carbonate is strongly enhanced by the branched NaYF(4):Yb(3+)/Er(3+) nanoparticle (NP) in the presence of aqueous ammonia. It was explained that ammonia catalyzes the decomposition of peroxymonocarbonate, which is the product of hydrogen peroxide mixing with bicarbonate, making the formation of (CO(2))(2)*, (O(2))(2)*, and (1)O(2). The excitation energy, carried by these emitter intermediates, can be transferred to NaYF(4):Yb(3+)/Er(3+) NP. The CL intensity is directly proportional to the concentration of ammonia present in the solution. A flow-injection CL system with high sensitivity, selectivity, and reproducibility is proposed for the determination of aqueous ammonia. The proposed method exhibited advantages in a larger linear range from 0.5 μmol L(-1) to 50 μmol L(-1) and a lower detection limit of 1.1 × 10(-8) mol L(-1) (S/N = 3). This method has been successfully applied to the evaluation of ammonia in water samples with recoveries from 95% to 108%. The relative standard deviations are 1.8% and 4.1% for intra-assay and inter-assay precision, respectively.

Abstract:Simultaneous determination of 2,4-, 2,6-toluenediamine (2,4- and 2,6-TDA), aniline, o-toluidine, benzidine, p-chloroaniline, 4,4'-diaminodiphenyl methane (4,4'-DADPM), m-nitroaniline and 1-naphthlamine by ion chromatography with direct current-amperometric detection is described. The DC-amperometry detection, was fixed in voltage of 0.80 V. The CG12 (2mm) and CS12 (2mm) column used for separation, 5% CH3CN+35 mmol/L H2SO4 (0.5 min) to 27% CH3CN and 35 mmol/l H2SO4 (35 min) were were used as gradient eluent with flow rate of 0.25 mL/min. The detection limits of 2, 6- and 2, 4-TDA, aniline, o-toluidine, benzidine, p-chloroaniline, 4,4'-DADPM, m-nitroaniline and 1-naphthlamine are 46 mug/L, 7.25 mug/L, 4.99 mug/L, 7.43 mug/L L, 21.21 mug/L, 16.3 mug/L, 2.60 mug/L, 201.47 mug/L, 22.60 mug/L (S/N=3), respectively. Experiment results show that linear relationship and repeatability are good. The method has been used to determine benzidine, p-chloroaniline and 1-naphthlamine in waste water. The results are satisfactory.
An All-Solid-State Ammonium Ion-Selective Electrode Based on Polyaniline As Transducer and Poly (O-Phenylenediamine) As Sensitive Membrane

Abstract:An all-solid-state ammonium ion-selective electrode, with silver/nano-silver wire as the substrate, polyaniline as transducer and poly (o-phenylenediamine) as sensitive membrane, is presented. The nano-silver was prepared by electrodeposition in 0.1 M AgNO3 standard solution with the half-sine wave at amplitude of 500 mV, frequency of 50 HZ and action time of 1 min. The polyaniline and poly (o-phenylenediamine) were electropolymerized on the surface of the silver/nano-silver substrate by cyclic voltammetry with the scanning potential from 0 to 0.45 V in solutions of 1.0 M aniline hydrochloride and 0.08 M o-phenylenediamine hydrochloride, respectively. Under the SEM, poly (o-phenylenediamine) with 0.5-5 mu m in size was evenly and firmly embedded on the surface of the PANI. This ammonium electrode exhibited low impedance of approximately 70 Ohm. It can detect as low as 1.2 x10(-5) M of ammonium ion, with the slopes of 54.99-55.70 mV per decade (R-2>0.99) within a linear range between 2 x 10(-5) M to 0.1 M. It responds as quickly as 0.5-2 s, as well as its lifespan extends to at least 147 days. The electrode showed a superior selectivity toward NH4+, and was successfully applied to detect the NH4+ concentration in real samples. In addition, this electrode is miniature and convenient for package and assembly, which will enable in-situ measurement and long-term monitoring of NH4+ concentration in rivers, lakes and industrial sewage.
The Reversible Ammonium Detection Based on the Coupled Microfluidic Chipand the Investigation of the Impact Factors

Abstract:The spectroscopic detection of ammonium (NH4+) has great significance. With the development of microfluidics, microfluidic spectroscopic analysis aiming at rapid, portable and multi-component detection has realized tremendous achievements. However, the excessive consumption of the indicating material which exists all the time has not been settled yet. Zinc porphyrin, being a natural chromophore, can reversibly detect NH4+ and settle the problem. But it suffers from the lack of selectivity. In allusion to these issues, a coupled microfluidic chip which consists of a reaction chip, a gas-diffusion chip and a detection chip is fabricated in our experiments. The reaction chip made of polydimethylsiloxane can convert NH4+ into NH3. The gas-diffusion chip which is made of two glass chips and a piece of polydimethylsiloxane gas-diffusion membrane allows NH3 diffuses into the detection solution. The detection chip immobilizes the indicating material tightly through the multi-layer structure. The indicating material fabricated by dying zinc porphyrin on the surface of the cation exchange resin permanently will turn from green to purple when it meets NH3 and will turn reversely into green when pure water is around. Using the coupled chip as the analytical platform, we set up a miniaturized spectroscopic detection system. With a portable spectrometer being the analytical instrument, we detect NH4+ by the transmission spectrum intensity change at the wavelength of 450 nm and study three factors that could affect the detection performance: the thickness of the gas-diffusion membrane, the flow rate and the dosage of the indicating material. First, we confirm the reversibility of the indicating process and settle the issue of indicating material consumption by the spectrum change intensity. The time responses show the rapidity of the indicating process. Then through the comparison to the contrast experiment, the high selectivity of the indicating process is demonstrated because the interference has no influence and the spectrum change is attributed merely to NH3. The relationship between the membrane thickness and the spectrum intensity change is obtained by changing the thickness of the gas-diffusion membrane in the gas-diffusion chip. Results show that the increase of the membrane thickness leads to a worse detection performance, but the outcome remains stable when the thickness is less than 10 mu m. So 10 mu m is selected as the optimal thickness in consideration of the mechanical strength. The impact of the flow rate is investigated afterwards. The results exhibit that the increase of the flow rate will decrease the spectrum intensity change, but the performance becomes unchanged when flow rate is less than 5 mu L . min(-1). Last, how the dosage of the indicating material impacts the results is studied. It is demonstrated that both excessive and inadequate indicating material will deteriorate the performance. So 5 mg is selected as the optimal amount. The spectroscopic detection system which uses the coupled chip as the analytical platform has the advantages of small volume, high economy, rapid response and realizes highly selective and reversible NH4+ detection.

Abstract:A bidirectional selection experiment was conducted to measure 5-day antibody titers to sheep erythrocytes in White Leghorn chickens. There was an immediate response to selection with significant differences between lines for the selected trait found in the S1 and all subsequent generations. Comparisons of S6, S7 and S8 generation females revealed differences between lines in disease resistance and in certain reproductive traits such as age at first egg, percentage hen-day egg production, percentage fertility and duration of fertility. The implications of these correlated responses are important to selection programs for general disease resistance.

Abstract:A flow injection analysis system (FIA) for the simultaneous determination of salinity, carbonate and ammoniacal nitrogen has been developed and reported in this paper. FIA incorporating membrane units was used, not only for the separation of the gaseous carbon dioxide and ammonia, but also for on-line dilution in the salinity measurement. The sample was injected via a 10-port valve with two sample loops. One loop was used for salinity and carbonate measurements and the second loop for ammoniacal nitrogen determination. A dual-channel capacitively coupled contactless conductivity detector was assembled in a single shielding box. Input voltage from the same AC power supply was fed to the input electrodes of both C4D cells. One channel of the C4D was used to monitor the change in conductivity of an acceptor stream that carried a zone of the water sample that has passed through the on-line dilution unit. Conductivity of this zone relates directly to the salinity of the sample. The same sample zone was next acidified to generate carbon dioxide gas that diffused through a hydrophobic membrane of the first gas diffusion (GD) unit. The zone of dissolved carbon dioxide in acceptor stream of water flowed into the same C4D cell as for the salinity measurement, but arriving at a later time. Concurrently, the second channel of the C4D monitored the change in conductivity of the acceptor stream in the second GD unit due to the diffusion of ammonia gas generated by the reaction of base with the sample injected from the second sample loop. The change in conductivity at this second C4D cell correlates with the concentration of ammoniacal nitrogen present in the sample. The proposed method is low cost, simple, rapid and sensitive. The limit of quantitation for salinity, carbonate and ammoniacal nitrogen are 0.31mmolL-1, 1.85 µmol L-1, respectively. Throughput of 20 samples h-1 for simultaneous analysis can be achieved with RSD of less than 3.8%. The system had been applied to the determination of salinity, carbonate and ammoniacal nitrogen in 15 water samples, with results in agreement with those obtained using comparison methods.
