Abstract:A sustainable, cyanide-free synthesis of alkyl nitriles via the aerobic oxidative deconstruction of unstrained cycloalkanones with ammonium salts has been developed. Using inexpensive and stable ammonium salts as the nitrogen source, a variety of alkyl nitriles containing a distal carbonyl group were obtained in good yields under visible-light-promoted iron catalysis. This protocol is characterized by mild conditions, abundant and environmentally benign materials, and high atom and step economy with minimal waste generation. The primary mechanism study revealed that 1O2 is likely to be involved in this reaction.

Abstract:A photoredox-catalyzed sequential decarboxylative/defluorinative aminoalkylation of CF3-alkenes with N-arylglycines is described. This metal-free and redox-neutral protocol provided efficient access to the monofluoroalkenyl-1,5-diamines in good yields with excellent functional group compatibility. Mechanistic studies revealed that the reaction proceeds via a radical pathway with the gem-difluoroalkenyl amine as an intermediate.

Abstract:We report a Pt-catalyzed double deoxygenative transformation of N-acyloxyamines, employing PtCl 2 catalyst without external ligand under mild conditions. This method facilitates efficient C–N bond formation and can be used to synthesize a broad spectrum of alkylated secondary and tertiary amines with excellent yields. Abstract:A deoxygenative radical alkylation of aromatic alkynes with alkyl aldehydes for the preparation of allylarenes has been successfully achieved. This transformation is accomplished through the reaction of alkyl aldehydes with alkynes in the presence of dipropylamine and Hantzsch ester catalyzed by a P/N heteroleptic Cu(I)-based photosensitizer under photoredox catalysis conditions. Preliminary mechanistic studies reveal that this aldehyde-alkyne coupling process comprises a regioselective radical addition of in situ-generated alkyl-substituted alpha-amino radicals to alkynes with subsequent 1,5-proton transfer, C-N bond cleavage, and concomitant isomerization of the resulting allyl radical species. Thus, in net result, dipropylamine serves as a traceless linker agent for the deoxygenative radical cross-coupling of alkyl aldehydes with alkynes under photoredox catalysis reaction conditions.

Abstract:An iron-catalyzed diastereoselective intermolecular olefin amino-oxygenation reaction is reported, which proceeds via an iron-nitrenoid generated by the N-O bond cleavage of a functionalized hydroxylamine. In this reaction, a bench-stable hydroxylamine derivative is used as the amination reagent and oxidant. This method tolerates a range of synthetically valuable substrates that have been all incompatible with existing amino-oxygenation methods. It can also provide amino alcohol derivatives with regio- and stereochemical arrays complementary to known amino-oxygenation methods.

Abstract:Deoxygenative difunctionalization of carbonyls represents a convenient route to construct complex molecules considering the readily available aldehyde and ketone compounds. The present approaches typically rely on strategies via carbene, carbanion, or carbocation equivalents. Herein, combined with dual nickel/photoredox catalysis regime, we developed a strategy through a radical intermediate to achieve the deoxygenative arylboration of aldehydes. Compared with the known patterns of carbon–oxygen (C–O) bond transformations, this coupling of C-OBPin unit opens a direction for this chemistry. A wide variety of substrates bearing a diverse set of functional groups were compatible with this method under very mild conditions (visible light, ambient temperature, no strong base) to afford the benzylic boronic esters, which have important versatilities in organic synthesis.

Abstract:A metal-free and 4-methyl aniline mediated method for the oxidative C-C bond cleavage has been developed. The reaction proceeds in air using molecular oxygen as the oxidant, affording one-carbon shortened esters in moderate to good yields within a short time. Moreover, it provides a model reaction for the highly enantioselective synthesis of (D)-serine esters by combining with a l-proline catalyzed Mannich reaction.

Abstract:A novel Pd/Cu-catalyzed chemoselective aerobic oxidative N-dealkylation/carbonylation reaction has been developed. Tertiary amines are utilized as a "reservoir" of "active" secondary amines in this transformation, which inhibits the formation of undesired by-products and the deactivation of the catalysts. This protocol allows for an efficient and straightforward construction of synthetically useful and bioactive (E)-α,β-unsaturated amide derivatives from easily available tertiary amines, olefins and CO.

Abstract:An approach to the vicinal oxyazidation of alkenes has been achieved under mild and transition metal-free conditions. This method utilizes NaN3 as the azidation agent and 2,2,6,6-tetramethylpiperidine-1-oxoammonium tetrafluoroborate (TEMPO+BF4-) as the single-electron oxidant as well as the oxygen source. By using this protocol, various β-aminooxy azides were synthesized and several complex bioactive molecules were functionalized.

Abstract:The use of molecular oxygen as the terminal oxidant in transition metal catalyzed oxidative process is an appealing and challenging task in organic synthetic chemistry. Here, we report a Ni-catalyzed hydroxylarylation of unactivated alkenes enabled by a beta-diketone ligand with high efficiency and excellent regioselectivity employing molecular oxygen as the oxidant and hydroxyl source. This reaction features mild conditions, broad substrate scope and incredible heterocycle compatibility, providing a variety of beta-hydroxylamides, gamma-hydroxylamides, beta-aminoalcohols, gamma-aminoalcohols, and 1,3-diols in high yields. The synthetic value of this methodology was demonstrated by the efficient synthesis of two bioactive compounds, (+/-)-3 '-methoxyl citreochlorol and tea catechin metabolites M4.

Abstract:Abstract The direct coupling of olefins and alkyl amines represents the most efficient and atom-economical approach to prepare aliphatic allylamines which are fundamental building blocks. However, the method that achieves this goal while exhibiting exquisite control over the site at which the amine is introduced remains elusive. Herein, we report that the combination of a photocatalyst and a cobaloxime enables site-selective allylic C–H amination of olefins with secondary alkyl amines to afford allylic amines, eliminating the need for oxidants. This reaction proceeds by a radical-based mechanism distinct from those of existing allylic amination reactions. It affords the product resulting from cleavage of the stronger, primary allylic C–H bonds over other weaker allylic C–H bond options. DFT calculations reveal that this selectivity originates from a cobaloxime-promoted hydrogen atom transfer (HAT) process. Our method is compatible with a broad scope of alkenes, and can be extended to achieve a site- and diastereoselective amination of natural terpenes.

Abstract:It is of great significance to realize the alkynylation of the reaction since alkynes are widely present in natural products and biologically active molecules. And at the same time alkynes have very rich transformations in synthetic organic chemistry. It is also attractive to functionalize molecules by cleaving inert carbon-carbon bonds albeit with great challenge due to their inherent stability. Recently, significant progress has been made in the cleavage of alkynyl carbon-carbon single bonds for enabling the alkynylation of organic molecules. This review focuses on the recent advances in transition-metal-catalyzed cleavage and transformation of alkynyl carbon-carbon bonds with alkynoic acids, propargylic alcohols, alkynyl esters, and alkynyl ketones.

Abstract:Alkylamine structures represent one of the most functional and widely used in organic synthesis and drug design. However, the general methods for the functionalization of the shielded and deshielded alkyl radicals remain elusive. Here, we report a general deoxygenative amination protocol using alcohol-derived carbazates and nitrobenzene under electrochemical conditions. A range of primary, secondary, and tertiary alkylamines are obtained. This practical procedure can be scaled up through electrochemical continuous flow technique. Secondary arylalkylamines represent important building blocks for organic synthesis, and as such their straightforward synthesis from readily available chemicals remains a priority in organic chemistry. Here, the authors report a synthesis of arylalkylamines from nitroarenes and carbazates via paired electrolysis.

Abstract:Given the natural abundance and industrial accessibility, inert amides have gained considerable attention in homologous amine synthesis and the development of novel catalytic strategies for their deoxygenative functionalization is highly desirable. Although organic electrosynthesis has become an uprising research area by virtue of its sustainability and atom economy, the combination of amide reduction with an electrochemical process in value-added amine synthesis is still undeveloped. Herein, a strategy merging iridium-catalyzed activation of amides with electrochemical radical generation to access deoxygenative arylation of secondary amides is developed. This methodology is featured by broad substrate scope, mild reaction conditions, and operational simplicity, giving corresponding benzylic amines with high efficiency. Notably, the successful application of electrochemical method in the deoxygenative functionalization of amides would provide an efficient strategy for quick access to value-added amines from inert amides.

Abstract:Aliphatic allylamines are widely used for the synthesis of diverse building blocks for agrochemicals and pharmaceuticals; there is therefore considerable interest in developing versatile and direct routes to aliphatic allylamines using common chemical feedstocks—olefins and amines. However, examples of such coupling reactions remain limited. It is even more challenging to achieve this goal with precise site control. Here we report that the combination of a photocatalyst and cobaloxime enables site-selective amination of olefins with secondary alkyl amines to afford allylic amines, eliminating the need for oxidants. This method is compatible with a broad scope of olefins and can be extended to achieve a site- and diastereoselective amination of terpenes. Mechanistic studies disclose that the reaction proceeds via a cobaloxime-promoted hydrogen atom transfer pathway to afford the product that results from cleavage of the stronger, primary allylic C–H bonds over other weaker allylic C–H bond options. Allylic amination of unactivated alkenes with aliphatic amines is a long-standing synthetic challenge in organic chemistry. This is now accomplished in an oxidant-free, site-selective process by using a combination of a photocatalyst and cobalt complex for the coupling of olefins and alkyl amines with hydrogen evolution.

Abstract:A transition-metal-free deacylative C(sp(3))-C(sp(2)) bond cleavage for the synthetically practical oxidative amination of ketones and aldehydes to nitriles is first described, using cheap and commercially abundant NaNO2 as the oxidant and the nitrogen source. Various nitriles bearing aryl, heteroaryl, alkyl, and alkenyl groups could be smoothly obtained from ketones and aldehydes in high yields, avoiding highly toxic cyanides or transition metals.

Abstract:A synthetic idea for the general and efficient utilization of (-chiral) alkyl aldehydes as deoxygenative (chiral) alkyl radical equivalents based on photocatalytic traceless C-N bond formation/cleavage strategy has been proposed and successfully applied in the alkylation/cyclization of 2-biphenylisonitriles with (-chiral) alkyl aldehydes in the presence of amines to give a broad range of 6-alkyl-substituted phenanthridines catalyzed by 4-CzIPN under mild reaction conditions. In this transformation, -chiral alkyl aldehydes, e.g. chiral -amino aldehydes, have been enabled for the first time as deoxygenative chiral alkyl radical equivalents in most cases without erosion of the optical purity of the chiral centers inherited from the corresponding mother aldehyde.
