Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Multicomponent catalytic processes that can generate multiple C(sp3)–C(sp3) bonds in a single step under mild conditions, particularly those that employ inexpensive catalysts and substrates, are highly sought-after in chemistry research for complex molecule synthesis. Here, we disclose an efficient Ni-catalyzed reductive protocol that chemoselectively merges alkenyl amides with two different aliphatic electrophiles. Starting materials are readily accessible from stable and abundant feedstock, and products are furnished in up to >98:2 regioisomeric ratios. The present strategy eliminates the use of sensitive organometallic reagents, tolerates a wide array of complex functionalities, and enables regiodivergent addition of two primary alkyl groups bearing similar electronic and steric attributes across aliphatic C═C bonds with exquisite control of site selectivity. Utility is underscored by the concise synthesis of bioactive compounds and postreaction functionalizations leading to structurally diverse scaffolds. Abstract:Development of a catalytic multicomponent reaction by orthogonal activation of readily available substrates for the streamlined difunctionalization of alkynes is a compelling objective in organic chemistry. Alkyne carboalkynylation, in particular, offers a direct entry to valuable 1,3-enynes with different substitution patterns. Here, we show that the synthesis of stereodefined 1,3-enynes featuring a trisubstituted olefin is achieved by merging alkynes, alkynyl bromides and redox-active N-(acyloxy)phthalimides through nickel-catalyzed reductive alkylalkynylation. Products are generated in up to 89% yield as single regio- and E isomers. Transformations are tolerant of diverse functional groups and the resulting 1,3-enynes are amenable to further elaboration to synthetically useful building blocks. With olefin-tethered N-(acyloxy)phthalimides, a cascade radical addition/cyclization/alkynylation process can be implemented to obtain 1,5-enynes. The present study underscores the crucial role of redox-active esters as superior alkyl group donors compared to haloalkanes in reductive alkyne dicarbofunctionalizations.

Abstract:Abstract The direct coupling of olefins and alkyl amines represents the most efficient and atom-economical approach to prepare aliphatic allylamines which are fundamental building blocks. However, the method that achieves this goal while exhibiting exquisite control over the site at which the amine is introduced remains elusive. Herein, we report that the combination of a photocatalyst and a cobaloxime enables site-selective allylic C–H amination of olefins with secondary alkyl amines to afford allylic amines, eliminating the need for oxidants. This reaction proceeds by a radical-based mechanism distinct from those of existing allylic amination reactions. It affords the product resulting from cleavage of the stronger, primary allylic C–H bonds over other weaker allylic C–H bond options. DFT calculations reveal that this selectivity originates from a cobaloxime-promoted hydrogen atom transfer (HAT) process. Our method is compatible with a broad scope of alkenes, and can be extended to achieve a site- and diastereoselective amination of natural terpenes.

Abstract:Enantioselective transformations of hydrocarbons to three-dimensional chiral molecules remain a significant challenge in synthetic chemistry. This study uses asymmetric paired oxidative and reductive catalysis (APORC) to promote the enantioselective alkylarylation of olefins through the functionalization of C(sp3)−H bonds in alkanes. This asymmetric photoelectrocatalytic approach enables the facile construction of a wide range of enantioenriched α-aryl carbonyls with excellent enantioselectivity (up to 96% ee) from readily accessible starting materials. Notably, aryl bromides, aryl iodides, and even aryl chlorides were compatible with the developed catalytic system. Mechanistic studies reveal that alkanes and electrophiles are simultaneously activated on the electrodes.

Abstract:Highly regio- and enantioselective allylic alkylation has been achieved enabled by the merger of photoredox and palladium catalysis. In this dual catalytic process, alkyl radicals generated from 4-alkyl-1,4-dihydropyridines act as the coupling partners of the π-allyl palladium complexes. The generality of this method has been illustrated through the reaction of a variety of allyl esters with 4-alkyl-1,4-dihydropyridines. This mechanistically novel strategy expands the scope of the traditional Pd-catalyzed asymmetric allylic alkylation reaction and serves as its alternative and potential complement.

Abstract:Despite tremendous efforts aimed at devising methods for stereoselective alkene synthesis, critical challenges are yet to be addressed. Direct access to a diverse range of 1-aryl(boryl)-1-methyl-functionalized tri- and tetrasubstituted trans alkenes, entities that are prevalent in many important molecules, through a catalytic manifold from readily available α-olefin substrates remains elusive. Here, we demonstrate that catalytic amounts of a non-precious N-heterocyclic carbene–Ni(I) complex in conjunction with a sterically bulky base promote site- and trans-selective union of monosubstituted olefins with a wide array of electrophilic reagents to deliver tri- and tetrasubstituted alkenes in up to 92% yield and >98% regio- and stereoselectivity. The protocol is amenable to the preparation of carbon- and heteroatom-substituted C=C bonds, providing distinct advantages over existing transformations. Utility is highlighted through concise stereoselective synthesis of biologically active compounds.

Abstract:Enantio-enriched amines are found in a wide range of bioactive natural products and pharmaceutical agents. However, radical-based asymmetric cross-dehydrogenative amination via electrochemical oxidative C–H/N–H coupling to facilitate catalytic C( sp 3 )–N bond formation remains a largely unsolved challenge in organic synthesis. In the present study, we present nickel-catalysed, anodically coupled electrolysis for the stereoselective, cross-dehydrogenative amination of acylimidazoles, with commercially available nitrogen nucleophiles as coupling partners to access structurally diverse α-amino carbonyls. This method involves the coupling of an electrogenerated nickel-bound α-keto radical species and an aminyl radical to provide a stereoselective approach for the cross-dehydrogenative amination. The utility of this anodic oxidative strategy has been highlighted through the stereoselective synthesis of (+)-γ-secretase inhibitor, (+)-flamprop-methyl and (+)-flamprop-isopropyl.

Abstract:Direct utilization of allylic alcohols as allylic reagents without metal reductants has drawn significant attention. Herein, we report a practical, diastereoselective photoredox/Ni(II)-dual-catalyzed reductive allylation reaction of aldehydes with readily accessible allylic alcohols in the presence of CO2. This methodology allows the branched regioselective direct allylation of a wide range of aldehydes with various allylic alcohols in good to high yield and up to >20:1 d.r. ratio. Accurate DFT calculations revealed the rationale behind the diastereoselectivity differences via Zimmerman-Traxler transition state for different substrates, while also delineated the energetically most favorable mechanistic pathway for this allylation reaction.

Abstract:Chiral nickel complexes promoted enantioselective reductive alkenylation of a range of conjugated enones, using alkenyl bromides, triflates, and tosylates. The neutral condition was compatible with sensitive groups and azaheteroaryl rings. Importantly, alkenyl groups in products can be readily converted to functionalized alkyl groups via iron-catalyzed reductive hydrofunctionalization. Some examples of asymmetric alkenylation of N-enoyl pyrroles and indoles were also included.

Abstract:(Z)-Enamides are important motifs, due to their prevalence in bio-active molecules. However, few strategies are known to obtain them stereoselectively. Herein we report a nickel catalyst which, combined with simultaneous photoredox and energy transfer catalysis, enables the selective formation of (Z)-enamides. Di- and tri- functionalized functionalized olefin products can be obtained in high yields and stereoselectivity, respectively, from a redox-modulated nickel intermediate. Selective and challenging nickel-catalyzed β-H abstraction is attainable through kinetic control facilitated by an external additive.

Abstract:Radical-mediated Hydrogen Atom ion of Csp3-H bonds has become a powerful tool for the asymmetric functionalization of organic feedstocks. Here, we present an asymmetric synthesis of α-aryl amides via carbamoylation of alkylarenes with isocyanates as electrophiles. The synergistic combination of a photoredox and a chiral nickel-catalyst, enables the use of readily available and neutral reagents under mild reaction conditions and provides straightforward access to pharmacologically relevant motifs in enantiomerically pure form.

Abstract:The merger of photoredox and transition-metal catalysis has evolved as a robust platform in organic synthesis over the past decade. The stereoselective 1,4-functionalization of 1,3-enynes, a prevalent synthon in synthetic chemistry, could afford valuable chiral allene derivatives. However, tremendous efforts have been focused on the ionic reaction pathway. The radical-involved asymmetric 1,4-functionalization of 1,3-enynes remains a prominent challenge. Herein, we describe the asymmetric three-component 1,4-dialkylation of 1,3-enynes via dual photoredox and chromium catalysis to provide chiral allenols. This method features readily available starting materials, broad substrate scope, good functional group compatibility, high regioselectivity, and simultaneous control of axial and central chiralities. Mechanistic studies suggest that this reaction proceeds through a radical-involved redox-neutral pathway.

Abstract:A general and practical approach for diverse dealkenylative functionalization of olefin-containing substrates has been developed through the one-pot formation and utilization of pro-aromatic 1,4-dihydropyridazines using tetrazine as the key cycloaddition reagent. Triggered by either excitation or oxidation, the targeted C-C bonds in the 1,4-dihydropyridazine intermediates could be readily cleaved to generate alkyl radicals for subsequent transformations. Diverse carbon-carbon and carbon-hetero bond forming protocols, including Giese-type addition, hydrazination, borylation, Minisci-type alkylation, copper-catalyzed NH alkylation, acylation, alkynylation, cyanation, and azidation, are achieved in a highly modular fashion.

Abstract:Herein, we report a mild and operationally simple photoredox/NHC dual catalysis strategy for the α-carboxylation of tertiary amine C(sp3)-H bonds using diethyl pyrocarbonate. This method offers a novel approach for synthesizing α-amino acid derivatives. The protocol features a broad substrate scope, accommodating both N-aryl tetrahydroisoquinolines (THIQ) and N-methyl aniline and is scalable to gram quantities. Additionally, it is suitable for the late-stage derivatization of certain pharmaceutical compounds.

Abstract:The construction of diverse alkylamines is of importance in drug discovery and natural product synthesis. Herein, we report an efficient photoredox/palladium dual catalysis for the construction of protected 1,2-amino alcohols and 1,2-diamines via asymmetric allylation of α-silyl amines with heteroatom-substituted allenes and 1,3- dienes. This protocol is characterized by its mild conditions, excellent regio-, enantioselectivity, and wide substrate scope, including aliphatic amines or N-heterocyclic substrates especially. Computational and experimental mechanistic studies indicate that the reaction proceeds through a sequence of oxidative addition of Pd(0) catalyst by PPTS, allene insertion, outer-sphere radical substitution-type C–C bond formation, and reduction of Pd(I) by Ir(II). Thereinto, the outer-sphere radical substitution is the enantio- and regio-selectivity determining step.
