Abstract:Purpose: BATF2, a novel IFN-stimulated gene, inhibits tumor cell proliferation, invasion, and migration. The objectives of this study were to determine how BATF2 expression is associated with colorectal cancer progression and patient outcome, to investigate how BATF2 overexpression inhibits hepatocyte growth factor (HGF)/MET signaling, and to elucidate the rationale for combining MET inhibitors with IFN.Experimental Design: BATF2 expression in colorectal cancer tissues was determined and correlated with colorectal cancer patient prognosis. Cultured colorectal cancer cells were used to investigate the effects of BATF2 overexpression on the malignant phenotype of colorectal cancer cells and HGF/MET signaling. Tumor xenograft models were used to validate the effects of BATF2 on colorectal cancer xenograft growth and assess the efficacy of the combination of MET inhibitors with IFNs in colorectal cancer.Results: In colorectal cancer tissues, BATF2 was found to be significantly downregulated, and its expression negatively correlated with MET expression. Decreased BATF2 expression was associated with progression and shorter patient survival in colorectal cancer. BATF2 overexpression promoted apoptosis and inhibited proliferation, migration, and invasion in colorectal cancer cells, as well as dramatically blunted tumor xenograft growth. In addition, MET inhibitors in combination with IFN beta produced synergistic cytotoxicity both in vitro and in vivo.Conclusions: Together, these novel findings suggest that BATF2, a tumor suppressor gene, is a potent negative regulator of HGF/MET signaling in colorectal cancer and may serve as a prognostic tumor marker. Abstract:Colorectal cancer (CRC) is one of the leading causes of cancer death worldwide. Better understanding of the molecular mechanisms of CRC pathogenesis is crucial to identify new therapeutic targets and facilitate treatment decisions. The recent work from our laboratory found that BATF2 deficiency promotes progression in CRC via activation of HGF/MET signaling, which potentially provides a promising therapeutic strategy: IFNs in combination with MET inhibitors [1]. Decreased BATF2 expression predicts poor prognosis in CRC. It is well known that many oncogenes are hyperactivated and suppressor genes are inactivated when non-tumor tissue progressed to invasive carcinoma. BATF2, a novel tumor suppressor and type I IFN-inducible early response gene, has a broader expression profile that includes hematopoietic and non-hematopoietic tissues [2]. Recent evidence suggests that decreased BATF2 expression is associated with an inferior outcome in hepatocellular carcinoma and oral tongue squamous cell carcinoma [3,4]. Our findings that decreased BATF2 correlates with tumor progression and poor CRC patient prognosis strongly support the notion that BATF2 plays a critical role in controlling tumor initiation and subsequent progression and could be used as a candidate marker for CRC diagnosis as well as prognosis [1]. BATF2 inactivates HGF/MET signaling in CRC. HGF/MET signaling is frequently activated in diverse human malignancies; yet the underlying mechanism of HGF/MET activation is not fully elucidated. We first identified BATF2 as an independent factor in cell proliferation inhibition in vitro and xenograft formation blockage in vivo in CRC. Our findings are consistent with the results of Su et al., showing that BATF2 is a potent inhibitory factor that arrests cell growth in prostate cancer, glioma, and melanoma cells [2]. Mechanistically, BATF2 overexpression dramatically impairs activator protein (AP)-1 function via the direct binding with c-Jun (a critical component of AP-1). Coincidently, another report documented that overexpression of AP-1 (c-Jun plus c-Fos) dramatically increased MET promoter activity [5], which was also confirmed in our study. Therefore, a novel pathway comprising BATF2 and c-Jun/AP-1 that inhibits canonical HGF/MET signaling is established. Several studies have shown that activation of HGF/MET signaling promotes the progression of many tumors via PI3K/Akt and/or MAPK/ERK. Our data showed that BATF2 overexpression downregulates MET expression by inhibiting transcription and thereby dephosphorylates Akt in CRC cells. Additionally, the results from immunohistochemistry (IHC) further confirmed our previous findings. MET protein expression detected by IHC was gradually enhanced in tissues from non-tumors that progressed to invasive carcinomas. Furthermore, there was a negative correlation between BATF2 and MET protein expression in CRC [1]. BATF2 is an IFN-stimulated gene. Interferons (IFNs), apart from their function as anti-viral infection agents, exert a variety of inhibitory effects on cell growth, apoptosis, and angiogenesis. IFNs exert potent anti-tumor effects and are used clinically either as a mono-therapy or as an adjuvant to chemotherapy for a variety of human cancers. For example, in hairy cell leukaemia and chronic myelogenous leukemia (CML), IFN-α2 reduced marrow infiltration with malignant cells and normalized peripheral haematological parameters. In addition to hairy cell leukaemia and CML, therapeutic efficacy of IFN-α2 in causing at least partial disease regression has been identified in other diverse human malignancies including melanoma, renal cell and bladder cancer, lymphoma, myeloma, and Kaposi's sarcoma [6]. IFNs induce growth inhibition by a variety of pathways that involve many IFN-stimulated genes. BATF2 is one of these genes and can be induced by IFNβ, which indicates that BATF2 may be a key component involved in IFN signaling. Both Su et al. and we confirmed that BATF2 expression is induced after IFNβ treatment in melanoma cells and CRC cells. Inhibition of BATF2 by an antisense approach in HeLa cells significantly abrogated IFNβ-induced growth inhibition, thus establishing the role of BATF2 in mediating IFN anti-proliferative action [1,2]. MET inhibitors and IFNs in CRC therapeutic strategy. Currently, many targeted approaches to cancer therapy have focused on the reliance of cancer cells on HGF/MET signaling for cell proliferation. Although approaches targeting MET, for instance diverse specific MET inhibitors, monoclonal antibodies and siMET, were developed, and have yielded promising results in preclinical studies, few clinical trials of MET inhibitors have demonstrated the expected therapeutic benefits [7]. This inconsistency raises the possibilities that there are different regulatory mechanisms of HGF/MET signaling in different cancers. Based on our findings that BATF2 can dramatically suppress MET expression and an earlier report that MET inhibitor PF-04217903 only partially inhibited HGF-mediated cell proliferation in CRC cells, we evaluated whether the BATF2 inducer IFN-β in combination with the MET inhibitor PF-04217903 would produce a synergistic effect on cell apoptosis and proliferation in CRC cells. Then, our work validated the efficacy of the combination in inhibiting proliferation in vitro and blocking xenograft formation in vivo in CRC [1]. Taken together, these findings indicate that BATF2 status could be served as a potential biomarker to predict prognosis and IFNs in combination with MET inhibitors may serve as a novel therapeutic strategy for CRC patients.

Synergistic Antitumor Effects of Cmet Inhibitor in Combination with Anti-Vegf in Colorectal Cancer Patient-Derived Xenograft Models

Abstract:cMet signaling pathway is involved in the resistance to anti-VEGF therapy and cMet overexpression is associated with tumor progression and poor prognosis. In this study, the expression of cMet in 146 Chinese colorectal cancer (CRC) patients was examined by immunohistochemistry staining. Our data demonstrated that cMet overexpression rate was 42.5% (62/146) and cMet overexpression was closely correlated with distant metastasis of CRC. Using CRC patient-derived xenograft (PDX) mouse models we investigated antitumor activity of a novel selective cMet inhibitor volitinib alone or in combination with anti-VEGF inhibitor apatinib in vivo. Our results showed that combination treatment significantly inhibited tumor growth in two PDX models. While volitinib treatment alone induced moderate improvement in tumor growth inhibition, combination treatment synergistically reduced microvessel density, suppressed proliferation, and increased apoptosis in PDX models. Further analysis showed synergistic inhibition of MAPK and PI3K/Akt pathways by volitinib and apatinib. Taken together, our data provide a rationale to targeting both cMet and VEGF in the treatment of cMet overexpressing CRC in clinical trials.

Abstract:Abstract Background BATF2, also known as SARI, has been implicated in tumor progression. However, its role, underlying mechanisms, and prognostic significance in human gastric cancer (GC) are elusive. Methods We obtained GC tissues and corresponding normal tissues from 8 patients and identified BATF2 as a downregulated gene via RNA-seq. qRT-PCR and western blotting were applied to examine BATF2 levels in normal and GC tissues. The prognostic value of BATF2 was elucidated using tissue microarray and IHC analyses in two independent GC cohorts. The functional roles and mechanistic insights of BATF2 in GC growth and metastasis were evaluated in vitro and in vivo. Results BATF2 expression was significantly decreased in GC tissues at both the mRNA and protein level. Multivariate Cox regression analysis revealed that BATF2 was an independent prognostic factor and effective predictor in patients with GC. Low BATF2 expression was remarkably associated with peritoneal recurrence after curative gastrectomy. Moreover, elevated BATF2 expression effectively suppressed GC growth and metastasis in vitro and in vivo. Mechanistically, BATF2 binds to p53 and enhances its protein stability, thereby inhibiting the phosphorylation of ERK. Tissue microarray results indicated that the prognostic value of BATF2 was dependent on ERK activity. In addition, the N6-methyladenosine (m 6 A) modification of BATF2 mRNA by METTL3 repressed its expression in GC. Conclusions Collectively, our findings indicate the pivotal role of BATF2 in GC and highlight the regulatory function of the METTL3/BATF2/p53/ERK axis in modulating GC progression, which provides potential prognostic and therapeutic targets for GC treatment.

Abstract:Abstract Background: The type-I interferon (IFN-I) system appeared before adaptive immunity during evolution as the primary defense system for metazoans. Initiating cancers frequently disable this first line of defense before establishing T-cell exhaustion programs. However, the molecules linking IFN-I, danger signals, and unique metabolic conditions in the tumor microenvironment (TME) remain to be discovered. Methods: We performed centered log-ratio transformation on the cluster composition data using the R-package ALDEx2 to compare the clusters in single-cell data sets. IFN-I signatures were quantified using qPCR and ELISA. ChIP was performed to examine the H3K27me3 epigenetic marker on the promoters of BATF2 and IFN-I genes in bone marrow-derived macrophage (BMDM). For the cigarette smoke carcinogen-induced head and neck squamous cell carcinoma (HNSCC) model, experimental mice were fed with water containing 50 μg/mL of 4-NQO for 16 weeks and then switched to regular feeding water. We started monitoring 10 weeks post-treatment for the number, location, and diameter of the lesions. TME analysis was performed using single-cell RNA-Seq and multispectral imaging. Results: We analyzed three large independent cohorts of patients and found that BATF2 strongly correlated with the immunogenicity of HNSCC, with a stronger correlation coefficient than STING. We generated Batf2−/− BMDM and found that Batf2 deficiency rendered cells insensitive to STING agonists. However, the dsRNA-induced IFN-I activation was not affected by Batf2 deletion. BATF2 interacted with STING, promoting its phosphorylation and oligomerization. Glutamine is enriched in the TME. We found that glutamine inhibited the BATF2-STING pathway by increasing a repressive epigenetic marker, H3K27me3, on the promoters of BATF2 and IFNB1. The expression of BATF2 in cancer cells resulted in IFN-I- and γδ T-cell-dependent tumor rejection. Single-cell immune profiling showed that Batf2-expressing tumors contained significantly expanded Cxcl10+ myeloid cells and γδ T-cells. Utilizing the 4-NQO-induced HNSCC model, we found that BATF2 limited suppressive myeloid cells and expanded effector cells in the TME, serving as a potent tumor suppressor. Conclusion: BATF2 is a glutamine-responsive tumor suppressor that limits tumor initiation by engaging the STING-γδ T-cell pathway. Abstract:Objective: Transcription factors have emerged as primary regulators in colon cancer. Basic Leucine Zipper Transcription Factor (BATF) was found to be differentially expressed in colon cancer. This study aimed to explore the impact of BATF on the malignant phenotype and epithelial-mesenchymal transition (EMT) process. Methods: Based on The Cancer Genome Atlas (TCGA) data, the correlation between BATF and patients' overall prognosis was analyzed. BATF expression in epithelial and colon cancer cells was evaluated. By knocking down its levels in colon cancer cells, its effects on the malignant phenotype, apoptosis, EMT progression, and ERK/PD-L1 were evaluated. Cells were treated with ERK/PD-L1 agonists, and the BATF cell regulation was re-examined. Results: BATF levels were negatively correlated with patients' overall survival. BATF is upregulated in colon cancer cell lines, and BATF knockdown in HCT116 cells suppressed the malignant cellular phenotypes (proliferation, migration, and invasion) and increased apoptosis. BATF knockdown inhibited EMT and ERK/PD-L1 signaling activation, whereas upon agonist treatment, BATF potency was disrupted. Conclusion: This study revealed that BATF is involved in the malignant phenotype and EMT of colon cancer cells, and this process may be mediated by ERK/PD-L1 signaling.

Abstract:Hepatocyte growth factor (HGF)/MET signaling is implicated in the development of colorectal cancer (CRC) and possesses therapeutic value for various types of cancer. However, inhibition of MET alone has been demonstrated to have limited efficacy. The present study examined the combined inhibition of MET and SRC kinase activity in colon cancer cells. Furthermore, the role of the HGF/MET pathway in ligand-dependent and-independent activation was demonstrated. The single inhibition of MET by knockdown small interfering RNA or inhibitor indicated a limited anti-viability effects without inhibiting the basal phosphorylation levels of SRC, protein kinase B (AKT) or extracellular signal-regulated kinase (ERK). In view of the strong association between MET and SRC identified by direct regulation, growth factor-induced MET activation was suppressed by pretreatment with the SRC inhibitor, dasatinib, and downstream phosphorylation of AKT and ERK partially decreased, which suggested that SRC activation was essential for ligand-dependent and independent activation of MET. Considering that both the activation of MET and SRC was required in ligand-dependent and -independent MET activation, the antitumor effect of concurrent inhibition of MET and SRC was examined, and it was demonstrated that combination treatment exerted increased viability inhibition and apoptosis enhancement in mutant and wild type RAS colon cancer cells. Therefore, combinational inhibition of MET and SRC may be a promising strategy for the treatment of CRC.

We enrolled 227 patients with pancreatic cancer in the study. RON and MET expression was analyzed by immunohistochemical staining. Four human pancreatic cancer cell lines expressing variable levels of RON or MET and four MET superfamily inhibitors (BMS777607, PHA665752, INCB28060, Tivantinib) were used. The effect of the four tyrosine kinase inhibitors on cell viability, migration, and apoptosis were determined using cell viability, scratch wound healing, and Caspase-Glo 3/7 assays. Cellular signaling was analyzed by immunoprecipitation and western blotting. The therapeutic efficacy of the tyrosine kinase inhibitors was determined with mouse xenograft pancreatic cancer models in vivo. There was wide aberrant RON and MET expression in the cancer tissues. In 227 pancreatic cancer samples, 33% had RON overexpression, 41% had MET overexpression, and 15.4% had RON and MET co-overexpression. RON and MET expression were highly correlated. RON and MET expression levels were significantly related to OS. Patients with RON and MET co-overexpression had poorer OS. BMS777607 and PHA665752 inhibited pancreatic cancer cell viability and migration, and promoted apoptosis by inhibiting RON and MET phosphorylation and further inhibiting the downstream signaling pathways in vitro. They also inhibited tumor growth and further inhibited phosphorylated (phosphor)-RON and phospho-MET expression in the mouse xenograft models in vivo effectively. INCB28060, which inhibits the MET signaling pathway alone, was not effective. RON and MET can be important indicators of prognosis in pancreatic cancer. Tyrosine kinase inhibitors targeting RON and MET in pancreatic cancer are a novel and potential approach for pancreatic cancer therapy.

Abstract:Immune checkpoint blockades (ICBs) have made huge breakthrough among some cancer types including lung cancer. However, only a small proportion of patients will benefit from ICBs, other patients has no or minor response to immunotherapy. The underlying mechanisms and efficient biomarkers to predict immunotherapy resistances remains unclear and lacking. In this study, BATF2 knockout mice, human xenograft mice were used for in vivo studies. Relevant RNA and protein levels were analyzed by RT-qPCR and western blotting. As a result, we found the expression of BATF2 is negatively correlated with expression of PD-L1 in the plasma of patients. Mechanically, we showed that BATF2 inhibits PD-L1 expression in cancer cells by inhibiting the PI3K/AKT pathway where ZEB2 plays an important role in this process. Based on bioinformatics analysis, we found the function of BATF2 in promoting anti-tumor immune response in patients with NSCLC, which is mediated by BATF2 enhances CD8<sup>+</sup> T cells infiltration as well as activation. BATF2 from circulating and tissue can serve as an efficient biomarker to predict diagnosis, prognosis and immunotherapy efficacy.

Background During the tumourigenesis and development of colorectal cancer (CRC), the inactivation of tumour suppressor genes is closely involved, although detailed molecular mechanisms remain elusive. Accumulating studies, including ours, have demonstrated that basic leucine zipper transcription factor ATF (activating transcription factor)‐like 2 (BATF2) is a capable tumour suppressor that localises in the nucleus. However, its different subcellular localisation, potential functions and underlying mechanisms are unclear. Methods The translocation of BATF2 and its clinical relevance were detected using CRC samples, cell lines and xenograft nude mice. Candidate BATF2‐binding proteins were screened using co‐immunoprecipitation, quantitative label‐free liquid chromatography–tandem mass spectrometry proteomic analysis, Western blotting and immunofluorescence. Recombinant plasmids, point mutations and siRNAs were applied to clarify the binding sites between BATF2 and chromosome region maintenance 1 (CRM1). Results The present study found that BATF2 was mainly localised in the cytoplasm, rather than nucleus, of CRC cells in vitro and in vivo, while cytoplasmic BATF2 expression was inversely correlated with the prognosis of CRC patients. Furthermore, we identified the nuclear export and subsequent ubiquitin‐mediated degradation of BATF2 in CRC cells. Mechanistically, a functional nuclear export sequence (any amino acid) was characterised in BATF2 protein, through which BATF2 bound to CRM1 and translocated out of nucleus, ultimately enhancing CRC growth via inducing activator protein 1 (AP‐1)/cyclin D1/phosphorylated retinoblastoma protein (pRb) signalling pathway. Additionally, nuclear export of BATF2 can be retarded by the mutation of NES in BATF2 or the knockdown of CRM1, whereas CRM1 expression was negatively associated with nuclear BATF2 expression and the prognosis of CRC patients. Conclusion These findings revealed the biological effects and underlying mechanisms of cytoplasmic localisation of BATF2. Furthermore, suppressing nuclear export of BATF2 via mutating its NES region or inhibiting CRM1 expression may serve as a promising therapeutic strategy against CRC.
