Abstract:Folliculin interacting protein 1 (FNIP1) primarily participates in regulating cellular energy metabolism and is associated with Birt-Hogg-Dube (BHD) syndrome. Although FNIP1 has been demonstrated to function as both a tumor suppressor and promoter, its role in colorectal cancer (CRC) remains unclear. Our study demonstrated a significant downregulation of FNIP1 in CRC, correlating with shorter overall and disease- specific survival. FNIP1 may potentially serve as an independent prognostic factor in CRC. Moreover, FNIP1 inhibited CRC progression in vitro and in vivo. . Mechanistically, FNIP1 bound to phosphorylated signal transducer and activator of transcription-3 (p-STAT3) and downregulated its expression. FNIP1 deletion increased STAT3 phosphorylation and nuclear localization, thereby promoting CRC progression. The use of p-STAT3-specific chemical inhibitors successfully mitigated excessive tumorigenesis resulting from FNIP1 absence. Thus, our results suggest that FNIP1 hinders CRC progression by suppressing STAT3 phosphorylation and nuclear translocation. FNIP1 may be a candidate prognostic indicator and a therapeutic target for intervention in CRC.

Abstract:Occurrence of Colorectal cancer (CRC) is relevant with gut microbiota. However, role of IRF3, a key signaling mediator in innate immune sensing, has been barely investigated in CRC. Here, we unexpectedly found that the IRF3 deficient mice are hyper-susceptible to the development of intestinal tumor in AOM/DSS and Apcmin/+ models. Genetic ablation of IRF3 profoundly promotes the proliferation of intestinal epithelial cells via aberrantly activating Wnt signaling. Mechanically, IRF3 in resting state robustly associates with the active β-catenin in the cytoplasm, thus preventing its nuclear translocation and cell proliferation, which can be relieved upon microbe-induced activation of IRF3. In accordance, the survival of CRC is clinically correlated with the expression level of IRF3. Therefore, our study identifies IRF3 as a negative regulator of the Wnt/β-catenin pathway and a potential prognosis marker for Wnt-related tumorigenesis, and describes an intriguing link between gut microbiota and CRC via the IRF3-β-catenin axis.

Abstract:BACKGROUND. Expression microarrays are widely used for investigating the nature and extent of global gene-expression changes in human cancer. Accurate genomewide gene-expression profiles have not been conducted in colon tumor and normal colon tissue specimens obtained from Chinese patients. METHODS. in the present study a pure population of colon cancer and normal colon cells was obtained and the global gene-expression differences were compared in the 2 cell types using combined experimental and bioinformatic approaches. Various categories of genes that were differentially expressed in those 2 types of cells were identified, including a novel candidate tumor marker, IFITM3. RESULTS. Elevated IFITM3 expression in colon cancer cells was first confirmed using quantitative real-time polymerase chain reaction. IFITM3 protein expression in human colon cancer specimens was further analyzed using both tissue microarray and standard tissue sections by immunostaining analyses. It was found that there was a significant IFITM3 increase in adenoma as compared with that in normal colon tissue. CONCLUSIONS. The data suggest that IFITM3 plays an important role in early colon cancer development.

Abstract:Background: Humankind have been struggling with colorectal cancer (CRC) for long period with its rapid progression and invasive metastasis. By hyperactivating IL-6/STAT3 signaling, CRC facilitates the capacity of angiogenesis to plunder massive nutrients and develops gradually under harsh condition. Methods: The Cancer Genome Atlas database was analyzed for acquiring interferon-γ inducible protein 10 (IFITM10) expression levels and their correlation with clinical outcomes. The cell angiogenic ability were assessed by Cell Counting Kit-8 (CCK-8) and tube formation assay. Immunofluorescence, Western blot, and enzyme-linked immunosorbent assay (ELISA) assay were using to assess potential mechanism. Results: In our study, we find that IFITM10 is upregulated in CRC and is positively related with tumor angiogenesis. We also find that IFITM inhibition decreased STAT3 phosphorylation level and IFITM10-mediated angiogenesis depends on STAT3 activation. Furthermore, our data suggests that IFITM10 may be a key prognostic biomarker in colorectal cancer. Conclusion: Together, our study suggests that IFITM10 enhance angiogenesis through STAT3 activation during CRC progression, which highlighting its potency as a therapeutic target for colorectal cancer.

Abstract:Objective·To analyze the relationship between the expression level of interferon regulatory factor 3(IRF3)in colorectal cancer and its clinicopathological features and prognosis,and to observe the effects of IRF3 overexpression on the proliferation and invasion ability of colorectal cancer cells and the related protein molecular pathways.Methods The Cancer Genome Atlas(TCGA)data were downloaded and used to analyze the correlation between expression levels of IRF3 and the prognosis of patients(including renal cell carcinoma,colorectal cancer,hepatocellular carcinoma,and prostate cancer).Immunohistochemistry was used to detect the differences in the expression levels of IRF3 between cancerous tissue and adjacent normal tissues of 10 patients with colorectal/renal cancer.The C-terminal residue sites of the IRF3 protein were modified to construct HEK-293T cells overexpressing the phosphorylated IRF3-5D(396/398/402/404/405-D).At 12 and 24 h of cell culture,treatment with TANK-binding kinase 1(TBK1)inhibitor was performed,and Western blotting was used to detect the expression levels of IRF3 and p-IRF3(Ser386)in the cells.RNA sequencing(RNA-seq)was employed to explore the correlation between high expression of IRF3-5D and the expression levels of tumor-related proteins.Colorectal cancer cells CT26 and COLON26 overexpressing wild-type IRF3(IRF3-WT)and IRF3-5D were construct,and cell proliferation and migration ability were assessed by using cell counting,scratch assay,and clonogenic assay.Results·Analysis of TCGA data suggested that the expression level of IRF3 protein in cancer tissues was positively correlated with poor prognosis in patients.Immunohistochemical analysis of pathological tissues from patients with cancer showed that the expression level of IRF3 was significantly upregulated in colorectal cancer tissues and renal cancer tissues,with protein expression concentrated in the cell nucleus.After treatment with TBK1 inhibitors for 12 and 24 h in cell culture,the expression of p-IRF3(Ser386)protein in HEK-293T cells decreased.The results of RNA-seq and Western blotting showed that the expression levels of multiple proteins associated with poor prognosis[such as IRF9,programmed cell death 1-ligand 1(PD-L1),etc.]were significantly upregulated under conditions of high expression of IRF3-5D.Overexpression of IRF3-5D in colorectal cancer cells could significantly enhance the proliferation and migration capabilities of cancer cells.Conclusion·The expression level of IRF3 in colorectal cancer is positively correlated with poor patient prognosis.High expression of IRF3-5D protein in colorectal cancer cells can promote malignant biological behavior of cancer cells.Additionally,IRF3-5D is dependent on the TBK1-mediated activation of the IRF3 activation pathway and upregulates the expression levels of multiple tumor-related proteins.

Abstract:Abstract Targeting tumor-infiltrating regulatory T cells (Tregs) is an efficient way to evoke an anti-tumor immune response. However, how Tregs maintain their fragility and stability remains largely unknown. IFITM3 and STAT1 are interferon-induced genes that play a positive role in the progression of tumors. Here, we showed that IFITM3-deficient Tregs blunted tumor growth by strengthening the tumor-killing response and displayed the Th1-like Treg phenotype with higher secretion of IFNγ. Mechanistically, depletion of IFITM3 enhances the translation and phosphorylation of STAT1. On the contrary, the decreased IFITM3 expression in STAT1-deficient Tregs indicates that STAT1 conversely regulates the expression of IFITM3 to form a feedback loop. Blocking the inflammatory cytokine IFNγ or directly depleting STAT1-IFITM3 axis phenocopies the restored suppressive function of tumor-infiltrating Tregs in the tumor model. Overall, our study demonstrates that the perturbation of tumor-infiltrating Tregs through the IFNγ-IFITM3-STAT1 feedback loop is essential for anti-tumor immunity and constitutes a targetable vulnerability of cancer immunotherapy.
