Abstract:A catalytic annulation cascade is achieved that affords an efficient access to phthalides from readily available benzoic acids and saturated ketones. The reaction is catalyzed by a bimetallic Ir/Cu system and proceeds via the combination of Cu-catalyzed dehydrogenation of ketones and Ir-catalyzed direct C-H functionalization under the assistance of the weakly coordinating carboxyl followed by the beta-H elimination and intramolecular Michael addition. This method shows a broad scope of substrates and good functional group tolerance. The protocol furnishes an alternative strategy for the construction of diverse 3-substituted phthalides.

Abstract:A photocatalytic acyl lactonization of unsaturated carboxylic acids using simple and inexpensive acyl chlorides has been developed for a modular synthesis of acyl lactones. Significantly, the simple protocol could allow an efficient construction of biologically important phthalide framework. The transformation could be extended to a wide variety of unsaturated carboxylic acids, including substituted 2‐vinyl benzoic acids and different types of alkenoic acids. Moreover, a series of aroyl, heteroaroyl chlorides could serve as coupling partners. Notably, functional groups including MeO, F, Cl and Br could survive. It is believed that acyl radicals generated from acyl chlorides under photoredox catalysis reacted with alkenes via atom‐transfer radical addition (ATRA) and triggered subsequent lactonization in the process. Preliminary mechanistic investigations revealed that the transformation probably proceed through a free radical pathway.

Abstract:A convenient approach for the synthesis of 3-difluoroalkyl phthalides has been developed from phthalaldehydic acids and difluoroenoxysilanes by using relatively inexpensive p-toluenesulfonic acid monohydrate (PTSA) as a catalyst. A series of 3-difluoroalkyl phthalides and cyclic difluoroalkyl ethers were obtained in up to 99% yield. The products obtained could be readily converted into difluoroalkyl phthalide derivatives by simple modifications.

Abstract:Herein, we report an efficient one-pot iridium-catalyzed reductive hydroamination of terminal alkynes with amines. A variety of primary amines and secondary amines are amenable to this catalytic system, leading to the complicated aliphatic amines in excellent yields. More broadly, this straightforward and convenient transformation do not just employ inexpensive and easy to handle HCO 2 H as reducing agent, but also employs H 2 O as reaction media, which makes this strategy be in accordance with green and sustainable chemistry principles.

Abstract:Reductive functionalization between two electrophiles represents a straightforward protocol to access carbon‐carbon bonds without generating organometallic reagents beforehand. Here, we developed a manganese‐mediated reductive alkylation of heterocyclic thiazolyl‐phosphonium salts with benzyl bromides to form C2‐alkylated benzothiazoles. The method features a broad substrate scope, high‐functional‐group tolerance, and can be utilized in late‐stage modification of complex natural products and bioactive molecules.

Abstract:Abstract Organophosphines are an important class of ligands widely used in organic chemistry. Although great progress has recently been made in the rapid construction of new phosphines through Rh- or Ru-catalyzed C–H bond functionalizations, synthetic access to more diverse phosphines remains a challenge. We describe an efficient process for the rhodium-catalyzed phosphorus(III)-directed hydroarylation of internal alkynes to generate various alkenylated and 2′,6′-dialkenylated biarylphosphines with high selectivity. A range of diverse alkynes and phosphines were effectively prepared with broad functional-group compatibility under the optimized conditions. In addition, the developed protocol can be extended to modify chiral phosphine ligands, providing enantioenriched alkenylated phosphines without erosion of the enantiomeric excess.

Abstract:The reductive carbonylation of aryl iodides to aryl aldehydes possesses broad application prospects. We present an efficient and facile Rh-based catalytic system composed of the commercially available Rh salt RhCl3·3H2O, PPh3 as phosphine ligand, and Et3N as the base, for the synthesis of arylaldehydes via the reductive carbonylation of aryl iodides with CO and H2 under relatively mild conditions with a broad substrate range affording the products in good to excellent yields. Systematic investigations were carried out to study the experimental parameters. We explored the optimal ratio of Rh salt and PPh3 ligand, substrate scope, carbonyl source and hydrogen source, and the reaction mechanism. Particularly, a scaled-up experiment indicated that the catalytic method could find valuable applications in industrial productions. The low gas pressure, cheap ligand and low metal dosage could significantly improve the practicability in both chemical researches and industrial applications.

Abstract:An iridium photoredox catalyst in combination with either a stoichiometric amount of Bronsted acid or a catalytic amount of Lewis acid is capable of catalyzing regioselective alkylation of N-heteroarenes with N-(acyloxy) phthalimides at room temperature under irradiation. A broad range of N-heteroarenes can be alkylated using a variety of secondary, tertiary, and quaternary carboxylates. Mechanistic studies suggest that an Ir-II/Ir-III redox catalytic cycle is responsible for the observed reactivity.

Abstract:The reductive lactonization strategy provides an efficient access to stereoenriched polycyclic γ-lactones. However, it is still a formidable challenge to develop an efficient and versatile protocol with excellent levels of diastereocontrol. Herein we provide a highly diastereoselective and efficient route to diastereopure bi- and polycyclic γ-lactones, by means of an iridium-catalyzed hy-dride transfer strategy. This method features high levels of stereocontrol, broad substrate scope, and high catalyst efficiency (S/C = up to 5000). Mechanistic studies suggested that the iridium hydride formation be the rate-determining step, and that the hydride transfer step be the diastereo-determining step. The large steric hindrance of the iridium hydride species and intramolecular hydro-gen bonding are critically key to the diastereocontrol of the hydride transfer process. From the perspectives of configurational anal-ysis and Duniz angles of attack, the nature of diastereocontrol is well rationalized. A more general empirical rule based on facial se-lectivity analysis for explaining and predicting the stereochemistry is also proposed.

Abstract:A functionalization of benzylic C(sp(3))-H bonds was established through a mild metal-free intramolecular lactonization protocol in the presence of DDQ/tert-butyl nitrite as photocatalysts, allowing practical and low-cost access to a series of phthalide products in moderate to excellent yields. Compared with many existing methodologies, this visible-light-driven process exhibits an excellent substrate scope and fascinating features, including the formation of water as the sole byproduct, an abundant and green energy source, commercially available catalysts, and room-temperature reaction. Finally, detailed mechanistic investigations clearly revealed the role of the photocatalysts and molecular oxygen.

Abstract:A diverse array of bidentate P-containing ligands has been developed for Ir-catalyzed asymmetric hydrogenation, but chiral monodentate phosphine ligands remained comperatively underexplored. We herein report a novel iridium catalyst with XuPhos as a chiral monodentate phosphine ligand for the direct asymmetric reductive amination of ketones with secondary amines. This catalytic system tolerates a wide range of substrates, providing a series of chiral tertiary amines efficiently with high enantioselectivities. The high catalytic activity attributed to the presence of the aliphatic cyclohexyl group on the P-atom and the relative configuration of the ligand, which is crucial for the XuPhos to act as a monodentate P-ligand for iridium.

Abstract:We report a practical method for synthesis of 3‐gem‐difluorovinyl lactams and their dideuterated derivatives with excellent deuterium (D) incorporation. This zinc‐mediated sequential reductive hydrodehalogenation proceeds via a radical process. The in‐situ generated zinc cation complex plays a key role in facilitating the transformation due to its strong Lewis acidity. Comprehensive Summary Herein, we describe a direct route for the synthesis of 3‐gem‐difluorovinyl lactams through Zn‐mediated reductive hydrodehalogenation. Importantly, by using inexpensive deuterium oxide (D2O), the high value‐added vicinal dideuterated gem‐difluoroalkenes with excellent deuterium (D) incorporation were prepared. Mechanism studies indicated a successive single electron transfer process: the reaction initially undergoes hydrodechlorination to give the intermediate α‐trifluoromethylidene lactams, which are then activated by the in‐situ generated zinc cations and reduced to the desired product via hydrodefluorination.

Abstract:Tandem one‐pot regioselective synthesis of C4‐difluoroalkylated phthalazin‐1(2H)‐ones from readily available hydrazines, 2‐formylbenzoic acids and bromodifluoroacetate derivatives under visible‐light photocatalysis is reported. This mild protocol provides straightforward access to various C4‐difluoroalkylated phthalazin‐1(2H)‐ones, and displays broad substrate scope and wide functional group tolerance. Moreover, this process is also suitable for the synthesis of C4‐trifluoromethylated and perfluorobutylated phthalazin‐1(2H)‐ones. The use of fac‐[Ir(ppy)3] photocatalyst and the introduction of an organic base additive are pivotal to catalysis.

Abstract:An electrochemical approach in promoting directed C-H alkynylation with terminal alkyne via iridium catalysis is reported. This work employed anodic oxidation of Ir(III) intermediate (characterized by X-ray crystallography) to promote reductive elimination, giving the desired coupling products in good yields (up to 95%) without the addition of any other external oxidants. This transformation is suitable for various directing groups with H2 as the only by-product, which warrants a high atom economy and practical oxidative C-C bond formation under mild conditions.

Abstract:A direct palladium-catalyzed γ-lactonization of free carboxylic acids via C–O reductive elimination as a key step is described. Notable aspects of this protocol include the use of convenient and easily available sodium percarbonate as an oxidant and the development of a new beta-alanine derived ligand. The reported method enables the functionalization of a wide range of aliphatic acids including previously inaccessible β-non-quaternary acids. The regioselectivity and consequently substrate scope of this protocol proved complementary to established routes for the direct lactonization of aliphatic carboxylic acids. A modification of this method furthermore enables a direct β-C(sp3)–H hydroxylation of challenging α-non-quaternary carboxylic acids with considerable functional group tolerance. Through this report valuable compound libraries of γ-lactones and β-hydroxy acids with potential applications in various fields become available.

Abstract:The novel cascade photoredox/iodide catalytic system enables the alkene to serve as a radical acceptor capable of achieving aminodifluoroalkylation of alkenes. Cheap iodide salts play a vital role in this reaction, which could tune carbocation reactivity through reversible C–I bond formation for controlling reaction selectivity, and a series of competitive reactions are completely eliminated in the presence of multiple reactivity pathways. The present dual catalytic protocol affords a very convenient method for direct synthesis of various difluoro-γ-lactams from simple and readily available starting materials under mild reaction conditions.
