Abstract:As an important building block for the synthesis of angiotensin-converting enzyme inhibitors, ethyl (R)-2-hydroxyl-4-phenylbutanoate [(R)-HPBE] has attracted increasing attention. The key to industrial biosynthesis of (R)-HPBE is a biocatalyst that efficiently reduces ethyl 2-oxo-4-phenylbutanoate (OPBE) with high R-enantioselectivity. This paper proposed a strategy for identifying key residues involved in enantioselectivity control based on per-residue free energy decomposition and sequence conservatism analysis. Using this strategy, 4 nonconservative sites with high energy contribution to binding of OPBE were chosen as engineering targets, generating variant Mu27 with 99% conversion and 98% (R) ee value at substrate loading of up to 500 mmol/L. MD simulations suggested the higher stability and formation probability of Mu27-OPBEproR prereaction state as key reasons for the excellent R-enantioselectivity of Mu27 towards OPBE. The success in this study provides a viable approach for rational design of alcohol dehydrogenases with high enantioselectivity towards unnatural substrates.

Abstract:The methyl diazoacetate and aryl (N-tosyl)imines can be transformed into syn or anti α-hydroxy β-amino esters with high diastereoselectivities in three steps: the base promoted nucleophilic condensation of the methyl diazoacetate and aryl (N-tosyl)imines to give β-(N-tosyl)amino α-diazoesters, followed by oxidation with Oxone® to generate α-oxo esters, which were reduced with NaBH4 to yield the anti-N-tosyl-α-hydroxy β-amino ester, or hydrogenated with Pd/C (10%) as the catalyst to yield corresponding syn isomer, both in high diastereoselectivity.

Abstract:Saturated alpha-substituted beta-ketoesters are important building blocks in the synthesis of pharmaceuticals and agro-chemicals. Herein, we report a one-pot biosynthesis of alpha-substituted beta-ketoesters via Knoevenagel condensation and reduction of the obtained unsaturated alkenes in situ, catalyzed by a single ene-reductase (NerA). A series of inexpensive and readily available aldehydes and 1,3-diketones were condensed and reduced by NerA in aqueous solutions at room temperature. We also noted that low loadings (3 mg/mL) of NerA were sufficient to facilitate the cascade process; both E and Z isomeric intermediates could be reduced effectively, and the overall yield was improved up to 95%. Meanwhile, the method could be applied to a gram preparative-scale synthesis of pharmaceutical intermediates. This process conformed to the concepts of green chemistry and showed advantages for the synthesis of high value saturated alpha-substituted beta-ketoesters.

Abstract:Development of an efficient, stereoselective, sustainable synthesis of chiral aryl β‐hydroxy α‐amino acids and their derivatives is of paramount importance, owing to the broad utility of these molecules in pharmaceutical application and asymmetric synthesis. We report the first systematic study on ketoreductase (KRED)‐catalyzed dynamic reductive kinetic resolution (DYRKR) of aryl α‐amino β‐keto esters 6 , furnishing 20 structurally diverse chiral syn ‐aryl β‐hydroxy α‐amino esters ( syn ‐ 7 ) in moderate‐to‐excellent isolated yield (up to 93%), along with moderate‐to‐excellent diastereoselectivity (up to >99: 1 dr) and excellent enantioselectivity (mostly >99% ee). The practical synthesis potential of our developed method was showcased by the asymmetric, chemo‐enzymatic total synthesis of thiamphenicol ( 1 ).

Abstract:Reduction of 16-(hydroxymethylidene)-3-methoxy-13α-estra-1,3,5(10)-trien-17-one yielded a mixture of two diastereomeric diols in the 6:1 ratio. The configurations of the newly formed stereogenic centres were determined by X-ray crystallography and NMR spectroscopy (NOE experiments) on the compounds in their cyclic acetaldehyde acetal forms.

Abstract:We report here a generic, green synthesis of 17 valuable syn-aryl-(2S,3R)-2‑chloro-3‑hydroxy esters (syn-(2S,3R)-1) in 73%-99% isolated yields along with 6.1:1–83:1 dr and 31%∼>99% ee, through dynamic reductive kinetic resolution of racemic aryl α‑chloro β-keto esters (2) catalyzed by an engineered ketoreductase which was obtained via epPCR-based directed evolution. The hectogram scale synthesis of syn-(2S,3R)-1b at a substrate concentration of 120 g/L showcased the application potential of the biocatalytic method developed presently.

Abstract:Highly stereo- and enantioselective synthesis of (E)-δ-hydroxymethyl-anti-homoallylic alcohols is reported. Abstract:Highly stereoselective access to β-glycosyl esters was disclosed, employing 1-hydroxybenzotriazole (HOBt) mediated esterification of glycosyl hemiacetals in the presence of EDCI and 1,4-diazabicyclo[2.2.2]octane (DABCO). Mechanistic studies indicated a dynamic kinetic acylation pathway. In addition, a stereoretentive esterification of glycosyl hemiacetals with tert-butyloxycarbonyl ortho-hexynylbenzoate and DMAP was also reported.

Abstract:The naturally occurring (4R,5R)-4-hydroxy-γ-decalactone from the Japanese orange fly and the antipode of (4S,5R)-4-hydroxy-γ-dodecalactone from the harvestmen arachnid and their stereoisomers are synthesized from the chiral pool material d-glucono-δ-lactone in a few steps. The one-pot conversion of the latter to γ-vinyl-β-hydroxy-γ-lactone, cross-metathesis with requisite olefin, and hydrogenation enabled the synthesis of syn-lactones in just a two-pot operation. An additional efficient Pd-catalyzed allylic isomerization of γ-vinyl-β-hydroxy-γ-lactone led to the anti-lactones in high yields.
