Abstract:Copper(II)-catalyzed synthesis of sulfones from sulfinate salts and aryl halides was investigated by means of a combination of experiment and DFT calculation. Experimental results demonstrated the wide applicability of the title approach. The reaction mechanisms are revealed by in-situ IR and theoretical study. It reveals remarkable ligand effect the bidentate amine plays in the reaction, that is, it initially activats the C-I bond of iodobenzene so as to enhance the whole catalytic process. Abstract:A new type of Rh2(OAc)4-catalyzed [2,3]-sigmatropic rearrangement of sulfur ylides is reported. A series of cyclopropenes were successfully employed for [2,3]-sigmatropic rearrangement by a reaction with either allylic or propargylic sulfides. Under the optimized conditions, the reaction afforded the products in moderate to excellent yields. In these transformations, the vinyl metal carbenes generated in situ from the cyclopropenes were effectively trapped by sulfides, resulting in the formation of corresponding products upon [2,3]-sigmatropic rearrangements.

Abstract:A highly stereoselective [2,3]-sigma rearrangement of sulfur ylide derived from Cu(I) carbene and allyl/propargyl sulfides (Doyle-Kirmse reaction) is reported. High stereocontrol is achieved by a dual asymmetric induction approach which involves a chiral auxiliary on diazo substrate, and steric bulky ligand of the Cu(I) catalyst. From mechanistic study, we suggest the [2,3]-sigma rearrangement process occurs through free sulfur ylide intermediate. The reaction was further applied in kinetic resolution of allyl sulfide with a chiral center.
Studies on the Reactivity of Migrating Group in [2,3]-Sigmatropic Rearrangement of Sulfur Ylides

Abstract:The [2,3]-sigmatropic rearrangement of sulfur ylides is unique and useful reaction in organic synthesis. In this study, the reactivity of sulfides containing three different migrating groups (propargyl, allyl, and allenyl) in [2,3]-sigmatropic rearrangement of sulfur ylides has been compared. The competition reactions of phenylethyldiazoacetate with sulfides through sulfonium ylide [2,3]-sigmatropic rearrangement are designed under Rh(II)- or Cu(I)-catalyzed reaction conditions. Both intra- and intermolecular competitions of sulfides bearing two different migrating groups have been carried out. The ratio of products has been determined by H-1 NMR in order to compare the reactivity of different sulfides bearing allyl, propargyl or allenyl groups. Obvious disparity of the reactivity of these sulfides in [2,3]-sigmatropic rearrangement has been observed. Experimental data indicate that the tendency of preferential [2,3]-sigmatropic rearrangement has the following order: propargyl sulfide>allyl sulfide>allenyl sulfide. Catalysts such as Rh-2(OAc)(4), Rh-7(O2CCF3)(4), and Cu(CH3CN)(4)PF6 ligated with a series of diimine ligands have been investigated for these reactions. Rh(II) complexes are found more efficient than Cu(I) complexes, and Rh-2(O2CCF3)(4) is more efficient than Rh-2(OAc)(4). The efficiency of different catalytic system has been explained based on the proposed reaction mechanism. The reaction catalyzed by Rh(II) complexes is suggested to be different from that catalyzed by Cu(I) complexes. In the case of Cu(I)-catalyzed reaction, Cu(I)-bonded sulfur ylide is considered as the predominant intermediate, while the [2,3]-sigmatropic rearrangement is suggested to proceed through free ylide. Both steric hindrance and electronic properties of ligands influence the ratio and selectivity in Cu(I)-catalyzed reactions. This study provides useful information for further investigation of [2,3]-sigmatropic rearrangement of sulfur ylides.

Abstract:We report a stereoretentive cross-coupling reaction of configurationally stable nucleophiles with disulfide and N-sulfenylsuccinimide donors promoted by Cu(I). We demonstrate the utility of this method in the synthesis of thioglycosides derived from simple alkyl and aryl thiols, thioglycosides, and in the glycodiversification of cysteine residues in peptides. These reactions operate well with carbohydrate substrates containing common protective groups and reagents with free hydroxyl and secondary amide functionalities under standardized conditions. Competition experiments in combination with computational DFT studies established that the putative anomeric intermediate is an organocopper species that is configurationally stable and resistant to epimerization due to its short lifetime. The subsequent reductive elimination from the Cu(III) intermediate is rapid and stereoretentive. Taken together, the glycosyl cross-coupling is ideally suited for late stage glycodiversification and bioconjugation under highly controlled installation of the aliphatic carbon-sulfur bonds.

Abstract:Although α-chiral C( sp 3 )–S bonds are of enormous importance in organic synthesis and related areas, the transition-metal-catalysed enantioselective C( sp 3 )–S bond construction still represents an underdeveloped domain probably due to the difficult heterolytic metal–sulfur bond cleavage and notorious catalyst-poisoning capability of sulfur nucleophiles. Here we demonstrate the use of chiral tridentate anionic ligands in combination with Cu(I) catalysts to enable a biomimetic enantioconvergent radical C( sp 3 )–S cross-coupling reaction of both racemic secondary and tertiary alkyl halides with highly transformable sulfur nucleophiles. This protocol not only exhibits a broad substrate scope with high enantioselectivity but also provides universal access to a range of useful α-chiral alkyl organosulfur compounds with different sulfur oxidation states, thus providing a complementary approach to known asymmetric C( sp 3 )–S bond formation methods. Mechanistic results support a biomimetic radical homolytic substitution pathway for the critical C( sp 3 )–S bond formation step.

Abstract:In this paper, Rh2(OAc)4-catalyzed [2,3]-sigmatropic rearrangement of sulfur ylides derived from N-tosylhydrazones and sulfides is reported. A series of tosylhydrazones derived from aldehydes were successfully used for [2,3]-sigmatropic rearrangement by reaction with either allylic phenyl sulfides or propargyl phenyl sulfides. The reaction conditions were optimized and afforded the products in moderate to good yields. In addition, a novel and convenient approach for the synthesis of cyclobutenones and cyclopropanes has been developed through direct oxidation of the rearrangement products.
Catalytic enantioselective synthesis of chiral sulfonium ylides with S-stereogenic center

Abstract:A sulfur stereogenic center is found in naturally occurring compounds and constitutes an important design element for pharmaceuticals and chiral auxiliaries. In view of the broad applications of S-stereogenic compounds, many methods were developed to enantioselectively construct the sulfur stereocenters. By contrast, the synthesis and isolation of S-stereogenic ylides was elusive although sulfonium ylides are important synthetic building blocks and such species have been proposed in the Doyle-Kirmse reaction. Using judicious substrate design, we have successfully developed the first catalytic enantioselective synthesis of sulfonium ylides with a chiral sulfur center. They are constructed from a broad array of sulfides in high yields (up to 98%) and enantioselectivities (up to 99%) through copper-catalyzed intramolecular carbene transfer reaction. The obtained ylides could be converted into alkylidenecyclopropanes with efficient sulfur-to-carbon chirality transfer.

Abstract:A mild and general procedure for the difluoromethylation of organochalcogen compounds has been accomplished via sulfoxide and selenoxide-directed [3,3]-sigmatropic rearrangement/Haller-Bauer reaction with difluoroenoxysilanes. The reactions showed good functional group compatibility, wide substrate applicability, and facile scalability. The synthetic utility is highlighted by the iterative use of this protocol to construct multi-CF2H-containing chalcogenides and the late-stage modification of pharmaceuticals.
