Abstract:Treatment of acid chlorides with a carbamoylstannane under mild conditions (mostly rt for a few hours) affords alpha-oxo amides in high yields. Vicinal polycarbonyl compounds are also obtained, although spontaneous decarbonylation occasionally occurs.

Abstract:In sharp contrast to the well researched and well developed aminocarbonylation of alkenes, the dearomative carbamoylation of abundant arene pi-systems has scarcely been studied despite its great potential to enrich the diversity-oriented synthesis of high-value amides. The formidable challenges associated with such dearomatization include the low reactivity of stable aromatic pi-systems and intricate selectivity issues. Herein, we disclose a general approach toward highly selective dearomative carbamoylations of arene pi-bonds under CO-gas-free strated by tolerating a broad range of nucleophilic partners with high yields and excellent selectivities, thus providing modular access to the divergent synthesis of beta-functionalized primary amides. In addition, diverse downstream derivatizations including a for-mal C-H 1,2-olefination/carbamoylation reaction were conducted, exhibiting great potential in syn-thetic and medicinal chemistry.

Abstract:A facile and direct oxidative reaction for the synthesis of vicinal tricarbonyl amides in moderate to excellent yields (53-88%) was developed starting from readily available β-ketoamides in the presence of phenyliodine(III) bis(trifluoroacetate). The resulting products possess significant synthetic potential, making this approach a valuable addition to the group of traditional methods already available for the preparation of these molecules.

Abstract:Rh(acac)(CO)(2) catalyzes addition of (N,N-diisopropylcarbamoyl)trimethylstannane to terminal alkynes to give (Z)-beta-stannyl-alpha,beta-unsaturated amides with terminal attachment of the amide group, while the use of Ni(COd)(2) results in the reversal of the regioselectivity. A mechanism initiated by the oxidative addition of the carbamoylstannane was proposed on the basis of the isolation of (trimethylstannyl)(N,N-diisopropylcarbamoyl)[1,2-bis(dimethylphosphtno)ethane]pallidium.

Abstract:The preparation of the carborane acid chloride [1-(COCl)-CB11H11]- from the carboxylic acid [1-(COOH)-CB11H11]- is reported. This acid chloride exhibits remarkable inertness towards moisture and can be stored under ambient conditions for several months. Reaction with amines affords secondary and tertiary carborane amides [1-(CONR1R2)-CB11H11]- in moderate to high yields under mild conditions. Two of the amide products were characterized by X-ray crystallography in addition to spectroscopic analysis. Preliminary studies show that the amides can be reduced to the corresponding amines and that the acid chloride has the potential to serve as a starting material for carborane ester formation.

Abstract:We report a strong Brønsted acid-catalyzed three-component oxy-aminomethylation of styrenes with sym-trioxane and sulfonamides or carbamates. Abstract:Stannous chloride efficiently catalyzes condensations between alpha-isocyanoacetamides (1) and a variety of aldehydes to afford the corresponding 5-amino-2-(1-hydroxyalkyl-)oxazoles (3) in good to excellent yields. The [Sn-(R)-Ph-PyBox](OTf)(2)-catalyzed reaction between la and 2-(benzyloxy)acetaldehyde (2g) shows an isoinversion effect, with the maximum enantiomeric excess of oxazole 3g (80 %) being obtained at -40 degrees C. Abstract:Thioamides are widely used structures in pharmaceuticals and agrochemicals, as well as important synthons for the construction of sulfur-containing heterocycles. This report presents a series of visible-light-driven multicomponent reactions of amines, carbon disulfide, and olefins for the mild and versatile synthesis of linear thioamides and cyclic thiolactams. The use of inexpensive and readily available carbon disulfide as the thiocarbonyl source in a radical pathway enables the facile assembly of structurally diverse amine moieties with non-nucleophilic carbon-based reaction partners. Radical thiocarbamoylative cyclization provides a practical protocol that complements traditional approaches to thiolactams relying on deoxythionation. Mechanistic studies reveal that direct photoexcitation of in situ formed dithiocarbamate anions as well as versatile photoinduced electron transfer with diverse electron acceptors are key to the reactions.

Abstract:Oxalic acid monothioester (OAM), an easily accessible and storable reagent, was reported herein as a thioester synthetic equivalent for palladium-catalyzed decarboxylative thiocarbonylation of organohalides and hydrothiocarbonylation of unsaturated carbon-carbon bonds at room temperature with high chemo- and regioselectivity. The reaction is applicable to the synthesis of cysteine-derived thioesters, thus allowing chemical modification of cysteine-containing peptides. Decarboxylation of OAM proceeds through oxidative addition of Pd(0) to the acyl-S bond, which accounts for the very mild reaction conditions.

Abstract:The novel reactivity of O-silylthionoesters with amine nucleophiles to generate oxoamides (rather than thioamides) is described. A straightforward first-generation trimethylsilylation protocol using bistrimethylsilylacetamide (BSA) combined with the unique reactivity of the O-silylthionoesters toward 1° and 2° amines to generate oxoamides provides the simplest means of activating a thiol acid for peptide bond formation at neutral pH. Excellent stereoretention is observed.

Abstract:Amides are ubiquitous in physical and life sciences. Given the significant abundance of arenes, dearomative aminocarbonylation of arenes would lead to a large and underexplored chemical space for amide discovery. However, such reactions are challenging due to the high degree of resonance stabilization and selectivity issues. Herein, we disclose an unprecedented dearomative trifluoromethylative aminocarbonylation of arenes via bifunctional coordination to chromium, providing a modular platform for the construction of amides possessing trifluoromethyl (CF3) groups and three-dimensional rings. Its versatility further enabled a switchable difluoromethylation or trifluoromethylation aminocarbonylation of arene C-H bonds. A possible mechanism was proposed based on control experiments. Finally, the synthetic utility was well demonstrated by diverse applications in the total synthesis of CF3-functionalized amide-type drugs, including praziquantel, nateglinide, maraviroc and alloyohimbane.
