Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:Reactions of acyl chlorides with hydroxylamine hydrochloride and NaHCO3 generate the corresponding hydroxamic acid products in ethyl acetate and water at room temperature for 5 min. This is a simple and efficient method to synthesize a wide range of hydroxamic acids from carboxylic acids in excellent yield and high purity after simple post-treatment without chromatographic purification. In this process, the highlights are the simple separation of products and cheaply available reagents.

Abstract:A copper‐catalysed (diacetoxyiodo)benzene‐promoted aerobic esterification reaction of acetoacetamides was developed for the synthesis of oxamates, which are useful precursors in synthetic organic chemistry. This practical and mild synthetic approach proceeded at 25 °C under open‐air conditions and afforded methyl 2‐oxo‐2‐(phenylamino)acetates in good to excellent yields combined with C−C σ‐bond cleavage and formal oxidative C−H bond functionalization. A mechanism is proposed.

Abstract:Adding an amine moiety to a carbonyl group poses a challenging synthetic task; nevertheless, it is a crucial step in developing numerous bioactive molecules. Here, we report a straightforward and efficient protocol for synthesizing various amides using deoxygenated amidation of abundant carboxylic acids. This modular one-pot method uses feedstock carboxylic acids as acyl surrogates, amines, and sulfonyl hydrazides as coupling reagents. This method is a simple and affordable way to introduce structural diversity. Several biologically relevant skeletons have been easily synthesized under mild conditions. We identified the crucial reaction intermediate, bromophosphonium ion and acyloxyphosphonium ion, through NMR spectroscopy. We have also used this protocol for the late-stage functionalization of pharmaceuticals and blockbuster drugs.

Abstract:An efficient and operationally simple method for the synthesis of α-acyloxy ketones through the readily available 2-methylimidazole-promoted reaction of α-hydroxy ketones and anhydrides is developed. In the reaction, the anhydrides act as both a substrate and a solvent. The new method features good substrate tolerance, mild reaction conditions, readily accessible starting materials, and excellent yields, providing facile and green access to the targets. Importantly, the reaction also avoids the use of reagents with pungent odors, such as pyridine, in traditional esterification, which may promote the development of organocatalysis using nitrogen-containing heterocyclic compounds as catalysts.

Abstract:The novel efficient procedure has been developed for the oxathioacetalization of carbonyl compounds and 2-mercaptoethanol at room temperature under solvent-free condition. The results showed that the novel multi-SO 3 H functionalized ionic liquid was very efficient for the reactions with the good to excellent yields in short time. Operational simplicity, without need of any solvent, room temperature, low cost of the catalyst used, high yields, applicability to large-scale reactions, reusability and chemoselectivity are the key features of this methodology.

Abstract:2-oxazolines are common moieties in numerous natural products, pharmaceuticals, and functional copolymers. Current methods for synthesizing 2-oxazolines mainly rely on stoichiometric dehydration agents or catalytic dehydration promoted by specific catalysts. These conditions either generate stoichiometric amounts of waste or require forcing azeotropic reflux conditions. As such, a practical and robust method that promotes dehydrative cyclization while generating no byproducts would be attractive to oxazoline production. Herein, we report a triflic acid (TfOH)-promoted dehydrative cyclization of N-(2-hydroxyethyl)amides for synthesizing 2-oxazolines. This reaction tolerates various functional groups and generates water as the only byproduct. This method affords oxazoline with inversion of α-hydroxyl stereochemistry, suggesting that alcohol is activated as a leaving group under these conditions. Furthermore, the one-pot synthesis protocol of 2-oxazolines directly from carboxylic acids and amino alcohols is also provided.

Abstract:Alkenes react with substituted N ‐hydroxysulfonamides and tetrabutylammonium periodate under metal catalyst free conditions to give oximes in good to high yields. Abstract:Herein, we report a highly efficient ZnI2-triggered oxidative cross-coupling reaction of P(O)-H compounds and amines. This operationally simple protocol provides unprecedented generic access to phosphinic amides/phosphoramidate derivatives in good yields and short reaction time. Besides, the reaction proceeds under mild conditions, which avoids the use of hazardous reagents, and is applicable to scale-up syntheses as well as late-stage functionalization of drug molecules. The stereospecific coupling is also achieved from readily available optically enriched P(O)-H compounds.

Abstract:An simple, efficient method for the construction of an array of α-ketoamides has been described from readily available O-benzoyl hydroxylamines and diazo compounds as starting materials by the combined use of CuI as a catalyst and H2O as oxygen source. The mechanism research reveals that the O-benzoyl hydroxylamines serve a dual role as both amine source and oxidant, thereby obviating the need for additional oxidants in the transformation process. This methodology could afford a wider range of products in good to excellent yields for most substrates, and provides a new idea for synthesis of α-ketoamide.

Abstract:Allylamines have long been recognized as valuable synthons because of their excellent reactivity in organic synthesis. Here, an efficient amination reaction of allenyl ethers via copper/Lewis acid synergistic catalysis has been established, providing straightforward access to diverse functionalized Z-oxalylamines and E -halogenated oxalylamines in good to excellent yields with high regio- and stereoselectivities. The developed method tolerates more than 100 examples that include late-stage functionalization of bioactive molecules, and features gram-scale synthesis of oxalylamines with high turnover number (TON > 1000) under mild and simple conditions. The applicability of the protocol is further demonstrated with the construction of drug molecules.

Abstract:Oxazoles are important five-membered heterocycles that contain both nitrogen and oxygen atoms. Due to their wide range of biological activities, many oxazoles demonstrate potential for extensive application in various fields, including medicinal chemistry. Trifluoromethyl carbinol, an important pharmacophore, contains both trifluoromethyl and hydroxyl groups and is common in molecules with important biological activities. Constructing oxazoles that contain a trifluoromethyl carbinol unit is undoubtedly important and valuable for expanding the chemical space in drug discovery. In this study, a simple and efficient method was developed for the synthesis of oxazoles containing a CF3-substituted alcohol unit via the tandem cycloisomerization/hydroxyalkylation of N-propargylamides with trifluoropyruvates through a rational Lewis acid catalytic mechanism. This Zn(OTf)2-catalyzed synthetic protocol is operationally simple and provides a series of oxazoles in moderate to good yields. The protocol demonstrates broad substrate scope, high functional group tolerance, and high atom economy and can achieve gram-level reactions, indicating the strong possibility of its practical application.

Abstract:Background: The development of simple yet efficient functionalization protocols has gained considerable importance for the discovery of useful heterocycle compounds, such as isoxazolo, isoindol and their derivatives. 3a-hydroxyisoxazolo[3,2-a]isoindol-8(3aH)-ones are a type of novel heterocycles first reported by Lu group involving Ph3P-catalyzed annulation process. In this work, we aim to explore the synthesis of this novel complex heterocycles using simple base catalyst from internal alkynoates with N-hydroxyphthalimide. Methods: The reactions were conducted in the Schlenk flasks, protected by N2. The products were identified by TLC, and isolated by column chromatography on silica gel (200-300 mesh) using petroleum ether (60-90 oC) and ethyl acetate. All products were characterized by 1H NMR, 13C NMR and mass spectroscopy. Results: A highly active base-promoted system based on NaOAc•3H2O has been designed for the oxa- Michael-aldol [3+2] annulation reactions of internal alkynoates with N-hydroxyphthalimide, giving a series of pharmaceutically attractive 3a-hydroxyisoxazolo[3,2-a]isoindol-8(3aH)-ones in synthetically useful yields of up to 98%. Only 10 mol% NaOAc•3H2O is needed for the reaction at room temperature within 6 h. Besides, a plausible base-promoted intermolecular [3+2] oxa-Michael-aldol type mechanism is proposed. Conclusions: We have demonstrated the simple cascade reaction between alkynoates and NHPI as a facile access to pharmaceutically attractive 3a-hydroxyisoxazolo[3,2-a]isoindol-8(3aH)-ones, with a base-promoted intermolecular [3+2] oxa-Michael-aldol type mechanism. Target heterocycles with various functional groups were achieved in good to excellent yields at room temperature for 6 h. Such a synthetic methodology provides an effective approach of the synthesis of the fused heterocyclics in combination with CO2 utilization. Abstract:The esterification of carboxylic acids is an important reaction for preparing esters which find wide applications in various research fields. In this manuscript, we report an acid/iodide cooperative catalytic method which enables highly efficient esterification of carboxylic acids with a wide range of equivalent O-H nucleophiles including both alcohols and weak nucleophilic phenols. Under the reaction conditions, both aromatic and aliphatic carboxylic acids including those bearing functional groups work well, furnishing the corresponding esters in good to high yields. Moreover, this reaction is scalable and applicable to the modification of bioactive molecules. These results demonstrate the synthetic value of this new reaction in organic synthesis.

Abstract:We developed a facile one-pot synthesis of 2-oxazolines from carboxylic acid and 2-chloroethyl isocyanate, involving amide bond formation and a following intramolecular cyclization using 4-dimethylaminopyridine as the catalyst. A large variety of functional groups are well tolerated by the mild reaction conditions to afford diverse 2-oxazolines in good to excellent yields. This reaction will keep the chirality of the isocyanate at position 1, the R-2 substituted carbon. Microwave-assisted synthesis can further enhance the reaction yield and reduce the reaction time to <5 min. This method facilities the synthesis of 2-oxazolines for diverse applications, such as 2-oxazoline derived polymers and materials. (C) 2022 Elsevier Ltd. All rights reserved.
