Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A general route to either cis- or trans-2, 5-disubstituted tetrahydrofurans was described by using the nucleophilic addition of organolithium derivatives to tricyclic lactones, followed by a highly stereocontrolled acid-assisted reduction with sodium cyanoborohydride of the hemiketals formed. The stereoselectivity observed can be rationalized by the preferential approach of the hydride on the less hindered face of an oxonium ion intermediate. The cis- or trans-2, 5-disubstituted tetrahydrofurans were provided by the hot decomposition and then hydrogenation of tricycle compounds produced.

Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:An effective method to synthesize alpha-functionalized furan and pyrrole derivatives was developed using 2-alkoxy-2,3-dihydrofurans as modular precursors. This protocol featured a previously unreported tandem nucleophilic substitution/heteroaromatization reaction. Nucleophiles such as indole, alpha-oxoketene dithioacetal, trimethoxybenzene, and dimethoxynaphthalene can react readily with 2-alkoxy-2,3-dihydrofurans to afford a-functionalized five-membered ring heterocycles in the presence of acid catalysts, such as copper bromide and iron chloride. The mechanism of the reaction was also discussed, in which the first step, nucleophilic substitution, is the key in triggering the succeeding heteroaromatization. This method can also be extended to the synthesis of dihydrothiophenes.

Abstract:A [1+4] annulation of 1,2-dicarbonyl compounds with a,ss-unsaturated ketones has been readily realized under the mediation of P(NMe2)3, producing polysubstituted 2,3-dihydrofurans bearing a quaternary carbon center in fair to good yields with a broad substrate scope. This reaction accordingly constitutes a new synthetic method for such compounds. In some cases, a [1+2] annulation reaction competitively occurs, delivering polysubstituted cyclopropanes. With the aid of density functional theory (DFT) calculation, a plausible mechanism for the [1+4] annulation and the chemoselectivity between [1+2] and [1+ 4] annulations has been discussed.

Abstract:In this paper, an efficient way to synthesize 2-(1'(Z)-iodoalkenyl)tetrahydrofurans from 4,5-alkadienols and 12 was developed. The reaction of the 4,5-allenols with a substituent in the 3-position afforded the trans-2,3-disubstituted tetrahydrofurans with very high diastereoselectivity. However, when the axially optically active 4,5-allenol was treated with 12 in n-hexane, the efficiency for chirality transfer was low. This problem was circumvented by conducting the reaction in CH2Cl2 at room temperature and applying N-iodosuccinimide as the electrophilic reagent; however, the Z/E ratio for the products is much lower. Highly optically active Z-products may be prepared via the kinetic resolution via a Sonogashira coupling reaction with propargyl alcohol.

Abstract:An unusual I2‐mediated triple C(sp3)−H functionalization reaction between aryl methyl ketones and dimethyl sulfoxide to form polysubstituted furans has been developed. In this transformation, dimethyl sulfoxide functions as a dual synthon via C(sp3)−H functionalization with formation of two C−C bonds, one C−O bond, and one C−S bond in a one‐step process. Iodine is crucial as promoter of the reaction, by which a number of ketones could be converted easily into 2,4,5‐trisubstituted furans. This provides an extremely simple and expeditious approach to high‐value‐added, biologically significant O‐heterocycles. Based on preliminary experiments, a plausible mechanism is proposed.

Abstract:An efficient route to 1,3,4-alkatrien-2-yldihydrofurans via a highly chemo- and regioselective palladium(0)-catalyzed coupling-cyclization reaction of 2-(2',3'-allenyl)acetylacetates with propargylic carbonates was reported. The reaction proceeded smoothly under neutral conditions, affording the O-attacked five-membered products with a 1,3,4-trienyl substituent exclusively in good to excellent yields. The products can be efficiently applied to the synthesis of polysubstituted 2-(dihydrofuryl)cyclopentenone derivatives via a catalytic Pauson-Khand reaction under ambient conditions. Abstract:Under different conditions, the reaction of propargyl alcohols and terminal alkynes leads to the selective formation of 1,4-diynes and polysubstituted furans/pyrroles. Water is the only byproduct in the selective synthesis of 1,4-diynes and pyrroles, and the strategy for the furan synthesis is of 100% atom economy.

Abstract:The cycloadditions of vinylidenecyclopropanes with aldehydes for the synthesis of tetrahydrofurans are detailed in this paper. The obtained polysubstituted tetrahydrofurans could serve as versatile intermediates to selectively produce furan, indene, and benzo[c]fluorene derivatives depending on the substituents at the tetrahydrofurans' rings and the varied reaction conditions, which clearly exhibit insights into the synthetic applications of tetrahydrofurans. The scope, limitations, and mechanisms of these transformations have been discussed in detail. Attractively, these cycloadditions of vinylidenecyclopropanes with aldehydes and the further transformations of the cycloadducts provide an efficient protocol for the construction of various medium- and large-size ring-fused tetrahydrofuran, furan, and benzo[c]fluorene derivatives.

Abstract:A facile access to 4-((trifluoromethyl) thio)-2,3-dihydrofurans from unprotected homopropargylic alcohols in high regioselectivity is reported. Abstract:In contast to the general [4+2] cycloaddition reactions of alkynylcyclopropane ketones reported in the literature, we report herein a Sc(OTf)3-catalyzed formal intermolecular [3+2] cycloaddition reaction of alkynylcyclopropane ketones with electron-rich aromatic aldehydes. 2,3,3,5-Tetrasubstituted tetrahydrofurans were obtained by this method, and the tetrahydrofuran skeleton was diastereoselectively constructed with a 2,5-trans configuration. This is also quite different to the 2,5-cis configuration usually obtained from typical [3+2] cycloaddition reactions of most donor–acceptor cyclopropanes. Additionally, the cycloadducts were subjected to a domino carbopalladation/C–H activation/C–C bond-formation process for the highly efficient construction of polycyclic skeletons.
