Abstract:In this paper, we have demonstrated a facile nucleophilic addition of 2-substituted diethyl malonate to various substituted 1,2-allenic ketones to afford β,γ-unsaturated enones using a catalytic amount of t-BuOK as the base. The reaction usually completes within 10min in acetone at room temperature. The stereoselectivity of the reaction with γ-substituted allenic ketones is very high affording the β,γ-unsaturated E-enones.

Abstract:The K2CO3 (10 mol %)-catalyzed 1,4-addition reaction of diethyl malonate with various substituted 1,2-allenic ketones leading to polyfunctionalized beta,gamma-unsaturated enones 3 or 4 was studied. With 3-unsubstituted 1-substituted-1,2-allenyl ketones, the highly selective formation of beta,gamma-unsaturated enones 4 was observed; with 1,2-allenyl ketones bearing one or two 3-substituents in the allenyl group, only beta,gamma-unsaturated enones 3 with an unmigrated carbon-carbon double bond were produced; with 3-monosubstituted-1,2-allenyl ketones Z-beta,gamma-unsaturated enones 3 were formed with excellent stereoselectivity (E:Z > 96:4); with propadienyl ketones, mixtures of beta,gamma-unsaturated enones 3 and 4 were formed. alpha-Pyrone derivatives were synthesized via the K(2)CO(3)-catalyzed or -promoted reaction of 1,2-allenic ketones with diethyl malonate via a sequential Michael addition-carbon-carbon double bond migration-lactonization process.

Abstract:Hochsubstituierte α,β-ungesättigte Cyclohexenone, ein zentrales Strukturmotiv in vielen Naturstoffen, sind mit der Titelreaktion zugänglich (siehe Schema). Die Diastereoselektivität bezüglich der beiden stereogenen Zentren (4- und 6-Position) ist hoch. Mit optisch aktiven Ausgangsmaterialien wurden die erwünschten Produkte ohne nachweisbare Racemisierung gebildet. Eine mögliche Erklärung der Stereoselektivität wird präsentiert.

Abstract:A catalytic enantioselective addition reaction of silyl enol ethers to ketomalonate in the presence of C-2-symmetric copper(II) bisoxazolines as catalysts is presented; the reaction has been studied for different ligands and substrates under various reaction conditions and proceeds in good yield and with ee's > 90% and a procedure for the preparation of an optically active hydroxy acid is shown.

Abstract:A highly enantioselective, organocatalytic Mukaiyama−Michael addition reaction of silyl ethers and α,β-unsaturated aldehydes has been developed. The process, catalyzed by MacMillan's chiral imidazolidinone, affords δ-keto aldehydes in high yields (56−87%) and high enantioselectivities (85−97% ee). Moreover, the reaction is applicable to a wide range of silyl ethers and α,β-unsaturated aldehydes and, as such, provides access to a range of important synthetic building blocks.

Abstract:Highly enantioselective Michael addition of malonates to enones catalyzed by dipeptide-derived multifunctional phosphonium salts has been developed. The newly established catalytic system was characterized with its wide substrate scope featured with aliphatic aldehyde-derived enones and substituted malonates. The gram scale-up synthesis of adducts can also be successfully achieved under optimal conditions with both excellent yield and enantioselectivity.

Abstract:An enantioselective Michael addition of ketones to alkylidenemalononitriles catalyzed by chiral primary amine I with (R)-5c as a co-catalyst in good yields (>90%) and with good enantioselectivities (85-96% ee) has been developed. Abstract:By employing a cinchonine-based thiourea as catalyst, highly enantioselective Michael addition reactions of 2-hydroxy-1,4-naphthoquinone to beta,gamma-unsaturated alpha-ketophosphonates were realized. The reaction afforded the corresponding beta-substituted carboxylates in excellent yields with high levels of enantioselectivities (94 -> 99% cc) upon quenching the generated parent structures with DBU and MeOH as a second nucleophile.

Abstract:The first chloramphenicol base-derived thiourea-catalyzed enantioselective Michael addition of malononitrile to α,β-unsaturated ketones is reported. The Michael adducts were obtained in good to excellent yields (up to 98% yield) and enantioselectivities (up to 94% ee). This reaction has a broad substrate scope to various α,β-unsaturated ketones. With this in mind, this methodology was successfully applied to the synthesis of a chiral piperidone, an advanced building block for dihydropyridinone P2X7 receptor antagonists.

Abstract:An efficient, organocatalytic enantioselective addition of cyclic diketones with beta,gamma-unsaturated a-keto esters has been developed that affords products in high yields (up to 95 %) and excellent enantioselectivity (up to >99?% ee) under mild conditions with a low catalyst loading (2.5 mol-%). The unsaturated a-keto esters are effectively coordinated and activated through hydrogen bonds with the squaramides and proved to be excellent hydrogen-bond acceptors in this asymmetric organocatalytic reaction. This reaction provides valuable and easy access to chiral Michael adducts, which are important moieties in the skeletons of biological and pharmaceutical molecules.
