Abstract:A series of biphenol-derived quaternary ammonium salts, originally developed as DNA-crosslinking agents, and carrying either two (i.e., 1) or four (i.e., 2) net positive charges, were investigated for their in vitro DNA-transcription- and acetylcholinesterase (AChE)-inhibitory activities. The effects of charge and type of linker between the two phenolic residues were systematically investigated. Several compounds showed good activities in both tests, which makes them potential lead candidates for drug design.

Abstract:Based on the structure characteristics of the lead compounds, 1, 1' octanedioyl-4, 4'-dimethyl-4, 4'-dibenzyl dipiperazinium dibromide (2) and 3, 8-disubstituted-3, 8-diazabicyclo [3.2.1]octanes (DBO), di-(3, 8-diazabicyclo [3.2.1]octane) diquaternary ammonium salts 3 a-c were designed and synthesized through a highly practical procedure. Target compounds 3 a-c and the hydrochloride salts of their precursors 10 a-c were evaluated for their in vivo analgesic and sedative activities. Interestingly, the introduction of an endoethylenic bridge in the piperazine of lead compound 2 causes loss of the analgesic activity and increases the toxicity dramatically. This result shows that the flexible conformation of piperazine in compound 2 is favorable for interaction with the receptor, and the quaternization of compounds 10 a-c is the main reason for the toxicity increase.

Abstract:A series of proline derivated spirocyclic quaternary ammoniums were synthesized and evaluated via in vivo analgesic activities. Compounds 9a showed the best analgesic effect with 84 % inhibition in mice. These compounds showed only micromolar Ki value by in vitro α7 nAChR binding test, which may indicate the form of active metabolite play a role in their in vitro analgesic activities.

Abstract:AIM:To design and synthesize novel AchE inhibitors.METHODS:The condensation of 2-bromo-5, 6-dimethoxy-indan-1-one with various aminoalkyl phenols in the presence of K2CO3 and acetonitrile gave the corresponding title compounds, and the in vitro AchE and BchE inhibitory activities were evaluated by the modified Ellman method.RESULTS:Sixteen novel target compounds 8a - p were synthesized, their structures were confirmed by 1H NMR, MS, IR and elemental analysis. Preliminary pharmacological test demonstrated that most of these compounds displayed high AchE inhibitory activities, the IC50 of the most potent inhibitor 8h was 50.0 nmol x L(-1), similar to that of Huperzine A (IC50 = 53.0 nmol x L(-1)), while all the compounds were almost inactive against BchE.CONCLUSION:2-Phenoxy-indan-1-one derivatives exhibit high activities of AchE inhibition and are worthy of further investigation.

Abstract:As a nonaddictive analgesic widely used in clinics, the LD50 of bulleyaconitine A is just only 0.92 mg/kg, which exhibits obvious toxicity. Therefore, 31 new non-natural C19-diterpenoid alkaloids (2a–w, 2′a–e, 3, 4a, and 4b) were designed and synthesized from bulleyaconitine A to develop nonaddictive analgesics with low toxicity. The chemical structures were characterized by 1H NMR, 13C NMR, and high-resolution mass spectrometry (HRMS) spectra. The analgesic activities were evaluated by a hot plate test in mice. At the dosage of 10 mg/kg, six compounds (2d, 2j, 2k, 2m, 2t, 2w) exhibited good analgesic activities (increased pain threshold >100%) with a long duration. Among them, 2w showed the best analgesic activity and the longest duration. Its pain threshold reached 166.35% in 15 min, peaked at 30 min (182.35%), and remained 82.59% even at 60 min.This article has not yet been cited by other publications.

Abstract:Twenty new 4,4′-dibenzoyl-1,1′-dibenzyl-1,1′-(decane-1,5-diyl)-piperazinium dihalides 5a-l and 1,1prime;-dibenzyl-1,1′-(decane-1,5-diyl) piperazinium dihydrochloride dihalides 6a-l were prepared and evaluated for their analgesic, sedative and anti-inflammatory activities. Structure-activity relationship studies indicated that the compounds 6c (Ar = 4-NO2C6H5) and 6k (Ar = 3-Me-C6H5) exhibited higher activities than others. Compared with the corresponding compounds 6k and 6l, the presence of benzoyl in the compound 5k and 5l exerted a contrary influence on the activities. 5h and 6h show the highest anti-inflammatory activity (59%, dose 20 mg/kg and 48%, dose 1 mg/kg) in the series 5 and 6, respectively.

Abstract:Natural product bufotenine (5) which could be isolated from Venenum Bufonis, has been widely used as a tool in central nervous system (CNS) studies. We present here its quaternary ammonium salt (6) which was synthesized with high yields using 5-benzyloxyindole as raw materials, and we firstly discover its analgesic effects in vivo. The analgesic evaluation showed that compounds 5 and 6 had stronger effects on the behavior of formalin induced pain in mice. Moreover, the combination of compound 6 and morphine has a synergistic effect. We intended to explain the molecular mechanism of this effect. Therefore, 36 analgesic-related targets (including 15 G protein-coupled receptors, 6 enzymes, 13 ion channels, and 2 others) were systemically evaluated using reverse docking. The results indicate that bufotenine and its derivatives are closely related to acetyl cholinesterase (AChE) or α4β2 nicotinic acetylcholine receptor (nAChR). This study provides practitioners a new insight of analgesic effects.

Abstract:Azo coupling reactions of ( Z )-2-(3,3-dimethyl-3,4-dihydroisoquinolin-1(2 H )ylidene)- N -(2,4-xylyl)-acetamide with diazonium salts were used to synthesize 2-(3,3-dimethyl-3,4-dihydroisoquinolin-1-yl)-2-(2-arylhydrazono)- N -(2,4-xylyl)-acetamides. All compounds showed analgesic effects in the hotplate test, exceeding the effect of metamizole sodium. In the case of the most active hydrazone, the defensive reflex time in mice was 27.10 min (compared with 16.60 min for metamizole sodium).

Abstract:Four new hemorphin‐4 peptides containing steric‐restricted nonnatural amino acids were synthesized and characterized. All peptides demonstrated anticonvulsant, antinociceptive, and antioxidant activity with low neurotoxicity. They behave as weak protolytes and are in a soluble, stable molecular form at physiological pH values. Some new hemorphin‐4 analogs with structures of Xxx‐Pro‐Trp‐Thr‐NH2 and Tyr‐Yyy‐Trp‐Thr‐NH2, where Xxx is 2‐amino‐3‐(4−hydroxy‐2,6‐dimethylphenyl)propanoic acid or 2‐amino‐3‐(4‐dibenzylamino‐2,6‐dimethylphenyl)propanoic acid, and Yyy is (2S,4S)‐4‐amino‐pyrrolidine‐2‐carboxylic acid, were synthesized and characterized by electrochemical and spectral analyses. In vivo anticonvulsant and antinociceptive activities of peptide derivatives were studied after intracerebroventricular injection in mice. The therapeutic effects of the modified peptides on seizures and pain in mice were evaluated to provide valuable insights into the potential applications of the novel compounds. Electrochemical characterization showed that the compounds behave as weak protolytes and that they are in a soluble, stable molecular form at physiological pH values. The antioxidant activity of the peptides was evaluated with voltammetric analyses, which were confirmed by applying the 2,2‐Diphenyl‐1‐picrylhydrazyl method. The compounds showed satisfactory results regarding their structural stability, reaching the desired centers for the manifestation of biological activity without hydrolysis processes at 37°C and physiological pH. Dm‐H4 and H4‐P1 exhibited 100% and 83% potency to suppress the psychomotor seizures in the 6‐Hz test compared to 67% activity of H4. Notably, only the H4‐P1 had efficacy in blocking the tonic component in the maximal electroshock test with a potency comparable to H4. All investigated peptides containing unnatural conformationally restricted amino acids showed antinociceptive effects. The analogs Db‐H4 and H4‐P1 showed the most pronounced and long‐lasting effect in both experimental models of pain induced by thermal and chemical stimuli. Dm‐H4 produced a dose‐dependent thermal antinociception and H4‐P2 inhibited only formalin‐induced pain behavior.

Abstract:7α-Phenyl-6α,14α- endo -etheno-tetrahydrothebaine is an analogue of morphine but with much weaker affinity and efficacy to opioid receptors. Three compounds with o -, m - and p -amino substitution on its 7α-phenyl group were designed and synthesized to evaluate their κ-opioid receptor agonistic activity and antinociceptive effect. The introduction of amino group greatly increased the binding activity to κ-opioid receptor but only compound with p -substitution showed agonistic activity with potency similar to the marketed κ agonist butorphanol. In vivo antinociceptive test showed that compound with p -amino substitution displayed highest antinociceptive activity while compounds with o -, m -amino substitution showed partial or little analgesia effects. All these data indicate that p -amino substitution conferred κ agonist activity, suggesting that 7α-phenyl-6α,14α- endo -etheno-tetrahydrothebaines with a p -amino substitution may contain a novel pharmacophore component and could be considered as a leading compound for novel antinociceptive agents.

Abstract:The synthesis and structural characterization of new derivatives of [B12H12]2− is of fundamental interest and is expected to allow for extended applications. Herein we report on the synthesis of a series of amidine, amide, urea and isocyanate derivatives based on the amino-closo-dodecaborate anion [B12H11NH3]−. Their structures have been confirmed by spectroscopic methods, and nine crystal structures are presented.

Abstract:A series of dispirocyclopiperazinium analogues were synthesized by the reaction of hexanedioyl chloride, piperazine analogues and dibromides. The structures of target compounds were characterized by H-1 NMR, C-13 NMR spectra and elemental analysis. Preliminary bioassay test indicated that most of the compounds showed significant analgesic activity, and the structure-activity relationship was revealed.

Abstract:AIM: To look for the new kind of analgesic compounds with higher activity and lower toxicity. METHODS: Six 1, 1′-diacyl-4, 4, 4′, 4′-tetraalkyl dipiperazinium dibromides (IVa ∼ f) were synthesized from acylation of 1-methyl piperazine with corresponding diacide chloride (IIa ∼ d), and then quaternization by different alkyl bromide. RESULTS: Preliminary biological activity test showed that all the tested compounds IVa∼f showed obvious analgesic and sedative activity, especially for IVe (dose 20 mg·kg-1, analgesic activity 80.3%, and sedative activity 95.3%) and IVf (dose 0.5 mg · kg-1, analgesic activity 75.5%, sedative activity 53.8%). CONCLUSION: The distance between two nitrogen cations affects the biological activities and the toxicity distinctively. When n=4, the analgesic and sedative activities are best in IVa∼d, the toxicity decreases as the n decreases. The introduction of benzyl group increases the analgesic and sedative activities and decreases toxicity obviously.

Abstract:Tertiary amines 3-(dialkylamino)-2-hydroxypropyl 4-[(alkoxycarbonyl)amino]benzoates and their quaternary ammonium salts were synthesized. The final step of synthesis of quaternary ammonium salts was carried out by microwave-assisted synthesis. Software-calculated data provided the background needed to compare fifteen new resulting compounds by their physicochemical properties. The acid dissociation constant (pKa) and lipophilicity index (log P) of tertiary amines were determined; while quaternary ammonium salts were characterized by software-calculated lipophilicity index and surface tension. Biological evaluation aimed at testing acetylcholinesterase and butyrylcholinesterase-inhibiting activity of synthesized compounds. A possible mechanism of action of these compounds was determined by molecular modelling study using combined techniques of docking; molecular dynamics simulations and quantum mechanics calculations.

Abstract:Two new hybrids of local anesthetic analogs and polyoxometalates {(Bu4N)(2)[MO6O18NC6H4COOCH3] (1) and (Bu4N)(2)-[Mo6O18NC6H4COOC2H5] (2)} have been prepared in high purity and moderate yields using the easy reaction route of [alpha-Mo8O26](4-) with corresponding aromatic amine hydrochlorides in the presence of a N,N'-dicyclohexylcarbodiimide (DCC), and have been characterized by H-1 NMR, IR, UV/Vis, ESI-MS, and single-crystal X-ray diffraction study. Compound I crystallizes in the triclinic space group Pi with dimer structure, while Compound 2 crystallizes in the monoclinic space group P2(1)/n with dimer and chain structure. These two hybrids have the typical Mo equivalent to N triple bond that links a phenylimido group in local anesthetic analogs to the Mo atom of a hexamolybdate cluster. Because the formed Mo equivalent to N bond is not stable in basic solution, this property is useful and could be developed as a novel controlled-release technology for drug delivery. Abstract:A novel series of 2-substituted-quinazolin-4(3H)-ones were synthesized by reacting 3,5-disubstituted-anthranilic acid with acetic anhydride/benzoyl chloride, which were further reacted with different primary amines to obtain 2,6,8-substituted-quinazolin-4(3H)-ones 6a-f, 7, 8. All the synthesized compounds were characterized and screened for analgesic and anti-inflammatory activities. Compounds 6,8-dibromo-2-phenyl-3-(4'-carboxyl phenyl)quinazolin-4(3H)-one 7 and 6,8-dibromo-2-phenyl-3-(2'-phenylethanoic acid)quinazolin-4(3H)-one 8 displayed good analgesic and anti-inflammatory activity in comparison to the reference standards acetyl salicylic acid and indomethacin, respectively.

Abstract:Esters of N,N-diethylaminoacetic acid and hydroxyamines, obtained from structurally different natural monoterpenes, were pharmacologically examined. It was proved that salts of the obtained compounds had local anesthetic properties in infiltration anesthesia, compounds 9, 6 and 8 having been more potent than lidocaine. Compounds 7-9 slightly increased the arrhythmogenic dose, and compound 12 - the lethal dose of strophanthin. All the examined compounds transiently decreased the arterial blood pressure and displayed a cardiopressive activity.
