Abstract:This paper reported the investigation of structure-activity relationship of the strong analgetic alkaloid epibatidine and it's analogues by using AM1 method. Some information was given according to the calculation results of HOMO electron density and electron charge distribution etc. It is also indicated that the biophores were the bridge nitrogen and exo-pyridine moiety of these alkaloids.

Abstract:According to the theoretical calculation of epibatidine and nicotine by using AM1 method, we made a good explaination for the phenomenon that the analgetic action of epibatidine doesn't be reversed by naloxone as usual analgetics for the first time, and this result supports the results of Daly's experiment. In addition, as epibatidine and micotine have many similarities such as structure, activity and other qualities, so we believe that they also have the same action receptor-nicotine receptor.

Abstract:μ opioid receptor (MOR) agonists have been widely applied for treating moderate to severe pain. However, numerous adverse effects have been associated with their application, including opioid-induced constipation (OIC), respiratory depression, and addiction. On the basis of previous work in our laboratory, NAP, a 6β- N-4'-pyridyl substituted naltrexamine derivative, was identified as a peripheral MOR antagonist that may be used to treat OIC. To further explore its structure-activity relationship, a new series of NAP derivatives were designed, synthesized, and biologically evaluated. Among these derivatives, NFP and NYP significantly antagonized the antinociception effect of morphine. Whereas NAP acted mainly peripherally, its derivatives NFP and NYP actually can act centrally. Furthermore, NFP produced significantly lesser withdrawal symptoms than naloxone at similar doses. These results suggest that NFP has the potential to be a lead compound to treat opioid abuse and addiction.

However, their prolonged use was associated with undesirable side effects. Many new strategies to reduce side effects have been proposed, but not without disadvantages. Using a hot plate model as a phenotypic screening method, our studies identified (3R,4S)-9d with a new scaffold as a potent analgesic with ED50 values of 0.54 mg/kg and 0.021 mg/kg in hot plate and antiwrithing models, respectively. Mechanistic studies showed that it elicited its analgesic effect via the active metabolite (3R,4S)-10a. The mechanism of (3R,4S)-10a-induced activation of the μ opioid receptor (MOR) was proposed by means of molecular dynamics (MD) simulation.

Abstract:Opioid analgesics are potent pain-killer and are widely used in clinical practice.Opioid receptors are classified into μ,δ,κ subtypes.Most of the currently available opioid analgesics are μ opioid receptor agonists, but they cause undesired side effects such as physical dependence, respiratory depression and constipation and so on.The antalgic activity of δ receptor agonist is weaker than that of μ agonist, but it can modulate the activity of μ receptor and prevent its addiction. Therefore, discovering and designing compounds with μ/δ dual action is a new way for finding analgesics with low toxicity and less side effects.This review summarizes the development of compound with dual μ/δ activities and provides a new strategy for designing analgesics with low toxicity and less side effects.

Abstract:AIM: To look for the new kind of analgesic compounds with higher activity and lower toxicity. METHODS: Six 1, 1′-diacyl-4, 4, 4′, 4′-tetraalkyl dipiperazinium dibromides (IVa ∼ f) were synthesized from acylation of 1-methyl piperazine with corresponding diacide chloride (IIa ∼ d), and then quaternization by different alkyl bromide. RESULTS: Preliminary biological activity test showed that all the tested compounds IVa∼f showed obvious analgesic and sedative activity, especially for IVe (dose 20 mg·kg-1, analgesic activity 80.3%, and sedative activity 95.3%) and IVf (dose 0.5 mg · kg-1, analgesic activity 75.5%, sedative activity 53.8%). CONCLUSION: The distance between two nitrogen cations affects the biological activities and the toxicity distinctively. When n=4, the analgesic and sedative activities are best in IVa∼d, the toxicity decreases as the n decreases. The introduction of benzyl group increases the analgesic and sedative activities and decreases toxicity obviously.

Abstract:7α-Phenyl-6α,14α- endo -etheno-tetrahydrothebaine is an analogue of morphine but with much weaker affinity and efficacy to opioid receptors. Three compounds with o -, m - and p -amino substitution on its 7α-phenyl group were designed and synthesized to evaluate their κ-opioid receptor agonistic activity and antinociceptive effect. The introduction of amino group greatly increased the binding activity to κ-opioid receptor but only compound with p -substitution showed agonistic activity with potency similar to the marketed κ agonist butorphanol. In vivo antinociceptive test showed that compound with p -amino substitution displayed highest antinociceptive activity while compounds with o -, m -amino substitution showed partial or little analgesia effects. All these data indicate that p -amino substitution conferred κ agonist activity, suggesting that 7α-phenyl-6α,14α- endo -etheno-tetrahydrothebaines with a p -amino substitution may contain a novel pharmacophore component and could be considered as a leading compound for novel antinociceptive agents.

Abstract:Dezocine, a synthetic opioid, introduced in 1970s as an analgesic, was redeveloped for relieving moderate to severe pain by Yangtze River Pharmaceutical Group in China in 2009. To date, dezocine occupies 45% of China’s opioid analgesic market. Along with dezocine being a dominated painkiller, a certain amount of research was conducted to elucidate dezocine’s action. In this review we summarize the current knowledge on the receptor, preclinical and clinical pharmacology of dezocine. Briefly, preclinical data show that dezocine is effective under varying pain conditions, particularly chronic neuropathic pain and cancer pain, through activation of opioid receptors, and inhibition of norepinephrine reuptake. Clinical data establish the effectiveness of dezocine either as a primary analgesic for postoperative pain management or a supplement for balanced analgesia. The receptor profile of dezocine is different from known pure μ agonists, and allows it to be used in combination with other opioids for additivity in efficacy or lower incidence of adverse effects.

Abstract:Peripheral selective μ opioid receptor (MOR) antagonists could alleviate the symptoms of opioid-induced constipation (OIC) without compromising the analgesic effect of opioids. However, a variety of adverse effects were associated with them, partially due to their relatively low MOR selectivity. NAP, a 6β-N-4'-pyridyl substituted naltrexamine derivative, was identified previously as a potent and highly selective MOR antagonist mainly acting within the peripheral nervous system. The noticeable diarrhea associated with it prompted the design and synthesis of its analogues in order to study its structure-activity relationship. Among them, compound 8 showed improved pharmacological profiles compared to the original lead, acting mainly at peripheral while increasing the intestinal motility in morphine-pelleted mice (ED(50) = 0.03 mg/kg). The slight decrease of the ED(50) compared to the original lead was well compensated by the unobserved adverse effect. Hence, this compound seems to be a more promising lead to develop novel therapeutic agents toward OIC.

Abstract:As a nonaddictive analgesic widely used in clinics, the LD50 of bulleyaconitine A is just only 0.92 mg/kg, which exhibits obvious toxicity. Therefore, 31 new non-natural C19-diterpenoid alkaloids (2a–w, 2′a–e, 3, 4a, and 4b) were designed and synthesized from bulleyaconitine A to develop nonaddictive analgesics with low toxicity. The chemical structures were characterized by 1H NMR, 13C NMR, and high-resolution mass spectrometry (HRMS) spectra. The analgesic activities were evaluated by a hot plate test in mice. At the dosage of 10 mg/kg, six compounds (2d, 2j, 2k, 2m, 2t, 2w) exhibited good analgesic activities (increased pain threshold >100%) with a long duration. Among them, 2w showed the best analgesic activity and the longest duration. Its pain threshold reached 166.35% in 15 min, peaked at 30 min (182.35%), and remained 82.59% even at 60 min.This article has not yet been cited by other publications.

Abstract:P2X3 receptors (P2X3R) are ATP-gated ion channels predominantly expressed in C- and Aδ-fiber primary afferent neurons and have been introduced as a novel therapeutic target for neurological disorders, including neuropathic pain and chronic cough. Because of its localized distribution, antagonism of P2X3R has been thoroughly considered, and the avoidance of issues related to CNS side effects has been proven in clinical trials. In this article, benzimidazole-4,7-dione-based derivatives were introduced as a new chemical entity for the development of P2X3R antagonists. Starting from the discovery of a hit compound from the screening of 8364 random library compounds in the Korea Chemical Bank, which had an IC50 value of 1030 nM, studies of structure–activity and structure–property relationships enabled further optimization toward improving the antagonistic activities as well as the drug’s physicochemical properties, including metabolic stability. As for the results, the final optimized compound 14h was developed with an IC50 value of 375 nM at P2X3R with more than 23-fold selectivity versus P2X2/3R, along with properties of metabolic stability and improved solubility. In neuropathic pain animal models evoked by either nerve ligation or chemotherapeutics in male Sprague-Dawley rats, compound 14h showed anti-nociceptive effects through an increase in the mechanical withdrawal threshold as measured by von Frey filament following intravenous administration.

Nicotinic acetylcholine receptors (nAChR), specifically nAChR containing α4β2* subunits (* denotes additional receptor subunits), are known for their role in nicotine dependence and addiction (1). These receptors are found throughout the central and peripheral nervous systems on neuronal, glial, and immune cells (2). Although varenicline, an α4β2 nAChR* partial agonist, is a useful smoking cessation pharmacotherapy, the largest individual cause of preventable death continues to be cigarette smoking. (1)
Computer‐Aided Design of Opioid Receptors Dual Action Ligands with Mitigated Side Effects As a Potential New Avenue for Analgesic and Substance Abuse Treatment

Abstract:Opioid Receptors have long been targeted for analgesic and substance abuse treatment. However, current drugs targeting opioid receptors, e.g., morphine, have side effects such as tolerance, dependence and respiratory depression that put the public health at high risk. Thus, there is an unmet need for the discovery of new opioid receptor ligands that have improved profiles than the current marketed drugs. We previously identified that dual action ligands of mu opioid receptor agonist/delta opioid receptor antagonist could exert analgesic activity with mitigated side effects, and thus may have an advantage over the current drugs. Subsequently, we have explored the structure-activity relationship of a 5′-aryl-14-alkoxypyridomorphinan scaffold that exhibits such a dual action profile. In addition to evaluating the pharmacokinetic properties of this scaffold, we also have performed computational docking studies of mu, delta, and kappa opioid receptors in their agonist and antagonist bound states. Docking results and structural analysis have unveiled key residues that may explain the agonist/antagonist mechanism as well as variant residues among different opioid receptor isoforms that can be targeted for selectivity. Encouragingly, the results from in vivo animal studies showed analgesic activity and less side effects versus morphine, such as tolerance, rewarding effect, and respiratory depression. These results suggest that this chemical series possessing the pyridomorphinan scaffold is worthy for further exploration for either novel analgesic agents or substance abuse treatment.

Abstract:Management of moderate to severe pain by clinically used opioid analgesics is associated with a plethora of side effects. Despite many efforts have been dedicated to reduce undesirable side effects, moderate progress has been made. In this work, starting from Tramadol, a series of 3-((dimethylamino)methyl)-4-hydroxy-4-(3-methoxyphenyl)piperidine-1-carboxamide derivatives were designed and synthesized, and their in vitro and in vivo activities were evaluated. Our campaign afforded selective μ opioid receptor (MOR) ligand 2a (KiMOR: 7.3 ± 0.5 nM; KiDOR: 849.4 ± 96.6 nM; KiKOR: 49.1 ± 6.9 nM) as potent analgesic with ED50 of 3.1 mg/kg in 55 °C hot plate model. Its antinociception effect was blocked by opioid antagonist naloxone. High binding affinity toward MOR of compound 2a was associated with water bridge, salt bridge, hydrogen bond and hydrophobic interaction with MOR. The high selectivity of compound 2a for MOR over δ opioid receptor (DOR) and κ opioid receptor (KOR) was due to steric hindrance of compound 2a with DOR and KOR. 2a, a compound with novel scaffold, could serve as a lead for the development of novel opioid ligands.

Abstract:A series of highly sterically hindered secondary amine analogues of pyridylmethylamine (7a-f, 8a-c) and positional isomeric analogues of ABT-594 (9a-c) were synthesized and evaluated for their in vivo analgesic activity. The compounds 7a and 7d show potent analgesic activity and lower toxicity. Some interesting structure-activity relationships have been revealed.

Abstract:Fifty‐three analogs of lappaconitine, a natural analgesic, were synthesized and their biological activities were studied. Several analgesic agents with potential for development were identified. The structural characteristics and structure–activity relationships of these lappaconitine analogs were analyzed and discussed through pharmacophore modeling and 3D‐QSAR. Alleviating pain is crucial for patients with various diseases. This study aimed to enhance the analgesic properties of lappaconitine, a natural drug, through structural modifications. Specifically, carbamate analgesic active fragments were innovatively introduced at multiple sites on the benzene ring of lappaconitine. A total of 53 lappaconitine analogs were synthesized and evaluated. Compounds 5a, 5c, 5e, 6, and 15j addressed the narrow therapeutic window of lappaconitine, enhancing drug safety. Notably, carbamate analogs exhibited significantly enhanced analgesic activity, with compounds 5a and 5c having ED50 values of 1.2 and 1.6 mg/kg, respectively, indicating higher potency than lappaconitine (3.5 mg/kg). A metabolic analysis of compound 5e was conducted in mice, revealing its primary metabolic processes and metabolites, and providing preliminary exploration for the druggability. Given the multiple analgesic targets of lappaconitine, its analgesic mechanism remains inconclusive. This study, for the first time, analyzed the pharmacological activity characteristics of the lappaconitine analogs using a pharmacophore model and established a three‐dimensional quantitative structure–activity relationship (3D‐QSAR) to elucidate the quantitative relationship between the structures of the synthesized compounds and their analgesic activities. These findings provide valuable guidance for future structural modification and optimization of analgesic drugs.

The use of opioid drugs, which are the strongest painkillers, is associated with several side effects, such as tolerance, addiction, overdose, and even death. Since the G protein-coupled receptors (GPCRs) play a crucial role in pain modulation, the discovery of new peptide ligands for GPCRs is a significant challenge for novel drug development. The aim of this review is to present peptides of human and animal origin with antinociceptive potential and to show the possibilities of their modification, as well as the design of novel structures. The study presents the current knowledge on structure-activity relationship in the design of peptide-based biomimetic compounds, the modification strategies directed at increasing the antinociceptive activity, and improvement of metabolic stability and pharmacodynamic profile. The procedures employed in prolonged drug delivery of emerging compounds are also discussed. The work summarizes the conditions leading to the development of potential morphine replacements.

Abstract:The opioids have been used for more than a thousand years and are not only the most widely prescribed drugs for moderate to severe pain and acute pain, but also the preferred drugs. However, their non-analgesic effects, especially respiratory depression and potential addiction, are important factors that plague the safety of clinical use and are an urgent problem for pharmacological researchers to address. Current research on analgesic drugs has evolved into different directions: de-opioidization; application of pharmacogenomics to individualize the use of opioids; development of new opioids with less adverse effects. The development of new opioid drugs remains a hot research topic, and with the in-depth study of opioid receptors and intracellular signal transduction mechanisms, new research ideas have been provided for the development of new opioid analgesics with less side effects and stronger analgesic effects. The development of novel opioid drugs in turn includes selective opioid receptor ligands, biased opioid receptor ligands, and multi-target opioid receptor ligands and positive allosteric modulators (PAMs) or antagonists and the single compound as multi-targeted agnoists/antagonists for different receptors. PAMs strategies are also getting newer and are the current research hotspots, including the BMS series of compounds and others, which are extensive and beyond the scope of this review. This review mainly focuses on the selective/biased/multi-targeted MOR/DOR/KOR (mu opioid receptor/delta opioid receptor/kappa opioid receptor) small molecule ligands and involves some cryo-electron microscopy (cryoEM) and structure-based approaches as well as the single compound as multi-targeted agnoists/antagonists for different receptors from 2019 to 2022, including discovery history, activities in vitro and vivo, and clinical studies, in an attempt to provide ideas for the development of novel opioid analgesics with fewer side effects.
