Abstract:Two novel classes of monospirocyclopiperazinium salts were designed, synthesized, and evaluated for their in vivo analgesic activities. Some interesting structure–activity relationships are revealed: (1) Spirocyclopiperazinium moiety is favorable to improve the analgesic activity; (2) The size and conformation of spirocyclopiperazinium moiety significantly affects the analgesic activity; (3) Phenylethyl group of 3d is a crucial pharmacophore. Among the compounds synthesized, 3d exhibited the most potent activity with low toxicity. Further antinociceptive mechanism studies of 3d showed that these compounds will be a new kind of analgesics.

Abstract:In order to improve the analgesic activity of lead compound 7a, two series of dispirocyclopiperazinium (DSPZ) salts 9a-h, 10a-e and compounds 14, 15 were synthesized and evaluated for their in vivo analgesic activity both by acetic acid induced writhing test and hot plate test. Compounds 9h, 14, and 15 exhibited better analgesic activities than 7a. Several important structure-activity relationships were revealed from this study: (1) the introduction of aryl group would obviously improve the activity; (2) it was favorable to enhance the analgesic activity and reduce the toxicity to incorporate alkyl group with suitable length in the molecule; (3) carbamate analogues displayed lower toxicity than carboxylic ester analogues; (4) hydroxylation and chlorination of lead compound could increase the analgesic activity in hot plate test.

Abstract:A series of monospirocyclopiperazinium salts were designed and synthesized to search for a peripherally-acting analgesic drug with low side effects. Extensive SAR studies revealed that a suitable NR2R3 was critical for the analgesic activity, which might be beneficial to expose the cationic nitrogen to bind to the receptor, and possibly interact with the receptor via π–π interaction. Introduction of substituting group on the N4-phenyl ring could improve the activity, and the best position was the 4-position. Compound 14n showed more potent analgesic activity (63%, 20μM/kg, sc) and holds promise for development as a mechanically new analgesic drug.

Abstract:A series of dispirocyclopiperazinium analogues were synthesized by the reaction of hexanedioyl chloride, piperazine analogues and dibromides. The structures of target compounds were characterized by H-1 NMR, C-13 NMR spectra and elemental analysis. Preliminary bioassay test indicated that most of the compounds showed significant analgesic activity, and the structure-activity relationship was revealed.

Abstract:Based on the structure characteristics of the lead compounds, 1, 1' octanedioyl-4, 4'-dimethyl-4, 4'-dibenzyl dipiperazinium dibromide (2) and 3, 8-disubstituted-3, 8-diazabicyclo [3.2.1]octanes (DBO), di-(3, 8-diazabicyclo [3.2.1]octane) diquaternary ammonium salts 3 a-c were designed and synthesized through a highly practical procedure. Target compounds 3 a-c and the hydrochloride salts of their precursors 10 a-c were evaluated for their in vivo analgesic and sedative activities. Interestingly, the introduction of an endoethylenic bridge in the piperazine of lead compound 2 causes loss of the analgesic activity and increases the toxicity dramatically. This result shows that the flexible conformation of piperazine in compound 2 is favorable for interaction with the receptor, and the quaternization of compounds 10 a-c is the main reason for the toxicity increase.

Abstract:Four novel cycloartanes, named sphaerophysone A (1), B (2), C (3) and D (4), were isolated from the ethanol extract of Sphaerophysa salsula (Pall.) DC. The structures were elucidated on the basis of spectral evidence, and the stereochemistry of compound 1 was defined by X-ray crystallographic analysis. Sphaerophysone B (2) may be an artifact formed in the isolation procedure.

Abstract:Seven new 1, 1-dialkyl-4-(3-bromopropionyl) piperazinium bromides were synthesized. Their structures were elucidated by IR, 1H NMR and elemental analysis. The compounds were screened for antitumor, analgesic and antioxide SOD activities. Though none of them showed antitumor activity, most of them showed distinct analgesic activity and antioxide SOD activity.

Abstract:A series of proline derivated spirocyclic quaternary ammoniums were synthesized and evaluated via in vivo analgesic activities. Compounds 9a showed the best analgesic effect with 84 % inhibition in mice. These compounds showed only micromolar Ki value by in vitro α7 nAChR binding test, which may indicate the form of active metabolite play a role in their in vitro analgesic activities.

Abstract:Management of moderate to severe pain relies heavily on opioid analgesics such as morphine, oxycodone, and fentanyl in clinics. However, their prolonged use was associated with undesirable side effects. Many new strategies to reduce side effects have been proposed, but not without disadvantages. Using a hot plate model as a phenotypic screening method, our studies identified (3R,4S)-9d with a new scaffold as a potent analgesic with ED50 values of 0.54 mg/kg and 0.021 mg/kg in hot plate and antiwrithing models, respectively. Abstract:Twenty new 4,4′-dibenzoyl-1,1′-dibenzyl-1,1′-(decane-1,5-diyl)-piperazinium dihalides 5a-l and 1,1prime;-dibenzyl-1,1′-(decane-1,5-diyl) piperazinium dihydrochloride dihalides 6a-l were prepared and evaluated for their analgesic, sedative and anti-inflammatory activities. Structure-activity relationship studies indicated that the compounds 6c (Ar = 4-NO2C6H5) and 6k (Ar = 3-Me-C6H5) exhibited higher activities than others. Compared with the corresponding compounds 6k and 6l, the presence of benzoyl in the compound 5k and 5l exerted a contrary influence on the activities. 5h and 6h show the highest anti-inflammatory activity (59%, dose 20 mg/kg and 48%, dose 1 mg/kg) in the series 5 and 6, respectively.

Abstract:The fragmentation patterns of a series of dispirocyclopiperazinium dibromides with strong analgesic activity were analyzed by positive ion electrospray ionization mass spectrometry in conjunction with tandem mass spectrometry (ESI-MSn). The [C2+Br−]+ ions showed the characteristic isotopic peaks with high intensity. In each of their MS2 spectra, only the [C2+Br−HBr]+ ion peak was observable. Further analysis indicated that a selective rearrangement occurred in the unsaturated spirocyclopiperazine ring to achieve dihydropyrrole moiety. Meanwhile, the [C]2+ ions were unique and always the base peaks. The ions [C2+Br−]+ and [C]2+ were formed from the equilibrium of precursor molecules 1 in solution, and the latter ions could not be observed in the MS2 spectra of ions [C2+Br−]+. The related fragmentation mechanisms were proposed.
Icetexane diterpenoids as Cav3.2 T-type calcium channel inhibitors from Salvia prattii and analgesic effect of their Semi-synthesized derivatives

Abstract:Ten new icetexane diterpenoids, salpratins E–N (1–10) and a known analogue (11) were characterized from Salvia prattii Hemsl. Structurally, 1 is the first 19(4 → 3)-abeo-icetexane diterpenoid featuring with a 6/7/6 ring system. The structures of isolated compounds were determined by comprehensive analyses of spectroscopic data, ECD calculation, and single-crystal X-ray diffraction. Biological studies initially revealed that 1, 7, 10, and 11 are notable Cav3.2 T-type Ca2+ channel (TTCC) inhibitors with IC50 values of 2.9, 5.1, 2.3, and 3.2 μM, respectively. Five icetexane related derivatives (13–17) were synthesized from an abietane type precursor, (+)-carnosic acid (12), for the purpose of overcoming the poor water solubility of aforementioned active compounds and further investigating diverse diterpenes with valuable activity. Among them, 13 and 14 showed potent inhibitions on Cav3.2, having IC50 values of 6.7 and 2.4 μM, respectively. Significantly, they exhibited dose-dependent (1, 3, and 10 mg/kg) and comparable analgesic effects as that of Z944, a TTCCs inhibitor under clinical trial for pain management, in the mouse acetic acid writhing test. These findings further enrich structural diversity and bioactivity of Salvia diterpenoids, as well as provide promising structural templates for the development of Cav3.2 analgesics.

Abstract:AIM: To look for the new kind of analgesic compounds with higher activity and lower toxicity. METHODS: Six 1, 1′-diacyl-4, 4, 4′, 4′-tetraalkyl dipiperazinium dibromides (IVa ∼ f) were synthesized from acylation of 1-methyl piperazine with corresponding diacide chloride (IIa ∼ d), and then quaternization by different alkyl bromide. RESULTS: Preliminary biological activity test showed that all the tested compounds IVa∼f showed obvious analgesic and sedative activity, especially for IVe (dose 20 mg·kg-1, analgesic activity 80.3%, and sedative activity 95.3%) and IVf (dose 0.5 mg · kg-1, analgesic activity 75.5%, sedative activity 53.8%). CONCLUSION: The distance between two nitrogen cations affects the biological activities and the toxicity distinctively. When n=4, the analgesic and sedative activities are best in IVa∼d, the toxicity decreases as the n decreases. The introduction of benzyl group increases the analgesic and sedative activities and decreases toxicity obviously.

Abstract:This study introduces a novel approach for synthesizing Benzoxazine-Centered Polychiral Polyheterocycles (BPCPHCs) via an innovative asymmetric carbene-alkyne metathesis-triggered cascade. Overcoming challenges associated with intricate stereochemistry and multiple chiral centers, the catalytic asymmetric Carbene Alkyne Metathesis-mediated Cascade (CAMC) is employed using dirhodium catalyst/Brønsted acid co-catalysis, ensuring precise stereo control as validated by X-ray crystallography. Systematic substrate scope evaluation establishes exceptional diastereo- and enantioselectivities, creating a unique library of BPCPHCs. Pharmacological exploration identifies twelve BPCPHCs as potent Nav ion channel blockers, notably compound 8g. In vivo studies demonstrate that intrathecal injection of 8g effectively reverses mechanical hyperalgesia associated with chemotherapy-induced peripheral neuropathy (CIPN), suggesting a promising therapeutic avenue. Electrophysiological investigations unveil the inhibitory effects of 8g on Nav1.7 currents. Molecular docking, dynamics simulations and surface plasmon resonance (SPR) assay provide insights into the stable complex formation and favorable binding free energy of 8g with C5aR1. This research represents a significant advancement in asymmetric CAMC for BPCPHCs and unveils BPCPHC 8g as a promising, uniquely acting pain blocker, establishing a C5aR1-Nav1.7 connection in the context of CIPN.

Abstract:This paper presents a comprehensive evaluation of novel 8-piperazinylcaffeine carboxylate ionic liquids, including their design, synthesis, characterization, analgesic and antibacterial properties, as well as docking studies. These unique salts were produced by combining 8-piperazinyl caffeine ( 8-PC ) with various carboxylic acids, some of which are commonly used nonsteroidal anti-inflammatory drugs (NSAIDs). Through in vivo experiments on female mice using the formalin test, the analgesic efficacy of different 8-PC salts with various NSAIDs was assessed. Results demonstrated that a majority of these salts exhibited significant analgesic activity when compared to NaIBP, a standard reference drug. Particularly noteworthy was the enhanced analgesic effect of the 8-PC 's NSAIDs salts ( 11a , 11c–e , and 11k ) compared to their corresponding sodium salts, which was attributed to the presence of the 8-PC cation (synergistic effect). Furthermore, all synthesized salts were subjected to in vitro testing against Gram-positive Staphylococcus aureus (PTCC 1133), Gram-negative Pseudomonas aeruginosa (ATCC 27853), and Escherichia coli (PTCC 1330) bacteria. Among them, salt 11k displayed notable antibacterial activity, especially against P. aeruginosa , a dangerous opportunistic pathogen. Additionally, docking analysis revealed strong binding of the synthesized 8-PC and NSAID salts to the active site of the COX-2 enzyme.

Abstract:1-[1-Phenylcyclohexyl]piperidine (Phencyclidine, PCP ) is a tricyclic compound with an aromatic ring that can create a negative charge area, a fundamental heterocyclic nitrogen ring that bears a local positive charge and a cycloalkyl ring that has a lipophilic character. This compound and its analogs are very potent and vastly abused psychotomimetic drugs that affect the central nervous system. Some analogs of PCP are prepared by replacements in substitution on this tricyclic structure for obtaining new ones with extra analgesic activities. New derivatives ( II-IV ) were synthesized with changing of substituting on aromatic, cyclohexyl, and amine moieties of PCP . In vivo evaluation of acute and chronic pain activities of the new compounds were carried out by applying tail immersion and formalin tests on mice. Then the results were compared with control and PCP groups at 10 mg/kg dosage. Obtained results suggested compounds II and IV showed better activity to decrease acute thermal and chemical pains, respectively, compared with other compounds ( III ) and PCP animal groups which may concern more nociception opioid receptor (NOP) antagonistic characterizes and a close affinity for NMDAantagonists in the family. It is concluded that compound II with three and compound IV with two changing moieties of PCP on phenyl, cyclohexyl, and amine rings were suitable candidates to reduce acute thermal and chronic chemical pains in Tail immersion and formalin tests, respectively.
