Abstract:A novel cycloartane, named sphaerophysone A, 9, 19-cycloart-7beta, 24beta, 25-triol-1-en-3-one, was isolated from the ethanol extract of Sphaerophysa salsula DC. The structure was elucidated on the basis of spectral evidences and confirmed by X-ray analysis, the stereochemistry of the compound was also defined by X-ray analysis.

Abstract:The cytotoxicity of three cycloartane triterpenoids, 9,19-cycloart-7beta, 24R, 25- triol-1-en-3-one (1), 9,19-cycloart-7beta, 24R, 25-triol-1-en-3-one 25-O-beta-D-glucopynanoside (2), and 25-O-beta-D-arabinopyranosyl-(1-->4)- beta-D-glucopyranosyl-9,19-cycloart-7beta, 24R, 25-triol-1-en-3-one (3), isolated from Sphaerophysa salsula was investigated on SF188, U87wt, MCF-7, and H460 cancer cell lines. Compound 1 showed the strongest activity. Cell cycle analysis was employed to elucidate the cytotoxicity on the tested U87wt cells, which led to G2/M arrest. In addition, from the Western blotting experiments, the expression of P21 is increased.

Abstract:Phytochemical study on rhizomes of Beesia calthifolia resulted in the isolation of five new (1–5) and three known (6–8) cycloartane triterpenoids possessing a hemiketal or ketal group at C-24 from the EtOAc fraction of 95% ethanol extract. Their structures were determined by spectroscopic and chemical methods, especially HRMS and 2D NMR techniques. Compounds 3 and 4 showed potential hepatoprotective activities against d-galactosamine induced human hepatic L02 cell damage.

Abstract:Six cycloartanes were isolated from ethanol extract of marine green algaCladophora fascicularis by column chromatography. Procedure of isolation and description of these compounds are given in this paper. The structures were elucidated as (1). 24-hydroperoxycycloart-25- en-3β-ol; (2). cycloart-25-en-3β 24-diol; (3). 25-hydroperoxycycloart-23-en-3β-ol; (4). cycloart-23-en-3β, 25-diol; (5). cycloart-23, 25-dien-3β-ol; and (6). cycloart-24-en-3β-ol by spectroscopic (MS, ID and 2D NMR) data analysis. Cycloartane derivatives are widely distributed in terrestrial plants, but only few were obtained in the alga. All these compounds that have been isolated from terrestrial plants, were found in the marine alga for the first time.

Abstract:The 70% ethanol extract of the whole plant of Souliea vaginata was purified by multi-chromatographic methods including macroporous resin,silica gel,Sephadex LH-20,and C18-reversed-phase column chromatography. A new spirocyclic cycloartane triterpenoid was isolated and identified as( 16 R*,20 R*,23 S*,24 R*,25 S*)-16,23: 23,26-diepoxy-15α,24,25-trihydroxy-9,19-cycloart-3β-O-β-D-xylopyranoside( 1),and named as soulieoside S. Its planar structure and relative configuration were determined by spectroscopic techniques including 2 D NMR and HRESI-MS. As one of the main components of S. vaginata,compound 1 was evaluated for its anti-inflammatory activity by a lipopolysaccharide( LPS)-stimulated NO production model in RAW264. 7 macrophages,but it didn't show NO production inhibitory effect.

Abstract:Two new cycloartane glycosides were isolated from the whole herbs of Camptosorus sibiricus Rupr.. By means of chemical and spectroscopic methods (IR, 1D- and 2D- NMR, HRMS, ESI-MS), the structures were established as (24 R)-3β,7β,24,25,30-pentahydroxycycloartane 3- O-β-D-glucopyranosyl-24- O-β-D-glucopyranosyl-30- O-β-D-glucopyranoside (1), and (24 R)-3β,7β,24,25,30-pentahydroxycycloartane 3- O-β-D-glucopyranosyl-(1→2)-β-D-glucopyranosyl-24- O-β-D-xylopyranosyl-(1→2)-β-D-glucopyranoside (2). The two new compounds were tested for their cytotoxicity in vitro against human tumor cell lines (A375-S2, Hela) using the MTT method, but both compounds were inactive.

Abstract:Two new cycloartane triterpenoids, (23R)-21,23:23,27-diepoxycycloarta-1,24-diene-3,27-dione (1) and (3 alpha)-(alpha-L-arabinopyranosyloxy)-1 alpha-hydroxy-23-oxocycloartan-28-oic acid (2), together with six known pentacyclic triterpenoids, 3-8, and five known C-29 steroids, 9-13, were isolated from Kleinhovia hospita. The structures of these compounds were determined by analysis of their spectroscopic data. Moreover, the absolute configuration of 1 was confirmed by quantum-chemical TDDFT calculation of its ECD spectrum. All compounds were evaluated for their cytotoxic activities against human colon carcinoma (HCT116) and gastric carcinoma (SGC7901) cell lines, and compounds 6, 7, 8, 11, and 12 exhibited antiproliferative activities with IC50 values ranging from 23.0 to 91.8 mu m.

Abstract:Phytochemical study of the aerial parts of Cassiaitalica Mill. (family: Fabaceae) growing in Saudi Arabia afforded one new cycloartane triterpene, named (22E)-3-β-hydroxycycloart-22-en-24-one (2), together with eight known compounds: β-sitosterol (1), uvaol (3), daucosterol (4), methyl 3,4-dihydroxybenzoate (5), emodin (6), 4-hydroxypheny-O-β-D-glucopyranoside (7), aloin (8) and rutin (9). The structure of the isolated compounds was determined by physical, chemical and spectral data (UV, IR, MS, 1D ((1)H, (13)C and DEPT) and 2D ((1)H-(1)H COSY, HSQC and HMBC) NMR), as well as by comparing with authentic samples. Compounds 3-5 and 7-9 were isolated for the first time from the plant. Compound 2 was evaluated for its cytotoxic activity against the L5178Y and PC12 cell lines. The total methanolic extract and compounds 5-9 exhibited free radical-scavenging activity using DPPH assay.

Abstract:To investigate the eembranoid diterpenes from the soft coral Sarcophyton sp. collected in the South China Sea. Re-peatedly column chromatography was performed for the isolation and purification. The structures were elucidated on the basis of extensive spectral data (IR, MS, ID- and 2D-NMR) analysis by comparing with literature data. A new eembranoid diterpene namely sareophyolide A (1), together with 7a, 8β-dihydroxydeepoxysareophine (2) were isolated from this species. The cembra-noid diterpenes with γ-lactone could be chemotaxonomic markers ofSarcophyton sp.

Abstract:Four new cycloartane triterpenoids, together with the known gardenolic acid B, were isolated from Kleinhovia hospita. The triterpenoids (1-3) contain a unique 21,23-diacetal side-chain, while compound 4 contains two alpha,beta-unsaturated ketone moieties. Their structures and relative configurations were determined by spectroscopic methods, including 2D NMR and IR. These compounds showed promising hepatoprotective effects on nitrofurantoin-induced cytotoxicity in human liver-derived Hep G2 cells.

Abstract:Fourteen 20,24-epoxy-cycloartane triterpenoids, including eight new ones (1–8), were isolated from 95% ethanol extract of the rhizomes of Beesia calthifolia. Their structures were determined by spectroscopic and chemical methods, especially 2D NMR and HRMS techniques. Among them, four new compounds (1–4) possess carbonyl groups at C-16, which were rarely found in cycloartane triterpenoids from this genus. Relative configuration at C-12 in beesioside III (9) and its aglycone (10) was revised to be 12α-OH rather than the reported 12β-OH. Some of the compounds showed potential hepatoprotective activities against human hepatic L02 cell damage induced by d-galactosamine.

Abstract:The chemical screening of an algae-containing marine sponge identified as Lendenfeldia species has isolated two 24-homoscalarane sesterterpenoids, including a known compound, felixin A ( 1 ) and a new analogue, lendenfeldarane V ( 2 ). The structure of 1 , obtained in a previous study, was cited for the first time in this study via single-crystal X-ray diffraction analysis. The structure of 2 was ascertained via 2D NMR experiments and a literature review. The absolute configurations of 1 and 2 were delineated with DP4+ computation and specific optical rotation. Homoscalarane 1 exhibited cytotoxicity towards MG63 human osteosarcoma cells.
