Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:Facile construction of 1,3-dienes building upon cross-electrophile coupling of two open-chain vinyl halides is disclosed in this work, showing moderate chemoselectivities between the terminal bromoalkenes and internal vinyl bromides. The present method is mild and tolerates a range of functional groups and can be applied to the total synthesis of a tobacco fragrance solanone.

Abstract:A direct ethynylation reaction between N-vinylacetamides and (bromoethynyl)triisopropylsilane catalyzed by palladium is reported. A broad scope of cyclic N-vinylacetamide derivatives was tolerated and the corresponding products could be obtained in acceptable yields. This work provides a general tool for the synthesis of conjugated enyne products using palladium(II) acetate as catalyst. The mild reaction conditions and reasonable results should make this method a useful synthetic protocol. Unfortunately, other bromo-substituted alkynes are found to be unreactive and could not afford the desired products under the current reaction conditions.

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:"Through space" palladium/hydrogen shift is an efficient strategy to achieve selective functionalization of a specific remote C-H bond. Compared with relatively extensive exploited 1,4-palladium migration process, the relevant 1,5-Pd/H shift was far less investigated. We herein report a novel 1,5-Pd/H shift pattern between a vinyl and an acyl group. Through the pattern, rapid access to 5-membered-dihydrobenzofuran and indoline derivatives has been achieved. Further studies have unveiled an unprecedented trifunctionalization (vinylation, alkynylation and amination) of a phenyl ring through 1,5-palladium migration relayed decarbonylative Catellani type reaction. A series of mechanistic investigations and DFT calculations have provided insights into the reaction pathway. Notably, it was unveiled that the 1,5-palladium migration in our case prefers a stepwise mechanism involving a PdIV intermediate.
