Abstract:Within the past few years electrospinning has been paid much attention due to the increasing interests of the researchers in the nanotechnology and the unique properties of the nanomaterials. This review focused on the electrospinning behaviors of biomacromolecules including synthetic biomedical polymers, natural-occurring biopolymer and the composite nanofibrous membranes. The electrospinning parameters of the biomacromolecules and the properties of the electrospun membranes such as the morphological stability, mechanical properties and degradation behavior were discussed in details. In addition, the application of the electrospun membranes for controlled drug release was also included. The incorporation method and the release behavior of the drugs were discussed and summarized, according to the solubility of the drugs.

Abstract:The local delivery and controllable release profiles make electrospun ultrafine fibers as potential implantable drug carriers and functional coatings of medical devices. Till date there is no literature report on drug delivery from acid-labile electrospun fibers, whose degradation and drug release behaviors respond to the local pathological pH environment. Acid-labile groups have been incorporated into nonbiodegradable backbones as crosslinkers or linkers of the side chains. A novel strategy was developed in this study to synthesize acid-labile polymers by introducing acetal groups into biodegradable backbone of poly(dl-lactide)–poly(ethylene glycol). In vitro release study showed that the total amount of drug released from acid-labile polymeric fibers was accelerated on account of pH-induced structural and morphological changes of fibrous mats and the degradation of matrix polymers, and the burst release was significant higher for polymers with higher contents of acid-labile segments. During the investigational period, almost no molecular weight reduction and mass loss was detected in neutral buffer solutions, but the degradation was enhanced in acid buffers with a two-stage degradation profile. Surface erosion mechanism was initially detected for fibrous mats with distinct fiber morphologies, and bulk degradation was determined during the following incubation for polymeric films resulting from the morphological changes.

Abstract:Biodegradable polymers,such as polylactic acid (PLLA),poly-ε-caprolactone (PCL),polyvinylpyrrolidone (PVP),polyacrylonitrile (PAN),polyethylene glycol (PEG),etc.with different ratios were intermittent electrospun to obtain nanofibers.The morphologies,structures,and properties of those nanofibers were characterized by SEM,XRD,FT-IR,etc.The degradation behavior of nanofibers was investigated in different solution.By analyzing the use of different degradation behaviors,the carrier for different types of DDS was preliminarily discussed for choosing suitable controllable-degradation carrier for different drugs to construct drug composite nanofibers.
Coaxial Electrospun Poly(L‐Lactic Acid) Ultrafine Fibers for Sustained Drug Delivery

Abstract:A coaxial electrospinning technique to fabricate core-shell ultrafine fiber mats for drug delivery application is described in this paper. Poly (L-lactic acid) (PLLA) and tetracycline hydrochloride (TCH) were employed as the shell and core materials, respectively. To investigate the feasibility of the resulting fiber mats for use as drug release carriers, these electrospun fibers were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), differential scanning calorimetry (DSC), and tensile testing. In vitro drug release behavior was also examined by ultraviolet-visible (UV-VIS) spectroscopy. Results indicated that a reservoir-type drug release device can be conveniently obtained through encapsulating TCH in the PLLA ultrafine fiber. The size of the ultrafine fibers had a significant effect on their physical-chemical properties. Furthermore, a sustained TCH release from these fiber mats was also observed. Consequently, the electrospun ultrafine fiber mats containing drugs may be used as drug release carriers or made into biomedical devices such as sutures and wound dressings.
Fabrication, Characterization and Drug Release Characteristics of Drug Loaded Poly (L-Lactic Acid) Fiber

Abstract:To develop a long-term released and implantable biodegradable fiber carrier for hydrophobic drug incorporation, fibers were fabricated by organic phase separation method. The structure of the fiber was observed by scanning electron microscopy and the state of the drug dispersed in the polymer was measured by differential scanning colorimetry (DSC) and Fouvier transform infrared spectroscopy (FTIR). The drug loading content and release profiles were determined by high performance liquid chromatography (HPLC) and ultra-violet spectrophotometry, respectively. Blank and drug loaded fibers were successfully fabricated and the drug was entirely encapsulated into the fiber. The drug was dispersed in the polymer with minicrystal and noncrystal form. The drug release profile was long-term sustained and could be regulated. The PLLA fibers in micrometer range successfully fabricated by organic phase separation method could be taken as sustained release and implantable drug carrier.
Electrospun Core-Shell Plla Nanofiber And Its Potential For Drug Release

Abstract:Electrospinning has been developed to fabricate composite nanofibers for biomedical applications such as drug delivery systems and tissue engineering scaffolds. The objective of this study was to evaluate the physical-chemical properties and drug release properties of electrospun core-shell poly (L-lactic acid) (PLLA) fibrous membranes used as a controlled release carriers for tetracycline hydrochloride (TCH). The fibrous membranes were fabricated by coaxial electrospinning in which PLLA (2:1 mixture of chloroform and acetone by weight as a solvent) and TCH/PLLA (2:1 mixture of methanol and chloroform by weight as a solvent) were used as the shell and the core materials, respectively. The electrospun fibers and their membranes were characterized by scanning electron microscope (SEM), transmission electron microscopy (TEM), and differential scanning calorimetry (DSC), whereas the mechanical performances of fibrous membranes were tested through tensile test. The drug release behavior was investigated by UV-VIS spectroscopy. Results indicated that the morphology of fibers and distribution of fiber diameter relied on the processing parameters, and the uniform composite fibers with eminent mechanical properties could be obtained when the core and shell solution concentrations were kept at 5 wt% (0.3 ml.h-1 flow rate) and 8-10 wt% (5.0 ml.h-I flow rate), respectively. TCH could be encapsulated in the PLLA ultrafine fibers and maintain a sustaining release from the fibrous membranes, suggesting that the thus obtained core-shell composite fibrous membranes can be a promising candidate for drug release carrier.

Abstract:This work is to develop novel electrospun poly(l-lactic acid) (PLLA) fiber mats for controllable delivery of hydrophobic and hydrophilic drugs. For this aim, bovine serum albumin (BSA, used as a hydrophilic model drug) was firstly enveloped into chitosan microspheres by spray drying. Benzoin (used as a hydrophobic model drug) was directly dissolved in PLLA solution and then the chitosan microspheres were suspended into the PLLA solution, which was used to prepare PLLA fiber mats by electrospinning. Polyvinylpyrrolidone (PVP) was added into the PLLA solution to tune the drug release behaviors. The results showed that the chitosan microspheres were uniformly distributed in the fibers. BSA had a short-term release while benzoin had a long-term and sustained release in all the dual drug delivery systems, and the release of both hydrophobic and hydrophilic drugs could be adjusted by changing the ratio of PVP/PLLA.
The Influence of Fiber Diameter of Electrospun Poly(lactic Acid) on Drug Delivery

Abstract:Electrospinning is a simple process for the production of fibers with diameters in the range from submicron to micron. Herein we aim to explore the influence of fibrous diameter on the drug delivery. The feasible methods by making choice of solvents and changing flow rate were used to prepare 5-fluorouracil-loaded polylactide (PLA) fibers with a large diameter gap. The drug release behavior in vitro was investigated and analyzed in phosphate buffer solution. The drug distribution and fiber diameter both affected the initial burst release. The results showed that all the asspun fibers could not avoid of burst release. The coarse fibers exhibited slight burst release as compared to fine fibers. During the second stage, the fine fibers released faster than that of the coarse fibers. For the whole release stage, the large-diameter fibers seemed to be beneficial for drug release in the long term and smoothly. The MTT results showed that the cytotoxicity of drugs was maintained.

Abstract:BACKGROUND:Drug delivery systems based on electrospun fibers have been under development for many years. However, studies of controllable long-term drug release from electrospun membrane systems and the underlying release mechanisms have seldom been reported.METHODS:In this study, electrospun membrane drug delivery systems consisting of the antibiotic ciprofloxacin hydrochloride and FDA-approved polymers are fabricated. Different second-component polymers are introduced to change the properties of a poly(d,l-lactide-co-glycolide) (PLGA) matrix, thereby altering the drug release behavior. On the basis of observations of morphology, cumulative release profiles, and determinations of release duration, the drug release kinetics and critical characteristics influencing drug release behavior are discussed.RESULTS:It is found that the drug release profiles can be divided into three stages according to the rate of drug release. Stage I is controlled by fiber swelling and diffusion according to Fick's second law. Stage II is controlled by diffusion through a fused membrane structure, which results in very slow drug release. Stage III is controlled by polymer degradation and involves release of the remaining drug.CONCLUSIONS:The results of this study of release mechanisms should provide a basis for adjustments of drug release dosage and duration, thereby contributing to the development of drug delivery systems satisfying clinical requirements.

Abstract:Wound dressing, which can release anti‐infectives in a controlled way, is taking an important role in the treatment and recovery of the open wound. An adequate release of antibiotics can prevent infections from microorganisms effectively. Among the new candidates of fabricating base materials for wound dressing, electrospinning fiber mats are attracting numerous attentions for their excellent performance in controlled drug delivery. The drug release behavior of electrospinning fiber mats can be tuned by changing the chemical components and the geometric structures of the mats. In this study, fiber mats with different geometric structures, which composed of poly‐ε‐caprolactone (PCL), polyethylene glycol (PEG), and ciprofloxacin (Cip) with different blending ratios, were successfully fabricated by direct‐writing melt electrospinning, and the release behavior of Cip were subsequently investigated in vitro. The results showed that the addition of PEG improved the hydrophilicity of the mats, which in turn affected the manner of drug release. The presence of PEG changed the releasing mechanism from a non‐Fickian diffusion into Fickian diffusion, which indicated that the diffusion of Cip from the composite fiber mats became the main factor of drug release instead of polymer degradation. Besides, with the same composition but different geometric structures, the drug release behavior is of significant difference. Therefore, all the Cip‐loaded composite fiber mats showed antibacterial activities but with different efficiency. In summary, the release of the drug could be controlled by adding PEG and changing the geometric structures according to the different requirement of wound dressings.
Investigation of Drug Release and Matrix Degradation of Electrospun Poly(<scp>dl</scp>-lactide) Fibers with Paracetanol Inoculation

Abstract:This study was aimed at assessing the potential use of electrospun fibers as drug delivery vehicles with focus on the different diameters and drug contents to control drug release and polymer fiber degradation. A drug-loaded solvent-casting polymer film was made with an average thickness of 100 mu m for comparative purposes. DSC analysis indicated that electrospun fibers had a lower T-g but higher transition enthalpy than solvent-casting polymer film due to the inner stress and high degree of alignment and orientation of polymer chains caused by the electrospinning process. Inoculation of paracetanol led to a further slight decrease in the Tg and transition enthalpy. An in vitro drug release study showed that a pronounced burst release or steady release phase was initially observed followed by a plateau or gradual release during the rest time. Fibers with a larger diameter exhibited a longer period of nearly zero order release, and higher drug encapsulation led to a more significant burst release after incubation. In vitro degradation showed that the smaller diameter and higher drug entrapment led to more significant changes of morphologies. The electrospun fiber mat showed almost no molecular weight reduction, but mass loss was observed for fibers with small and medium size, which was characterized with surface erosion and inconsistent with the ordinarily polymer degrading form. Further wetting behavior analysis showed that the high water repellent property of electrospun fibers led to much slower water penetration into the fiber mat, which may contribute to the degradation profiles of surface erosion. The specific degradation profile and adjustable drug release behaviors by variation of fiber characteristics made the electrospun nonwoven mat a potential drug delivery system rather than polymer films and particles.

Abstract:A drug delivery system with sustainable controlled drug release can improve the quality of life of a patient by reducing the side-effects and better absorption of the drug locally. However, the main disadvantageous of this delivery model is the burst release of the drug, which can result in severe health problem, such as toxicity. Here in this study, a new coaxial microfiber has been developed with encapsulated anti-inflammatory drug, ibuprofen, inside the core structure of the coaxial fibre. The core consisting of polyethylene oxide (PEO) carrying the drug was covered with the polylactic acid (PLA)/PEO and shell to prevent the burst release of the drug and provide sustainable release over a prolonged time. The release profiles showed that the burst release was reduced from 20% in control scaffold, core only, to 5% in core-shell structure after 6 hrs. The higher percentage of PLA in the shell composition provides a slower release of ibuprofen, due to the slower degradation of PLA in comparison with PEO. The result indicates the developed structure can be a potential system for the localized release of the various drug system, which leads to a more sustainable and controlled release of the drug over the more extended period and deliver a better outcome along with side-effect prevention.

Abstract:Currently drug-controlled release technology has gained a great deal of attention on the field of biomedicine. Nevertheless, most drug-controlled release systems usually require complex modification. Herein, the monolayer and multilayer structure nanofibers for drug delivery of acetylsalicylic acid (ASA), as a model drug, were fabricated using the one-step electrospinning technique for the first time, which was composed with the poly(lactic-co-glycolic acid) (PLGA) and silk fibroin (SF). Meanwhile, the effects of the mass radio of PLGA to SF on the properties of the electrospun monolayer nanofiber membrane was further investigated. The results of the electrospun monolayer PLGA-SF nanofiber membrane with mass radio of 2:1 displayed significant improvement in hydrophilicity as well as mechanical properties compared to the other mass radio of nanofibers membranes. Moreover, drug release studies revealed that nanofiber membrane with multilayer structure was able to promote a much more sustained release of ASA in comparison with monolayer PLGA-SF nanofibers membranes. More remarkable, the nanofiber membrane with multilayer structure presented good biocompatibility. Overall, these results demonstrate the potential of using PLGA-SF nanofibers with multilayer structure as a scaffold for drug-controlled release.

Abstract:Alginate is a natural polymer with good biocompatible properties and is a potential polymeric material for the sustainable development and replacement of petroleum derivatives. However, the non-spinnability of pure alginate solutions has hindered the expansion of alginate applications. With the continuous development of electrospinning technology, synthetic polymers, such as PEO and PVA, are used as co-spinning agents to increase the spinnability of alginate. Moreover, the coaxial, parallel Janus, tertiary and other diverse and novel electrospun fiber structures prepared by multi-fluid electrospinning have found a new breakthrough for the problem of poor spinning of natural polymers. Meanwhile, the diverse electrospun fiber structures effectively achieve multiple release modes of drugs. The powerful combination of alginate and electrostatic spinning is widely used in many biomedical fields, such as tissue engineering, regenerative engineering, bioscaffolds, and drug delivery, and the research fever continues to climb. This is particularly true for the controlled delivery aspect of drugs. This review provides a brief overview of alginate, introduces new advances in electrostatic spinning, and highlights the research progress of alginate-based electrospun nanofibers in achieving various controlled release modes, such as pulsed release, sustained release, biphasic release, responsive release, and targeted release.
Encapsulating Drugs in Biodegradable Ultrafine Fibers Through Co-Axial Electrospinning

Abstract:This article describes an electrospinning process to fabricate double-layered ultrafine fibers. A bioabsorbable polymer, Polycaprolactone (PCL), was used as the outer layer or the shell and two medically pure drugs, Resveratrol (RT, a kind of antioxidant) and Gentamycin Sulfate (GS, an antibiotic), were used as the inner layers or the cores. Morphology and microstructure of the ultrafine fibers were characterized by scanning electron microscope (SEM) and transmission electron microscopy (TEM), whereas mechanical performance of them was understood through tensile test. In vitro degradation rates of the nanofibrous membranes were determined by measuring their height loss when immersed in pH 7.4 phosphate-buffered saline (PBS) mixed with certain amount of Pseudomonas lipase for a maximum of 7 days. The drug release behaviors of the RT and GS were measured using a high performance liquid chromatography (HPLC) and ultraviolet-visible (UV-vis) spectroscopy, respectively. It has been found that the drug solutions without any fiber-forming additive could be encapsulated in the PCL ultrafine fibers, although they alone cannot be made into a fiber form. Beads on the fiber surface influenced the tensile behavior of the ultrafine fibers remarkably. When the core solvent was miscible with the shell solvent, higher drug concentration decreased the bead formation and thus favored the mechanical performance. The situation, however, became different if the two solvents were immiscible with each other. The degradation rate was closely related to hydrophilicity of the drugs in the cores. Higher hydrophilicity apparently led to faster degradation. The release profiles of the RT and GS exhibited a sustained release characteristic, with no burst release phenomenon. Abstract:In order to realize controlled dual release of two hydrophobic drugs with distinct rates in a vehicle, novel poly(lactic-co- glycolic acid) (PLGA) composite nanofibers encapsulated with micelles were successfully fabricated by "emulsion-electrospinning." Brefeldin A (BFA) was firstly embedded in monomethoxy-poly(ethylene glycol)-b-poly(L-lactide) (MePEG-PLLA) micelles. By means of "emulsion-electrospinning," paclitaxel (PTX) and polymeric micelles contained BFA were successfully loaded into the electrospun PLGA composite nanofibers. The in vitro release results demonstrated that the location of the drugs in the electrospun fibers determined their release profiles. BFA had a long-term and sustained release while PTX had a relatively rapid release in the dual drugs delivery system. In vitro cytotoxicity studies revealed that the composite nanofibers with two drugs restrained HepG-2 cells more efficiently. These results strongly suggested that the electrospun composite nanofibers containing polymeric micelles can be used as an effective controlled dual release of hydrophobic drugs and were suitable for postoperative chemotherapy of cancers.
Strategies For Delivering Bioactive Molecules In Electrospun Fibers

Abstract:Electrospun fibers have been proposed for many biomedical applications. However, little is known about their release kinetics as bioactive molecules delivery systems and tissue scaffolds. The specific aim of this study was to compare the fiber morphologies and release kinetics between blend nanofibers and core-shell ones. Tetracycline hydrochloride (TCH) was selected as model compounds while poly( L-lactic acid) ( PLLA) was used as polymer carriers. Two kinds of medicated fibers and fibrous threads were fabricated by blend electrospinning and coaxial electrospinning, respectively. In vitro release studies were conducted to compare the release properties between two kinds of fibers. The results showed that the drug particles are likely to locate on the fiber surface owing to the rapid evaporation of the solvent and the high ionic strength of the drugs during blend-electrospinning, which is mostly to result in a burst release of drug in the initial stage, whereas coaxial electrospinning can provide reversely smooth release of bioactive agents. Two kinds of electrospun fibers should be found some different applications in specific fields.

Abstract:In the present study an innovative tissue engineering scaffold was developed which can be used on wound dressing with a controllable drug-releasing capability. By coaxial electrospinning a coaxial nanofibrous mats were obtained which was made from Poly(L-lactid-co-ε-caprolactone) (PLLACL) and Tetracycline Hydrochloride (TCH), with P(LLA-CL) as the shell and TCH as the core. The TCH release behavior of the coaxial nanofibrous mats was measured by ultraviolet-visible (UV-vis) spectroscopy in vitro environment of 37°C for a maximum 180 hours. To compare with the method of mix electrospinning, TCH without any other material could encapsulated in P(LLA-CL) nanofibers very well and released for a long time.

Abstract:In this study, electrospun membranes were developed for controlled drug release applications. Both uniaxial Polyvinyl alcohol (PVA) and coaxial fibers with a PVA core and a poly (L-lactic acid) (PLLA) and polycaprolactone (PCL) coating were produced with different coating structures. The best conditions for the manufacture of the fibers were also studied and their morphology was analyzed as a function of the electrospinning parameters. Special attention was paid to the fiber surface morphology of the coaxial fibers, obtaining both porous and non-porous coatings. Bovine serum albumin (BSA) was used as the model protein for the drug release studies and, as expected, the uncoated fibers were determined to have the fastest release kinetics. Different release rates were obtained for the coated fibers, which makes this drug release system suitable for different applications according to the release time required.
