Abstract:Allene moieties with axial chirality are featured in many natural products with biological activities. They also serve as unique building blocks in organic synthesis and as chiral ligands/catalysts. In recent years, chemists have made great efforts in the synthesis of chiral allenes, in particular with asymmetric catalytic methods. Significant progress has been achieved, although it still remains a challenging task. In this minireview article, we summarize the recent advances in the catalytic asymmetric synthesis of allenes.

Abstract:Allenes represent a distinctive class of organic molecules characterized by their unique physical and chemical properties. Among them, chiral allenes play a significant role in modern chemistry. They not only serve as pivotal frameworks for natural products and drug molecules but also are essential building blocks in organic synthesis. Over the years, numerous remarkable and efficient methods have been developed for the synthesis of chiral allenes. Notably, the catalytic synthesis of chiral allenes using transition metals in conjunction with chiral ligands has garnered considerable attention. This review paper aims to provide an overview of recent advancements in the synthesis of chiral allenes through asymmetric 1,4‐difunctionalization of 1,3‐enynes, in which two distinct strategies including non‐radical and radical processes are highlighted.

Abstract:The axially chiral allene framework has garnered significant attention from synthetic community, due to its wide applications in natural products, organic synthesis, pharmaceuticals, and materials science. Among the structurally diverse chiral allene compounds, heteroatom-substituted axially chiral allenes represent a large and versatile subclass. While the utility involving heteroatom-substituted chiral allenes has been extensively explored, the asymmetric synthesis of these valuable chiral frameworks has posed a long-standing challenging. Benefited from the development of asymmetric catalysis, many efficient synthetic methods in enantioselective catalysis as well as a variety of competent reaction partners recently have emerged. This review primarily summarizes the contributions made to date regarding the heteroatom-substitued axially chiral allenes in their catalytic asymmetric syntheses and their synthetic applications.

Abstract:Chiral allene and N -heteroaryl motifs are present in an ever-growing list of biologically active natural products and synthetic drugs. Although significant progress has been made in asymmetric syntheses of chiral allenes, general and practical protocols for enantioselective syntheses of chiral N -heteroaryl-substituted allenes from readily available starting materials still remain rare. Here we report a highly enantioselective synthesis of quinolinyl-substituted chiral allenes through a copper-catalyzed asymmetric allenylation of quinoline N -oxides with readily available 1,3-enynes. A variety of 1,3-enynes react with quinoline N -oxides, affording the corresponding quinolinyl-substituted allenes in high yields (up to 95%) and high enantioselectivities (up to 99% ee). This transformation tolerates a variety of functional groups, such as chloro, bromo, trifluoromethyl ether, tertiary amine, siloxy, carboxylic ester, imide, pyridine, and thiophene moieties. DFT calculations suggest a pathway involving an intramolecular nucleophilic addition of an allenyl copper intermediate with a coordinated quinoline N -oxide through a five-membered, rather than seven-membered, transition state.

Abstract:CuBr and ZnI2 have been developed as catalysts or subcatalysts for the efficient asymmetric synthesis of axially chiral allenols with up to 97% ee from readily available propargylic alcohols, aliphatic or aromatic aldehyde, pyrrolidine, and commerically available ligands. The alcohol unit in the terminal alkynes plays a very important role for ensuring high enantioselectivity via coordination.

Abstract:Allenes with two carbon–carbon double bonds belong to a unique class of unsaturated hydrocarbons. The central carbon atom of allene is sp hybridized and forms two σ-bonds and two π-bonds with two terminal sp2 hybridized carbon atoms. The chemistry of allenes has been well documented over the last decades. They are more reactive than alkenes due to higher strain and exhibit significant axial chirality, thus playing a vital role in asymmetric synthesis. Over a variety of organic transformations, allenes specifically undergo classical metal catalyzed cycloaddition reactions to obtain chemo-, regio- and stereoselective cycloadducts. This review briefly describes different types of annulations including [2+2], [2+2+1], [3+2], [2+2+2], [4+2], [5+2], [6+2] cycloadditions using titanium, cobalt, rhodium, nickel, palladium, platinum, gold and phosphine catalyzed reactions along with a mechanistic study of some highlighted protocols. The synthetic applications of these reactions towards the synthesis of natural products such as aristeromycin, ent-[3]-ladderanol, waihoensene(−)-vindoline and (+)-4-epi-vindoline have also been described.

Abstract:Allenes are valuable organic molecules that feature unique physical and chemical properties. They are not only often found in natural products, but also act as versatile building blocks for the access of complex molecular targets, such as natural products, pharmaceuticals, and functional materials. Therefore, many remarkable and elegant methodologies have been established for the synthesis of allenes. Recently, more and more methods for radical synthesis of allenes have been developed, clearly emphasizing the associated great synthetic values. In this Perspective, we will discuss recent important advances in the synthesis of allenes via radical intermediates by categorizing them into different types of substrates as well as distinct catalytic systems. The mechanistic studies and synthetic challenges will be highlighted.

Abstract:The cadmium iodide‐mediated reaction between an amine and two molecules of terminal alkynes for the synthesis of allenes has been investigated by using DFT calculations. On the basis of these calculations, a plausible mechanism has been proposed. The results also reveal that pyrrolidine serves not only as a reactant but also as a shuttle in a proton relay.

Abstract:AbstractThe stereodivergent synthesis of allene compounds bearing α,β‐adjacent central chiralities has been realized via the Pd/Cu‐catalyzed dynamic kinetic asymmetric alkylation of racemic allenylic esters. The matched reactivity of bimetallic catalytic system enables the challenging reaction of racemic aryl‐substituted allenylic acetates with sterically crowded aldimine esters smoothly under mild reaction conditions. Various chiral non‐natural amino acids bearing a terminal allenyl group are easily synthesized in high yields and with excellent diastereo‐ and enantioselectivities (up to >20 : 1 dr, >99 % ee). Importantly, all four stereoisomers of the product can be readily accessed by switching the configurations of the two chiral metal catalysts. Furthermore, the easy interconversion between the uncommonη3‐butadienyl palladium intermediate featuring a weak C=C/Pd coordination bond and a stable Csp2−Pd bond is beneficial for the dynamic kinetic asymmetric transformation process (DyKAT).

Abstract:Asymmetric synthesis of chiral allenes starting from prochiral substrates under mild reaction conditions promoted by Pd-SYNPHOS catalyst is reported. This protocol provides an efficient access to various enantioenriched aryl- and alkyl- substituted allenes, which are versatile building blocks of high utility to both organic and medicinal chemists, in excellent isolated yields (up to 96%) and high enatiomeric ratio values (up to 95:5). In addition, a comparative study using several C-2-symmetric atropisomeric diphosphine ligands revealed the overwhelming impact of the steric and electronic properties of the ligands for the catalytic efficiency of this process.

Abstract:Phosphine-catalyzed cascade isomerization/annulation has been developed to realize a diastereoselective synthesis of allenes installed on the hexahydropentalene skeleton, which contains five chiral centers (and one axial chirality). This reaction tolerated a broad range of allenoates and enynes. The allene products were transformed to various halogen-substituted fused-ring compounds.

Abstract:This review is focused on the recent advances in the functionalization of allenes via radical process. Different radical partners including carbon radicals and heteroatom radicals are discussed in the reactions of allenes. Generally, the radical formed in situ would attack the allene at the central carbon leading to allyl radical intermediate. Abstract:A photoexcited palladium-catalyzed synthesis of chiral allenes from alkynes via a sequence of isomerization and deracemization is disclosed. This method offers an efficient and cost-effective approach to produce a wide range of chiral allenes with good enantioselectivities and yields (up to 98% yield and 99% ee). The use of readily available and stable alkynes as starting materials simplifies experimental procedures and broadens the scope of the chiral allene synthesis. DBU plays a crucial dual role in this reaction to effectively facilitate the isomerization of alkynes to allenes and enhance the palladium-catalyzed deracemization under visible light excitation, both of which are vital for the success of the process.

Abstract:We herein describe a synergistic chiral primary amine/achiral palladium catalyzed enantioselective terminal addition to allenes with alpha-branched,beta-ketocarbonyls and aldehydes. The reactions afford allylic adducts bearing acyclic all-carbon quaternary centers with high regio- and enantioselectivity. A wide range of allenes including those aliphatic or 1,1'-disubstituted could be employed, thus expanding the scope of typical asymmetric allylic alkylation reactions.
