Abstract:In this account, we will summarize our recent progress on development of Pt‐ or Au‐catalyzed benzannulation of 1‐indolyl‐2,3‐allenols for the efficient construction of carbazole skeleton via a metal carbene intermediate and its application towards the synthesis of naturally occurring carbazole alkaloids. The strategy enjoys the outstanding regioselectivity, atom economy, mild reaction conditions, substrate scope, functional group tolerance.

Abstract:Ru3(CO)12 has been found to be the efficient catalyst for the addition reactions of catechols with both terminal and internal alkynes to selectively afford 2,2-disubstituted 1,3-benzodioxoles in good to high yields. The formation of 2,2-substituted 1,3-benzodioxoles results from the tandem addition of two O-H bonds of catechols to alkyne's triple bond.

Abstract:Propargyl cyclic carbonates have emerged as versatile precursors in synthetic chemistry. However, their reactivity has so far been limited to transition metal-catalyzed substitution and cyclization reactions. Herein, we illustrate the successful employment of propargyl cyclic carbonates as coupling partners in Ru(II)-catalyzed C-H annulation of benzoic acids and benzamides. This approach allowed us to access a broad range of biologically relevant isocoumarin and isoquinolinone derivatives in good to excellent yields, utilizing bench-stable and easily accessible precursors. Preliminary mechanistic studies indicated that the C-H metalation step is both reversible and rate-determining in the reaction pathway. Furthermore, the utility of the developed methodology has been illustrated by scale-up and postfunctionalization experiments.

Abstract:The sequential treatment of terminal alkynes or propargylic alcohols with n-BuLi and MOMCl afforded the corresponding propargylic methyl ethers, which would react with primary alkyl Grignard reagents under the catalysis of CuBr to afford 3-substituted 1,2-allenes or 2-substituted secondary 2,3-allenols, respectively. The reaction may be applied to the synthesis of optically active 2-substituted secondary 2,3-allenols with up to >99% ee without any protection to the free hydroxyl group in the starting 4-hydroxy-2-alkynyl methyl ethers.

Abstract:The cadmium(II) chloride‐catalyzed dehydrative C−P cross‐coupling reaction of propargyl alcohols with diarylphosphine oxides is reported. Several propargyl alcohols including those bearing the sterically demanding tert‐butyl group at the triple bond terminus can be used as good substrates in the reaction to produce the corresponding allenylphosphine oxides in good to high yields in acetonitrile at 100 °C. The reaction can also be easily scaled up to a gram‐scale synthesis. A mechanism study indicates that the reaction may proceed through a process of propargylic substitution to generate phosphonite intermediates followed by [2,3] sigmatropic rearrangement to produce the allenyl products, rather than through a common allenylative substitution resulting from P‐nucleophilicity.

Abstract:A novel methodology for the synthesis of trisubstituted allenes is reported. Lithiation of 1-aryl-3-alkylpropadienes and subsequent transmetalation with zinc bromide followed by Pd-catalyzed Negishi coupling reactions with halides afforded the corresponding trisubstituted allenes in a highly regioselective fashion with moderate to excellent yields. A plausible regioselective lithiation mechanism was proposed on the basis of deuterium labeling experiments.

Abstract:An unprecedented Ru(II)-catalyzed deoxygenative, site-selective formal [3 + 1 + 2] benzannulation reaction for the efficient synthesis of highly substituted benzene molecules is reported. This reaction between allyl alcohols and acetylenedicarboxylate esters proceeds via cascade C-H activation, consecutive double migratory insertion with alkynes, and cycloaromatization followed by an unusual specific C-C triple bond cleavage.

Abstract:The first examples of cross-metathesis between two different allenes is disclosed. First- and second-generation Ru complexes were found to be ineffective, at most affording only oligomeric products. The exception was a first-generation complex bearing a bidentate phenyl isopropoxy ligand (i.e., PCy3 is not released upon initiation), reactions with which afforded a 1,3-disubstituted allenyl boronate in 22% yield. On the basis of mechanistic studies designed to gain deeper understanding of the reasons for the ineffectiveness of different Ru catalysts, it was discovered that phosphine-free Ru-CAAC complexes have the steric and electronic attributes to be highly effective. The results of these investigations pave the way for development of additional olefin metathesis reactions that generate allenes.

Abstract:An efficient one-pot synthesis of trisubstituted allenes from readily available terminal alkynes and ketones is realized. A wide range of trisubstituted allenes may be synthesized efficiently via this method. Preliminary mechanistic studies revealed that CuI and Ti(OEt)4 are in charge of the formation of propargylic amine, while ZnBr2 is responsible for the transformation from propargylic amine to allene.

Abstract:The Doyle-Kirmse reaction, namely the [2,3]-sigmatropic rearrangement of sulfonium ylides generated from α-diazo carbonyl compounds and sulfides has been for the first time catalyzed by [RuCl2-(p-cymene)]2. The reaction with allyl sulfides afforded the corresponding [2,3]-sigmatropic rearrangement products in moderate to good yields. The reaction with propargyl sulfide gave a mixture of allenic sulfide and furan. The former is the [2,3]-sigmatropic rearrangement product, while the latter is derived from the further rearrangement of the allene product under Ru(II) catalysis.

Abstract:An allene to allene protocol for the synthesis of beta-allenyl butenolides in moderate to high yields from 2,3-allenoic acids and propargylic carbonates catalyzed by Pd(OAc)(2)-TFP has been developed; the products were applied successfully to the Diels-Alder reaction with electron-deficient alkynes to afford polysubstituted benzene derivatives with an excellent regioselectivity.

Abstract:The Ru-catalyzed meta-C-H benzylation of arenes bearing pyridyl, pyrimidyl, and pyrazolyl directing groups with toluene derivatives has been reported. Heptafluoroisopropyl iodide (iC3F7I) was employed as both a mild oxidant and radical initiator. This reaction showed excellent site-selectivity and tolerated a wide range of functional groups, providing a new strategy for the synthesis of various diarylmethane moieties.

Abstract:A copper-catalyzed efficient regioselective silylation reaction of chloro-substituted allenyl-Bdan was developed. Under mild reaction conditions, allenyl and propargyl silane compounds can be selectively obtained in moderate to high yields by adjusting the bases and solvents used in the reactions. This study offers direct and efficient methods for synthesizing multifunctionalized allenyl and propargyl silane compounds from the same initial material of chloro-substituted allenyl-Bdan.

Abstract:We have established an efficient ruthenium(ii)-catalyzed cis carbohalogenation and carbooxygenation cyclization of internal alkyne-tethered cyclohexadienones with halide and acetate ions, which produces a series of alkenyl halides and acetates with hydrobenzofuran skeletons in high yields and features high atom economy and regio- and stereoselectivity. Furthermore, the obtained hydrobenzofuran products can easily undergo aromatization and subsequent cross-coupling reactions, which provide a novel and stereoselective approach to synthesize tetrasubstituted alkenes, particularly diarylalkenes and triarylalkenes.
