Abstract:Sly silyl caught in the act: Protonation of a mesitylene ring by the strongly acidic arenium carborane [CH(3)C(6)H(6)]- [CHB(11)Me(5)Br(6)] initiates a cascade reaction that results in a stable beta-silyl allyl cation (see picture, H yellow, C blue, silyl allyl group red). Remarkably, the driving force in the reaction suffices to disrupt a stable aromatic ring in favor of a cationic reactive intermediate.

Abstract:Geminal bis(silyl) enal 2a is shown to be a useful scaffold for anion relay chemistry (ARC) aimed at the stereoselective synthesis of C(3),O(1)-disilylated allylic alcohols. The ARC reaction is initiated by the addition of an alkyllithium to the aldehyde and features a CuCN-promoted C(sp2)-to-O 1,4-silyl migration to generate a vinylcuprate that reacts with activated electrophiles.

Abstract:An efficient approach for the reversal of regioselectivity in the nucleophilic introduction of difluorinated carbanion into α,β-enones has been developed via a silylium catalysis. The strong electron-withdrawing properties and bulky substituents of in situ-generated silyl triflic imide catalyst is the key for the 1,4-addition reaction to proceed smoothly. The synthetic utility is highlighted by the further use of this method for the synthesis of 2,4,6-triarylsubstituted 3-fluoropyridines in a one-pot manner.

Abstract:Here, we report a cobalt-catalyzed sequential dehydrogenative Heck silylation/hydroamination of styrenes with hydrosilane and diazo compound to access 1-amino-2-silyl compounds with excellent regioselectivity. This difunctionalization reaction could undergo smoothly using 1 mol% catalyst loading with good functional group tolerance. Not only di- and tri-substituted hydrosilanes, but also alkoxysilane are suitable, which does explore the scope of the family of 1-amino-2-silyl compounds. The ligand relay phenomenon between neutral tridentate NNN ligand and anionic NNN ligand is observed for the first time via absorption spectra analysis in this one-pot, two-step transformations. The primary mechanism has been proposed based on the control experiments.

Abstract:Using terminal alkyne as a nontraditional one-carbon (C1) unit and silylborane as an external silicon pronucleophile, a relay palladium/copper-catalyzed silylative [5 + 1] benzannulation of 3-acetoxy-1,4-enynes for producing polysubstituted arylsilanes, especially including bioactive motif-based analogues, in a single reaction step through benzene ring skeleton assembly and silyl intermolecular incorporation cascades is developed. Abstract:Bissilyl enal, prepared by a Mannich reaction of 3,3-bissilyl aldehyde with formaldehyde, has proven to be a useful linchpin in an efficient three-component coupling process. The reaction features a [1,4]-Brook rearrangement to generate the silylallyl anion, which adopts a predominant endo-orientation and can undergo addition to electrophiles in a regio- and stereoselective manner, giving various E-vinylsilane species in good yields.

Abstract:A catalytic enantioselective addition reaction of silyl enol ethers to ketomalonate in the presence of C-2-symmetric copper(II) bisoxazolines as catalysts is presented; the reaction has been studied for different ligands and substrates under various reaction conditions and proceeds in good yield and with ee's > 90% and a procedure for the preparation of an optically active hydroxy acid is shown.

Abstract:The novel efficient procedure has been developed for the conjugate addition of amines to electron deficient alkenes. A novel multi-SO3H functioned ionic liquid has been synthesized for the conjugate addition. The results showed that the catalyst was very efficient for the reactions with the excellent yields in several minutes. Operational simplicity, without need of any solvent, low cost of the catalyst used, high yields, reusability, excellent chemoselectivity, applicability to large-scale reactions are the key features of this methodology.

Abstract:To date, the iron-catalyzed construction of C-heteroatom bonds has been less developed due to the difficulty of transmetalation with heteroatom anions and the sluggish reductive elimination. Herein we report an iron-catalyzed method for the silylation of (hetero)aromatic chlorides. It features high efficiency, a broad substrate scope, and excellent functional group compatibility. Moreover, this protocol enables the late-stage silylation of some pharmaceuticals, thus providing an excellent method to access valuable intermediates in medicinal chemistry.

Abstract:Although dirhodium-catalyzed multicomponent reactions of diazo compounds, nucleophiles and electrophiles have achieved great advance in organic synthesis, the introduction of allylic moiety as the third component via allylic metal intermediate remains a formidable challenge in this area. Herein, an attractive three-component reaction of readily accessible amines, diazo compounds, and allylic compounds enabled by a novel dirhodium(II)/Xantphos catalysis is disclosed, affording various architecturally complex and functionally diverse α-quaternary α-amino acid derivatives in good yields with high atom and step economy. Abstract:A highly selective cationic rearrangement of β-silylated cyclopropanes bearing an allyl group at the silicon atom is reported. The reaction cascade is initiated by trityl-cation-mediated deallylation to preferentially yield one of two diastereomeric cyclopropane-stabilized silylium ions. This is the starting point for a concerted [1,2]-hydride shift/ring expansion driven by the release of ring strain of the cyclopropane ring. The resulting tertiary carbenium ion is then captured by an intermolecular Si-to-C allyl transfer from another precursor molecule. By this, a quaternary carbon atom is formed. The computed reaction mechanism is fully consistent with the experimental findings.

Abstract:Abstract Facile formation of carbon-heteroatom bonds is a long-standing objective in synthetic organic chemistry. However, direct cross-coupling with readily accessible alkenyl acetates via inert C‒O bond-cleavage for the carbon-heteroatom bond construction remains challenging. Here we report a practical preparation of stereoselective tri- and tetrasubstituted alkenyl silanes and stannanes by performing cobalt-catalyzed C‒O silylation and stannylation of alkenyl acetates using silylzinc pivalate and stannylzinc chloride as the nucleophiles. This protocol features a complete control of chemoselectivity, stereoselectivity, as well as excellent functional group compatibility. The resulting alkenyl silanes and stannanes show high reactivities in arylation and alkenylation by Hiyama and Stille reactions. The synthetic utility is further illustrated by the facile late-stage modifications of natural products and drug-like molecules. Mechanistic studies suggest that the reaction might involve a chelation-assisted oxidative insertion of cobalt species to C‒O bond. We anticipate that our findings should prove instrumental for potential applications of this technology to organic syntheses and drug discoveries in medicinal chemistry.

Abstract:Multicomponent reactions that involve interception of onium ylides through Aldol, Mannich, and Michael addition with corresponding bench-stable acceptors have demonstrated broad applications in synthetic chemistry. However, because of the high reactivity and transient survival of these in situ generated intermediates, the substitution-type interception process, especially the asymmetric catalytic version, remains hitherto unknown. Herein, a three-component asymmetric allylation of α-diazo carbonyl compounds with alcohols and allyl carbonates is disclosed by employing a ternary cooperative catalysis of achiral Pd-complex, Rh2(OAc)4, and chiral phosphoric acid CPA. This method represents the first example of three-component asymmetric allylic alkylation through an SN1-type trapping process, which involves a convergent assembly of two active intermediates, Pd-allyl species, and enol derived from onium ylides, providing an expeditious access to chiral α,α-disubstituted ketones in good to high yields with high to excellent enantioselectivity. Combined experimental and computational studies have shed light on the mechanism of this novel three-component reaction, including the critical role of Xantphos ligand and the origin of enantioselectivity.

Abstract:A nonracemic 3-silyl-3-borylhex-4-enoate reagent has been developed. Its asymmetric crotylboration of aldehydes provides Z-anti-homoallylic alcohols possessing a trisubstituted vinylsilane in high yields with excellent stereo- and enantioselectivity. Diverse decoration of vinylsilane and ester groups, as well as formation of functionalized THF rings, showcase the potential of the approach in the synthesis of polyketide natural products.

Abstract:A general synthesis of chiral 1,3‐disubstituted allylsilanes is established through copper(I)‐catalyzed asymmetric 1,4‐conjugate silylation of α,β‐unsaturated sulfones and subsequent Julia‐Kocienski olefination. By modification of McQuade's NHC ligand, the catalytic asymmetric conjugate silylation with a broad substrate scope is achieved in high enantioselectivity. The following Julia–Kocienski olefination proceeds smoothly at room temperature to deliver an array of chiral allylsilanes in moderate yields. More interestingly, a one‐pot asymmetric synthesis with high synthetic efficiency is successfully realized. Utility of the prepared chiral 1,3‐disubsituted allylsilanes is demonstrated in the asymmetric allylation of both aldehyde and aldimine. Finally, an interesting "match and mismatch" phenomenon is observed in the asymmetric allylation of chiral aldehydes.This article is protected by copyright. All rights reserved.
