Relationship of the Lymphotoxin A Gene Polymorphism with Chronic Obstructive Pulmonary Disease

Abstract:Objective It has been proved that lymphotoxin A(LTA) plays key roles in the regulation of B and T cell activities,which is important for the pathogenesis of chronic obstructive pulmonary disease(COPD).The aim of present study was to explore the relationship between LTA gene polymorphism and the genetic susceptibility and clinical severity of COPD in Han populations living in northern China.Methods Six tSNPs sites(rs2844482,rs2071590,rs2239704,rs909253,rs2229094 and rs1041981) of LTA gene were selected for genotyping in 122 COPD patients and 163 healthy subjects(control),the age and sex of whom were well matched with COPD patients,by PCR-sequencing analysis.Chi-square test was used to perform the Hardy-Weinberg test,and unconditional logistic regression was used to calculate the odds ratio(OR) and 95% confidence interval(CI).Results The distribution of genotypes of 6 tSNPs sites in COPD patients showed no difference to that in healthy controls.While advanced analysis performed in the COPD patients revealed that the genotype distributions of rs2071590,rs2239704,rs909253 and rs1041981 in mild COPD patients were significantly different to that in patients with intermediate and serious COPD(P0.05).The rs2071590 was associated with severity of COPD in recessivation(OR: 2.270;95% CI: 1.133-5.022).The 2 SNPs(rs909253 and rs1041981) were associated with severity of COPD in dominance(OR: 0.370 and 0.364;95% CI: 0.116-0.923 and 0.111-0.965).The rs2239704 SNP was associated with severity of COPD in co-dominant model(OR: 2.817 and 2.199;95% CI: 1.137-9.744 and 1.107-4.588).Conclusion It is revealed by present study that LTA is not associated with the incidence of COPD,but significantly related with the progression of COPD course.
