Abstract:Objective To investigate the association between IL-4-590C/T gene polymorphism and the occurrence of chronic periodontitis.Methods The genotypes of IL-4 of 104 patients with chronic periodontitis and 106 healthy controls were analyzed by polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP) in a case-control study.Distributions of single alleles and genotypes were analyzed by the chi-square test.Results No significant difference in the allele and genotype frequencies of IL-4-590 between patients group and control group was found.ConclusionThe results suggest that the IL-4-590 gene polymorphisms are not associated with the susceptibility of Chinese Han population chronic periodontitis.

J Clin Periodontol 2009; 36: 198-203. doi: 10.1111/j.1600-051X.2008.01366.x.Several common single nucleotide polymorphisms (SNPs) of the cyclooxygenase-2 (COX-2) gene have been reported to be functional. The association between -1195GA, -765GC and 8473TC of COX-2, and severe chronic periodontitis (CP) in a Chinese population was investigated.148 cases of healthy controls (control group) and 146 cases of severe CP were recruited in this study. Genotypes of -1195GA, -765GC and 8473TC were determined by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). The distributions of genotypes and haplotypes were compared by chi(2) test and the odds ratios (ORs) were calculated by logistic regression analysis.The prevalence of the -1195A was more prevalent in CP group (60.62%) than control group (51.35%), and the distributions of the -765C and 8473C were higher in control group (6.76% and 21.96%) compared with CP group (3.08% and 15.07%). Only genotype distribution of -1195GA was significant when p-value was corrected for multiple testing (p(c)=0.033). The adjusted ORs for the -1195AA/GA, -765GC and 8473CC/TC were 2.49 (95% CI=1.33-4.69, p=0.005), 0.45 (95% CI=0.20-1.04, p=0.061) and 0.67 (95% CI=0.41-1.11, p=0.118). Subjects with the haplotype AGT had a significantly higher risk of periodontitis than those with the most common haplotype GGT (OR=1.91, 95% CI=1.32-2.76, p(c)< 0.001).It suggests the -1195A variant is associated with an increased risk for severe CP.
TLR4 Polymorphisms May Increase Susceptibility to Periodontitis in Pg-positive Individuals

Abstract:Objective To investigate the correlation between the single nucleotide polymorphisms (SNPs) in the toll-like receptor 4 (TLR4) gene and the susceptibility to chronic periodontitis. Design 241 Chinese subjects from the cohort of Beijing Shijingshan Community were recruited. Buccal swab samples, the whole unstimulated saliva and periodontal clinical parameters were collected. Human DNA extracted from buccal swab samples were used for genotyping eight SNPs of the TLR4 gene (rs11536889, rs1927906, rs1927911, rs2149356, rs4986790, rs4986791, rs2737190, rs787384) by the Sequenom MassARRAY system. Porphyromonas gingivalis (P. gingivalis) was detected from the deposition of the whole unstimulated saliva through polymerase chain reaction (PCR) method based on 16S rRNA. The correlation between SNPs of TLR4 and chronic periodontitis susceptibility in the whole subjects and the patients detected with P. gingivalis was investigated. Results The variants of rs4986790 and rs4986791 were not found in 241 Chinese subjects. Moreover, there was no significant difference in the distribution of theother6 SNPs of TLR4 between groups of none/mild -periodontitis and moderate/severe-periodontitis subjects. When combined with P. gingivalis infection, rs1927911 (TT/CC+CT), rs2149356 (TT/GG+GT) and rs2737190 (GG/AA+AG) were independent risk factors of chronic periodontitis. Conclusion Three SNPs of TLR4, i.e., rs1927911 (TT/CC+CT), rs2149356 (TT/GG+GT) and rs2737190 (GG/AA+AG), were associated with moderate/severe chronic periodontitis in Chinese population infected with P. gingivalis. P. gingivalis, which interacted with TLR4 gene plays an important role in the pathogenesis of periodontitis.
Functional variations of the TLR4 gene in association with chronic obstructive pulmonary disease and pulmonary tuberculosis

Abstract:Abstract Objective Chronic obstructive pulmonary disease (COPD) and pulmonary tuberculosis (PTB) share a number of common risk factors, including innate immunity-related genetic factors. In the present study, we compared the role of genetic variations of the TLR4 gene in susceptibility to COPD and PTB and illuminated the underlying molecular mechanism of functional single-nucleotide polymorphisms (SNPs). Methods A population-based case control study was performed in a Chinese Han population and included 152 COPD cases, 1601 PTB cases and 1727 controls. Five SNPs in the TLR4 gene (rs10759932, rs2737190, rs7873784, rs11536889, and rs10983755) were genotyped using TaqMan allelic discrimination technology. We estimated the effects of SNPs using the odds ratio (OR) together with 95% confidence interval (CI). Dual-luciferase reporter vectors expressing different genotypes of SNPs were constructed and transfected into the human HEK 293 T cell line to explore their effects on potential transcription activity. Results After Bonferroni correction, the genetic polymorphisms of all five SNPs remained significantly associated with COPD, while rs10759932 and rs2737190 were also associated with PTB. Compared with rs10759932-TT, individuals carrying TC (OR: 0.42, 95% CI: 0.28–0.64) or CC (OR: 0.24, 95% CI: 0.09–0.63) had a significantly reduced risk of COPD. However, individuals carrying TC (OR: 1.28, 95% CI: 1.11–1.49) or CC (OR: 1.26, 95% CI: 0.98–1.62) had an increased risk of PTB. The OR (95% CI) for allele rs10759932-C was 0.45 (0.32–0.62) for COPD and 1.18 (1.07–1.32) for PTB. For rs2737190, heterozygous AG was related to a decreased risk of COPD (OR: 0.32, 95% CI: 0.21–0.49) and an increased risk of PTB (OR: 1.30, 95% CI: 1.11–1.52). The dual-luciferase reporter assay showed decreased transcription activity caused by rs10759932-C and rs2737190-G. Conclusion Genetic polymorphisms of rs10759932 and rs2737190 in TLR4 are significantly related to both COPD and PTB but with inverse effects. The altered transcription activity caused by mutations in these two loci may partly explain the observed relationship.

Abstract:Background: Allergic rhinitis (AR) is a nasal inflammatory disease resulting from a complex interplay between genetic and environmental factors. The association between Toll-like receptor (TLR) signaling pathway and environmental factors in AR pathogenesis remains to be explored. This study aims to assess the genetic association of AR with single nucleotide polymorphisms (SNPs) in TLR signaling pathway, and investigate the roles of gene-gene and gene-environment interactions in AR. Methods: A total of 452 AR patients and 495 healthy controls from eastern China were enrolled in this hospital-based case-control study. We evaluated putatively functional genetic polymorphisms in TLR2, TLR4 and CD14 genes for their association with susceptibility to AR and related clinical phenotypes. Interactions between environmental factors (such as traffic pollution, residence, pet keeping) and polymorphisms with AR were examined using logistic regression. Models were stratified by genotype and interaction terms, and tested for the significance of gene-gene and gene-environment interactions. Results: In the single-locus analysis, two SNPs in CD14, rs2563298 (A/C) and rs2569191 (C/T) were associated with a significantly decreased risk of AR. Compared with the GG genotype, the GT and GT/TT genotypes of TLR2 rs7656411 (G/T) were associated with a significantly increased risk of AR. Gene-gene interactions (eg, TLR2 rs7656411, TLR4 rs1927914, and CD14 rs2563298) was associated with AR. Gene-environment interactions (eg, TLR4 or CD14 polymorphisms and certain environmental exposures) were found in AR cases, but they were not significant after Bonferroni correction. Conclusion: The genetic polymorphisms of TLR2 and CD14 and gene-gene interactions in TLR signaling pathway were associated with susceptibility to AR in this Han Chinese population. However, the present results were limited to support the association between gene-environment interactions and AR.
Polymorphism -509C/T in TGFB1 Promoter Is Associated With Increased Risk and Severity of Persistent Allergic Rhinitis in a Chinese Population.

Abstract:Background Polymorphism -509C/T in the promoter of transforming growth factor beta1 (TGFB1) gene is implicated in the pathogenesis of asthma. This polymorphism might also act to regulate the development of allergic rhinitis (AR). Objectives To investigate whether -509C/T is associated with AR susceptibility and severity in a Han Chinese population. Methods The study enrolled 263 patients with persistent AR and 249 healthy controls. AR patients were classified as mild or moderate/severe AR groups according to the Allergic Rhinitis and its Impact on Asthma classification. TGFB1 gene polymorphism -509C/T was genotyped with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Serum total Immunoglobulin E (IgE) and specific IgE levels were determined using an ImmunoCAP. Results Significant difference was found in the allele frequency of TGFB1 -509C/T between AR patients and healthy controls (P = .027) but not in the genotype frequency (P =.051). However, the genotype frequency of TGFB1 -509C/T showed significant difference between the mild AR group, the moderate/severe AR group, and the control group (P = .012); between the moderate/severe AR group and the control group (P =.036); between the mild AR group and the moderate/severe AR group (P = .038); but not between the mild AR group and the control group (P =.075). Conclusion TGFB1 promoter polymorphism -509C/T may be associated with the susceptibility and the severity of persistent AR of Han Chinese, but the functional relationship still needs clarification.
SMAD4 Rs10502913 is Significantly Associated with Chronic Obstructive Pulmonary Disease in a Chinese Han Population: A Case-Control Study

Abstract:Background: COPD is a respiratory disease caused by a combination of genetic and environmental factors. Polymorphism, as a genetic factor, can affect the susceptibility of the disease of COPD. In this study, we assessed the relationship between the polymorphisms of three genes and COPD risk in a Chinese Han population.Patients and Methods: A total of 376 patients diagnosed with COPD and 284 control subjects were enrolled in this study. Multivariate logistic regression analysis was used to analyze the association between three polymorphisms (SMAD4 rs10502913, IL-4 rs2070874, HSPA1L rs2227956) and COPD susceptibility.Results: The SMAD4 rs10502913 GG and AG genotype significantly increased COPD risk (adjusted OR = 2.235, 95% CI 1.198-4.104; adjusted OR = 2.218, 95% CI 1.204-4.151, respectively) compared with the AA genotype. In the stratification analyses, the GG genotype significantly increased the risk of COPD in subjects aged 60 and over (adjusted OR = 2.519, 95% CI 1.266-5.015) and with a smoking history of less than 30 years (p=0.009; adjusted OR = 3.751; 95% CI 1.398-10.062). This increased risk was more pronounced in the group of GOLD I and GOLD II (adjusted OR = 3.628, 95% CI 1.022-12.885; adjusted OR = 2.394, 95% CI 1.004-5.710, respectively). In addition, AG genotype was associated with an increased COPD risk in subjects aged 60 and over (adjusted OR = 2.599, 95% CI 1.304-5.176) and in smokers (p=0.021; adjusted OR = 2.269; 95% CI 1.132-4.548). This increased risk was more obvious in the group of GOLD III COPD (p=0.047; adjusted OR = 2.532; 95% CI 1.012-6.336). Conclusion: Our present study indicated that the genotype GG and AG of SMAD4 rs10502913 are associated with an increased risk of COPD in a Chinese Han population. Further validation studies with large-scale populations are needed to confirm our findings.
Association of Gene Polymorphisms of Tumour Necrosis Factor-Alpha and Interleukin-13 with Chronic Obstructive Pulmonary Disease in Han Nationality in Beijing.

Abstract:BACKGROUND:Genetic factors are believed to play a role in the individual susceptibility to chronic obstructive pulmonary disease (COPD). A single nucleotide polymorphism (SNP) of tumour necrosis factor-alpha (TNF-alpha) has been reported but inconsistent results may arise from different populations and phenotypes of COPD. There are only a few published studies of interleukin-13 (IL-13) SNPs on COPD. The SNPs of TNF-alpha and IL-13 have not been studied in the Chinese population. This research was conducted to study the frequencies of IL-13 gene promoter 1055 (IL-13-1055) and TNF-alpha gene-308 polymorphisms in the patients with COPD and to investigate the effect of those genetic polymorphisms on COPD in the Chinese population.METHODS:A cohort of COPD patients and age matched controls were recruited from an inpatient hospital service in Beijing. Venous blood was obtained and genomic DNA was extracted from peripheral blood monocytes using standard method. Genomic DNA was used as a template for amplification by polymerase chain reaction (PCR) to determine the polymorphism at -1055 in the IL-13 gene promoter region. PCR restriction fragment length polymorphism (RFLP) was used to determine polymorphisms in the TNF-alpha gene-308 position. The products were investigated by sequence analysis also.RESULTS:One hundred and eleven COPD patients and 97 controls were studied. Seventy-five cases were current smokers in COPD patients and 36 were current smokers in controls. The frequencies of TT genotype in the IL-13 gene promoter region were 11.7% (13/111) in the COPD group and 13.4% (13/97) in the controls (P = 0.713). However, the OR value of TT genotype was significantly increased to 6.4 (95% CI 1.62 - 25.39) in the smokers with COPD. TT genotype was also positively related to family history of COPD, OR = 7.7 (95% CI 1.37 - 43.80). The frequencies of A allele in the TNF-alpha gene were 5.9% in COPD and 3.1% in controls (P = 0.131). The OR value of A allele was 5.0 (95% CI 1.011 to 25.059) in smokers with COPD.CONCLUSIONS:There is no significant difference in the frequencies of the TT genotype of IL-13-1055 or the A allele of the TNF-alpha between Han Chinese patients with COPD versus control. Thus, it does not appear that these SNPs are independent factors in COPD for Han nationality in Beijing. However, these SNPs may increase the risk of COPD among smokers.

Abstract:BACKGROUND: Toll-like receptor 4 (TLR4) is considered to be involved in the pathogenesis and progression of atopic dermatitis (AD). In the present study, we evaluated the relationship between TLR4 gene polymorphisms and the susceptibility or severity of AD among Chinese Han children.METHODS: A total of 132 AD patients and 100 healthy controls were enrolled in this study. Four single-nucleotide polymorphisms (rs19277914, rs11536891, rs7869402, and rs11536889) of the TLR4 gene were genotyped by multiplex PCR combined with next-generation sequencing.RESULTS: Our results showed that a significantly reduced risk for AD was associated with C allele [p = 0.008; odds ratio (OR) = 0.41, C vs. T], TC genotype (p = 0.022; OR = 0.41, TC vs. TT), and TC + CC genotype (p = 0.010; OR = 0.39, TC + CC vs. TT) of TLR4 rs11536891. The frequency of the haplotype GCCG (rs1927914-rs11536891-rs7869402-rs11536889) in AD patients was lower than that in the controls (p = 0.010; OR = 0.38). Moreover, the results indicated that a higher risk of severe AD was related to the T allele (p = 0.019; OR = 2.97, T vs. C) and the TC genotype (p = 0.021; OR = 3.34, TC vs. CC) of TLR4 rs7869402. A risk haplotype of TLR4 (GTTG) was found in severe AD patients (p = 0.010; OR = 5.26).CONCLUSIONS: Our data suggested that TLR4 rs11536891 polymorphism was associated with the susceptibility to AD in Chinese Han children. And TLR4 rs7869402 might confer the severity of pediatric AD patients.

Abstract:Objective To investigate the distribution characteristics of the single nucleotide polymorphisms(SNPs) of the human Toll-like receptor 4(TLR4) gene and CD14 gene in Chinese Han children in Wenzhou,and their relation to atopic diseases.Methods A total of 113 cases diagnosed as atopic disease and 69 healthy children were enrolled.The related region of TLR4 gene containing Asp299Gly and Thr399Ile polymorphisms and CD14 gene were sequenced to identify and characterize the SNPs.The frequency of genotypes and alleles between two groups were compared.Results We did not find Asp299Gly and Thr399Ile polymorphisms in the toll-like receptor 4 gene in both groups;CD14/-159 SNP was present in Han population of Wenzhou.The frequency of each genotype was 57.0%(TT)、28.0%(TC)、15.0%(CC) in normal children.and 46.9%(TT)、35.4%(TC)、17.7%(CC) in atopic children.No significant difference was found in the distribution of CD14/-159 polymorphism between atopic children and healthy controls(χ2=1.918,P0.05) according to Hardy-Weinberg principle statistics.Conclusion Asp299Gly and Thr399Ile polymorphisms in the toll-like receptor 4 gene are not common in Han children in Wenzhou.CD14/-159 SNP is present in Han children in Wenzhou,and other SNP in CD14 gene is not found.TT genotype is the primary genotype in CD14/-159 SNP in Han children in Wenzhou.No relationship between CD14/-159 SNP and atopic disease is found.

Ctla4 Polymorphisms Are Associated With Severe Pulmonary Tuberculosis In A Chinese Han Population

Abstract:The cytotoxic T lymphocyte antigen‐4 (CTLA4) is a key negative regulator in T lymphocyte activation. Polymorphisms on CTLA4 are associated with susceptibility to many autoimmune diseases, resulting in down‐regulation of the inhibition of cellular immune response. The present study aimed to investigate potential associations of CTLA4 +49A>;G (rs231775), +6230G>;A (rs3087243), and 11430G>;A (rs11571319) polymorphisms with pulmonary tuberculosis (PTB) in a Chinese Han population. A case‐control study was performed, and the three SNPs were genotyped by PCR and DNA sequencing methods. We found the frequency of CTLA4 +49AG genotype in 274 PTB cases was significantly lower than that of the 265 controls ( P cor =0.025, 0R 0.613, 95% CI 0.401–0.937). However, no associations were found between the other 2 SNPs ( P >;0.05). After divided PTB cases into two groups: with or without cavities, we found a novel relation of +6230AG genotype to cavities ( P cor =0.046, 0R 2.677, 95% CI 1.029–6.965). Lately haplotype analysis showed that haplotype AGG was significantly associated with healthy controls (P=0.002, OR 0.286, 95% CI 0.124–0.657), while haplotype AAG was related to PTB with cavities ( P =0.047, OR 2.053, 95% CI 1.000–4.215). Our results suggest the CTLA4 polymorphisms can be associated with PTB in a Chinese Han population, and +49AG may reduce the risk of being PTB, while +6230AG may contribute to cavities formation. Abstract:PURPOSE To investigate the association of the single nucleotide polymorphism (SNP) in chromosome 9p21.3 and chronic periodontitis in the Han Chinese population. METHODS rs2891168,rs1333048 and rs1333042 genotypes of randomly grouped 172 healthy control subjects and 221 patients with chronic periodontitis(CP) were genotyped by TaqMan probe real-time fluorescent quantitative PCR. Statistical analysis of Chi-square test was performed using SPSS16.0 software package. RESULTS Rs2891168 was closely associated with chronic periodontitis(P=0.003), and the frequency of G allele was higher in the patients compared with the control(48.36% vs 40.00%, OR=1.209, 95% CI: 1.001-1.461, P=0.045). Rs1333048 and rs1333042 were not associated with chronic periodontitis. CONCLUSIONS There is SNP correlated with chronic periodontitis in chromosome 9p21.3, which indicates that the 9p21.3 locus may be the susceptible region. However, the mechanism remains unclear.

Abstract:Previous studies indicated that single-nucleotide polymorphisms (SNPs) of genes coding for toll-like receptors (TLRs) and toll-interacting protein (TOLLIP) may be involved in the pathogenesis of pulmonary tuberculosis (PTB). This study was designed to investigate potential associations between the polymorphisms in TLR2, TLR4, and TOLLIP and susceptibility to latent tuberculosis infection (LTBI) or subsequent PTB in a Chinese Han population. A total of 209 PTB and 201 LTBI patients and 204 healthy control subjects (HCS) were enrolled in our study. We performed a logistic regression including sex and age as covariates to test the effect of genotype. Genotyping was conducted using the improved multiplex ligase detection reaction (iMLDR). Eleven markers in TLR2, TLR4, and TOLLIP were assessed. No significant association between polymorphisms of TLR2 and TLR4 with PTB or LTBI was detected. For TOLLIP, rs5743899 (p=0.005, OR=1.83, 95% CI: 1.20-2.80) CC genotype were risk factors for PTB progression. Moreover, rs5743867 under addictive (p=0.005, OR=1.54, 95% CI: 1.14-2.07) and recessive model (p=0.004, OR=1.86, 95% CI: 1.22-2.83) were also risk factors for PTB susceptibility. Our results indicate that polymorphisms in TOLLIP affected the risk of PTB disease.
Association of Tumor Necrosis Factor Gene Promoter Polymorphism with Chronic Obstructive Pulmonary Disease

Abstract:Objective:To study whether gene polymorphism of tumor necrosis factor promoter is associated with chronic obstructive pulmonary disease (COPD). Methods:Polymerase chain reaction-based sequence analysis was applied to determine the frequencies of genotypes and allele of TNF-308 and TNF-238 in 66 COPD patients, 42 smoker control and 43 population-based healthy controls. Results:The frequencies of three genotypes (GG,GA and AA) of TNF-308 and TNF-238 gene were not significantly different between COPD patients, smoker control subjects and healthy controls. (P0.05). There were also no significant differences in the distribution of the TNF-α3082 and TNF-α2382 alleles between COPD patients, smoker control, and healthy controls, respectively (3.4% vs.6.1% and 5.1% vs.2.4%) (P0.05). Within the COPD group, the frequency of TNF-308 and TNF-238 gene allele was similar in the severe and non-severe subgroups(2.2% vs. 4.2% and 4.3% vs. 5.6%) ( P0.05) and was present only in the heterozygous state (TNF1/2); However, the blood concentration of the TNF-α in the COPD patients was significantly increased compared to smoker control and population healthy controls.( P0.01). Conclusion:Although there is no association of TNF-α3081/2 and TNF-α2381/2 alleles with susceptibility to smoking-related COPD and severity of COPD, the elevation of the TNF-αin COPD patients indicated that TNF-α might have a causal relationship with COPD and polymorphism in others gene regions of TNF-α may exist and need to be further demonstrated.

Abstract:BackgroundChronic obstructive pulmonary disease (COPD) has been recognized as a heterogeneous disease, which is caused by biological heterogeneity. The purpose of our study is to determine the association between the single nucleotide polymorphisms (SNPs) of miRNAs (miR-8079 and miR-5007) and the susceptibility to COPD in Chinese Han population.MethodsWe conducted a 'case-control' study involving 315 COPD patients and 314 healthy individuals. Three SNPs of miR-8079 (rs9533803, rs9525927, rs7981875) and three SNPs of miR-5007 (rs9527345, rs2252932, rs2997119) were selected, then we used logistic regression to analyze the association between candidate SNPs and COPD susceptibility under different genetic models. Multi-factor dimensionality reduction (MDR) was used to analyze the interaction of "SNP-SNP" in COPD risk studies. Finally, we used univariate and ANOVA to analyze the differences in clinical characteristics among different genotypes. Results: Our results showed that miR-8079-rs9525927 was significant associated with COPD susceptibility whether in overall and stratified analysis. miR-5007-rs2997119 was associated with the increased risk of COPD in women under multiple genetic models; miR-8079-rs9525927 and miR-5007-rs9527345 had a certain association with clinical indicator Fev_1 of COPD patients; rs9527345 or rs2252932 on miR-5007 was associated with the risk of COPD with wheezing dyspnea or wheezing.ConclusionOur results suggested that the genetic polymorphisms of miR-8079 or miR-5007 were potentially associated with COPD risk, of which miR-8079-rs9525927 was more prominent.

Abstract:Abstract Background: Periodontitis is a common disease with an unclear pathological mechanism. No precise consensus has been reached to evaluate the association between the IL-10 rs1800872 (- 592, -590, -597 C>A) polymorphism and periodontal disease. Thus, we performed this meta-analysis to collect more evidence-based information. Methods: Four online databases, PubMed, Embase, Web of Science, and China Biology Medicine disc (CBM), were searched in August 2018. An odds ratio (OR) with a 95% confidence interval (CI) was applied to evaluate the association of the rs1800872 with periodontitis susceptibility. Results: Twenty three case–control studies with 2714 patients and 2373 healthy controls were evaluated. The overall analyses verified that the IL-10 rs1800872 polymorphism was significantly associated with an increased risk of periodontitis in the allelic model, homozygote model, dominant model, and recessive model (A vs C: OR = 1.28, 95%CI = 1.11–1.49, P = .00, I 2 = 56.87%; AA vs CC: OR = 2.06, 95%CI = 1.32–3.23, P = .00, I 2 = 73.3%; AA + AC vs CC: OR = 1.42, 95%CI = 1.03–1.96, P = .03, I 2 = 76.2%; AA vs AC + CC: OR = 1.78, 95%CI = 1.26–2.56, P = .00, I 2 = 76.7%). Moreover, the subgroup analysis based on ethnicity, periodontitis type, and smoking status showed significant differences. Conclusions: The results of our meta-analysis demonstrate that rs1800872 is associated with periodontitis susceptibility in Caucasians and Asians. Moreover, A allele, AA genotype, CC genotype may be closely associated with chronic periodontitis (CP), while A allele, AA genotype may be closely associated with aggressive periodontitis (AgP).

Abstract:Chronic obstructive pulmonary disease (COPD) is a complex polygenic disease in which gene–environment interactions play a critical role in disease onset and progression. The gene encoding tumor necrosis factor (TNF) is one of several candidate loci for the pathogenesis of COPD and is highly polymorphic. A number of studies have investigated the association between the TNF-308 polymorphisms and COPD risk in different populations, and resulted in inconsistent results. A systematic review and meta-analysis of the published studies were performed to gain a clearer understanding of this association. The PubMed, Embase, Web of Science, and CNKI databases were searched for case–control studies published from 1966 to April 2009. Data were extracted and pooled odds ratios (OR) with 95% confidence intervals (CI) were calculated. Twenty-four eligible studies, comprising 2,380 COPD cases and 3,738 controls, were included in the meta-analysis. The pooled result showed that the TNF-308 polymorphisms were significantly associated with an increased risk of COPD (OR = 1.335, 95% CI: 1.172–1.522, for allele A carriers versus G/G; OR = 1.330, 95% CI = 1.174–1.505, for allele A versus allele G). Subgroup analysis supported the results in the Asian populations, but not in the Caucasian populations. When the analysis was limited to only those studies in which the COPD cases and controls were smokers/ex-smokers, the pooled results supported the conclusion. This meta-analysis suggested that the TNF-308 A allele is a more significant risk factor for developing COPD among Asian populations, but not among Caucasians.
The Assessment of TLR1 Gene Polymorphism Association with the Risk of Allergic Rhinitis in the Chinese Han Population from Northern China

Abstract:Background: Environmental factors and genetic predisposition can influence the occurrence and development of AR. Toll-like receptor 1 (TLR1) belongs to the TLR receptor family, which plays a fundamental role in the activation of innate immunity. This study aimed to explore the association between TLR1 genetic loci and AR susceptibility in the Han Chinese from northern China. Methods: Genotyping of three SNPs in the TLR1 has proceeded using the Agena MassARRAY platform. Odds ratio (OR) and 95% confidence interval (CI) were used to assess the correlation between candidate SNPs and AR susceptibility. Using FPRP (false-positive report probability analysis) to detect whether the positive results are noteworthy findings. The SNP-SNP interactions were detected by multifactor dimensionality reduction (MDR). Results: TLR1-rs72493538 (Allele "G": OR=0.77, p = 0.034) and -rs76600635 (Allele "G": OR=0.75, p = 0.024) were associated with reducing the risk of AR among Han Chinese in northern China. In addition, we found evidence that TLR1-rs72493538 (males, participants with aging > 43 years, or coming from the wind-blown sand region) and -rs76600635 (males, participants with BMI ≤ 24 kg/m2, or coming from the wind-blown sand region) were associated with AR risk in stratified analyses. FPRP showed that all positive results are noteworthy findings. MDR analysis showed that a two-loci genetic model composed of rs72493538 and rs76600635 can be chosen as the best genetic model to predict the risk of AR. Conclusion: TLR1-rs72493538 and -rs76600635 have a close association with reducing the risk of AR.

Abstract:Objective To investigate the correlation between IL-6、IL-6R and IL-4 single nucleotide polymorphisms(SNPs) and the susceptibility of chronic periodontitis among Shanghai patients of Han nationality.Methods Through the application of TaqMan probe real-time fluorescent quantitative PCR,128 health control subjects and 198 patients with chronic periodontitis(CP) above moderate degree were investigated from three single nucleotide polymorphisms sites,namely IL-6-572G/C,IL-6R 48892A/C and IL-4-590C/T.Statistical analysis of Chi square test was performed with SPSS 17.0 software package.Results In the screening of the 3 target SNPs,there were no statistically significant differences in distribution of the genotypes and alleles between healthy controls and CP patients.Conclusions The result suggests that the SNPs gene locus of IL-6-572G/C,IL-6R 48892A/C and IL-4-590C/T may not be significantly associated with the susceptibility of chronic periodontitis among Shanghai patients of Han nationality.
