Abstract:A broad range of N-sulfonyformamidines, widely used intermediates for drugs, were synthesized in moderate to excellent yields from 2-Pyridinemethanamine as N-source via Copper-catalyzed C-N cleavage. Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides, then reacted with N,N-dimethylformamide dimethyl acetal to obtain the corresponding N-Sulfonylformamidines analogs, during which pyridin-2-ylmethyl and sulfonyl groups were essential for the C-N bond cleavage. The current work presents a valuable complementarity to the synthesis of N-sulfonyformamidines as 2-pyridinemethanamine can provide the N source and sulfonyl chloride,s which could be original materials. Background: N-sulfonylamidines have gained considerable attention from schools and industries because of their unique bioactivity. Since Pinner's strategy, expanding the synthesis methods of N-sulfonylamidines has been the goal of many organic chemists over the past decades. Besides the crash reaction conditions and the participation of undesirable reagents, the production of N-sulfonylamidines commonly required unstable ammonia and azides as the source of nitrogen that hindered the further development and application of N-sulfonylamidine derivatives. Objective: The study aims to find a stable N source to replace NaN3 or NH3 to synthesize N-sulfonylamidines from sulfonyl chlorides. Methods: Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides. Then the reaction conditions of N-(2-pyridinylmethyl) benzenesulfonamides and N,N-dimethylformamide dimethyl acetal (DMF-DMA) were screened and optimized. The reaction was processed in glycol at 80 degrees C for 8 hours with the addition of 5 mol% Cu(OAc)(2)center dot H2O as a catalyst. Results: Taking advantage of pyridin-2-ylmethyl, a scope of N-Sulfonylformamidines were synthesized from those N-(2-pyridinylmethyl)benzenesulfonamides under copper-catalyzed C-N bond cleavage. Conclusion: This ready synthetic method will be more of a promising inspiration for bioactive compound synthesis and drug development than for an innovative approach to synthesizing N-sulfonylformamidines.

Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:N-Methyl-4-aminobenzyl sulfonamide, a key intermediate for synthesis of the pharmaceutical substance Sumatriptan, was prepared throught four steps. 4-Nitrobenzyl chloride was first converted to 4-nitrobenzyl sulphonic acid by reacting with sodium hydrosulfite, then the sulphonic acid obtained was oxidized to 4-nitrobenzyl sulfonyl chloride by chlorine, and N-methyl-4-nitrobenzyl sulfonamide was produced by reacting with methylamine, in the end, the title product was obtained by reducing the nitro group to amino group with hydrazine hydrate in the presence of iron trichoride on active carbon as the catalyst. The yields of the four steps are 90%, 93%, 60% and 77%, respectively. The intermediates and product were characterized by GC-MS and H~1NMR.

Abstract:An efficient synthesis of novel phenoxyimine ligands containing a difluorovinylbiphenyl group was based on the Suzuki coupling coupling of 4-bromo-2,6-difluoroaniline with 4-styrylboronic acid followed by a condensation reaction with various arylaldehydes. The structures were established by IR, 1H NMR, 13C NMR spectra and HRMS. The advantages of this synthetic route were its simple operation, mild reaction conditions and good yields.

Abstract:The construction of N-aryl sulfonamides is usually an important reaction in organic synthesis because these compounds are privileged in the structure of many natural products and pharmaceutical molecules. Also, N-aryl sulfonamides are well-known due to their antibacterial, antitumor, and anti-HIV biological activities. Therefore, research on the laboratory synthesis of N-aryl sulfonamides is an important challenge in organic synthesis. In this review, we discussed on methods reported up to now for the synthesis of N-aryl sulfonamides with the goal of stimulating further progresses in this field. We believe that this review will open up a new road for the preparation of pharmaceutically and biologically active sulfonamides.

Abstract:The amide compounds have high pharmaceutical activity. To further study the synthesis and properties of the amide compounds, N-(substituted phenoxyacetyl)-3-amino-dihydrothiophene-2-one derivatives were synthesized by the reaction of substituted phenoxyacetic acid with 3-amino-dihydrothiophene-2-one hydrochloride. The structures were confirmed by IR, 1HNMR and elemental analysis. The antifungal activity of 5 pathogens were tested. Phosphorus oxychloride was used as the condensation reagent in synthesis. The method has advantages with one-pot synthesis, simple and easy operation, cheap and easily-obtained raw material, high yield and high purity. The results provide the basis for synthetizing and screening new compounds.

It was found that many problems occurred preparing amino acid derived N-acylsulfonamides with the commonly used coupling approach in our previous studies. In this paper, we report an efficient approach to synthesize various amino acids derived N-acylsulfonamides in high yields without any racemization.

Abstract:Three new compounds were first synthesized by the reaction of N-benzoyl-N~α-solanesyl glutaminic acid with different amines or amino acid ester hydrochloride by the method of the activated N-hydroxysuccinimide ester in the presence of N, N-dicyclohexylcarbodiimide. Abstract:The development of novel HIV-1 non-nucleoside reverse transcriptase inhibitors (NNRTIs) offers the possibility of generating novel structures of increased potency. Based on the bioisosterism principle, two novel 2-(5-(naphthalen-1-yl)-1,2,3-thiadiazol-4-ylthio)-N-acetamides derivatives have been designed, synthesized and evaluated for their anti-HIV activities in MT-4 cells. The results indicate that these compounds show good activities against HIV-1. Especially, compound 9B (EC50 = 0.019 mu M) was more effective than the reference drugs nevirapine and delavirdine.

Abstract:A series of novel N-n-butyl-1,8-naphthalimide derivatives were synthesized via a three-step reaction involving nucleophilic substitution and acylation. All of the compounds were characterized by IR, 1H NMR, 13C NMR, MS, and elemental analysis, and the crystal structure of N-n-butyl-4-[N’,N’-bis(2`,4`-dichlorobenzoyl)ethylamino]-1,8-naphthalimide was determined. The π-π stacking interactions and hydrogen bonds between the two molecular core planes (naphthalimide ring) and the van der Waals forces between the flexible n-butyl groups resulted in a 3D long-chain structure. The UV-vis and fluorescence properties of the title compounds were investigated. The results indicated that the monosubstituted 1,8-naphthalimide derivatives bearing an electron-donating group on the benzene ring or a structure with a larger conjugative effect exhibited enhanced fluorescence properties.

Abstract:In this research, the functionalized 1,6‐naphthyridines were synthesized using multicomponent reaction of benzaldehydes, malononitrile, and 4‐aminocumarin in water at room temperature in the presence of SiO2/Fe3O4@MWCNTs as a recyclable catalyst in high yields. This study synthesized 1,6‐naphthyridine as a novel class of fused heterocyclic compounds with high efficiency using a multicomponent reaction. The reaction involved benzaldehyde or its derivatives, 2 mol of malononitrile, and 4‐aminocumarin in an aqueous solution at room temperature in the presence of SiO2/Fe3O4@MWCNTs as a recyclable catalyst. This study examines the antioxidant properties of 1,6‐naphthyridine in addition to the other research undertaken in this work. The production process of 1,6‐naphthyridine demonstrated several advantageous features, including rapid reactions, high yields of the final product, and straightforward separation of the catalyst and product from the reaction mixture.

Abstract:A triflate salt-catalyzed nonhydrolytic method for the deacylation of N-acylsulfonamides and subsequent one-pot condensation of the newly formed sulfonamides with N,N-dimethylformamide dimethyl acetal to provide N-sulfonylamidines is presented. A range of aliphatic and aromatic N-acylsulfonamides bearing various N-acyl groups such as acetyl, propionyl, butyrl, isobutyryl, octanoyl, benzoyl, 2-phenylacetyl, and sterically hindered pivaloyl are readily transformed into the corresponding N-sulfonylamidines in good to excellent yields. A variety of functional groups including halogeno, keto, nitro, cyano, hydroxyl, ether, and carboxylic ester are tolerated intact.

To full acylated norepinephrine 3 , different de-protection conditions will afford different products ( 4 or 5 ) selectively. Abstract:A new Schiff base was prepared by the condensation between the sulfa drug sulfadiazine and 5-nitrovanilline to form 4-((4-hydroxy-3-methoxy-5-nitrobenzylidene) amino)-N-(pyrimidin-2-yl) benzene sulfonamide (5NVDA). Spectral studies have been performed to confirm the structure of the compound. Electronic structure of the compound was studied using DFT B3LYP/aug-cc-pVDZ level. Detailed potential energy distribution and vibrational assignments were made with the help of scaled simulated spectrum. UV spectra in DMSO medium showed a strong peak at 335 nm, which is in consistent with TDDFT simulations using CAM-B3LYP functional. Frontier orbitals were analyzed and found that the compound is easily polarizable, but stable. The compound was stabilized by extensive delolcaltaion of electrons. Durg likeness studies proved that this compound is an ideal candidate to be used as a drug and PASS studies showed that it shows binding to HIV RNA activity, which is confirmed by molecular docking studies with docking score of −8.4 kcal/mol.

Abstract:The 2-Aminonicotinonitrile derivative was reacted with different bi-functional reagents such as formamide, thiourea, acetic anhydride, and phthalic anhydride under optimized conditions to give pyrimidine, thiourea, acetamide, and isoindoline derivatives, respectively, When it was treated with active methylene reagents as malononitrile, phenacyl bromide, and ethyl bromoacetate under varied experimental modulation, it afforded 1, 8-naphthyridine, ethyl, and methylamino nicotinonitrile derivatives, respectively. Also, it was reacted with p-toluene sulfonyl chloride, chloroacetyl chloride, and benzoyl chloride to give sulfonamide, 2-chloro-N-acetamide, and benzamide derivatives, respectively. Likewise, it was reacted with diethyl malonate, ethyl cyanoacetate, and cyano acetic acid to give ethylpropanoate, naphthyridine, and cyano acetamide derivatives, respectively. However, treatment of ethylpropanoate and cyano acetamide derivatives with hydrazine hydrate gave pyrazole and 5-amino-pyrazole nicotinonitrile derivatives, respectively. In addition, it was reacted with p-anisaldehyde, phenyl isocyanate, and triethyl orthoformate to give benzylamino nicotinonitrile, phenyl urea, and N-formamide derivatives, respectively. Furthermore, it was reacted with nitrous acid then coupled with aniline; it was also reacted with isatine and 1,3- dibromo propane to give oxoindoline derivative, and the dimer. Elemental analyses, together with spectroscopic data including IR, 1H-NMR in addition to 13C-NMR and mass spectra submit proofs for the chemical structures for all compounds.Graphical

Abstract:A new series of N -acyl derivatives ( 4a–4d ) and Azetidin-2-one derivatives ( 5a , 5b ) containing a 1,3,4-oxadiazole ring were prepared by sequence reactions. The new Schiff bases ( 3a–3d ) used as starting materials were prepared via condensation of (4-(5-phenyl-1,3,4-oxadiazole-2yl)aniline) with four replaced aromatic dialdehyde ( 2a–2d ). The newly synthesized Schiff bases ( 3a–3d ) reacted with, acetyl chloride in the dry benzene to get new derivatives ( 4a–4d ). While the second path was towards the formation of new Azetidin-2-one derivatives ( 5a , 5b ) from cyclization reaction of the Schiff bases with chloroacetylchloride in dioxan and triethylamine at 0–5°C. Synthesized compounds had been characterized by FT-IR and 1 H NMR spectra in order to elucidate their structures. Finally, the resulting products were evaluated for it is efficacy measurements against two types of bacteria showed excellent to good efficacy antibacterial activity.

Abstract:A simple and efficient method to synthesize N-chloro-N-sulfonylsulfinamides by the direct aminoxidation of arenethiols under aqueous and mild conditions is disclosed, geminally installing the oxo and amino groups on the sulfur atom of arenethiols. The products have been primarily developed as sulfinylation reagents to convert Grignard reagents into sulfoxides, and as amination reagents to convert secondary amines into hydrazine derivatives.
