Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:AbstractIn contrast to the well‐established [3+2] cycloaddition reactions, the catalytic enantioselective [3+n] (n≥3) cycloaddition reaction of activated isocyanides for the preparation of six‐membered or larger ring systems has remained underdeveloped. Herein, we report the first example of highly diastereo‐ and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. By employing silver catalysis, a wide range of biologically important bicyclic 1,2,4‐triazines were obtained in high yields (up to 99 %) with good to excellent stereoselectivities (up to >20 : 1 dr, 99 % ee). In addition, the same catalytic system could be applied to both the late‐stage functionalization of complex bioactive molecules and the kinetic resolution of racemic azomethine imines, further highlighting its versatility and synthetic utility.

Abstract:The first nitro-group-initiated redox-neutral [3+2] cycloaddition of nitrocyclopropanes with alkenes by using visible-light-absorbing transition-metal complexes was reported. High diastereoselectivities were observed for two quaternary carbon centers on the ring and validated by DFT calculations. Spiro- or polycyclic structures can be constructed smoothly. Cyclic γ-amino acid derivatives and polysubstituted cyclic amino alcohols can be obtained easily through reduction of the nitro group.

Abstract:A palladium-catalyzed tandem dehydrogenative [4 + 2] annulation of terminal olefins with N-sulfonyl amides via C(sp2)-H activation, allylic C(sp3)-H activation, and homoallylic C(sp3)-H elimination processes has been developed. Promoted by the DMSO ligand, various benzamides, heterocyclic arylamides, alkenyl carboxamides, and commercial olefins are found to be efficient substrates to construct important heterocyclic compounds bearing a vinyl substituent with high E stereoselectivity. Using air as the terminal oxidant also provides a great advantage regarding environmental friendliness.

Abstract:Graphical abstract This review collects the recent developments in enantioselective nickel-catalysed cycloadditions published since the beginning of 2015. It illustrates the power of these inexpensive and highly abundant catalysts to provide remarkably diverse chemical reactivity. Abstract:Cross-electrophile coupling reactions of two Csp3-X bonds remain challenging. Herein we report an intramolecular nickel-catalyzed cross-electrophile coupling reaction of 1,3-diol derivatives. Notably, this transformation is utilized to synthesize a range of mono- and 1,2-disubstituted alkylcyclopropanes, including those derived from terpenes, steroids, and aldol products. Additionally, enantioenriched cyclopropanes are synthesized from the products of proline-catalyzed and Evans aldol reactions. A procedure for direct transformation of 1,3-diols to cyclopropanes is also described. Calculations and experimental data are consistent with a nickel-catalyzed mechanism that begins with stereoablative oxidative addition at the secondary center.

Abstract:Herein, we report a general and practical nickel-catalyzed deaminative allenylation of amino acid derivatives with terminal alkynes. The well-designed, electron deficient, and sterically hindered amide-type NN2 pincer ligand was crucial to the success of this transformation, enabling the coupling to occur under mild conditions with high efficiency. The remarkable features of this chemistry are its good scalability, its broad substrate scope, functional group tolerance, and the efficient modification of peptides, drugs, and natural products.

Abstract:We have established a nickel-catalyzed domino reaction that harnesses sulfonamide, alkyl chloride, and alkyne functionalities in a multistep sequence to afford vinylcyclopropanes bearing tetrasubstituted olefins. The starting materials are prepared by iron-promoted aza-Prins reactions of ynals. This method provides rapid synthetic access to valuable building blocks with applications in medicinal chemistry. Experimental and computational results support the initiation of the catalytic cycle by oxidative addition of the propargylic sulfonamide, and a key ambiphilic allenylnickel intermediate leads to a bifurcated reaction pathway that generates olefin isomers.

Abstract:Abstract Metal-catalyzed [2+2+2] cycloaddition is a powerful tool that allows rapid construction of functionalized 6-membered carbo- and heterocycles in a single step through an atom-economical process with high functional group tolerance. Abstract:Efficient strategies to assemble enantioenriched pyridine derivatives are highly important, given their significance in both synthetic and medicinal chemistry. Here, we report an enantioselective nickel-catalyzed intermolecular [2 + 2 + 2] cycloaddition of alkyne-tethered malononitriles with alkynes for the synthesis of densely substituted pyridines. The α-all-carbon quaternary center adjacent to pyridine is introduced by desymmetrizing the two cyano groups of disubstituted malononitriles. Notably, terminal alkynes are also tolerated with good regioselectivity to afford tetrasubstituted pyridines. Zinc halide is essential to enable the occurrence of the transformation by promoting the hetero-cyclometallation step. The reaction uses bio-renewable 2-MeTHF as the solvent and features mild reaction conditions, good functional group compatibilities, and enantioselectivities. This study offers a straightforward approach to the valuable enantioenriched heteroarenes from feedstock chemicals by forming three bonds and one quaternary stereocenter simultaneously in a single reaction step.

Abstract:A methodology for radical cross-coupling with vinylsiloxanes and pyridinium salts under nickel catalysis is described. Easily implemented from inexpensive and abundant primary amines and terminal alkynes, this modern Hiyama coupling provides efficient access to (E), (Z), and 1,1’-alkenes with configuration retention. Operating under mild conditions, this robust strategy applies to a broad range of functional groups with diverse double bond stereochemistries. This versatile reaction is scalable and straightforward, accommodating both secondary and primary alkyl groups. A radical mechanism involving hypervalent silicon intermediates is proposed.

Abstract:Palladium-catalyzed asymmetric [3+2] cycloaddition reaction of vinyl cyclopropane and electron-deficient dienes was realized. The cycloaddition reaction proceeded regioselectively on the distant C=C double bond of electron-deficient dienes, and was mainly controlled by the steric hindrance of the 5-substituent of electron-deficient dienes. Chiral multi-substituted cyclopentanes bearing three functional groups (monosubstituted alkene, conjugated ester, and cyano) and three continuous stereocenters were obtained in moderate to high yields, diastereoselectivities, and enantioselectivities. Abstract:Reported herein is an example of highly regio‐, diastereo‐ and enantioselective Cu(I)‐catalyzed intermolecular [3+2] cycloaddition reaction of α ‐substituted iminoesters with α ‐trifluoromethyl α,β ‐unsaturated esters. This novel strategy provided a facile access to pyrrolidines with two skipped (aza)quaternary stereocenters including a CF 3 all‐carbon quaternary stereocenter. A broad substrate scope was observed and high yields (up to 94%) with excellent diastereoselectivity (up to >20 : 1 d.r .) and enantioselectivity (up to 98% ee ) were obtained.
